3.Clinical Observation on Geshanxiaoyao Decoction Combining Acupuncture Effect on the Life Quality of Patients with Irritable Bowel Syndrome

[Objective] To observe the therapeutic effect of diarrhea-predominant irritable bowel syndrome patients’ life quality with Geshanxiaoyao decoction combining acupuncture.[Methods] 300 patients were randomly divided into therapeutic group (n=150) and control 1,2,3 groups (n=50).150 cases were treated with Geshanxiaoyao decoction combined with pricking taichong,sanyinjiao,shaohai; control 1,2,3 groups were respectively treated with Li-Zhu-Chang-Le,Geshanxiaoyao decoction and acupuncture,4 weeks a course.Clinical symptoms and life quality before and after treatment were recorded and analyzed.[Results] The total effective ratio in therapeutic group(89.71%) was higher than that in control 1,2,3 groups (68.52%,74.31%,66.87%) respectively.There was significant difference between therapeutic group and control groups.[Conclusion]Geshanxiaoyao decoction combining acupuncture can effectively improve the life quality of patients with diarrhea-predominant irritable bowel syndrome.
4.SPATIOTEMPORAL PATTERNING OF N-cadherin EXPRESSION ON RAT CARDIOMYOCYTES

Objective To research the spatiotemporal pattern of N-cadherin expression during development of rat cardiomyocytes and age-related changes of N-cadherin expression. Methods Using immunohistochemical method,myocardial N-cadherins distribution was investigated in fetal rat and postnatal development(1 postnatal day to old rat),and quantitied by HIPAS-1000 computerized image analytical instrument. Results The expression of N-cadherin was located in myocardium of atrial and ventricule and septum interventriculare and papillary muscles.The N-cadherin immunolabeling was found in myocyte membranes and within cytoplasm in fetal rat heart.From neonatal to infant rat,the pattern of N-cadherin immunolabeling changed progressively,from being dispersed over the entire cell surface as in the fetal to the transverse terminals of the myocytes,toward the distribution within the intercalated disk.From young to old rat heart,the typical N-cadherin was located in transversely orientated intercalated disk.The percentage of N-cadherin immune postive area in rat ventricular myocardium showed a progressively changement with age.Conclusion The present paper demonstrated that the N-cadherin expression exists progressively with ages and the changing pattern of N-cadherin is closely related with development of the intercalated disk in rat myocardium.The mechanical coupling provided by adherens junctions is essential for the stable cell-cell contact.
5.Age-dependant expression of alpha-macula adherens protein in rat heart

BACKGROUND: Macula adherens protein is found closely associated with congenital cardiac malformation and myocardial differentiation. OBJECTIVE: To investigate the expression characteristics of α-macula adherens protein in rat heart, as well as the property of age-dependant expression during myocardial growth. DESIGN: Randomized controlled, observational comparative study. SETTING: Department of Cell Biology of Xinxiang Medical College; Department of Bioengineering and Agricultural Economics of Puyang Vocational Technical School. MATERIALS: This study was conducted at the Morphological Laboratory of Xinxiang Medical College between January and June 2003. Totally 28 Wistar rats of clean grade were divided into infant group, youth group,middle-age group, and old-age group with 7 rats in each group. METHODS: All rats were anaesthetized and then cardiac tissues were cut into consecutive coronal slices of 5 μm thick. The expression of α-macula adherens protein in rat myocardium of infant, youth, middle-age and oldage groups was detected using IHC method. The positive cells displayed brownish yellow granules on the surface, cytoplasm and intercalated disc. Routine HE staining was performed on all specimens for structural comparison. MAIN OUTCOME MEASURES: The expression of α-macula adherens protein in rat myocardium of different groups. RESULTS: All the 28 rats entered the final results analysis. ① α-macula adherens protein was found to be expressed in myocardium in atrium, ventricle, papilla muscles and interventricular septum. ② In infant rats, the expression of α-macula adherens protein was mainly observed in intercalated disc at the end of myocardium, with less expression on cell surface and in cytoplasm; in contrast, α-macula adherens protein in young, middleaged and old rats was found to be typically expressed in intercalated disc at the end of myocardium. CONCLUSION: The expression of α-macula adherens protein displays age-dependant manner during rat myocardial development; moreover, the expression of α-macula adherens protein is found to be consistent with the development of intercalated disc.

