1.Long term effect of calcium carbonate bone cement combined with bFGF in repairing pulp chamber perforation

Objective To investigate the long-term efficacy of calcium phosphate cement combined with bFGF in repairing pulp chamber perforation and to analyze the correlation between the diameter of the perforation and the curative effect.Methods 75 patients with pulp chamber perforation (82 teeth) were enrolled and divided into the observation group and the control group according to the repair material and method.A series of subgroups were also modeled according to the diameter of the perforation,which include the control group A (≤ 1.5 mm),control group B (1.6~3 mm) and control group C (>3 mm),as well as the observation group A (≤ 1.5 mm),observation group B (1.6~3 rmm) and observation group C (>3 mm).The observation group was treated with calcium phosphate cement combined with bFGF,and the control group was treated with calcium phosphate cement alone.Results The total effective rate of the observation group was 97.8%,which was significant higher than 80.6% in the control group (P<0.05).The cure rate of the observation group A was 100%,which was significant higher than 73.3% in the observation group B and 41.7% in the observation group C (all P<0.05).The total effective rates of the observation group A and B were significantly higher than 91.7% in the observation group C (all P<0.05).The cure rate of the control group A was 92.9%,which was significant higher than 60.0% in the control group B and 25.0% in the control group C (all P<0.05).The total effective rates of the control group A (100%) and B (90.0%) were significantly higher than 91.7% in the control group C (all P<0.05).Conclusions Calcium phosphate cement combined with bFGF in repairing the pulp chamber perforation was significantly better than calcium phosphate cement alone.The cure rate of perforation repairing is closely related to the perforation size.The perforation with small diameter may achieve a better repairing effect.

OBJECTIVE:To provide reference for the practice of monitoring on adverse drug reactions (ADRs) in secondary or inferior to secondary medical institutions.METHODS:The problems in the practice of ADR monitoring in secondary or inferior to secondary medical institutions were analyzed and the approaches to strengthen the ADR monitoring was discussed taking our hospital as an example.RESULTS & CONCLUSIONS:The problems in the practice of ADR monitoring in secondary or inferior to secondary medical institutions manifested as low general technical level,lacking of advanced information network,hospital leaders' negligence,medical staff's poor consciousness,pharmacy departments' failure in bringing their role into full play,etc.Therefore,it is urgent to attach great importance to the problems to facilitate the development of ADR monitoring by enlarging publicizing,raising awareness;improving the organizational structure and the related working rules,bringing clinical pharmacists and nursing staff's roles into full play and establishing the reward and punishment mechanism,etc.
3.Clinical Study of the Collagenase Injection Combined Oxygen-Ozone for the Treatment of Lumbar Disc Herniation

Objective To assess the therapeutic effect of oxygen-ozone combined collagenase injection for the treatment of lumbar disc herniation.Methods Oxygen-ozone and collagenase were injected into the lumbar disc or outside the lumbar disc to treat the lumbar disc herniation.After the treatment,the patients were followed-up for 6~12 months,then the results were assessed according to the Macnab criteria.Results Of 183 patients,the effect was excellent in 144,good in 18,fair in 16 and poor in 5.The curative rate was 88.5%,the effective rate was 97.3%.No serious complications occurred in this group.Conclusion Oxygen-ozone combined collagenase injection is very effective,safe,and cheap method for treating lumbar disc herniation.
4.Percutaneous treatment of lumbar disc herniation by oxygen-ozone injection-clinical study with indication

Objective To study the clinical effect and the range of indications of oxygen-ozone treatment for lumber disc herniation. Methods 6~15 ml of oxygen-ozone(35~45 ?g/ml) were injected percutaneously into lumbar disc.In case of multiple disc herniations,the proceduce could be taken with two discs for once. Results 323 patients with 433 discs were treated by oxygen-ozone injection procedure.Total effective rate was 77.7%. Conclusions The treatment of lumber disc herniation by oxygen-ozone injection is simple,safe and effective with mild trauma.Oxygen-ozone not only can oxidize the proteoglycan in the nucleus leading to the contraction of nucleus,but also provide anti-inflammation effect with pain relief and without complication yet.
5.miR-34c suppresses cell proliferation and invasion of prostate cancer PC3 cells

