1.Culture and evaluation of rat’s mesenteric lymphatic endothelium in vitro

Aim To establish the technology of isolation rat’s mesenteri c lymphatics and the culture method of their endothelium and confirm the ability o f synthesis nitric oxide and Factor Ⅷ related antigens in the endothelium in vitro. Methods With an operating microscope and microsurgical in struments, rat’s mesenteric lymphatics were isolated and the endothelium were p rimarily cultured and subcultured. Exerting immunohistochemical method, Factor Ⅷ related antigens and nitric oxide synthase were evaluationed. The contents of NO metabolite were measured. Results The rat’s mesenteric lympha tics were isolated and the endothelium were Fusiform shape or polygon, the monol ayer cultures displayed typical cobblestone or pavingstone morphology. Factor Ⅷ related antigens were expressed and their positive rate was 98%. The positive rate of nitric oxide synthase was 83%. Positive reaction products were buffy ar ound nuclear. NO metabolite was (10.26?2.17) mol?L -1(n=7). Conclusion The culture method we introduced was very practical, simple and reliable. The endothelium is in normal structure and function characters. It also certificated that the endothelium in vitro could express nitric oxide synthase and Factor Ⅷ related antigens.
2.Pseudomonas aeruginosa Nosocomial Infections after Open Heart Surgery:A Clinical Analysis

OBJECTIVE To investigate the clinical distribution and antibiotics resistance of Pseudomonas aeruginosa nosocomial infection after open-heart operation.METHODS The clinical data of 393 patients with nosocomial infection from Jan 2004 to Dec 2007 were analyzed retrospectively.RESULTS There were 57 infected cases caused by P.aeruginosa,and accounted for 14.5% of all infections during period.All infected patients had serious original heart diseases,and received broad-spectrum antibiotic therapy previously.Forty-two patients developed postoperative cardiorespiratory function failure,and 28 patients needed circulatory support.Thirty-six patients prolonged mechanical ventilation time for over 1 week.Results of susceptibility test showed that ciprofloxacin,levofloxacin and piperacillin/tazobactam were the most active antibiotics,followed by tobramycin,netilmicin,gentamicin,meropenem and imipenem/cilastatin.P.aeruginosa presented high resistance to ceftazidime and cefoperazone/sulbactam.CONCLUSIONS P.aeruginosa is one of the most common pathogenic bacteria after open-heart operations in our hospital and presented multidrug resistance.Rational use of antibiotics is important to reduce drug resistant strains.

4.Study on differentially expressed genes in papillary thyroid carcinoma by cDNA microarray

Objective To screen the differentially expressed genes between papillary thyroid carcinoma (PTC) and normal thyroid tissues using cDNA microarray. Methods mRNA from both PTC and normal thyroid tissues were reversely transcribed to cDNAs with the incorporation of fluorescent dUTP (Cy-5 or Cy-3) to prepare the hybridization probes. The mixed probes were hybridized to the cDNA microarray, which was scanned for the fluorescent signals and showed difference between two tissues. Two among the differentially expressed genes were verified by RT-PCR and immunohistochemistry. Results In PTC 48 genes were upregulated while 17 were downregulated. The results of RT-PCR and immunohistochemistry were consistent with that of the gene chips. Conclusion cDNA microarray technique is effective in screening the differentially expressed genes between 2 different kinds of tissue. The obtained genes are mainly related with extracellular matrix, cytokine, signal transduction and so on.
5.Design the family support scale for adolescent patients with nephrotic syndrome

Objective To design the family support scale for adolescent patients with nephritic syndrome. Methods 192 adolescent patients with nephritic syndrome were selected in the pilot investigation, and then formed the first questionnaire. The reliability and the validity of the questionnaire were evaluated by the experts. Results The questionnaire involved 15 items and its reliability and the validity were satisfied. There was a positive correlation between the family support and the self-nursing degree. Conclusion The reliability and the validity of this questionnaire are both up to the demands, it can used to study the condition of family support for adolescent patients with nephrotic syndrome.
6.Micro-macroscopical and Microscopic Identification of Cinnabaris

OBJECTIVE:To provide identification reference for the clinical use of Cinnabaris. METHODS:TCM micro-macro-scopical identification method and microscopic identification method were used. RESULTS:The micro-macroscopical characteristics were obtained:irregular granule or sheet block,different forms,wide bright red,opuque translucent with some luster; some sam-ples showed irregular lump with big shape,red scale on surface,dulling or gray-black. The were microscopic characteristics ob-tained:different forms of irregular granule,some sheet block,wide bright red,with some luster,occasionally with yellow gran-ules. CONCLUSIONS:The method for micro-macroscopical identification and microscopic identification of Cinnabaris is simple and convenient,and it can be used for the rapid verification of Cinnabaris.
7.Study on eutopic endometrial blood in endometriosis with contrast enhanced ultrasound

