1.Obstructive jaundice caused by hepatocellular carcinoma (a report of 16 cases)

Objective To investigate the diagnosis and treatment of obstructive jaundice (OJ) caused by hepatocellular cacinoma (HCC) invasion to bile duct. Methods The diagnosis and treatment of 16 cases of OJ caused by HCC in our hospital from January 1989 to December 1998 were retrospectively analysed. Results Correct diagnosis was made in 2 cases and misdiagnosis in 14 cases preoperatively. 14 cases were operated on, including hepatectomy, enucleation of the tumor in the common bile duct (CBD) and T tube drainage in 2 cases; enucleation of the tumor in CBD and internal stent of T tube drainage in 11 cases; tumor biopsy and T tube drainage in 2 cases; one case died without operation. 15 cases were followed-up for 1 to 14 months postoperatively. The results demonstrated that 14 patients died within 6 months, and only 1 case remained alive for 14 months after operation. Conclusions The correct diagnosis of this disease could be made for the patients with jaundice accompanied with positive of HbsAg and AFP, local lesions in the liver and the dilated bile duct. B-US, CT, PTC and ERCP are the main examination methods for the diagnosis. The best treatment of this kind of HCC is to remove the hepatic tumor and to recanalize the affected bile ducts.

Human gliomas are one of the most aggressive tumors in brain.Radiotherapy plays an important role for patients with gliomas,as well as surgery.The efficacy of radiotherapy is associated with radiosensitivity of human gliomas.Radiosensitive genes of gliomas and apoptosis、cell cycle transformation、DNA damage induced by irradiation and DNA repair promoted by them are suggested to be associated with glioma radiosensitivity.The rediosensitive genes associated with apoptosis and DNA repairment are becoming hot spots of study.The other radiosensitivity genes are paid attention too.This paper makes a summary of current situation and progress for radiosensitive genes of human brain glomas.

5.Construction and identification of a lentiviral vector harboring RNAi sequence targeting the human high mobility group A1 gene

Objective: To construct and identify a lentiviral vector harboring RNAi sequence targeting the human high mobility group A1(HMGA1) gene.Methods: The effective sequence of siRNA targeting the HMGA1 gene confirmed in our previous study,the complementary DNA containing both sense and antisense Oligo DNA of the targeting sequence was designed,synthesized and cloned into the pGCL-GFP vector diced by the restriction enzyme of HpaⅠ and XhoⅠ,which contained the U6 promoter and green fluorescent protein(GFP).The resulting lentiviral vector containing HMGA1 shRNA was named LV-sh HMGA1 and confirmed by PCR and DNA sequencing.A total of 293T cells were cotransfected with LV-sh HMGA1,pHelper 1.0 and pHelper 2.0.All the virus stocks were produced by Lipofectamine2000-mediated transfection.The titer of the virus was tested according to the expression level of GFP.Results: PCR analysis and DNA sequencing demonstrated that the RNAi sequence targeting the human HMGA1 gene was successfully inserted into the lentiviral vector.The titer of the recombinant lentiviral vector was 5?107 TU/ml.Conclusion: The successful construction of the lentiviral vector of HMGA1 has prepared the ground for further studies on the functions of the HMGA1 gene with the RNAi technique.
6.Effect of continuous renal replacement therapy on the levels of IL - 27 in peripheral blood of patients with chronic heart failure concurrence acute kidney failure

Objective To investigate the effect of continuous renal replacement therapy on the levels of IL-27 in peripheral blood of patients with chronic heart failure concurrence acute kidney failure. Methods 78 patients with chronic heart failure were enrolled from June 2012 to December 2013 in Zhujiang hospital affiliated Southern medical university. According to patient′s condition, 38 patients with chronic heart failure concurrence acute kidney failure undertaking CRRT, 40 patients with chronic heart failure undertaking conventional therapy. Heart function were determined by cardiac ultrasound, the levels of IL-27 and BNP were determined by ELISA. Results Compared with chronic heart failure patients, the levels of IL-27 were decreasing but BNP were increasing in patients with chronic heart failure concurrence acute kidney failure (P < 0.05). After CRRT, the levels of IL-27 were increasing but BNP were decreasing in patients with chronic heart failure concurrence acute kidney failure. Correlation analysis showed that the levels of IL-27 is negatively correlated with the levels of BNP , but positively correlated with LVEF (P<0.05). Conclusion IL-27 participated in the development of chronic heart failure concurrence acute kidney failure. CRRT regulating and controlling the heart and kidney function may partly through effecting on immune-inflammatory mechanism.
7.Study of Microwave-assisted extraction on flavonoids of Radix Glycyrrliza

Objective: To search for the best condition of microwave assisted extraction(MAE) for the flavonoids in Radix glycyrrliza and to compare with traditional boiling water extraction(WE). Methods: MAE and BWE were adopted, and its proper experimental conditions were examined and optimized by means of uniform design. The flavonoids in Radix glucyrrliza were determined by spectrophotometry. Results: Appropriate MAE conditions included extraction time of 1min, ethanol concentration of 38%, liquid/solid ratios of 8∶1(ml/g) and the power of microwave oven of 288W, the recovery of flavonoids from Radix glycyrrliza with MAE(24.6mg/g) was higher than BWE (11.4mg/g). Conclusion: Compared with traditional boiling water extraction(BWE), the experiment results demonstrated higher extracting rate and shorter extracting time. It indicated that MAE was more suitable to extraction of the flavonoids.
8.Role of CD46 and Nectin-4 on Measles virus (MV) infecting human pulmonary alveolar epithelial cells

