Objective To investigate the roles of phosphatase and tensin homolog deleted on chromosome 10(PTEN),insulin-like growth factor 1(IGF-1),and insulin-like growth factor receptor 1 (IGF-1R) in the tumorigenesis,development,invasion and metastasis,and prognostic value of gastric cancer.Methods Immunohistochemical assay was used to detect the expressions of PTEN,IGF-1,and IGF-1R in gastric cancer tissue samples from 74 gastric cancer patients and in normal gastric mucosa of 20 health individuals.All data of these patients were collected including the grading and differentiation of the tumor.Results In gastric cancer,expression of PTEN was decreased,and expressions of IGF-1,and IGF-1R were enhanced.The expressions of PTEN,IGF-1 and IGF-1R had correlation with the outcome and biological behavior of gastric cancer.PTEN expression was higher in no lymph node metastasis group than lymph node metastasis group,higher in stage Ⅰ–Ⅱgroup than stage Ⅲ–Ⅳgroup.IGF-1 and IGF-1R expressions were higher in progressive group than early gastric cancer group,higher in poor differentiation group than moderate and well differentiation group,higher in lymph node metastasis group than no lymph node metastasis group,higher in stage Ⅰ–Ⅱgroup than stage Ⅲ–Ⅳgroup.PTEN-negatively-expressed patients had poor prognosis compared with positively-expressed ones;so did those who expressed IGF-1 and IGF-1R.Conclusion The changes of PTEN,IGF and IGF-1R play roles in gastric cancer genesis,progression and prognostic evaluation.

Aim To express hepatitis C virus glycoprotein E (gE) deleting carboxy-terminal 31 amino acids, and detect anti-E antibody in HCV patients using expressed gE. Methods E gene derived from HCV H strain was inserted into baculovirus transfer vector containing a polyhedrin promotor. The recombinant plasmid was cotransfected into insect cell sf9 with a viral expression vector. The expression of gE was analyzed with Western blot, and the cells were used for dectecting antibodies against E1 and E2 in 35 hepatitis C patients by indirect immunofluorescence. Results A series of proteins with different relative molecular masses(Mr) could be detected by Western blot. Results from indirect immunofluorescence staining showed and only 4 patients were anti-E antibody positive gE was located in cytoplasm. Conclusion HCV gE is expressed successfully in insect cells, the study lay the foundation for further developing HCV vaccine.

OBJECTIVETo study the effect of solid phase transformation on the metal-ceramic compatibility of Co-Cr alloy during firing programs.

METHODS9 foils of Co-Cr and Ni-Cr alloy with the dimension of 25 mmx3 mmx0.5 mm were casted using lost wax technology respectively. Among them, 6 specimens were subjected to metal-ceramic bonding strength test (three point bending method), the ceramic layer of 3 specimens were removed for X-ray diffraction (XRD) analysis. One cylindrical specimen of each alloy was casted for the test of coefficient of thermal expansion, cooling curves were recorded.

RESULTSThe metal-ceramic bonding strength of Ni-Cr, Co-Cr alloy was (49.1 +/- 3.40), (40.9 +/- 2.02) MPa respectively. There was significant difference between the two groups' bonding strength (P = 0.00). The coefficient of thermal expansion in the 20-500 degrees C interval of Ni-Cr and Co-Cr alloy was 13.9 x 10(-6), 13.8 x 10(-6) x K(-1) respectively. XRD analysis indicated that the microstructure of Ni-Cr alloy was austenite. While Co-Cr alloy was constituted by fcc phase, hcp phase and sigma phase.

CONCLUSIONDuring the cooling procedure, the transformation of fcc phase to hcp phase and segregation of needle-like sigma phase intensify the linear contraction speed of Co-Cr alloy, which decreases the metal-ceramic adaptability.

Objective To explore the differences between schizophrenic patients and normal at prospective memory,and investigate the effect of personality intermediary between schizophrenia and prospective memory.Methods A case-control study was utilized in this research.It used the color matching task to test prospective memory and MMPI-2 scale to test personality differences between the two groups.Results Schizophrenia prospective memory score(19.29±2.30),lower than the normal group(44.74±2.06),the difference was significant(P＜0.01).In mediating effect on the personality inspection,the subscales within MMPI-2 of Social Scale(Si) and Anxiety and tension Scale(ANX) regression coefficients were tested(P＜0.05),which showed significant effect.Further analysis with sobel statistics showed that only one factor tested was significant.The test coefficients of depression scale (D),mental weakness scale (Pt) and schizophrenia scale (Sc) were significantly different (P＜ 0.05),which showed that these subscales also had significant mediating effect.Conclusion The prospective memory performance is low in schizophrenia.The personality characteristics that depression,mental weakness,schizo phrenia,social anxiety and tension play an intermediary role in schizophrenia and prospective memory.

Objective To investigate the clinical application value of laparoscopy-assisted subtotal colectomy with antiperistalsis cecorectal anastomosis for slow transit constipation (STC).Methods From September 2007 to October 2010,a total of 31 patients with STC underwent laparoscopic-assisted subtotal colectomy with antiperistalsis cecorectal anastomosis.A follow-up survey was completed at 3 and 12 months after the operation.Results No death or conversions to open operation occurred.The mean operation time,mean intraoperative blood loss and mean post-operative hospitalization were 260 min (180-310 min),60 ml (30-120 ml) and 8d (6-11 d),respectively.No postoperative infection,anastomotic stoma,adhesive intestinal obstruction or other perioperative complications occurred.In 3-month follow-up,constipation was significantly alleviated in 23 patients,mild diarrhea occurred in 5,diarrhea in 2 and mild recurrence in 1.Constipation was significantly alleviated in 25 patients,mild diarrhea occurred in 5 and mild recurrence in 1 at 12-month follow-up.Conclusion Laparoscopy-assisted subtotal colectomy with antiperistalsis cecorectal anastomosis is safe,effective and less invasive for STC.