Objective To evaluate eutopic endometrial blood supply and vessels in patients with endometriosis(EMs) with contrast enhanced ultrasound (CEUS),to explore a newmethod to estimate vascularization of eutopic endometrium in EMs patients.MethodsThirty patients with EMs were enrolled and 20 patients who were diagnosed as other gynecological diseases as control.Informed consent of each case was obtained. AllcasesunderwentconventionalultrasoundexaminationandCEUSexamination.Characteristics of CEUS in each patients were observed and analyzed using off-line Qontrast software.Parameters of CEUS including peak intensity(PEAK),time to peak(TTP),regional blood volume(RBV),regional blood flow(RBF) were obtained automatically.ResultsComparing with control group,the eutopic endometrium in EMs group presented higher enhancement.The parameters of CEUS,including PEAK,TTP,RBV,RBF were (41.18 ± 3.29) dB,(29.01 ± 4.46) s,(29.07 ± 4.59) ml,(48.61 ± 5.35) ml/min in EMs group and (13.36 ± 2.34)dB,(24.59 ± 2.29)s,(26.51 ±- 3.80)ml,(48.71 ± 3.80)ml/min in controls respectively,the value of PEAK in EMs group was higher than that of controls ( P =0.000).Conclusions CEUS can be regarded as a new method which can be used to assess vascularization of eutopic endometrium in endometriosis.
8.Nursing observation of adverse reactions of advanced lung cancer treated with Apa imatinib mesylate

Objective Study on 60 cases of advanced non-small cell lung cancer patients in the Apa imatinib mesylateadverse reactions after treatment , and to develop nursing methods for these adverse reactions. Methods Non-small cell lung cancer patients who were treated in our hospital from September 2015 to September 2016 were treated with Apa imatinib mesylate and observed the adverse reactions during the course of treatment. Results Adverse reactions occurred in 45 patients, and the incidence of adverse reactions was 75.00%.29 cases of patients with skin rash, 24 cases occurred in patients with hypertension, 30 cases of patients with diarrhea, nausea and vomiting occurred in 21 patients, 5 cases occurred in patients with proteinuria, 7 cases of patients with oral mucositis.Conclusion Non-small cell lung cancer patients had adverse reactions after treatment with Apa imatinib mesylate and nursing intervention can improve the quality of life of patients.
9.Acute promyelocytic leukemia combined with hereditary fibrinogen deficiency: report of one case and review of literature

Objective:To improve the clinical recognition of hereditary fibrinogen deficiency.Methods:The diagnosis and treatment process of a patient with acute promyelocytic leukemia (APL) complicated with hereditary fibrinogen deficiency who was admitted to the second Affiliated Hospital of Anhui Medical University in December 2018 was retrospectively analyzed, and the relevant literature was reviewed.Results:The patient was initially diagnosed as APL, and the complete remission was obtained after dual-induction therapy of all-trans retinoid acid and arsenous acid. During the first consolidation treatment, repeated reviews of fibrinogen fluctuated between 1.0-1.5g/L, and further improving the fibrinogen gene sequencing to diagnose APL combined with hereditary fibrinogen deficiency.Conclusion:For APL patients in remission who have decreased fibrinogen for many times and patients with hereditary fibrinogen deficiency who have significantly decreased fibrinogen in a short period, bone marrow biopsy and genetic testing should be further conducted to determine the pathogenesis.

Objective To investigate the mechanism of glycoprotein serglycin (SRGN) promoting metastasis of breast cancer cells and the possible mechanism of SRGN expression.Methods Real time quantitative polymerase chain reaction (PCR) and bioinformation retrieval were used to detect the expression of SRGN in lymph node metastasis and non-metastasis breast cancer.MDA-MB-231 shRNA and MCF-7-SRGN of breast cancer stable cell line were established by lentivirus shRNA interferencc and overexpression.Transwell assay was used to test the effect of SRGN on invasion and metastasis of breast cancer cell line in vitro.Western blot assay was used to detect the changes of epithelial-mesenchymal (EMT) related markers.The possible regulatory mechanism of SRGN expression was detected by Western blot assay.Results SRGN expression was significantly increased in lymph node metastasis of breast cancer in clinical specimens.SRGN interference inhibited the invasion and metastasis of tumor cells.SRGN promoted breast cancer cells EMT.Transforming growth factor β1 (TGFβ1) promoted the expression of beta SRGN transcription.Conclusions SRGN can induce the change of EMT in breast cancer cells and promote the invasion and metastasis of breast cancer cells.
