1.Species and drug resistance of Mycobacterium isolated from sputum smear positive patients

Objective To study the species and drug resistance of Mycobacterium isolated from patients with spu-tum smear positive for acid-fast bacillus in Wuj iang city,and provide reference for the prevention and control of tu-berculosis. Methods Sputum specimens with positive smear were cultured,isolated bacteria were identified and performed drug susceptibility testing,drug resistance among different species of strains and between patients with initial and repeated treatment were compared.Results A total of 1 03 Mycobacterium isolates were included in the study,13 of which were nontuberculous Mycobacterium,drug resistance rate was 100.00% ,multidrug resistance (MDR)rate was 84.62% ;90 isolates were Mycobacteriumtuberculosis,81(90.00% )of which were Mycobacteri-umhominis. Drug resistance rate of Mycobacteriumtuberculosiswas 35.56% ,MDR rate was 14.44% . Of 70 ini-tially treated tuberculosis patients with positive sputum smear,14(20.00% )were resistant to drugs,MDR rate was 4.28% (3/70);Of 20 repeatedly treated tuberculosis patients with positive sputum smear,18(90.00% )were resist-ant to drugs,MDR rate was 50.00% (10/20).Conclusion Mycobacteriumtuberculosisis the major isolated strain from patients with positive sputum smear. Drug resistance and MDR rates of nontuberculous Mycobacterium are very high. Drug resistance and MDR rates of Mycobacteriumtuberculosisin repeatedly treated patients are higher than initially treated patients.
2.Effect of hyperthermia on tight junctions of endothelial cells in the in vitro blood-brain barrier model and its molecular mechanism

Objective To discuss the effect of hyperthermia on tight junctions of the endothelial cells in the blood-brain barrier and explore the molecular mechanism. Methods An in vitro blood-brain barrier model was established by coculture of ECV304 and astrocytes. Transendothelial resistance (TER) of in vitro blood-brain barrier was determined by Millicell-ERS system. The morphological change of tight junctions of the endothelial cells in the in vitro blood-brain barrier was determined by the method of silver staining. The expression levels of zonula occluden 1 (ZO-1) and occludin were analyzed by means of semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western bloting. Results After two hours at 43℃, the mean value of TER was decreased from (321.30? 58.59) ??cm2 to (65.67?6.02) ??cm2. The integrity of tight junctions was destroyed and the expressions of ZO-1 and occludin decreased significantly. Conclusions Hyperthermia can destroy the tight junctions of the endothelial cells in the in vitro blood-brain barrier. The expression decrease of ZO-1 and occludin induced by hyperthermia is one of the most important molecular mechanisms.
3.Therapy of murine gastric cancer by 5-FC/CD suicide gene system combined with HSP-PC tumor vaccine

Purpose:To investigate the antitumor effect of 5-fluorocytosine/cytosine deaminase (5FC/CD) suicide gene system combined with heat shock protein 70 peptide complexes(HSP70-PC) tumor vaccine.Methods:5-FC was administrated intraperitoneally after recombinant adenoviruses carrying E.coli CD gene were injected into murine tumor tissue. At the same time,mice bearing tumor were inoculated s.c. with HSP70-PC. Results:The combined therapy led to reduction in tumor growth, and complete tumor regression was observed in 70% of the mice. After the combined therapy, the tumor tissue showed more CD + 4 and CD + 8T cells infiltration, and the murine CTL activities increased significantly. Conclusions:The therapy combining 5-FC/CD system with HSP-PC tumor vaccine is very effective in murine gastric cancer, and has potential for clinical usage.
4.A study on the deletion and mutation of Rb gene in lung cancer

Objective To investigate the deletion and mutation of Rb gene in human lung cancer.Methods Polymerace chain reaction (PCR ) and restriction enzyme analytic techniques were used to detect the exon 14-16 and 22-23 regions of Rb gene in 20 human lung cancer DNA samples and 3 normal human lung tissue DNA samples.Results Two deletions existed in the exon 14-16 regions of two lung cancer DNA samples.Conclusion Deletion of Rb gene is in existance in human lung cancer, especially in small cell lung cancer. This may be useful for studying the cause of lung cancer.
5.The relation between red blood cell volume distribution width and degree of liver fibrosis in patients with chronic hepatitis B

