1.Effect Evaluation of Spironolactone in the Treatment of Hypertensive Heart Disease by the Real Time 3 -dimensional Echocardiography

Objective To investigate the effect evaluation of spironolactone in the treatment of hypertensive heart disease by the real time 3-dimensional echocardiography .Methods Totally 104 patients with hypertensive heart disease were in the study from Jan 2011 to Jan 2012 of our department and divided into objective group and control group .Conventional treatments were put in all the patients and the spironolactone was used in objective group patients only .The results of ventricular morphology and function indexes were compared be -tween the two groups, and then investigate the relationship between cystatin C (CycC) and left ventricular remodeling index (LVRI) at the same time.Results There were significant differences in the comparison of ventricular morphology and function indexes .The left ventric-ular end-diastolic volume(LVEDV), left ventricular end systolic volume (LVESV) and left ventricular ejection fraction (LVEF) of ob-jective group were more advantageous than flowse of control group for prognosis .The same results were observed in the comparison of Cys C and LVRI.Moreover, Cyc C and LVRI had certain correlation in the study .Conclusion The spironolactone can play a positive role in the treatment for the patients with hypertensive heart disease .In the meantime, the RT-3DE is a good evaluation method for the develop-ment of ventricular morphology and function .
2.The clinical study of twice chemonucleolysis with collagenase for the treatment of lumbar disc herniation

Objective To study the feasibility and indication of twice chemonucleolysis with collagenase for the treatment of lumbar disc herniation.Methods Eighty two patients of lumber disc herniation were treated with twice collagenase chemonucleolysis.All patients were followed up for 3 to 12 months and then the clinical results were assessed according to the Macnab criteria retrospectively.Results Eighty two cases were followed up from 3 to 12 months postoperatively.Fifty one cases were excellent,13 cases good,8 as fair and 10 were poor.The rate of excellent plus good reached 78%,the effective rate was 88%.Conclusion Twice chemonucleolysis with strict indications together with prevention of allergic reaction before,during and after the operation;is safe and effective for treating lumbar disc herniation.(J Intervent Radiol,2007,16:258-259)
3.Estrogen promotes ER positive breast cancer cell growth with up-regulating chemokine CXCL12

Background and purpose:Exposure to estrogen is an important independent determinant of the risk of breast cancer.Moreover,chemokines and its corresponding receptors play an important role in breast cancer occurrence and development.Our study is to investigate the impact and related mechanism of estrogen on chemokines expression in breast cancer cells.Methods:Microarray was performed to analyse chemokine and corresponding receptors' mRNA expression in MCF-7 cells treated with estradiol.Real-time(RT)-PCR was applied to confirm the change of chemokine expression at different time point with different dose of estradiol in presence or absence of ICI182,780.ELISA was used to analyse the change of extracelluar secrete protein.Results:Physiological estradiol could obviously up-regulate CXCL12 mRNA expression in breast cancer cell MCF-7,which emerged from 4h after treatment and continued to 24 h.In addition,estradiol could stimulate CXCL12 protein secretion from 2h to 24h after treatment.These effects could be totally inhibited when MCF-7 was treated with ICI182,780(ER antagonist)prior to estradiol.Conclusions:Our study demonstrated that estrogen could up-regulate CXCL12 mRNA expression and its extracelluar protein secretion,which was mediated through ER signaling pathway.
4.Laparoscopic renal plication for treatment of giant hydronephrosis(report of 12 cases)

Objective To describe the initial experience of transperitoneal laparoscopic renal plication for giant hydronephrosis,and to evaluate the clinical feasibility of the procedure. Methods This series included 12 cases of giant hydronephrosis (5 men and 7 women;age ranged from 19 to 64 years with a mean of 42.6 years).Of them 4 cases had upper ureter calculi;2 had ureteropelvic junction(UPJ) obstruction;2,middle ureter calculi;3,nephrolithiasis and 1,middle ureter stricture. The upper urinary tract obstruction was relieved and plication was performed via laparoscope. Results All the 12 operations were completed successfully.The intraoperative blood loss was 50 to 200 ml,operative time was 75 to 210 min.The postoperative follow-up lasted 3 to 18 months.In all these cases,hydronephrosis was relieved and renal function was improved. Conclusions Laparoscopic renal plication is an effective and minimally invasive method and can be considered as an alternative for treatment of giant hydronephrosis.
5.Effect of nerve growth factor on recuperation of optic nerve after contusion by clamping in adult rabbits

