1.RIA OF HUMAN PLASMA PARATHYROID HORMONE_(1-34) AND ITS CLINICAL APPLICATION

A RIA method for determining human plasma parathyroid hormone (hPTH1-34) was established. This was possible mainly by the production of an antiserum of high liter raised in rabbit against hPTH1-34. The methodological investigation data were listed below: the affinity constant, Ka= 1,95?1010 mol/L; the cross reactivity with each of the three analogous peptides, i.e., hACTH1-34, hCT and hPTH44_68, being all less than 0.005%; the intra- and inter-batch coefficients of variation being 6.0% and 13.0% respectively; the recoveries for different levels of hPTH1_34 being ranged within 83.1 - 102.6%; and the sensitivity being 1.1 pmol/L. The reference value obtained by testing 50 male and 50 female normal adults was 11.7 ?4.4 pmol/L. It has been applied clinically for patients with various endocrine diseases including hyper- and hypoparathyroidisms and metabolic bone diseases especially in the aged population and the results fitted well with clinical diagnosis.
2.Effect of rivaroxaban in preventing deep venous thrombosis after large orthopedic operation of lower extremity

Objective To discuss the effect ofrivaroxaban in preventing deep venous thrombosis after large orthopedic operation of lower extremity.Methods Totally 80 patients after lower extremity of large orthopedic operation were selected,they were divided into two groups randomly.The patients in control group (39 cases) were sc injected with enoxaparin through.The patients in observation group (41 cases) were given rivaroxaban orally.The effect of rivaroxaban in preventing deep venous thrombosis after large orthopedic operation of lower extremity were evaluated by hemodynamic indexes,diagnosis index,and complications during 1 months follow-up.Results Before operation,there was no statistical significance on platelet,hemoglobin,and PT values between two groups.Compared with the value before operation,the value ofplatelet and hemoglobin had no significantly change after 3 d of operation.But the PT was increased (P < 0.05).After 3 d of operation,there was no statistical significance on platelet and hemoglobin between two groups.But the PT value of observation was higher than that of control group (P < 0.05).After 1 h operation,there was no statistical significance on D-dimer,Hcy,and hs-CRP between two groups.Compared with the value after 1 h of operation,the values of D-dimer,Hcy,and hs-CRP were decreased significantly (P < 0.05).And the indexes of observation group were lower than those of control group (P < 0.05).After 1 months of operation,although there was no statistical significance on complications between two groups,the incidence rate of secondary bleeding and DVT of control group was higher than that of the observation group.Conclusion The rivaroxaban could prevent DVT well,which is suitable for the anticoagulation therapy after lower extremity of large orthopedic operation with little influence on platelet and hemoglobin.It is worthy of clinical application.
3.STUDIES ON THE ENZYME CYTOCHEMISTRY OF TOXOPLASMA GONDII AND THE INFLUENCE OF ARTEMETHER

Male mice of Kunmlng strain were infected with 10 000 tachyzoites intraperitoneally. 2 hours after infection the mice were divided into 2 groups and administered 5 % of amylurn and 200 mg/kg of artemether by gavage for 8 consecutive days. The ultrastructural enzyme cytochemical studles on cytidine monophosphatase (CMP ase) and glucose-6-phosphatase (G-6-P-ase) of the parasites were carried out. CMP-ase was found scattered in the lysosomes of the parasites as well as in the macrophages. No differences were observed in the localization and intensity of CMP-ase activity between the nontreated and treated with drug parasites. G6-P-ase was found surrounding the parasite membrane and scattered in the parasitophorous vacuole in the nontreated parasites. After treatment with artemether, the intensity of G-6-P-ase activity was decreased compared with nontreated control parasites. It is suggested that artemether may exert some action on the G-6-Patase of T. gondii and thus influence the energy metabolism of the parasite.
4.MicroRNA-21-5p regulating p38 mitogen-activated protein kinases signaling pathway