Objective To explore the relationship between red blood cell volume distribution width (RDW) and degree of liver fibrosis in patients with chronic hepatitis B (CHB). Methods One hundred and sixty-four patients with CHB were consecutively selected. According to the fibrosis index based on the 4 factors (FIB-4) index, the patients were divided into 3 groups: low-FIB group (FIB-4 <1.45, 116 cases), mid-FIB group (FIB-4 1.45-3.25, 31 cases) and high-FIB group (FIB-4 > 3.25, 17 cases), and the clinical data and laboratory indexes (including RDW) were compared. The patients were divided into Ⅰ group (RDW<0.113, 43 cases), Ⅱ groups (RDW 0.113-0.119, 40 cases), Ⅲ group (RDW 0.120-0.127, 43 cases) andⅣgroup (RDW>0.127, 38 cases) according to the RDW, and the FIB-4 were compared. The correlation between RDW and FIB-4 was analyzed by Spearman correlation test. Results The RDW and aspartate aminotransferase (AST) in low-FIB group, mid-FIB group and high- FIB group were gradually increased: 0.117 ± 0.011, 0.122 ± 0.010, 0.134 ± 0.018 and (18.9 ± 7.3), (28.2 ± 10.4), (44.1 ± 13.3) U/L, but the platelet was gradually decreased:(211.3 ± 48.8), (159.3 ± 40.0) and (150.4 ± 31.2) ×109/L, and there were statistical differences (P<0.05). The FIB-4 inⅠgroup,Ⅱgroup,Ⅲgroup andⅣgroup was gradually increased:1.07 ± 0.74, 1.11 ± 0.56, 1.35 ± 0.70 and 2.03 ± 1.06, and there was statistical difference (P<0.05). Spearman correlation test result showed that FIB-4 and RDW was positively correlated (r=0.447, P<0.01). Conclusions The level of RDW is closely related with the degree of liver fibrosis in patients with CHB. RDW has a potential value in assessing the progress of liver fibrosis.
6.Clinical features of 68 cases of complicated upper urinary tract infection in children

Objective To study the clinical features of complicated upper urinary tract infection in children,aiming to provide a reference for clinical diagnosis and treatment of the disease.Methods The clinical data of 68 cases with complicated upper urinary tract infection hospitalized at the Department of Urinary Surgery NO.1,Children's Hospital of Shenzhen between January 2013 and August 2015 were retrospectively analyzed.Results A total of 68 cases,in which 44 were male,24 were female,and repeated infections were found in 15 cases.Fever was the most common clinical manifestation(48 cases,70.59%),while frequent urination and odynuria were rare(9 cases,13.24%).A total of 57 strains had been cultured form the submitted specimens which were obtained from the 68 cases,including 41 strains of gram-negative bacteria,15 strains of gram-positive bacteria and 1 strain of fungus.Two different strains were cultured in 3 children.Fourteen strains of Escherichia coli had 11 extended spectrum beta lactamases (ESBLs)-positive strains.In the 11 strains of klebsiella pneumoniae,8 trains were ESBLs-positive.The drug resistant rates of gram-negative bacteria to Ampicillin and Cefuroxime sodium were both more than 90%.The sensitive rates to Piperacillin/Tazobactam was more than 90%.Thirty-one cases of complicated upper urinary tract infection were cured by administering Piperacillin/Tazobactam,while 15 cases were cured by changing Cefuroxime sodium to Piperacillin/Tazobactam according to the drug sensitivity results.Conclusions Clinical manifestations of complicated upper urinary tract infection are untypical,and fever is the most common symptom.Repeated infection is common.The gram-negative bacteria is the dominant pathogen causing the complicated upper urinary tract infection.ESBLs-positive bacteria accounts for high proportion.The drug resistance rate to penicillin and the first,second generation of the cephalosporin is high.The drug sensitive rate of piperacillin tazobactam is high,with good prognosis.
7.Overexpression of von Hippel-Lindau protein inhibits the growth of colon cancer and liver metastasis in mice

Objective To investigate whether overexpression of VHL can inhibit the growth and metasta-sis of colon cancer .Methods Adenovirus vectors containing VHL were used to overexpress VHL protein in vitro and in vivo.The proliferation,apoptosis and migration of colon cancer cells CT 26 were examined in vitro.The ex-pressions of related proteins were detected by Western blot analysis .Subcutaneous and liver metastasis of colon cancer models were established and the therapeutic effects were evaluated .Results Overexpression of VHL pro-tein inhibited the proliferation and invasion of CT26,and promoted the apoptosis of CT26 via the regulation of COX-2,Bcl-2 and MMP-9.Overexpression of VHL protein inhibited the growth of subcutaneous colon cancer and liver metastasis in vivo .Conclusion Overexpression of VHL protien can inhibit the growth and metastasis of colon cancer ,and enhance the apoptosis of colon cancer ,indicating that VHL can be used as a potential target for colon cancer therapy .
8.Repair of high energy injury-induced soft tissue wound of the heel

