China Pharmacist 2017;20(3):426-428,437
Objective:To study the effect and underlying mechanism of rosmarinic acid in the apoptosis of HepG2 cells. Meth-ods:An MTT method was used to determine the inhibitory effect on the growth of HepG2 cells treated with rosmarinic acid for 48 hours. Flow cytometry was used to detect the cell apoptosis after treated with rosmarinic acid at different concentrations for 24 h and 48 h. Western Blotting was used to detect the expression of P53 and c-Myc protein after treated with rosmarinic acid for 48 h. Results:When HepG2 cells were treated with rosmarinic acid at the concentrations of 12. 50, 25. 00, 50. 00 and 100. 00 μg·ml-1 for 48 h, the cell growth was inhibited and the half-inhibitory concentration (IC50) was 43. 48μg·ml-1. By comparing HepG2 cells treated with rosma-rinic acid at different concentrations for 24 h and 48 h, rosmarinic acid had the effect on the early apoptosis of HepG2 cells after the 48-hour treatment. When treated with rosmarinic acid at the different concentrations for 48 h, the expression of c-Myc decreased with the concentration increase of rosmarinic acid, and the expression of P53 increased with the concentration increase of rosmarinic acid. Con-clusion:The inhibitory effect of rosmarinic acid on the proliferation of HepG2 cells is in a dose-dependent manner, and it can promote the apoptosis of HepG2 cells through the activation of P53 and the cleavage of proto-oncogene c-Myc protein.

Objective To compare the impact of conventional vs. zero-balanced ultrafiltration on serum pro-inflammatory factors, acute kidney injury and clinical prognosis after cardio-pulmonary bypass procedure. Methods Forty patients receiving cardiac surgery under cardio-pulmonary bypass procedures in Xiangyang Central Hospital during January 2013 to June 2013 were randomly divided into conventional ultrafiltration group(group A,n=20)and zero-balanced ultrafiltration group(group B,n=20). Blood and urine samples were collected on different time points( pre-operation,T0;pre-ultrafiltration,T1;immediately after ultrafiltration,T2;24 hours post-operation,T3;48 hours post-operation,T4;7 days post-operation, T5). TNF-α,IL-6,KIM-1,CysC,serum creatinine and urea nitrogen were detected and compared. Pre-and post-operative clinical data were also collected. Results There was no difference in baseline data or intra-operative data(p>0. 05). TNF-αand IL-6 started to increase when the operation began. Compared with conventional ultrafiltration,zero-balanced ultrafiltration alleviated the increase of TNF-α and IL-6,espe-cially on T2,T3,and T4(p<0. 05). Zero-balanced ultrafiltration also decreased CysC,Rbp,serum creati-nine and urea nitrogen on T2,T3,and T4 compared with conventional ultrafiltration(p<0. 05). A total of 11 patients experienced AKI( >grade I)in group A and 2 patients experienced in group B(p<0. 01). There were significant differences of ventilation time, total complication incidence and ICU stay time be-tween two groups. There was no difference in other complications,post-operative days in hospital or death rate within 30 days. Conclusion Though there is a trend of more patients receving renal replacement therapy,no statistical difference has been achieved. In conclusion,zero-balanced ultrafiltration can effec-tively decrease the concentration of serum pro-inflammatory factors, alleviate acute kidney injury and improve the clinical prognosis after cardio-pulmonary bypass procedures. It is a safe and reliable method valuable for promotion.

Objective To observe the relationship of liver ultrasound class and urinary albumin excretion ratio (UAER) in type 2 diabetes mellitus (T2DM) combined with nonalcoholic fatty liver disease (NAFLD). Methods One hundred and ninety-seven T2DM patients were divided into 3 groups according to the degree of hepar adiposum: group A (66 subjects without NAFLD), group B (63 subjects with mild NAFLD) and group C (68 subjects with moderate or severe NAFLD). Their clinical indexes,UAER and biochemical parameters were measured and compared, the relative analysis of blood fat, HOMA-IR and UAER was done. Results Compared with those in group A, the levels of UAER were significantly increased [(86.49 ± 65.19) mg/24 h vs. (115.16 ± 101.99) mg/24 h vs. (159.45 ± 149.08) mg/24 h,P < 0.05], and the levels of high density lipoprotein cholesterol decreased in group B and group C[(1.21 ± 0.37) mmol/L vs.(1.05 ± 0.38) mmol/L vs. (0.99 ± 0.21) mmol/L,P < 0.05]. Multiple stepwise regression analysis showed that triglyeride was the most important risk factor affecting UAER(P < 0.05). Conclusions There is a close relationship between NAFLD and UAER in T2DM. In the subjects with moderate or severe NAFLD, the UAER increases which indicates that these patients already have capillary vessel injury apparently.
