1.Long-term effect of ketamine on learning and memory function of postnatal rats

Objective To investigate the effect of ketamine on learning and memory function of adult rats after injection of ketamine at early development stage. Methods Twelve rats born in a week were randomly divided into ketamine anesthesia group(Ket group)and normal saline control group(Ns group).Rats in Ket group were intraperitoneally injected with 50 mg/kg ketamine,and those in Ns group were administrated with same amount of normal saline.After eight weeks,Morris water maze test was adopted to explore the latency in place navigation and spatial probe ability.The sections of hippocampus were obtained,and immunohistochemical staining was employed to detect the expression of Bax and Bcl-2.Results In Morris water maze test,the latency in place navigation of Ket group Was significantly longer than that of Ns group(P<0.05),and the spatial probe ability significantly decreased(P<0.05).It Was revealed by immunohistochemical staining that the expression of Bax in Ket group was significantly higher than that in Ns group,while there was no significant difference in the expression of Bcl-2 between these two groups. Conclusion Application of ketamine in neonatal rats may result in learning and memory impairment in adulthood,and the underlying mechanism may correlate to the apoptosis of neuron in hippocampus region.
2.RANTES, MCP-1, and SDF-1β levels in healthy people and AIDS patients: a comparative analysis

Objective To evaluate the differences of serum RANTES(regulated on activation, normal T cell expressed and secreted), MCP-1 (monocyte chemoattractant protein), and SDF-1β (stromal cell-derived factor-1β) in patients with acquired immunodeficiency syndrome (AIDS) and healthy people.Methods 38 AIDS patients who were admitted to a hospital between January 2010 and January 2015 were as AIDS groups, 38 healthy persons were as a healthy group, serum levels of RANTES, MCP-1, and SDF-1β in two groups were detected, and the subgroup analysis was carried out according to the viral load.Results Serum levels of RANTES, MCP-1, and SDF-1β in AIDS group were (1 392.55±227.69)pg/mL,(450.91±103.04)pg/mL, and(104.82±22.52)pg/mL respectively,all were significantly higher than those in healthy group([120.58±55.87] pg/mL, [74.25±33.62] pg/mL, and [39.04±11.43]pg/mL respectively)(all P<0.05).Among AIDS patients with HIV viral load 4≤Log(VL)<5 and Log(VL)≥5, serum RANTES were (1 470.34±155.01)pg/mL and (1 408.29±181.54)pg/mL respectively,which were both significantly higher than patients with HIV viral load Log(VL)<4([1 183.12±174.54]pg/mL);serum MCP-1 and SDF-1β levels in AIDS patients with HIV viral load 4≤Log (VL)<5 were (537.93±89.32)and(149.31±18.05)pg/mL respectively,which were significantly higher than patients with HIV viral load Log(VL)≥5([410.26±80.57] pg/mL, [81.53±20.31]pg/mL respectively) and HIV viral load Log(VL)<4([381.71±77.26] pg/mL, [72.90±21.62]pg/mL respectively), differences were both statistically significant(both P<0.05).Conclusion Serum levels of RANTES, MCP-1, and SDF-1β are significantly increased in AIDS patients, which are related to the level of viral load.
3.A randomized controlled prospective study on ambulation versus bed rest for the initial treatment of patients with acute deep venous thrombosis

Objective To evaluate the benefits and the risk of pulmonary embolism between walking exercises and bed rest in the acute stage of deep venous thrombosis (DVT). Methods From February 2008 to March 2009,40 acute primary DVT patients were arbitrarily randomized into controlled group and experimental group,with 20 cases in each group,who were hospitalized within seven days of onset.Patients in controlled group underwent bed rest for 7-10 days,wearing elastic stockings.While patients in experimental group were early (1-2 days after admission) off bed walking about 600-1200 m every day with elastic stockings for 7-10 days.During the process patients in both group received regular anticoagulation treatment,and continued warfarin anticoagulation therapy after out of hospital for at least 6 months.The clinical characteristics of the two groups were comparable.Primary end points were the reduction of pain assessed daily with the Visual Analogue Scale (VAS),the reduction of leg circumference at thigh,and cumulative pulmonary embolism incidences of two groups in three months. Results Symptoms in all patients of the two groups were improved,but the pain level showed a statistically significant reduction starting after the third day(58 ±8) mm in the experiment group and after the fifth day (58 ± 13) mm in the controlled group.The leg circumferences of the two groups after 7 days were statistically significantly different (P < 0.05),the cumulative pulmonary embolism incidences of the two groups for 3 months were not statistically significantly different. Conclusion Mobile patients with acute DVT should be encouraged to walk with medical compression stockings.The rate of resolution of pain and swelling is significantly faster when the patient ambulates with compression.The quality of life significantly improved.The risk of pulmonary embolism does not significantly increase by this approach.
4.The prognostic value of CRP,PCT and IL-6 in liver cirrhosis with systemic inflammatory response syndrome