Objective To explore the effect and molecular mechanism of miR-34c in cell proliferation and invasion of human prostate cancer PC3 cells.Methods miR-34c was synthesized and transfected into prostate cancer PC3 cells.The ability of cell pro liferation was examined with methyl thiazolyl tetrazolium (MTT),and invasion with Transwell assay.The level of c-met mRNA was analyzed by real-time reversing transcription polymerase chain reaction (RT-PCR).Western blot was used to detect the expressions of cmet,cyclin D1,and EMT-associated markers.Results Over-expression of miR-34c reduced PC3 cell proliferation,and inhibited the abilities of invasion.Bioinformatics analysis showed that 3'-UTR of c-met had two miR-34c matched sites,and miR-34c inhibited the expressions of c-met mRNA and protein.Meantime,miR-34c obviously decreased the expressions of cyclin D1 and N-cadherin,but increased the expression of E-cadherin in PC3 cells.Conclusions miR-34c may suppress cell proliferation and invasion in prostate cancer PC3 cells,by which mechanism is related to reduction of the expressions of c-met,cyclin D1,N-cadherin,and upregulation of E-cadherin.
6.Determination of Mercury in Blood by Atomic Fluorescence Spectrometry with a Thiourea Extraction Method

Objective To develop a method for determination of mercury in blood by atomic fluorescence spectrometry. Methods Using a thiourea extraction method, the mercury in blood is determined by atomic fluorescence spectrometry. Results The linearity of calibration curve of mercury was in the concentration of 0.000-10.00 ?g/L. The detection limit was 0.06 ?g/L. The recovery rates were 95.5%-100.3%.The RSDs were 4.1%-4.5%. Conclusion The method has the advantages of simple operation, high sensitivity, good repeatability and is applicable to the determination of mercury in blood.
7.The value of serum progranulin in the clinical diagnosis of adult sepsis patients

Objective:To investigate the value of serum progranulin (PGRN)in the clinical diagnosis of adult sepsis patients.Methods:One hundred and eighty-seveninfection patients admitted to the intensive care unit(ICU) of Affiliated Dongfeng Hospital, Hubei University of Medicinewere divided into non-sepsis group(94 patients) and sepsis group (93 patients)according to the third international consensus definitions for sepsis and septic shock. Patients in the sepsis group were divided into two subgroups according to the degrees of infection: septic shock group (46 patients) and non-septic shock group (47 patients). The levels of serum PGRN, procalcitonin (PCT), C-reactive protein (CRP), interleukin-6 (IL-6), tumor necrosis factor-α(TNF-α) and lactic acid (Lac) were compared between the two groups and subgroups within 1 h into the ICU, as well as the acute physiology and chronic health evaluation Ⅱ(APACHE Ⅱ) and sequential organ failure assessment (SOFA) scores within 24 h into the ICU. Logistic regression model was used to analyze the relationship between PGRN and sepsis. The receiver operating characteristic (ROC) curve of the subject was drawn. The diagnostic value of PGRN in sepsis was evaluated and compared with PCT, CRP, IL-6, TNF-α and Lac.Results:The levels of PGRN, PCT, CRP, IL-6, TNF-α, Lac and APACHEⅡ, SOFA scores in the sepsis group were higher than those in the non-sepsis group: (129.25 ± 17.81) μg/L vs. (43.17 ± 7.68) μg/L, (5.92 ± 0.82) μg/L vs. (1.34 ± 0.17) μg/L, (64.07 ± 10.51) mg/L vs. (37.18 ± 5.44) mg/L, (111.68 ± 13.17) ng/L vs. (32.41 ± 5.61) ng/L, (86.06 ± 12.19) ng/L vs. (46.44 ± 7.63) ng/L, (2.96 ± 0.45) mmol/L vs. (1.47 ± 0.22) mmol/L, (23.62 ± 4.24) scores vs. (11.74 ± 2.07) scores, (14.84 ± 2.42) scores vs. (1.36 ± 0.23) scores, and the differences were statistically significant ( P<0.05). The levels of PGRN, PCT, CRP, IL-6, TNF-α, Lac in the septic shock group were higher than those in non-septic shock group:(143.29 ± 13.54) μg/L vs. (116.59 ± 10.73) μg/L, (7.64 ± 1.17) μg/L vs. (4.24 ± 0.59) μg/L, (74.49 ± 10.46) mg/L vs. (53.89 ± 8.41) mg/L, (124.48 ± 14.37) ng/L vs.(99.16 ± 13.61) ng/L, (95.91 ± 14.75) ng/L vs. (76.42 ± 11.24) ng/L, (3.52 ± 0.46) mmol/L vs. (2.45 ± 0.39) mmol/L, and the differences were statistically significant ( P<0.05). Logistic regression analysis showed that SODA scores and serum PGRN, PCT, CRP, IL-6, Lac levels were independent risk factors for sepsis ( P<0.05). The ROC curve analysis showed that the area under the curve (AUC) of PGRN was higher than that of CRP, IL-6, TNF-α and Lac for predicting the occurrence of sepsis in infection patients ( P<0.05). The AUC of PGRN was higher than that of CRP, IL-6, TNF-α and Lac for predicting the occurrence of septic shock in infection patients ( P<0.05). Conclusions:The levels of serum PGRN is a good biomarker for the diagnosis of sepsis and could reflect the severity. It has certain clinical value.
8.The effect of valproic acid on behavior and senile plaques in the male and female APP/PS1 double transgenic mouse model