Objective To verify the targeting regulatory relationship between microRNA (miR)-21-5p and mitogen-activated protein kinase kinase 3 (MKK3).Methods miR-21-5p and MKK3 targeted relationship was analyzed by bioinformatics database predict.Constructing MKK3 3'UTR reporter vector (MKK3-3U) and mutation reporter vector (MKK3-3 U-M) was constructed by synthesizing miR-21-5p mimics (mimic),miR-2 1-5p inhibitor (inhibitor),nonsense miR (NC).NC (NC1 group),mimics (mimic1 group),inhibitor (inhibitor1 group) together with MKK3-3U cotransfected human embryonic kidney (HEK293) cells,respectively.Empty vector-pYr-MirTarget (NC2 group),MKK33U (Mt group),M KK3-3U-M (Mut group) together with mimic co-transfected HEK293,respectively.The fluorescence ratio was detected by dual luciferase assay.NC,mimics,inhibitor intervened human renal tubular epithelial (HK-2) cells,respectively.RT-PCR and Western blot were used to test cell MKK3 expression level in HK-2.Results 1.The fluorescence ratios of NC1 group,mimicsl group,and inhibitor1 group were 100.00%,67.99%,133.17%,and there was difference between mimic1 group and NC1 group(t =19.93,P < 0.05).2.The fluorescence ratios of NC2 group,Mt group and Mut group were 100.00%,65.30%,98.48%,and there was difference between Mt group and NC2 group (t =14.39,P < 0.05).There was no significant difference between NC2 and Mut group (t =0.63,P > 0.05).3.MKK3 mRNA relative expression levels of the control group mimic2 group and inhibitor2 group were 1.36 ± 0.02,1.01 ± 0.04,1.43 ± 0.06 ; relative protein expression levels were 0.97 ± 0.05,0.62 ± 0.06,1.36 ± 0.32.MKK3 mRNA and protein levels of mimic2 group were significantly lower compared with the control group (F =85.98,118.26,all P < 0.01).Conclusions MKK3 is the target gene of miR-21-5p.miR-21-5p can regulate the expression of MKK3 both in mRNA and protein levels.miR-21-5p may be an important regulatory molecule in p38 mitogen-activated protein kinase signaling pathway.
5.Concurrent PCF chemotherapy plus radiotherapy for middle and advanced cervical cancer

Background and purpose:Cervix cancer is the second most common cancer in gynecologic malignant tumors. Chemotherapy has been used more and more lately, and concurrent chemo-radiotherapy are receiving increasing attention. In this study, we investigated the clinical effect of concurrent PCF chemotherapy plus radiotherapy for moderate and advanced cervical cancer. Methods:One hundred and ninty-six patients with moderate and advanced cervical cancer were randomly divided into two groups: only radiotherapy group and chemo-radiotherapy group (98 cases for each group). Two groups underwent the same fractionation irradiation, the chemo-radiotherapy group was given PCF regimen that consisted of (DDP) at the dose of 20 mg daily intravenously for 1-5 days, (5-FU) at the dose of 500 mg daily for 1,3,5 days and (CTX) at the dose of 400 mg daily for 2,4 day for 2-4 cycles. Both groups received radiotherapy with 30 Gy first and were irradiated by the combination of external radiotherapy plus intracavitary irradiation.Results:The immediate response rates of the two groups were 100 %, there was no statistical difference. 7.Experience of Minimally Invasive Percutaneous Nephrolithotomy for the Treatment of 216 Cases Upper Urinary Tract Calculus

Objective To investigate effectiveness of treatment of Upper urinary tract calculus by MPCNL application.Methods From May 2000 to May 2007,causing artificial kidney hydronephrosis for the affected kidney,established renal percutaneous channels(F16~18),which established the single channels of 202 cases,dual-channel of 14 cases,by Ureteroscopic Lithotripsy and holmium laser lithotripsy.Results 183 cases of 216 patients were One-time stone;33 cases were second stone.The average clearance rate is 90.32%,the average operation time is 79min,average valume of about bleeding is 80ml,the average hospitalization days are 8 days,kidney ostomy average stay 7.5 days.Of the operation,9 cases of obviously bleeding(4%)before and after operation,1 case uncontrollable bleeding after interventional treatment failure with Nephrectomy,others to stop beeding by symptomatic treatment,2 cases find urinary extravasation postoperative and cured by adjusting location of renal ostomy.Conclusion Minimally invasive percutaneous nephrolithotomy for the treatment of Upper urinary tract calculus is safe and effective for patients with less trauma,quicker recovery and high learance rate,the efficacy is satifactory.
8.Interventional Therapy for Left Iliac Vein Compression Syndrome

Objective To evaluate the efficacy and safety of interventional therapy for left iliac vein compression syndrome (LICS). Methods A total of 27 patients with LICS that were diagnosed by colour Doppler or venography,were included in this study. Among the cases,the disease course was shorter than 3 weeks in 13 patients,who presented with acute iliac-femoral vein thrombosis; in the other 14 patients,the disease course was longer than 3 weeks,showing the symptoms of chronic venous thrombosis. Venous puncture was performed via the contralateral femoral vein in 13 cases,and through the ipsilateral popliteal vein in 14. Venous filter was placed in 11 cases before thrombolysis,while in the other 16 cases,no filter was used. All of the patients received catheter-directed thrombolysis,afterwards,7 of them underwent percutaneous transluminal angioplasty,and the other 20 received angioplasty plus stent placement. Results In the 13 patients,who received venous puncture via the contralateral femoral approach,the guide wire could advance through the stenosis successfully in 8 patients but failed in 5. Ipsilateral popliteal vein puncture were then employed and succeeded in all the patients. Ipsilateral popliteal puncture were performed successfully in 14 patients. In 11 patients,inferior vena cava filter was placed before the procedure. In this series,the thrombolysis time was (85?16) h with a dose of (3000?320) thousand units urokinase. The thrombosis was dissolved completely in 13 acute patients and partial dissolution was attained in 14 patients in whom blood flow signal were detected color Doppler. After the operation,venogram showed complete patent iliac veins in all of the 27 cases. The patients were followed up for 6 to 26 months (mean,11 months). During the period,19 patients obtained patent iliac-femoral vein with the symptoms disappeared; 8 patients met the standard of improvement. Conclusions Interventional technique can be the first-line treatment for left iliac vein compression syndrome because of its favorable result and minimal invasion.
9.Differentiation of Multiple System Atrophy and Parkinson's Disease Using 11C-CFT PET/CT

