1.Expressions of the common β subunit and specific α subunit of IL-5, IL-3, and GM-CSF receptor mRNA in eosinophils in BALF from guinea pigs with asthma

Objective To investigate the relationship of the expressions of common β (βcR) and specific α chains of IL-5, IL-3 and GM-CSF receptors with the eosinophils(Eos) apoptosis for the roles of these receptors in asthma. Methods All of 12 guinea pigs were equally randomized into normal and ovalbumin sensitized asthmatic group. The apoptosis of hypodense and normodense Eos in bronchoalveolar lavage fluid (BALF) was detected by TUNEL method. The expression of common β and specific α chains of the 3 receptor mRNA were measured with RT-PCR and in situ hybridization. Results More Eos were observed in asthmatic group than in normal group, especially those in hypodense (P<0.01). The apoptotic rate of Eos in BALF were significantly lower in asthmatic group than in normal animals (P<0.01), but no difference was found between hypodense and normodense Eos. Compared with normal group, the expressions of the 3 receptor mRNA α chains were decreased in asthmatic group than in normal group (P<0.01), but βcR expression was increased significantly. More IL-3 Rα and GM-CSFRα mRNA were expressed in hypodense Eos than in normodense Eos. The contents of IL-5Rα and IL-3Rα mRNA were lower in asthmatic group than in normal group. Conclusion The apoptosis of Eos is inhibited in BALF from guinea pigs with asthma. The expressions of IL-5, IL-3 and GM-CSF receptors in Eos are regulated through 2 mechanisms: the reduced expressions of their specific α chains which attenuating the negative regulation on Eos activation and inhibited apoptosis and the increased expressions of their βcR which enhancing the positive regulation on the 2 aspects. These data suggest that IL-5, IL-3 and GM-CSF receptors might be involved in the pathogenesis of asthma through regulating the apoptosis of Eos.

Objective To explore the inhibitory effects of SB203580, a specific inhibitor of p38 mitogen activated protein kinase (p38 MAPK) on hyper secretion of airway mucus in mice with asthma. Methods Forty BABL/C mice were randomly allocated into asthma group, SB203580 treatment group, dexamethasone treatment group and control group (sham-challenged group). Mice in SB203580 treatment group were intraperitoneally injected with SB203580 (10mg/kg), while those in dexamethasone treatment group were injected with dexamethasone (1mg/kg) before provocation. The number of goblet cells in small bronchi of all groups was determined after Alcian blue-periodic acid Schiff (AB-PAS) staining. The contents of MUC5AC in bronchoalveolar lavarge fluid (BALF) were evaluated with ELISA asssy. The levels of MUC5AC mRNA expression were determined with reverse transcription-polymerase chain reaction (RT-PCR) assay, and gray-scale analysis was done with image analysis method. AIM To investigate the effects of aminophylline (AM) on expression of Fas mRNA and apoptosis in eosinophils(Eos) of bronchalveolar lavage fluid(BALF) in asthmatic guinea pig, and the mechanism of AM promoting Eos apoptosis. METHODS After guinea pigs were stimulated by ovalbumin(OVA) 48 h, hypodense Eos(HEos) and normodense Eos(NEos) were purified from BALF by gradients of percoll. Purified Eos were incubated with different concentration of AM (10 -6 ~10 -3 mol?L -1 ) in RPMI 1640 medium 24 h, the mRNA of Fas in Eos was measured by hybridization and apoptosis was detected by TdT mediated dUTP nick end labeling. RESULT Eos cultured under the condition of AM administration in vitro, apoptotic Eos increased and the expression of Fas mRNA increased in a dose dependent manner. The expression of Fas mRNA was correlated with Eos apoptosis. CONCLUSION AM promote Eos apoptosis and expression of Fas in dose dependent manner in vitro. Fas might be one of mechanism that AM promoting apoptosis of Eos.

6.Effects of KMT5A on cell migration and invasion in human hepatocellular carcinoma and its mechanisms

Objective To investigate the clinical roles of lysine methyltransferase 5A(KMT5A)in human hepatocellular carcinoma(HCC)and its functions in cell migration and invasion. Methods The expression levels of KMT5A of 60 cases were detected by immunohistochemistry(IHC). KMT5A siRNA was used to down-regulate the expression of KMT5A in SMMC-7721 cells. Cell migration and invasion were measured by wound healing assays and transwell assays,respectively. Immunoblotting was used to detect the expression of MMP-2 after siRNA trans-fection. miR-186 mimics were transfected into SMMC-7721 cells and the mRNA levels of KMT5A was detected by qRT-PCR after transfection. Results High expression of KMT5A was associated with large tumor diameter (>5 cm,P=0.047)and advanced TNM stage(Ⅲ+Ⅳ,P=0.035). The expression of KMT5A was knocked down by siRNA in SMMC-7721 cells. Down-regulation of KMT5A suppressed cell migration(P=0.031,P=0.006)and invasion(P=0.010),and impaired MMP-2 expression(P=0.040). Overexpression of miR-186 could significantly inhibit the expression of KMT5A(P = 0.007). Conclusions Over-expression of KMT5A in HCC tissues associ-ates with poor clinical features. KMT5A knockdown inhibits the migration and invasion on HCC cells.

