Insulin resistance(IR) is a independent risk factor of ichemic angiopathy .The abnormal state of thrombosis and fibrinolysis is a critical pathology component of IR,and play a vital role in initiating and developing cardiovasculer disease. Under IR state, the,inhancement of plasminogen-activator inhibitior-1(PAI-1)synthesis by adipose tissue and the reduced generation of nitric oxide with endothelial dysfunction contribute to the hypo-fibrinolytic/pro-thrombotic state and promote atherosclerotic. Accompanied with IR developing, when B cell function can't keep normal glycemia, the advanced glycosylation end products(AGEs) are increased as a result of hyperglycemia. AGEs can aggravate endothelial injury and accelerate the course of atherosclerotic. Fully comprehending the pathogenesis of thrombosis and fibrinolysis in IR and taking therapy for the pathology have clinical significance.

3.Arthroscopic anterior cruciate ligament reconstruction by artificial ligaments: A preliminary experience

Objective To study the feasibility and short-term effects of arthroscopic anterior cruciate ligament (ACL) reconstruction by using the Ligament Advancement Reinforcement System (LARS) artificial ligaments. Methods Arthroscopic ACL reconstruction was carried out in 16 patients with ACL injuries by using the LARS artificial ligaments. Bone tunnels of the femur and the tibia were prepared with an identical interval between each other. The artificial tendon was inserted into the tunnels, and then, fixed with 2 interface screws after being tightened, with the free end of the tendon within the articular cavity. The complicated injuries were treated simultaneously. Results The operation time was 51~86 min (mean, 64 min). No postoperative complications such as synovitis, ligament rupture, or movement restriction occurred. A follow-up checkup was made in the 16 patients for 1.5~6 months (mean, 3.8 months). The International Knee Documentation Committee (IKDC) scores showed 6 cases of grade C and 10 cases of grade D before the surgery, while 6 cases of grade A, 9 cases of grade B, and 1 case of grade C after the surgery ( ? 2 =6.264, P
4.THE HISTOLOGICAL OBSERVATION OF THE STEPWISE DEVELOPMENT OF NEONATAL MOUSE TESTIS IN IMMUNODEFICIENT MICE

Objective To investigate the development of neonatal mouse testis in castrated immunodeficient mice by monitoring the graft survival and weight and observing the structure of seminiferous epithelium and the composition of spermatogenic cells in grafts. Methods Neonatal Kun-ming mouse testis were grafted under the skin of castrated nude mice(7-12week-old).Grafts were then taken out at different time intervals(namely 16 time points: 3 days,1-11 weeks respectively and 3-6 months respectively).The survival rate of grafts was calculated and the wet weight was measured.Histological analysis was performed for the observation of the structure of seminiferous epithelium and the composition of spermatogenic cells in grafts. Results Four hundrcd and five grafts recovered out of 450 testis grafted, resulting in a recovery rate of 90.0%.The graft weight increased more than 40 times.The developmental pattern of seminiferous tubules and the appearance time of various spermatogenic cells in grafts were similar as seen in intact mice.Eight weeks after the grafrting,an increasing degradation of seminiferous epithelium was observed.Conclusion When neonatal mouse testis were grafted into nude mice,the developmental course was similar as that in normal donors.The optimal retrieval time of round spermatids and sperms was around the end of the first spermatogenesis wave,about 5-7weeks after the grafting procedure.
5.Development of Kun-ming mouse primordial spermatogenic cells grafted into nude mice

Objective To observe the growth and the development of primordial spermatogenic cells by allografting Kun-ming neonatal mouse testes into athymic nude mice.Methods Neonatal mouse testes were grafted under the back skin of nude mice (n=29). Grafts were taken out at the post-grafting day 30-110 and observed on the growth and the appearance, the structure of seminiferous tubules as well as the development of spermatogenic cells and the stage of seminiferous epithelium.(Results) The graft growth in all 29 recipient animals was observed, and the mass of grafts was significantly increased from (0.73)?(0.05) mm in diameter and (5.67)?(0.72) mg in wet weight pre-grafting to (6.75)?(0.73) mm and (113.12)?(78.23) mg post-grafting, respectively. The structure of seminiferous (tubules) inside grafts was clearly seen under a light microscope. And the histological analysis (demons)-(trated) a complete spermatogenesis with all levels of spermatogenic cells including sperm and stage I-XII of the seminiferous epithelium could also be identified.Conclusion Neonatal mouse testes could continuously grow and develop under the back skin of castrated nude mice, which would therefore be a useful tool for investigations on the proliferative and developmental pattern of spermatogenic cells and the regulation mechanism of spermatogenesis.
6.New pathological ultrastructure observation of compressive spinal cord injury in rats

[Objective]To observe the characteristic ultrastructure changes of compressive spinal cord injury.[Method]Animal model of compressive spinal cord injury was made by modified Allen's method.Pathological changes of spinal cord tissues were observed with transmission electronic microscopy after 4 weeks.[Result]The characteristic ultrastructure changes of compressive spinal cord injury were observed obviously.It was necrosis without inflammatory cell reaction.The single-direction break of myelin sheath and the traumatic cell could be observed.They were all formed by the reason of biomechanics.These two characters were the characteristic ultrastructure changes of compressive spinal cord injury.[Conclusion]These new ultrastructure observations can provide certain materials for the basal explore of compressive spinal cord injury.