Objective To detect the serum levels of C-reactive protein (CRP),procalcitonin (PCT),and interleukin-6(IL-6) and evaluate their prognostic values in liver cirrhosis with systemic inflammatory response syndrome(SIRS).Methods A total of 56 patients with liver cirrhosis and systemic inflammatory response syndrome were divided into two groups,improved group(n =39)and death group(n=1 7).Serum CRP、PCT、IL-6 levels were measured at the first day,third day and seventh day after SIRS. Results CRP,PCT and IL-6 levels of two groups were no statistically significant difference in the first day (P >0.05).PCT and IL-6 levels of two groups were no statistically significant difference in the third day (P >0.05).CRP levels in improved group were lower than those in death group in the third day (P <0.05).CRP,PCT and IL-6 levels in improved group were lower than those in death group in the seventh day (P <0.05).There was no statistically significant difference of CRP,PCT and IL-6 at the first day, third day and seventh day in death group (P >0.05).CRP levels at the third day and seventh day were lower than those at the first day in improved group (P <0.05).PCT and IL-6 levels at the seventh day were lower than those at the first day in improved group (P <0.05).Conclusion Poor prognosis was showed at sustained high levels of CRP,PCT and IL-6.Compared with PCT and IL-6, CRP might be more rapid in showing condition improved.
5.The Role of IκBα in TNF-α-induced Apoptosis in Hepatic Stellate Cell Line HSC-T6

To investigate the role of NF-κB in TNF-α induced apoptosis in HSC-T6, a mutant IκBα was transfected into HSC-T6 cells by lipofectin transfection technique and its transient effect was examined 48 h after the transfection. The activation of NF-κB was detected by immune fluorescence cytochemistry and Western blotting with anti-p65 antibody. The apoptosis and the rate of inhibition by TNF-α in both transfected and untransfected HSC-T6 cells were measured respectively by FAC-Scan side scatter analysis and MTT methods. Our results showed that TNF-α could activate NF-κB in untransfected cells but not in transfected HSC-T6 cells. The percentage of apoptosis in transfected cells were significantly higher than that in the untransfected ones (P<0.01) and it was also true of the inhibition rate (P<0.01). It is concluded that the resistance of HSC-T6 towards apoptosis induced by TNF-α can be mediated by NF-κB activation. The inhibition of NF-κB activation by mutant Iκ Bα can attenuate the resistance of HSC-T6 cells and increase its sensitivity to TNF-α.
6.Measles in pediatric liver transplant recipients: A report of 4 cases

Objective To summarize and explore the clinical characteristics,treatment and outcome of measles in pediatric liver transplant recipients.Method A total of 56 recipients underwent pediatric liver transplantation from June 2013 to April 2014,and 4 recipients suffered from measles post-liver transplantation with the incidence being 7.1%.The data of 4 recipients were retrospectively analyzed.All of the 4 cases were subjected to liver transplantation at our center for biliary atresia and cholestasis liver cirrhosis.All the patients received the combined immunosuppressive regimen on the basis of tacrolimus.All the patients did not accept the immunization with measles vaccine because of the primary liver diseases.All the cases presented the clinical manifestations of cough,high fever (T >38.5℃C) and conjunctiva congestion before the rashes came out.Maculopapular rashes appeared about 4 days after the fever.The rashes first appeared from the skine post aurem,face,neck,and then spread to trunk and extremities.Koplik's spots on the buccal mucosa were observed in all the 4 cases.The serological test of IgM antibodies to measles was done.The treatment was adjusted as soon as the diagnosis of measles was clear.Immunosuppressants were decreased or stopped,intravenous immunoglobulin (IVIG) combined with anti-infection drugs was given.Result The clinical manifestation of measles in pediatric liver transplant recipients was serious.There were 3 cases complicated with pneumonia,and one case with laryngitis.Two cases presented severe measles pneumonia and developed into severe respiratory failure requiring mechanical ventilation.Three eases recovered from the therapy and 1 case died of respiratory failure.Conclusion Pediatric liver transplant recipients who suffered from measles are at high risk of severe pneumonia.Measles pneumonitis is frequently fatal in immunocompromised pediatric patients.The treatment shoule be given as soon as possible.
7.Study on the Toxicity of Jianpi Shengxue Granule on Perinatal Rats