Objective To investigate the methods and effects of different flaps for repair of high energy injury-induced soft tissue wound of the heel.Methods From January 2002 to June 2012,the patients including 12 males and 9 females aged 18-57 years (mean,32 years) underwent heel soft tissue defect reconstruction.Causes of injury were traffic injury in 11 case and mechanical injury in 10 cases.Dimension of soft tissue defect ranged from 5 cm × 3 cm to 8 cm × 6 cm.Soft-tissue defect was repaired with sural neurovascular flaps at the posterolateral heel in 9 cases (Group A),with posterior tibial artery flaps at the posterolateral heel in 5 cases (Group B),and with medial plantar flaps at the loading area of heel in 7 cases (Group C).Sensory recovery and two point discrimination motion of the ankle joint were observed and compared among groups 12 month after operation.Heel pain was observed during weight bearing and joint activity was evaluated using the visual analogue scale (VAS).Results All the flaps survived,except for one with epidermal necrosis over the distal part,which healed after partial changing medication.Duration of follow-up was 12-24 months.There were no differences in the appearance,texture and contour between the flaps and recipient sites.Flaps showed no ulcer in the weight-bearing area and recovered their protective sense.Patients could walk normally after surgery.At postoperative 1 year,sensory recovery rate of the flaps in Groups A,B and C was 0,20% and 100% respectively (P <0.01).Appearance of the heel in all groups recovered to almost normal.Cases that could start nil weight-bearing exercise without pain accounted for 8 (89%) in Group A,4 (80%) in Group B,and 6 (86%) in Group C (P > 0.05).While heel pain existed in weight-bearing exercise.Difference in VAS was significant among the three groups (P < 0.05),but ankle range of motion was not (P >0.05).Conclusion Medial plantar flaps are suitable for tissue defect of 5-8 cm in length but sural neurovascular flaps and posterior tibial artery flaps should be considered for over 8 cm defect in order to elevate survival rate of the flaps and reconstruct limb function.
9.The effect of drug dependence severity on the relationship between impulsivity construct and cue-elicited craving

Objective To investigate the effect of drug dependence severity on the relationship between impulsivity and craving.Methods 36 abstiuent drug-dependent individuals were recruited in the study.The participants were divided into the heavy depeudence group (HDG) or the low dependence group (LDG) according to the scores of Addiction Severity Index (ASI).The Barratt Impulsiveness Scale (BIS-11) and classical Stroop task were used to measure the trait impulsivity and state impulsivity.Block designed cue-induced craving paradigm was presented to measure cue-elicited craving.Results For the HDG,a significant positive correlation was found between trait impulsivity (the mean value of BIS-11-CI scale was (39.03± 16.50)) or state impulsivity (the difference of reaction time between congruent and incongruent situation was (87.77±36.95)ms)and cue-elicited craving (0.83± 1.91)(r=0.487,0.500,P<0.05).However,for the LDG subjects,the impulsivity was not found significantly correlated with the cue-elicited craving(r=-0.261,0.081,P>0.05).Conclusion The addiction severity influences the relationship between impulsivity and craving,and impulsivity can only be used as a predictor of relapse in HDG.The findings suggest that the drug may influence the shared brain mechanism between impulsivity and craving.
10.Involvement of p38 kinase signaling in PC12 cell death induced by nitric oxide

AIM: To study signal transduction pathway of nitric oxide-induced PC12 cell death. METHODS: After PC12 cells were incubated with sodium nitroprusside(SNP), caspase-3 inhibitor Ⅱ plus SNP or p38 inhibitor-SB203580 plus SNP, cell survival rate was quantified by MTT assay and caspase-3 activity was measured with caspase-3 assay kits. RESULTS: SNP induced PC12 cell death in a dose- and time-dependent manner, and increased caspase-3 activity gradually. Both caspase-3 inhibitorⅡand SB203580 reduced cell death significantly, but SB203580 reduced caspase-3 activity significantly. CONCLUSION: NO might induce PC12 cell death through the activation of p38 and caspase-3.