7.Effect of protein tyrosine phosphatase non-receptor type 12 (PTPN12) downregulation on the radiosensitivity of non-small cell lung cancer

Objective: To identify the relationship between the expression of protein tyrosine phosphatase non-receptor type 12 (PTPN12) and radiotherapy effect in non-small cell lung cancer (NSCLC) tissues and to determine whether PTPN12 deficiency can sensi-tize lung cancer cells to irradiation. Methods: From September 2013 to October 2014, 92 NSCLC patients undergoing radiotherapy with or without platinum-based combination chemotherapy were analyzed retrospectively. Before the treatment, PTPN12 expression was detected through immunohistochemistry. After the completion of radiotherapy, the patients' responses were assessed and radio-therapeutic efficacy analyzed. The human NSCLC cell line H1299 was infected with shPTPN12 knockdown, and colony survival assay was analyzed after irradiation. Chi-square test was used to examine the correlation between PTPN12 expression and clinicopathologi-cal characteristics. Univariate analyses and Logistic regression test were used to analyze the relationship between clinicopathological characteristics and radiotherapeutic response. Results: Patients with low PTPN12 expression were more sensitive to radiotherapy than those with high PTPN12 expression (80.0%vs. 57.1%, P=0.018). Multivariate analysis showed that PTPN12 expression was the on-ly independent predictor of radiotherapeutic response in NSCLC. The H1299-shPTPN12-knockdown cells were sensitive to irradiation. Conclusions:The results of the study indicated that downregulation of PTPN12 improved the radiosensitivity of NSCLC cells.
8.Effect of warming yang and benefiting marrow recipe on the expression of collagen type II and proteoglycans in the articular cartilage

BACKGROUND:Col agen type II and proteoglycan loss are two most obvious manifestations of cartilage damage in the onset of osteoarthritis. Changes in col agen type II and proteoglycan as the main components of cartilage matrix directly lead to cartilage degeneration and subsequently result in osteoarthritis. How to reverse or prevent the development of this process becomes the focus of medical research. OBJECTIVE:To observe the effect of warming yang and benefiting marrow recipe on the expression of col agen type II and proteoglycan in the articular cartilage of knee osteoarthritis rabbits as wel as to further explore the mechanism underlying chondrocyte protection. METHODS:Ninety-six New Zealand rabbits, aged 9 months old, male and female, were selected to prepare osteoarthritis models in extension position using cast immobilization method, and were randomly divided into four groups:blank group (untreated), model group (simple modeling), Chinese medicine group (intragastric administration of extracts of warming yang and benefiting marrow recipe, 24 mL/kg/d) and western medicine group (intragastric administration of glucosamine hydrochloride, 24 mL/kg/d). Intragastric administration was done once a day for 6 weeks. RT-PCR technology was used to observe the effect of warming yang and benefiting marrow recipe on the expression of col agen type II and proteoglycans in the articular cartilage, and pathological examination was also done. RESULTS AND CONCLUSION:The cartilage surface was smooth in the blank group and Chinese medicine group, with uniform toluidine blue staining, but in the model group and western medicine group, the cartilage surface was rough and the toluidine blue staining was extremely uneven with obvious loss of surface and middle layer dying. The expressions of cartilage proteoglycan and col agen type II in the model group were significantly lower than those in the blank group (P<0.01) as wel as in the Chinese medicine group and western medicine group (P<0.05). In addition, the expressions of cartilage proteoglycan and col agen type II in the Chinese medicine group were higher than those in the western medicine group (P<0.05). These findings indicate that the recipe of warming yang and benefiting marrow can enhance the expressions of col agen type II and proteoglycan, which can maintain the normal col agen phenotype and protect the articular cartilage.
9.Influence of difference ileocecal junction length on the efficacy of laparoscopic subtotal colectomy antiperistaltic cecorectal anastomosis in treatment of slow transit constipation