Objective To investigate the effect of nerve growth factor (NGF) on recuperate of optic nerve after contusion by clamping in adult rabbits. Methods Sixteen adult rabbits were randomly divided into NGF and the control group with 8 rabbits in each group. After the optic nerve of the right eyes was clamped,tissue engineering nerve containing (0.06 ml) NGF(concentration: 5?10~(-4) g/L, NGF group) and 0.06 ml of PBS (control group) was immediately transplanted into the injured eyes respectively, and 0.02 ml NGF(concentration: 5?10~(-4) g/L, NGF group)and 0.02 ml of PBS (control group) were injected into the vitreous of right eyes respectively. Flash visual evoked potential (FVEP) test was performed on the eyes 1 day, 2 weeks and 8 weeks after the injury. The number of retinal ganglion cells (RGCs) and changes of optic nerves were observed by light microscopy and electron microscopy at the 8th week after contusion,and a computer-image-analysis system was used to count the optic nerve axons. AIM: To investigate whether the expression of integrin ?_3 and the activation of focal adhesion kinase(FAK) are involved in neointima formation after de-endothelium.METHODS: The model of intima hyperplasia was prepared by balloon injury.The levels of osteopontin(OPN),integrin ?_3 and FAK in vascular tissue were detected by Western blotting and immunohistochemistry.RESULTS: There were similar expression patterns in OPN,integrin ?_3 and FAK following balloon injury.The levels of three proteins were markedly increased 3 days after operation and reached the peak at 7th day.The increased FAK was mainly the phosphorylated form.The migration and proliferation of vascular smooth muscle cells was associated with the increase in the expression of integrin ?_3 and FAK,and was parallel with rapid turnover of extracellular matrix(ECM).CONCLUSION: The interaction of cells with ECM mediated by OPN and integrin ?_3 is essential for migration.The integrin ?_3-FAK pathway is involved in neointima formation.
7.The clinical experience of treating second degree burn with WS-frequency spectrum

Objective To observe the efficacy of WS-frequency spectrum in treating second degree burn wounds. Methods 350 cases of second degree burn were treated in the whole couse with WS-frequency spectrum in association with systemic treatment including anti-shock,anti-infection and expecant treatment. Results All the patiens healed,only 23 cases with deep burn area over 20% TBSA resulted in superficial pigmentation and scar formation without dysfuntion. Conclusion WS-frequency spectrum is a good remedy for treating second degree burn. This method is easy and simple to apply and has very good efficacy.
8.Serum levels of macrophage migration inhibitory factor in patients with vitiligo vulgaris

Serum levels of macrophage migration inhibitory factor (MIF) were measured with enzyme linked immunosorbent assay (ELISA) in 60 patients with vitiligo vulgaris (case group) and 30 healthy individuals (control group).The case group was further divided into two subgroups:progressive stage (n =30) and stable stage (n =30).The association of MIF level with vitiligo area and severity index (VASI) score of disease was analyzed.The results showed that the serum MIF levels in patients with progressive stage were significantly higher than those in stable stage and in healthy controls (79.8 ± 38.0) μg/L vs.(48.4 ± 17.6) μg/L vs.(29.6 ± 22.1) μg/L,respectively,both P < 0.001.Serum MIF levels were positively correlated with VASI scores of patients (r =0.48,P <0.001).The results indicate that MIF may be involved in the pathogenesis of vitiligo vulgaris.

Objective To observe the clinical efficieney and safety of Rosiglitazone(Avandia)Monotherapy or combi- nation with oral hypoglycemic agents in treating type 2 diabetes mellitus with Metabolic syndrome.Methods 68 type 2 diabetic patients with Metabolic syndrome were assigned to receive Avandia treatment at doses of 4mg for 12 weeks.Re,. sults Avandia used alone or in combination with oral hypoglycemic agents significantly reduced fasting plasma glucose (FBG)and fasting plasma insulin(FINS).Mean glycosylated hemoglobin values were significantly deereaseel.Home- ostasis model assessmen indicate that RSG reduced insulin resistance.There were statistically significiant decrease in systolic blood pressure(SBP)and diastolic blood pressure(DBP).No significant side-effects on kidney and liver function were found.Conclusion Avandia is an insulin sensitizer that is effective and safe on the treatment of Type 2 Diabetes with Metabolic syndrome by decreasing blood glucose and blood lipid,improving insulin resistance,and main- taining the function of pancreatic island cells.
10.Reduction of inflammatory-related factor expression in experimental acute pancreatitis in Egr-1 knockout mice

AIM: To observe the effects of Egr-1 gene knockout on the expression of inflammatory-related factors in pancreatic tissue in a mouse acute pancreatitis model.METHODS: The experimental pancreatitis was induced by high-dose of cearulein in wildtype mice and Egr-1 knockout mice.The pancreatitis indexes,such as serum amylase,pancreata edema,and myeloperoxidase(MPO) levels in pancreata and lungs were recorded.The mRNA levels of tissue factor(TF),plasminogen activator inhibitor(PAI-1),monocyte chemoattractant protein(MCP-1),Gro-1,IL-6 and ICAM-1 were measured by quantitative PCR.RESULTS: Contrary to wildtype mice,typical pancreatitis was not induced by high-dose cearulein in the Egr-1 knockout mice,not only markedly reduced edema in pancreata and lungs,but decreased MPO levels in lungs as well were found.Furthermore,the mRNA of TF,PAI,MCAP,ICAM-1 and IL-6 in pancreata were significantly decreased in Egr-1 knockout mice.CONCLUSION: The severity of pancreatitis and lung damage is ameliorated in Egr-1 knockout mice stimulated by high-dosage of cearulein,which was probably mediated by decreasing expression of inflammatory-related factors in pancreata,such as TF,PAI,MCP-1,ICAM-1 and IL-6.