Background and purpose:It was reported that zinc ifnger protein 139 (ZNF139) was expressed aberrantly in gastric cancer. But the relationship between ZNF139 and multidrug resistance (MDR) of gastric cancer is still not clear. The purpose of this research was to investigate the expressions and signiifcance of ZNF139, MRP-1, MDR1/P-gp, GST-π in human gastric carcinoma cell lines SGC7901 and SGC7901/ADR. Methods: The expressions of ZNF139, MRP-1, MDR1/P-gp, GST-πwere determined with RT-PCR and Western blot in SGC7901 and SGC7901/ADR cell lines. Then siRNA recombinant plasmid of targeting ZNF139 gene was constructed and imported into gastric cancer cell line SGC7901/ADR, and the expressions of MRP-1, MDR1/P-gp, GST-πwere tested simultaneously. Results:The expressions of ZNF139, MRP-1, MDR1/P-gp, GST-πwere higher in SGC7901/ADR than in SGC7901(P<0.05). ZNF139 was inhibited obviously after siRNA-ZNF139 was transfected into SGC7901/ADR, and expression of MRP-1, MDR1/P-gp, GST-πdecreased(P<0.05). Conclusion:ZNF139 may be invovled in multidrug resistance (MDR) of gastric cancer by up-regulating MRP-1, MDR1/P-gp and GST-π.

Objective To evaluate the influence of orthokeratology lenses on vision,intraocular pressure and biometric measurement parameters in adolescent myopia.Methods A total of 72 adolescent myopia patients (136 eyes) with orthokeratology lens,aged 8-16 years old,in Hebei Provincial Eye Hospital from Junuary 2013 to December 2015 were randomly selected.The vision,intraocular pressure,axial length,corneal topography,corneal thickness and anterior chamber depth were observed before wearing glasses,at 1 week,1,3,6 months after wearing glasses.Results After orthokeratology wearing,the uncorrected visual acuity was obviously improved (P＜0.05),the average refraction diopter was declined (P＞0.05) and the non-contact intraocular pressure was decreased (P＜0.05).The axial length after orthokeratology wearing had little change (P＞0.05).The curvature of the patient's cornea at one week after wearing glasses was deceased (P＜0.05) and the posterior corneal curvature tended towards stability.The corneal thickness at 1 week after wearing glasses had no obvious change compared with before wearing glasses(P＞0.05),which at 1 month after wearing glasses was decreased(P＜0.05) and which at 3,6 months after wearing glasses trended to be stabilized.The anterior chamber depth after wearing glasses had no obvious change.Conclusion Orthkeratology lens can decrease the myopia degree,increases the uncorrected visual acuity and has obvious effect for controlling adolescent myopia.The intraocular pressure and biometric measurement parameters have corresponding change after wearing orthkeratology lens.

OBJECTIVETo study the electrochemical corrosion behavior of Co-Cr, Ni-Cr and Ni-Cr-Be based porcelain alloys in NaCl solution.

METHODSFive samples of each alloy were made respectively, electric polarization curve of each alloy was obtained using potentiodynamic polarization technique. Self-corrosion potential (E(corr)), self-corrosion current density (I(corr), passive region and transpassivation potential were tested. Microstructure and constituent was examined using scanning electron microscopy and energy dispersive spectroscopy.

RESULTSCo-Cr alloy possessed the most desirable corrosion resistance because of its integrated, homogeneous and compact passive film. The poor compactness of Ni-Cr alloy's passive film decreased its corrosion resistance. Ni-Cr-Be alloy exhibited the worst corrosion resistance due to the Cr and Mo depleted Ni-Be eutectic phases in the alloy.

CONCLUSIONTaking biological security into consideration, it is necessary to avoid the application of porcelain alloys with Be element. Co-Cr alloy with better biocompatibility possesses much broader prospect in the field of dental restoration.

Objective To study the effects of inhibing AKT2 by siRNA on SMMC7721 liver cancer cells proliferation,apoptosis,migration and invasion.Methods The siRNA targeting AKT2 was designedandthe SMMC7721AKT2- siRNAplasmidwasconstructed andtransfected into SMMC7721 cells.The stable cell lines were screened by G418.The effects of AKT2 by siRNA on SMMC7721 liver cancer cells,growth inhibition was evaluated by MTT assay.Cell cycle was analyzed by flowcytometry (FCM).Protein of P27 and CyclinD1 was evaluated by Western-blot.The ability of migration and invasion was evaluated by wound healing and Transwell assay.ResultsThe growth of SMMC7721 cells was significantly inhibited by siRNA (P＜0.05).Flow cytometry display that AKT2 by siRNA can induce G1 phase arrest,the ratio of G1 phase increased homologously and S phase declined homologously.The protein of CyclinD1 was declined and the protein of P27 was increased by Western-blot.Wound healing and Transwell assay show that the ability of cells,migration and invasion was inhibited by AKT2 by siRNA.Conclusion AKT2 by siRNA can significantly inhibit the growth of SMMC7721 cells,arrest cell cycle.AKT2 by siRNA can inhibit the ability of invasion and migration of SMMC7721 cells.