Objective To investigate the influence of difference ileocecal junction length on the efficacy of laparoscopic subtotal colectomy combined with antiperistaltic cecorectal anastomosis (LSCACRA) in treatment of slow transit constipation (STC). Methods Ninety- two STC patients undergoing LSCACRA were divided into 2 groups by random digits table method:10-15 cm group (10-15 cm ascending colon preserved above ileocecal junction, 46 cases) and 2-3 cm group (2-3 cm ascending colon preserved above ileocecal junction ,46 cases). All the patients were followed up for 24 months, and the Wexner incontinence score (WIS), ileocecal junction emptying time of barium enema, Wexner constipation score (WCS), abdominal pain intensity score (NRS), gastrointestinal quality of life index (GIQLI), abdominal pain frequency score and abdominal bloating frequency score were compared between 2 groups. Results All the patients successfully completed LSCACRA, with no conversion to open surgery and death occurred. There were no statistical differences in amount of bleeding, operative time, hospitalization time, exhaust time, incidences of postoperative intestinal obstruction and pulmonary infection, defecation time, WIS and abdominal bloating frequency score between 2 groups (P>0.05). The WCS, GIQLI, NRS and abdominal pain frequency scores 6, 12 and 24 months after operation in 2-3 cm group were significantly better than those in 10 - 15 cm group, WCS: (1.7 ± 1.3) scores vs. (4.2 ± 2.3) scores, (1.7 ± 1.1) scores vs. (4.1 ± 1.9) scores, (1.2 ± 0.5) scores vs. (3.9 ± 2.5) scores;GIQLI:(116.8 ± 6.2) scores vs. (98.5 ± 14.7) scores, (122.9 ± 7.5) scores vs. (104.7 ± 16.5) scores, (124.3 ± 5.9) scores vs. (108.3 ± 15.1) scores; NRS: (1.3 ± 0.5) scores vs. (2.5 ± 1.7) scores, (0.9 ± 0.3) scores vs. (2.3 ± 1.4) scores, (0.8 ± 0.3) scores vs. (2.2 ± 1.5) scores;abdominal pain frequency score:(0.9 ± 0.3) scores vs. (1.6 ± 1.2) scores, (0.7 ± 0.3) scores vs. (1.4 ± 1.1) scores, (0.7 ± 0.2) scores vs. (1.2 ± 1.0) scores, and there were statistical differences (P<0.05). The ileocecal junction emptying time of barium enema 24 months after operation in 2-3 cm group was significantly shorter than that in 10-15 cm group: (17.6 ± 8.4) h vs. (21.3 ± 10.8) h, and there was statistical difference (P<0.05). Conclusions STC patients with LSCACRA is safe and effective. The shorter length of ascending colon preserved above ileocecal junction can improve the efficacy of LSCACRA in the treatment of STC and the prognosis of patients. The 2-3 cm length of ascending colon preserved above the ileocecal junction should be recommended.
10.Optimal animal model of osteoarthritis

BACKGROUND:There are a variety of methods for establishing osteoarthritis animal model, and different methods and different animals have their own characteristics.
OBJECTIVE:To review the status of the research on osteoarthritis animal models.
METHODS:The relevant articles to the osteoarthritis animal models were searched in Medline database from January 2002 to October 2011, with the key words of“animal model, osteoarthritis”in English by the computer. Similarly, Chinese Journal Ful-text Database was retrieved for related articles published in Chinese from January 2002 to October 2011, with the key words of“animal model, osteoarthritis, research development”in Chinese. Total y 423 relevant articles were col ected and 128 of them conformed the standard. At last 40 articles were included to review after ful-text reading.
RESULTS AND CONCLUSION:The artificial induced animal models are the principal means to establish animal models of osteoarthritis and they have been widely used in the study of osteoarthritis pathogenesis, drug efficacy, cartilage physiology and pathology. Spontaneous osteoarthritis animal models have great advantages in the study addressing the initial mechanism of osteoarthritis, the biochemical changes of articular cartilage and the comparison of prevention and cure effects. Transgenic animal models have great application prospects on studying cartilage repairing mechanisms and testing a gene to prevent, delay or reversal the morphological changes. Accordingly the choice of models should be based on research needs.