To investigate the role of calcineurin dependent signal pathway in cardiac hypertrophy of cold stress induced hypertensive rats, 3 months old Wistar rats were exposed to cold (4?2℃) for 4 hours per day for 8 weeks to produce cold stress induced hypertension and cardiac hypertrophy. The systolic blood pressure(SBP), heart rate and hemodynamic parameters were measured using a force transducer.The heart/body weight ratio, CaN activity, CaN, c fos expression protein of cardiac tissure were also investigated. 9.Hormone regulation of the expression of vascular endothelial growth factor and its receptors in mouse uterus

Objective: To investigate the effects of estrogen and progesterone on the expression of vascular endothelial growth factor (VEGF) and its receptors (VEGFR) in mouse uterus. Methods: 3-week-old immature female mice were randomly divided into 7 groups and treated with corn oil, estradiol (E2) of 1.5, 3.0, 10, 25 ng, progesterone (P) of 100 ?g and (E_2 10 ng + P 100 ?g)/mouse, respectively. After the treatment for 48 h, mouse uterus was collected to isolate total RNA. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of mRNA isoforms of VEGF and its receptors in mouse uterus. Results: Compared with control, both E_2 and P significantly increased the expression of VEGF164 and VEGF120 mRNA in mouse uterus. The expression of VEGFR2 mRNA, not VEGF1 mRNA, was decreased by E_2 treatment in a dose-independent manner. Conclusion: Both estradiol and progesterone up-regulated the expression of VEGF, but estradiol down-regulated the expression of VEGFR2 in mouse uterus.
10.Effect of bone marrow mesenchymal stem cells transplantation on intestinal mucosa barrier of acute pancreatitis

Objective To investigate the role of bone marrow mesenchymal stem cells (MSCs) transplantation in repairing injured intestinal mucosa of acute pancreatitis.Methods MSCs were harvested and cultured from femurs of male SD rats.Twenty female SD rats were divided into three groups,and serve acute pancratitis (SAP) model was induced by intraperitoneal injection of L-arginine (2 g/kg) twice.Twelve hours after SAP model established,MSC transplantation group (n=8) were injected MSCs (5 × 106 cell/rat) through tail vein for three days,and SAP group (n=6) were injected the same volume of saline through tail vein as control.Control group (n=6) were only injected the same volume of saline without any treatment.All the rats were sacrificed at 72 hours after model established.The small intestinal tissues were taken for HE staining and pathological score,the TNF-α mRNA and IL-1β mRNA expression level in small intestine and pancreas were tested by RT-PCR.Y chromo-some (Sry) gene in pancreatic and intestinal tissue was examined by polymerase chain reaction (PCR).Results The relative expression quantity of TNF-a mRNA and IL-1β mRNA in pancreas was significant higher in SAP group and MSC transplantation group than in control group (7.22 ± 1.99,3.46± 1.75 vs 1.32 ± 1.04 ; 2.71 ± 0.56,1.92 ± 0.28 vs 0.61 ± 0.45 ),the difference was statistically significant (F=18.375,F=22.701; P<0.05).Compared with SAP group,the expression quantity of TNF-α mRNA and IL-1β mRNA in pancreas was significantly decreased in MSC transplantation group,the difference was statistically significant (P<0.05).The relative expression quantity of TNF-α mRNA and IL-1β mRNA in small intestine was significantly higher in SAP group and MSC transplantation group than in control group (3.93 ± 1.08,2.13 ± 0.53 vs 0.68 ± 0.42 ; 2.44 ± 1.54,1.02±0.44 vs 0.60±0.14),the difference was statistically significant (F=21.772,F=6.132; P<0.05).The expression of TNF-αmRNA and IL-1β mRNA in MSC transplantation group was lower than that in SAP group,the difference was statistically significant (P<0.05).Compared with SAP group,pathological score indicated that small intestine injure was slighter in MSC transplantation group (3.83±0.28 vs 2.83±0.56),the difference was statistically significant (F=12.013,P<0.05).Sry gene could be detected in the pancreatic and intestinal tissue of MSC transplantation group.Conclusion Allogeneic MSC transplantation group can inhibit Pro-inflammatory cytokines expression in acute pancreatitis,relieve the intestinal mucosa injury and may involve in the intestinal tissue repair.