Purpose To investigate the value of 11C-CFT PET/CT dopamine transporter (DAT) imaging in differential diagnosis of multiple system atrophy (MSA) and Parkinson's disease (PD).Materials and Methods The 11C-CFT PET/CT images of clinically confirmed MSA patients (21 cases),PD patients (24 cases) and healthy adults (10 cases as normal control) were analyzed retrospectively.The volume of interest (VOI) were drawn manually,and the DAT binding indexes and asymmetry indexes of different regions of striatum,including caudate and putamen,were calculated.The differences of DAT binding indexes and asymmetry indexes among the above three groups were analyzed.Results Compared with the normal control group,the striatal DAT binding indexes of MSA group or PD group were significantly reduced (P<0.05).There were no significant differences in DAT binding indexes between the MSA group and the PD group (P>0.05).Compared with the normal control group,the DAT binding asymmetry indexes were significantly increased in the PD group (P<0.05),but the indexes showed no significant differences in MSA group (P>0.05).The DAT asymmetry indexes of caudate and putamen in the PD group were higher than those in the MSA group (P<0.05).Conclusion 11C-CFT PET/CT imaging can detect the degeneration of dopaminergic neurons in striatum.The number of striatal dopamine transporters declines in both MSA and PD patients,but the asymmetry of striatal dapamine transporter in PD patients is higher than that in MSA patients.11C-CFT PET/CT can differentiate MSA and PD.
10.Comparison of blood selenium content between cardio-cerebrovascular disease patients and healthy persons in Dalian City

BACKGROUND: Selenium is one of necessary microelement of human body. It acts as the active center of selenium-contained enzyme with antioxidant function, which can clean up internal free radical, effectively catalyze harmful perhydride deoxidizing into innocuous hydroxyl compound,reinforce immunity of human body, prevent and cure cardio-cerebrovascular diseases and cancer.OBJECTIVE: To provide evidence for preventing and treating cardiocerebrovascular diseases via comparison of blood selenium content between patients and healthy persons.DESIGN: Case-control analysis.SETTING: Dalian Railway Medical School, Dalian Railway Hospital, Department of Applied Chemistry, Dalian Railway College.PARTICIPANTS: Totally 319 blood samples (169 males, 150 females)were collected from patients with cardio-cerebrovascular diseases from Dalian Railway Hospital from March 2000 to May 2001. Among them,there were 164 with hypertension, 97 with coronary atherosclerotic heart disease (CAHD), 41 with cardiac dysrhythmia and 17 with cerebral vascular accident. 300 healthy patients (159 males and 141 females) were in the healthy group.METHODS: The blood selenium content of patients with cardio-cerebrovascular diseases was detected with neopolarography.MAIN OUTCOME MEASURES: ①Total level of blood selenium of testees of the two groups. ②Comparison of level of blood selenium between males and females. ③Comparison of level of blood selenium of patients with different diseases.RESULTS: A total of 619 testees were involved in the result analysis. ①The total level of blood selenium of the patient group was lower than that of the healthy group [(114.0±52.5), (146.5±51.0) μg/L,P < 0.001],which was only 76% of that of the healthy persons. ②Content of blood selenium of males and females was normal distribution. There was insignificant difference between males and females (P > 0.10), but the level of blood selenium in females was slightly higher than that of males. ③The levels of blood selenium of patients with hypertension, CAHD, cardiac dysrhythmia were 82.7%, 68.6% and 66.0% of that of healthy persons, respectively, but the difference between males and females was insignificant(P > 0.10). The level of blood selenium of patients with cerebrovascular accident (CVA) was the lowest, (84.4±28.9)μg/L, which was 57.6% of healthy persons.CONCLUSION: The content of blood selenium of patients with cardiocerebrovascular diseases is lower than that of healthy persons. Organic selenium and inorganic selenium should be supplemented moderately.
