1.Diagnosis and treatment of extranodal NK/T-cell lymphoma, nasal type

Extranodal natural killer (NK)/T-cell lymphoma, nasal type (ENKL) is an uncommon subtype of non-Hodgkin's lymphoma, which is aggressive and has poor prognosis. ENKL occurs predominantly in the nasal cavity and less frequently in the skin and in the gastrointestinal tract. The pathology of this disease is characterized by vascular invasion and destruction. ENKL is strongly associated with the Epstein–Barr virus, which aids in the diagnosis of ENKL. The characteristic markers on the surface of an NK/T cell and the spe-cific genetic variations are also useful in the diagnosis. At present, the treatment of ENKL is still under discussion. Although radiothera-py combined with chemotherapy on patients with early-stage ENKL and L-ASP-based chemotherapy and hematopoietic stem cell trans-plant on patients with advance-stage ENKL have achieved favorable effects, further studies are still necessary to develop the principles and methods of a standardized treatment.
2.Osteogenesis in transplantation of tissue engineered bone to repair segmental defect of long bone

Objective To investigate the ability of osteogenesis, repaired effects and possible mechanism of tissue engineered bone made in an approach of bionics as a transplantation biomaterial to repair a segmental defect of long bone. Methods HA/?-TCP was composed with PDLLA and then composed with rhBMP-2 and collagen of typeⅠ. The combined biomaterial was put in common culture with osteoblasts harvested from periosteum of rabbit and vascular endothelial cells from kidney of rabbit then transplanted this tissue engineered bone to total segmental periosteum-bone defect of 1.5 cm in the rabbits radius which were investigated 4, 8 and 12 weeks after operation respectively. Investigation of the bone defect was made by means of gross observation, X-ray examination, histology of HE and Masson staining, image pattern analysis, scanning electron microscopy, EDAX. Results In gross observation, the implantations were adhered to the host bone well in four weeks, the implantations was bony healed with host bone in eight weeks, and some of the implantations were replaced by new formation bone in 12 weeks. In histological examination of four weeks after operation, lamellar bone was found, and eight weeks after operation, implant was incorporated to host bone end by end through cortical bone, and new bone marrow was found to invade into the implant. Furthermore, the outer part of implant was completely substituted by new cortical bone 12 weeks after operation. In addition, the histological study pointed out that the new bone arranged in type of various bands which were in subsequent transition. There is significant difference between 4 weeks and 8 weeks, and 4 weeks and 12 weeks, but no significant difference between 8 weeks and 12 weeks of the quantity of new bone. The ratio of calcium to phosphorus in the transplants tended to approach that in the host cortical bone along with the period of time after operation. Conclusion Satisfied effects of remodeling appeared after this tissue engineered bone composed by bionics was transplanted to the segmental defect of long bone. The mechanism of bone regeneration was endochondral ossification.
3.Critical role of Dengue Virus NS1 protein in viral replication

Dengue virus (DENV) nonstructural protein 1 (NS1) is a highly conserved 46-kDa protein that contains 2 glycosylation sites (Asn-130 and Asn-207) and 12 conserved cysteine (Cys) residues. Here, we performed site-directed mutagenesis to generate systematic mutants of viral strain TSV01. The results of the subsequent analysis showed that an alanine substitution at the second N-linked glycan Asn-207 in NS1 delayed viral RNA synthesis, reduced virus plaque size, and weakened the cytopathic effect. Three mutants at Cys sites (Cys-4, Cys-55, Cys-291) and a C-terminal deletion (ΔC) mutant signiifcantly impaired RNA synthesis, and consequently abolished viral growth, whereas alanine mutations at Asn-130 and Glu-173 resulted in phenotypes that were similar to the wild-type (WT) virus. Further analysis showed that the Asn-207 mutation slightly delayed viral replication. These results suggest that the three conserved disulifde bonds and the second N-linked glycan in NS1 are required for DENV-2 replication.

5.Quantitative Determination of Selenium Species in Se-enriched Yeast Using Liquid Chromatography-Hydride Generation Atomic Fluorescence Spectrometry

A rapid and sensitive method has been developed for the simultaneous determination of four selenium species Se(Ⅵ), Se(Ⅵ), selenomethionine, and Se-methylselenocysteine in Se-enriched yeast by liquid chromatography-hydride generation atomic fluorescence spectrometry (HPLC-HG-AFS). The isolation of the analytes from yeast samples was accomplished by proteaseⅩⅣ and trypsin enzymatic digestion. The target compounds were separated on a PRP-X100 anion exchange column and analyzed by HG-AFS. The mobile phase was 20 mmol/L (NH4)2HPO4. Good linearity was obtained for all the selenium species, with linear correlation coefficients higher than 0. 9996. The LODs of the four species were between 0. 5 and 5. 0 μg/kg. Average recoveries for the four analytes were in the ranges of 82 . 5%-101 . 2%, with intra-and inter-day RSD lower than 8. 6% and 14. 5, respectively. The proposed analytical method is simple, sensitive, with low operation cost, making it applicable for the determination of the selenium species in Se-enriched feeds.
6.Application of computer system based on artificial neural network and artificial intelligence in diagnosing child mental health disorders