OBJECTIVE:To investigate the toxicity effect of Jianpi shengxue granule on perinatal rats. METHODS:Based on body mass,pregnant rats were randomly divided into negative control group and Jianpi shengxue granule low-dose,medium-dose, high-dose groups(0.77,2.31,6.93 g/kg),21 in each group,and intragastrically given relevant medicines from the 15th d of preg-nancy until 21st d after delivery,once a day. Effects of Jianpi shengxue granule on general toxicity and fertility of maternal rats were observed,as well as the effects on athletic ability,learning and memory ability,fertility of F1 offspring and early viability of F2 offspring. RESULTS:In terms of toxicity effect on maternal rats,compared with control group,the days of pregnancy in Jianpi shengxue granule high-dose group was significantly prolonged(P<0.05),initial body mass of administration was significantly re-duced (P<0.01). In terms of toxicity effect on offspring,compared with negative control group,the initial body mass of F1 off-spring in Jianpi shengxue granule high-dose group was significantly reduced(P<0.05),the time of 4th and 5th time for founding underwater platform was significantly shortened(P<0.01),and there was no obvious effect on fertility of F1 offspring and early vi-ability of F2 offspring (P>0.05);there was no obvious effect on the indexes of offspring in Jianpi shengxue granule medium-dose,low-dose groups. CONCLUSIONS:The no toxic dose of Jianpi shengxue granule in maternal and offspring rats is 2.31 g/kg.

9.Effects of Electro-acupuncture on Insulin Resistance and Hypothalamic Agouti Gene-related Protein and Neuropeptide Y in Obesity Rats

Objective To observe the effects of electro-acupuncture (EA) on body weight, insulin resistance, hypothalamic agouti gene-related protein (AGRP) and neuropeptide Y (NPY) in diet induced obesity (DIO) rats; To discuss its mechanism for DIO.Methods Fifty SD male rats were randomly divided into low fat diet groups (LFD) and high fat diet group. After the DIO models were established, the successful model rats were randomly divided into model group, EA group and Orlistat (OLST) group. EA was applied to “Zusanli” (ST36) and “Quchi” (LI11) for 20 minutes. The treatment was done once a day for 28 days. OLST was treated with orlistat by gavage. LFD and model did not receive treatment. Body weight was recorded every day. FPG and FINS were detected. The expressions of AGRP and NPY were detected by RT-QPCR. Morphological changes of adipocyte and liver were examined by HE staining.Results The body weight, HOMA-IR, AGRP, NPY and diameter of adipocyte of EA group and OLST group were significantly lower than model group (P<0.05), difference between EA group and OLST group. The states of hepatic steatosis were improved in EA group and OLST group.Conclusion EA has the effects on weight loss by inhibiting the expressions of AGRP and NPY by improving IR.
10.Effects of 'Living High-Training Low' on the Proliferative Capacity of Endothelial Progenitor Cells from Peripheral Blood of Rats

Objective We investigated if the proliferative capacity of endothelial progenitor cells was affected by hypoxia and exercise. Methods Twenty four male Sprague-Dawley rats were randomly and averagely divided into 4 groups: (1) living at low altitude (LL), (2) living and training at low altitude (LLTL), (3) living at high altitude (LH), and (4) living at high altitude and training at low altitude (LHTL). Eight-week incremental treadmill exercise and hypoxic simulation were used to establish LHTL animal model. Mononuclear cells from peripheral blood were obtained by density gradient centrifugation 12 hours by the end of 8-week experiment. The cells were suspended in conditioned medium 199 for culturing in vitro. Their phenotypes were confirmed by uptake of acetylated LDL and binding of fluoresce in isothiocyanate (FITC)-labeled Ulex europaeus agglutinin 1 (UEA-1) lectin. Inverted microscopic observation was used to identify the morphological changes in endothelial progenitor cells and measure the cell count. Results Adherent cells and early CFUs in groups LLTL, LH and LHTL increased more obviously than in group LL(P<0.01), whereas the number of attached cells between group LH and LHTL was not different (P>0.05). Conclusion Proliferative capacity of endothelial progenitor cells can be promoted by both hypoxic stimulation and exercise, and the promotion is more significant if combination of hypoxia and exercise was employed simultaneously.
