1.Protective effect of fentanyl on cultured cardiomyocyte against injury induced by anoxia-reoxygenation

ve To assess the protective effect of fentanyl on primary cultured myocardial cells against injury induced by anoxia-reoxygenation. Methods Ventricular myocardial cells enzymatically isolated and cultured in 1640 culture medium for 6 days were randomly divided into 5 groups: group A received no anoxia served as control; group B received 2 hours anoxia followed by half an hour reoxygenation; and group C, D and E received 10ng ? ml-1, 30 ng ? ml-1 and 50 ng? ml-1 fentanyl respectively before anoxia-reoxygenation. The cell viability, myocardial intracellular content of malondialdehyde (MDA) and the activities of lactic dehydrogenase (LDH) and creatine kinase(CK) were measured at the end of the experiment. Results Anoxia-reoxygenation caused dramatic decrease in cell viability, and increases in myocardial intracellular MDA content and the LDH and CK activities as compared with those in control group. Fentanyl 30 ng?ml-1 and 50 ng?ml-1 significantly attenuated the increases in LDH, CK activities and MDA content, and decrease in cell viability caused by anoxia-reoxygenation. Fentanyl 10ng?ml-1 did not produce any significant changes. Conclusions Fentanyl can produce protective effects on primary cultured cardiomyocytes against anoxia-reoxygenation injury.

3.Construction and identification of recombinant adenovirus encoding mouse FGFR3 cDNA

Objective To construct the recombinant adenovirus encoding mouse wild type FGFR3 cDNA. Methods Mouse FGFR3 cDNA obtained from MoFR3/SV was subcloned into plasmid pBluescript KS and further cloned into plasmid pAdTrack-CMV. The plasmid (pAdTR3) was transferred into BJ5183 cells which contained the adenovirus plasmid (pAdeasy-1) to produce recombinant adenovirus vector encoding FGFR3 cDNA (pAdE-FGFR3). The recombinant adenovirus vector was identified and transfected into the adenoviral packaging cell HEK293 by lipofectamine 2000 to get recombinant adenovirus particles. The adenovirus was confirmed by polymerase chain reactin (PCR) and its titer was determined. Then the recombinant vector was transfected into HT29 cells. The expression of FGFR3 mRNA and protein in HT29 cells was assayed by RT-PCR and Western blotting. Results The recombinant adenovirus vector encoding FGFR3 cDNA was correctly constructed and confirmed by restriction endonuclease analysis and DNA sequencing analysis. The transfected HEK 293 cells were lysed by freeze-thawing to obtain the recombinant adenovirus in the lysate. Further, PCR product of the lysate confirmed the presence of recombinant adenovirus. The viral titer was 3?109pfu/ml. RT-PCR and Western blotting showed that pAdE-FGFR3 transfected HT29 group expressed FGFR3 higher than that of GFP controlled group. Conclusion Infective recombinant adenovirus encoding FGFR3 cDNA was successfully obtained by plasmid homogenous recombination in bacteria, and highly expressed in HT29 cells after transfection, which paves a way for studying the effect of FGFR3 in bone fracture healing.
4.Effect of FGFR3 transfection on CbfaⅠ and Collagen Ⅰ expression in mouse mesenchymal stem cells

Objective To explore the effect of fibroblast growth factor receptor 3(FGFR3)on the expressions of CbfaⅠ and Collagen Ⅰ in mesenchymal stem cells(MSCs)after Ad-FGFR3 transfection.Methods After Ad-FGFR3 and Ad-GFP were respectively transfected into mouse mesenchymal stem cells,the expression of FGFR3,CbfaⅠ and Collagen Ⅰ mRNAs and proteins were detected by RT-PCR and Western blot.Results The expressions of FGFR3,CbfaⅠ and Collagen Ⅰ were significantly higher in Ad-FGFR3 transfected MSCs than in Ad-GFP group.Conclusion FGFR3 can up-regulate the expression of CbfaⅠ and Collagen Ⅰ.