BACKGROUND: It is not a report concerning utilization of artificial intelligence combined with artificial neural network in mental health domain, rather than that combining artificial intelligence, artificial neural network and simulating human brain thinking mode for diagnosing child mental health disorders. OBJECTIVE: Using computer to simulate thinking modes of human brain, to establish an artificial intelligence expert system for diagnosis and treatment of child mental health disorders based on artificial neural network and expert system.METHODS: The expert system involves in child psychiatry, child psychology, psychological measurement, psychological therapy, and computer science, and so on. The diagnosis system combines the diagnosis standard of ICD 10, DSM IV, CCMD-2, the epidemiological data, and the clinical data with senior psychiatrist knowledge. The clinical data were obtained from 14 epidemiological and outpatient department, and 1 125 valid data were harvested for the system compilation.RESULTS AND CONCLUSION: The system can diagnose 61 kinds of child mental health disorders. It includes more than 95% child mental health disorders. After diagnosis, the computer will give a treatment suggestion. Comparing the diagnosis by computer with diagnosis by the senior child psychiatrists, the consistent rate is 99%. The research can help the younger doctors to learn abundant clinical experiences of senior child psychiatrists and can help children of mental health disorders throughout the country.
7.Analysis of Cobalt Phthalocyanine Sulfonates byhin Layer Chromatography-Flame Ionization Detector

A new method for analysis of cobalt phthalocyamine sulfonates(CoSPC) by thin layer chromatography/flame ionization detector (TCL/FID) was investigated. Different compounds in CoSPC can be separated by TLC, and structure can be further identified by matrix-assisted laser desorption/ionization-time-of-flight-muss spectrometry (MALDI-TOF-MS). Experimental results indicated that commercial CoSPC has three compounds, cobalt phthalocyanine (CoPC) mono-sulfonates with two isomers, CoPC di-sulfonates with five isomers and CoPC- tri-suflonates, TCL/FID could be used to determine the amount of different compounds. This method is featured by its ease operation, quick response, good separation and satisfactory results.
8.Assessment of endocardial and epicardial rotation of left ventricle in patients with acute myocardial infarction before and after revascularization by speckle tracking imaging

Objective To evaluate peak rotation in the endocardium(endo)and epicardium(epi)and peak mural torsion in patients with anterior wall acute myocardial infarction(AMI)before and after revascularization by speckle tracking imaging(STI).Methods The study comprised 20 normal controls and 21 consecutive xpatients with a first-episode anterior wall AMI with stenosis of the left anterior descending artery.All patients underwent Successful percutaneous coronary intervention within 12h onset of AMI.During one month, two patients were ruled OUt because one died of heart failure and the other suffered from persistent atrial fibrillation.Conventional echocardiography and STI were performed in normal controls and AMI patients before and one month after revascularization.Results In the left ventricle(LV)apical and basal level,peak endo-and epi-rotation and peak mural torsion in AMI patients were significantly reduced relative to those of normal control group(all P<0.01).One month after revascularization,there were significant changes in these three variables,especially in peak endorotation(P<0.01).Peak apical endo-rotation positively correlated with LV ejection fraction(LVEF)(r=0.717,P<0.01)and inversely correlated with LV end-diastolic volume(EDV)(r=-0.694,P<0.05).However,tight relations were not found among other variables at the two planes.Conclusions Peak endo-and epi-rotation and peak mural torsion can make the sensitive assessment of abnormal myocardial contractile performancein AMI patients using STI.
9.Surface Display of Domain Ⅲ of Japanese Encephalitis Virus E Protein on Salmonella Typhimurium by Using an Ice Nucleation Protein

A bacterial cell surface display technique based on an ice nucleation protein has been employed for the development of live vaccine against viral infection.Due to its ubiquitous ability to invade host cells,Salmonella typhimurium might be a good candidate for displaying viral antigens.We demonstrated the surface display of domain III of Japanese encephalitis virus E protein and the enhanced green fluorescent protein on S.typhimurium BRD509 using the ice nucleation protein.The effects of the motif in the ice nucleation protein on the effective display of integral protein were also investigated.The results showed that display motifs in the protein can target integral foreign protein on the surface of S.typhimurium BRD509.Moreover,recombinant strains with surface displayed viral proteins retained their invasiveness,suggesting that the recombinant S.typhimurium can be used as live vaccine vector for eliciting complete immunogenicity.The data may yield better understanding of the mechanism by which ice nucleation protein displays foreign proteins in the Salmonella strain.

Objectives:To study the postoperative nutritional support in patients with pancreatic disease. Methods:Based on the kind of pancreatic disease,twenty two patients were randomly divided into PN+EN group and TPN group during 20 days after operation.Body weight,the restoring time of intestinal movement,the complications,the blood amylase,the serum glutamic pyruvic transaminase,the blood urea nitrogen,the blood sugar,the serum albumin(Alb),transferritin(TFN) and prealbumin(PAB) were compared on the first,fifth,tenth and twentieth day after operation.