AIM: To investigate the effects of dexamethasone (DM) on expression of fas and bcl-2 mRNA and apoptosis in eosinophils (Eos) of bronchoalveolar lavage fluid (BALF) in asthmatic guinea pig. METHODS: The guineas pigs were stimulated with ovalbumin (OVA) for 48 hours. Hypodense Eos (HEos) and normodense Eos (NEos) were purified from BALF by gradients of Percoll. Eos was cultured in PRMI-1640 medium and DM (10 -10-10 -5 mol/L) was added. The mRNA of fas and bcl-2 in Eos was measured by hybridization. Apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL). RESULTS: Eos was cultured under the condition of DM administration in vitro. Apoptotic Eos increased, fas mRNA expression increased and the bcl-2 mRNA expression decreased in a dose-dependent manner. There was significant correlation between Eos apoptosis and fas mRNA expression. The expression of bcl-2 mRNA was inversely correlated with Eos apoptosis. CONCLUSION: DM promoted Eos apoptosis, increased expression of fas and inhibited expression of bcl-2 in a dose-dependent manner in vitro. fas and bcl-2 might be one of mechanisms of DM promoting apoptosis in Eos.
9.Follow-up study on pedicle fixation for treatment of thoracolumbar burst fractures in patients with osteoporosis

Objective To explore the effect of pedicle fixation for treatment of thoracolumbar burst fractures in patients with osteoporosis,and to provide more evidence for the treatment.Methods Retrospectively analyzed the clinical data of 121 patients with osteoporotic vertebral burst fracture from June 2012 to October 2015.And these patients were divided into two groups according to different operation methods, namely the control group (n=56) who were given short segment fixation and the observation group (n=65) who were given single segment fixation.The visual analogue scale(VAS),Oswestry disability index(ODI),vertebral height,kyphotic angle and bone mineral density of the two groups were analyzed before surgery and 3 days,1 month,3 months and 12 months after surgery.Results The VAS score,ODI score,vertebral height,and Cobb angle of the injured vertebra were significantly improved in both of the two groups,and the difference was statistically significant (P<0.05).The VAS score of the observation group was better than that of the control group 3 days after surgery with statistically significant difference (P<0.05).But there was no significant difference 3 months,6 months and 12 months after surgery(P>0.05).The ODI score of the observation group was better than that of the control group 3 days and 3 months after surgery with statistically significant difference (P<0.05),and there was no significant difference between the two groups till the end of follow-up.Pedicle fixation at the injured vertebra significantly improved the vertebral height and Cobb angle with statistically significant difference (P<0.05).And the anti-osteoporosis treatment significantly increased the bone mineral density (P<0.05).Conclusion Pedicle fixation at the injured vertebra is useful in pain relief as well as function and anatomical structure restoring.And anti-osteoporosis treatment is necessary for the bone mineral density increase.
10.Effect of asymptomatic hyperuricemia on the postoperative functional recovery in patients with knee joint re-placement

Objective To evaluate the effect of asymptomatic hyperuricemia on the postoperative functional recovery in osteoarthritis (OA)patients with knee joint replacement.Methods Among the 294 patients who recieved knee joint replacement in our hospital from Feb-ruary 2013 to September 2015,a total of 187 patients were included in this study with 79 cases in the hyperuricemia group and the other 108 cases in the control group.The WOMAC index of the two groups were analyzed before the surgery,2 weeks,3 months,and 6 months after the knee joint replacement surgery.Multivariable linear regression was performed to test the effect of impact factors on the variation of WOMAC index.Results The WOMAC index of the two groups significantly increased 2 weeks after surgery,and the difference was statistically signifi-cant(P <0.05).And the WOMAC index of the control group was higher than that in the hyperuricemia group 2 weeks and 3 months after sur-gery with statistically significant difference(P <0.05).Conclusion The uric acid level before surgery was a influence factor of WOMAC index, and high uric acid level before knee joint replacement has negatively impact for the postoperative functional recovery in the osteoarthritis patients.
