Objective To investigate whether RNA interference in mediated Bcl-2 gene silence can decrease the gallbladder carcinoma xenograft formation rate and inhibit growth of xenograft in nude mice. Methods In the xenograft formation ability group, the Bcl-2-siRNAs were transfected into the gallbladder carcinoma cell.Transfected gallbladder carcinoma and gallbladder carcinoma were injected into nude mice hypodermically to form a gallbladder carcinoma tumor. The formation rate and volume of the tumors were measured and the tumor growth rates were calculated. In the therapy group, the animal models of human gallbladder carcinoma in nude mouse body were constructed. The nude mice were randomly divided into pSilencerTM-EGFP sh515 group,pSilencerTM-EGFP shCon group, and normal control group. After the formation of tumor knot,siRNA expressing plasmids pSilencerTM-EGFP sh515 and pSilencerTM-EGFP shCon were injected into the knots of pSilencerTM-EGFP sh515 group and pSilencerTM-EGFP shCon group. The tumor volume was measured and tumor growth rate was calculated. All mice were sacrificed after 6 weeks. The distribution of Bcl-2 in the tumors was detected by immunohistochemistry. Results The nude mouse model of human gallbladder cancer xenograft tumor was constructed successfully.Xenograft formation ability:The xenograft formation rate of gallbladder carcinoma and Bcl-2 siRNA transfected gallbladder carcinoma was 100% and 60%,respectively;the average tumor volume in gallbladder carcinoma group and transfected gallbladder carcinoma group was(1 914.6?125.0)mm3 and(629.7?78.9)mm3;the growth rate was 45.58% and 14.99%,and the average tumor weight was(2.24?0.33)g and(0.77?0.12)g,respectively.The tumor volume, weight and growth rate in the transfected gallbladder carcinoma group decreased significantly when compared with those in control group(P

Objective To investigate the association of glyoxalase Ⅰ (GLO1) gene polymorphisms with susceptibility to atherosclerotic cerebral infarction (ACI).Methods Single nucleotide polymorphisms (SNPs) rs1130534 and rs1049346 in the GLO1 gene were genotyped in 300 ACI patients and 300 healthy controls using the SNaPshot technique.Additionally, quantitative real-time PCR was employed to determine the GLO1 expression levels in 50 ACI patients and 50 healthy controls, respectively. Results In terms of the rs1049346 polymorphism, the respective frequencies of CC,CT and TT genotypes were 9.3%,42.7% and 48.0% in the ACI patients, and 14.0%,47.7% and 38.3% in the control group. The respective frequencies of C and T alleles were 30.7% and 69.3% in the ACI patients, and 37.8% and 62.2% in the control group. There were statistically significant differences in the genotype and allele frequencies of rs1049346 between the ACI patients and controls (genotype:x2 =6.877,P =0.032;allele:x2 =6.842,P=0.009).For rs1130534,the respective frequencies of AA,AT and TT genotypes were 52.0%,42.0% and 6.0% in the ACI patients, and 60.0%,33.7% and 6.3% in the control group. The respective frequencies of A and T alleles were 73.0% and 27.0% in the ACI patients, and 76.8% and 23.2% in the control group. However,no statistically significant differences were found in the distribution of genotypes or alleles of the rs1130534 SNP between the ACI patients and controls (all P > 0.05).The results of haplotype analysis revealed that the frequencies of the A-T and T-T (rs1130534-rs1049346) haplotypes in the ACI patients were all significantly higher compared to the controls (42.3% vs 39.0%,x2 =4.733,P =0.030;27.0% vs 23.2%,x2 =5.699,P =0.017).Additionally,the GLO1 expression levels in the ACI patients were significantly lower than that in the healthy controls (Mann-Whitney U =911.5,P =0.020).Conclusion The results indicate that the rs1049346 polymorphism of GLO1 gene is associated with the susceptibility to ACI.

Acute respiratory tract infection is the most common infectious disease in children, which seriously threatens children′s health.Rapid and accurate etiological diagnosis is of great significance for the clinical treatment and control of these diseases.Pathogen nucleic acid test was applied and became the main method of respiratory tract infection diagnosis for its high sensitivity and specificity. To regulate the application of pathogen nucleic acid amplification test in respiratory tract infection in children, improve the diagnosis level, expert consensus on nucleic acid amplification test of respiratory pathogens in children was prepared to guide the application and promote pathogens diagnosis ability.

Objective:To investigate the knowledge, attitudes, and practices (KAP) of pulse oximetry among pediatric healthcare providers in China and analyze the factor influencing the KAP.Methods:A self-developed questionnaire was used for an online research on the KAP of 11 849 pediatric healthcare providers from 31 provinces, autonomous regions, and municipalities of China from March 11 to 14, 2022.The factors influencing the KAP of pulse oximetry among pediatric healthcare providers were examined by Logistic regression. Results:The scores of KAP, of pulse oximetry were 5.57±0.96, 11.24±1.25 and 11.19±4.54, respectively. The corresponding scoring rates were 69.61%, 74.95%, and 55.99%, respectively. Logistic regression results showed that the gender and working years of pediatric healthcare providers, the region they were located, and whether their medical institution was equipped with pulse oximeters were the main factors affecting the knowledge score (all P<0.05). Main factors influencing the attitude score of pediatric healthcare providers included their knowledge score, gender, educational background, working years, region, medical institution level, and whether the medical institution was equipped with pulse oximeters (all P<0.05). For the practice score, the main influencing factors were the knowledge score, gender, age, and whether the medi-cal institution was equipped with pulse oximeters (all P<0.05). Conclusions:Chinese pediatric healthcare providers need to further improve their knowledge about and attitudes towards pulse oximetry.Pulse oximeters are evidently under-used.It is urgent to formulate policies or guidelines, strengthen education and training, improve knowledge and attitudes, equip more institutions with pulse oximeters, and popularize their application in medical institutions.