Objective To investigate whether valproic acid (VPA) affect spatial learning memory and senile plaques in the APP/PS1 double transgenic AD mouse model of different gender. MethodsTwenty 3-month old APP/PS1 double transgenic AD mice,male and female mouse evenly,were randomly divided into VPA-treated and saline-treated groups ( 10 for each group). 30 mg· kg-1 · d-1 of VPA and the same amount of saline were peritoneally injected into mice for 4 weeks. Morris water maze was conducted to check the effect of VPA on the capability of spatial learning and memory of AD mouse model. Immunohistochemical staining was used to examine the effect of VPA on the morphological changes in the brains of mice. ResultsVisible platform test showed that VPA-treated and saline-treated mice had similar escape latency (P>0.05) and path length (P>0.05) ,the swimming speed between male and female mice had no difference (P>0.05). Hidden platform test showed that VPA treated mice had a significantly shorter latency (P<0.01) and path length (P<0.01) to reach the platform compared with saline-treated mice. Meanwhile, both in VPA-treated and control groups, the male mice had a shorter correlation escape latency and path-length than female mice had(P<0.05 ). Immuohistochemical staining showed that the number (11.23±3.78) of senile plaques (SP) in the cerebral cortex and hippocampus of VPA-treated male mice were notably decreased than that(28.17 ±3.46) in the control group ( t= 14.67, P<0.01 ),furthermore,the number of SP in the cerebral cortex and hippocampus of VPA-treated male mice was significantly reduced,as compared with which (20.36 ±4.21)in the VPA-treated female mice(P<0.05). ConclusionVPA can significantly lower formation of SP, and remarkably improve the capability of spatial learning and memory of APP/PS1 transgenic mice,which have gender difference.
9.Efficacy of itopride in functional dyspepsia patients overlapping constipation-predominant irritable bowel syndrome

Objective To assess the efficacy of itopride on treatment of patients with functional dyspepsia (FD) overlapping constipation-predominant irritable bowel syndrome (C-IBS). Methods Patients who met criteria for FD and FD overlapping C-IBS were randomly assigned into FD treatment group (group A), FD control group (group B), FD overlapping C-IBS treatment group (group C) and FD overlapping C-IBS control group (group D). The patients in group A and group C received 100 mg of itopride 3 times daily for 8 weeks. Dyspeptic symptoms including abdominal pain, bloating, early satiety and constipation, were evaluated before and after treatment. Ultrasonic monitoring of gastric emptying function was performed in group A and group C before and two weeks after treatment.ResultsThe symptoms of FD were relieved in both group A and group C (P<0.05), while better results were shown in group C. The significant improvement of constipation was seen in group A and group C. Besides, after medication, gastric emptying was improved in group A and group C in comparison with group B and group D. Conclusion Itopride is an effective therapeutic option in the treatmentping of patients with overlapping of FD and C-IBS.
10.Effects and mechanisms of histone deacetylase inhibitors on invasion and metastasis of Her2 positive breast cancer cell

Objective To investigate the effect of histone deacetylase inhibitor on Her2 positive breast cancer cell line BT474 and SKBR3 in apoptosis and metastasis.Methods Histone deacetylase inhibitor MS-275,suberoylanilide hydroxamic acid (SAHA)(4 μmol/L,and 50 μmol/iL,respectively) treated the cell lines BT474 and SKBR3 cells.Flow cytometer examined the apoptosis ratio.Transwell tested their metastatic activity.Western blot assay was performed to detect the associated proteins.Results SAHA and MS-275 inhibited the cell survival.The BT474 cell survival was (39 ± 11) %,(54 ± 8) %,and the SKBR3 survival was (62 ± 6) %,(71 ± 9) %,according to the fluorescence-activated cell sorting (FACS) result.SAHA and MS-275 induced the BT474 cell apoptosis 8.46± 0.28 (P <0.01),4.15 ± 0.71 (P <0.01) fold change,respectively;and upregulated the SKBR3 cell apoptosis ratio 5.51 ± 1.24 (P <0.01),4.04 ±0.69 (P <0.01) fold.The Transwell result showed that SAHA,MS-275 inhibited the Transwell ability of BT474 from the control 184.7 ± 18.8 to 104.3 ± 7.1,131.3 ±9.1 per view,and the SKBR3 from control 60 ± 16.7 per view to 14.3 ± 6.5,34.3 ± 8.7 per view.The Western blot result showed that SAHA,MS-275 inhibited the protein level of vimentin,Her2,β-catenin,histone deacetylase inhibitor (HDACi),and upregulated the acetylation level of histone 3.The E-caherin protein was regulated in BT474 and SKBR3 cells.Conclusions MS-275,SAHA can induce BT474 and SKBR3 apoptosis significantly,also inhibit their metastatic activity.