Objective:To investigate the role of CD46 and Nectin-4 on Measles virus (MV) infecting human pulmonary alveolar epithelial cells (HPAEpiC),and the interaction between CD46 and Nectin-4.Methods: Measles virus was divided into pre-infection group and 2 h-infection group,HPAEpiCs treatment with anti-CD46 antibody and/or anti-Nectin-4 antibody as experimental groups,and untreated HPAEpiCs as a control.The variation of viral replication level was detected.A Co-immunoprecipitation assay (Co-IP) was used to explore whether CD46 and Nectin-4 had interactive relationship in MV infection.Results: Compared with the control group,MV titers were reduced in HPAEpiCs of the pre-infection group treated with anti-CD46 and anti-Nectin-4 respectively (48.03% and 49.53%).Furthermore,virus titers showed a more reduction in which treated with anti-CD46 and anti-Nectin-4 antibodies (27.15%,P<0.01).Western blot and Real-time PCR showed that anti-CD46 antibody and anti-Nectin-4 antibodies decreased the rate of MV infection.In the 2 h-infection group,however,the treatment with anti-CD46 and anti-Nectin-4 could significantly reduce the MV titer and NP protein in HPAEpiCs.The Co-IP assay showed that there were interaction between CD46 and Nectin-4.Conclusion: CD46 and Nectin-4 mediated MV infecting HPAEpiCs.Moreover,CD46 and Nectin-4 may play a synergetic role in MV infection,which could enhance the infection effect.
9.Diterpene ginkgolides meglumine injection inhibits oxidative stress induced by oxygen-glucose deprivation by activating Akt/Nrf2 pathway in SH-SY5Y cells

OBJECTIVE To investigate the protective effects and mechanism of diterpene ginkgolides meglumine injection (DGMI) against oxidative stress induced by oxygen-glucose deprivation (OGD) in SH-SY5Y cells. METHODS SH-SY5Y cells were divided into five groups: normal control, model control (OGD group) and drug(25 mg · L- 1) administration groups including DGMI group, extract of ginkgo biloba leaves injection group (EGBLI) and lactones ginkgo biloba injection group (LGBI). The cells suffered from oxygen-glucose deprivation (OGD) for 4 h, followed by reoxygenation with drugs for 6 h. Then, cell viabilities were detect using CCK-8 assays, reactive oxygen species (ROS) levels using fluorescence probe DCFH-DA and superoxide dismutase (SOD) activities using WST-1 test. Western blotting was used to detected protein levels of hemeoxygenase-1(HO-1), NAD(P)H, quinone oxidore?ductase l (Nqo1), protein kinase B (Akt), phosphorylated Akt (p-Akt), nuclear factor-E2-related factor2 (Nrf2) and phosphorylated Nrf2 (p-Nrf2). The cells were induced by OGD for 4 h, followed by reoxygen?ation and DGMI for 1 h, combined with different concentrations of PI3K inhibitor (LY294002) (at the final concentration of 12.5, 25 and 50 μmol · L-1) before the protein levels of AKT, p-AKT, Nrf2 and p-Nrf2 were detected by Western blotting. RESULTS SH-SY5Y cells induced by OGD for 4 h resulted in an increase in ROS(P<0.01), but a decrease in cell viabilities(P<0.01), SOD activities(P<0.01), and antioxidant protein levels ( Akt, p-Akt, Nrf2, p-Nrf2, HO-1 and Nqo1) (P<0.01). Compared with OGD group, treatment with reoxygenation and drugs (DGMI,EGBLI and LGBI respectively) for 6 h resulted in a decrease in ROS (P<0.01), but an increase in cell viabilities, SOD activities and antioxidant protein levels of p-Nrf2, HO-1, Nqo1 and p-Akt(P<0.05,P<0.01). DGMI group showed the best efficiently. Moreover, after OGD for 4 h, compared with DGMI group, combining reoxygenation and DGMI with LY294002 for 1 h resulted in a concentration-dependent inhibition of the protein levels of p-AKT and p-Nrf2(P<0.01). CONCLUSION DGMI 25 mg · L-1 can inhibit oxidative stress in SH-SY5Y cells induced by OGD by increasing the activity and expression of Nrf2 through PI3K/Akt pathway, which may be one of the mechanisms by which DGMI protects neurons from stroke.
10.Evaluation of the therapeutic effect of arterial PVA embolization for mid and advanced pulmonary carcinoma

Objective To explore the therapeutic effect of arterial PVA perfusion embolization in treating mid and advanced pulmonary carcinoma. Methods Thirty-one patients with pulmonary carcinoma in the mid or late stage diagnosed pathologically underwent selective angiographic studies of the bronchial arteries and intercostal arteries to provide information of the hypervascular feeding vessels, and then performed the perfusion with 5-FU? THP? DDP; followed by PVA grain embolization under fluoroscopic control. Results Abnormal tumor feeding vessels of pulmonary carcinoma were found deriving from bromehial arteries in 21 cases, and from intercostal arteries in 10. After arterial infusion of chemotherapy combined with embolization of PVA,alleviation of clinical symptoms was observed in all 31 cases(100%)without severe complication. The total effectiveness(CR plus PR) reached 80.6%(25/31). Conclusions Arterial embolization of PVA is safe, effective with less complication for treatment of mid or advanced pulmonary carcinoma.