7.Meta-analysis of the effect of flupentixol and melitracen combined with conventional medicine on irritable bowel syndrome

Objective To evaluate the efficacy difference between flupentixol and melitracen combined with conventional medicine and conventional drug alone in the treatment of irritable bowel syndrome (IBS).Methods Studies on flupentixol and melitracen combined with conventional medicine (pinaverium bromide and trimebutine maleate) and conventional medicine alone in the treatment of IBS were collected and reviewed through searching the Chinese Academic Journals Full-text Database,Chinese VIP Full-text Database and PubMed database.After that,the test for homogeneity and the combined effect test were estimated. Results A total of 12 clinical studies on flupentixol and melitracen combined with conventional medicine for four to twelve weeks in the treatment of IBS were collected six studies indicated the homogeneity test of flupentixol and melitracen combined with pinaverium bromide (x2 =1.90,df=5,P=0.86,I2 =0).For the fixed-effect model,the odds ratio was 7.92 and 95% CI was between 4.85 and 12.95,which suggested the combined therapy was better.The other six studies indicated the homogeneity test of flupentixol and melitracen combined with trimebutine maleate (x2 =5.60,df=5,P =0.35,I2 =10.6 %).For the fixed-effect model,the odds ratio was 5.91 and 95% CI was between 4.06 and 8.61,which suggested the combined therapy was better.After merging the 12 studies it indicated the homogeneity test of flupentixol and melitracen combined with conventional medicine (x2 =8.40,df =11,P =0.68,I2 =0).For the fixed effect model,the odds ratio was 6.63 and 95% CI was between 4.92 and 8.93.Conclusion The efficacy of flupentixol and melitracen combined with conventional medicine in the treatment of IBS was better than that of conventional medicine alone,however,still more high quality trials are needed for further confirmation.
8.A self-designed double J tube for ureteral stenting

Objective To devise an improved double J tube for ureteral stenting so as to simplify its postoperative removal. Methods One end of the double J tube was made blind and got through the urinary tract via the ureteral wall or via a weak point of the renal cortex.The blind end was then embeded in the subcutaneous tissue and the ureteral stent was left behind for 4~12 weeks.A small incision (0.5 cm) was made in order to withdraw the stent and cystoscopy could be avoided.The self devised tube has been clinically employed for 60 cases. Results The stent has been satisfactory in all the 60 cases and its postoperative removal has been easy with no need of cystoscopy. Conclusions The subcutaneous embedding of a blind end of double J tube is simple and effective and its postoperative withdrawal is easy with no need of cystoscopy.
9.Residue and Degradation of Profenofos in Soil

Objective To investigate the degradation rate of profenofos in soil. Methods The residues of profenofos at different times after spraying on soil were determined by GC-FPD method. Results Detection limit of profenofos in soil was 0.04 mg/kg. The recovery rates and relative standard deviation obtained by adding standard assay were 95.2%~98.7% and 3.4%~4.5% respectively. According to the dose recommended, degradation of profenofos sprayed on soil was consistent with one range dynamics. The half-life of profenofos in soil was 1.09 days. At the 13th day after spraying on soil, 98.3% of profenofos had degraded. Conclusion This method was simple, rapid, accurate and precise. The degradation of profenofos was more rapid and profenofos contamination in soil was not a serious problem.
10.Management of bladder cancer near ureteral orifice

Objective To evaluate the surgical treatment of bladder cancer near ureteral orifice. Methods 42 cases of bladder cancer requiring ureteral reimplantation were divided into two groups at random:20 cases in group A:tongue-shaped incision of bladder wall around ureteral orifice was performed to modify Huctch-Ⅱ ureter implantation and 22 cases in group B:severing the ureter just outside the bladder,the ureter was then implanted into the bladder after partial cystectomy.Duration of the procedure,blood loss,amount of post-operation drainage,early complication,bladder ureter reflux,ureter obstruction,and cancer recurrence rate were studied and compared. Results In group A,the surgery took 60.5?18.5 min,blood loss amounted to 50.6?12.8 ml and the post-operative drainage 50.6?17.8 ml.In group B,the operation needed 150.9?22.4 min,the blood loss amounted to 220.5?28.2ml and post-operative drainage 280.6?58.9 ml.All the above three indexes showed siginificant difference between the 2 groups (P0.05). Conclusions Tongue-shaped incision of bladder wall around the ureteral orifice better with less complication,technically easier and worth recommendation.
