Objective To establish a noninvasive method for prenatal genetic analysis by using maternal serum and apply the method in fetal sex determination, paternity testing. Methods Samples of maternal serum from 53 pregnant women (11 to 36 weeks of gestation) were collected. The DNA extracted from each sample was amplified by using"Y-PLEX 6" amplification kit .which enabled the simultaneous analysis of six Y-STR loci including DYS393.DYS19.DYS389 II, DYS390, DYS391 and DYS385. The PCR products were detected by using ABI PrismTM 377 Sequencer and genotyped by related analysis software. Results (1) Y-STR specific alleles were detected in the maternal sera of all 29 mothers bearing male babies. Among the six Y-STR loci, specific alleles were detected in 29/29 at DYS393 locus,in 18/29 at DYS19 locus and in 10/29 at DYS390 locus. (2) Y-STR specific alleles were not detected in maternal sera of 24 pregnant women bearing female babies. (3) According to the presence of specific alleles at DYS393 locus and the value of allelic peak height and peak area, the accuracy of fetal sex determination was 100% . (4)The observed Y-STR alleles of each prenatal specimen from pregnant women with male fetuses were the same as the results of their husbands. Conclusion The assay of highly polymorphic Y-STR genotyping system developed by the authors provided a sensitive, accurate and non-invasive method to prenatal diagnosis. Our results demonstrate that fetal sex can be accurately determined and imply that paternity testing could be performed for pregnant women carrying male fetuses.

Objective To summarize the keypoints of clinical diagnosis of chondroblastoma and the experience on the treatment Methods The clinical materials of 16 cases(11 males, aged from 12 to 26 years)from June 1992 to February 2003 of pathologically diagnosed chondroblastoma were analyzed retrospectively. The tumor located at distal femur in 6 cases, great trochanter in 1 case, proximal tibia in 6 cases, proximal humerus in 2 cases, talus in 1 case. The main symptoms were swelling and painful joint. All the 16 cases were treated with curettage and autologous bone graft from iliac crest. Results Twelve cases were followed up for 6 months to 5 years .averaging 3. S years. All the bone graft healed well. One case., recurred 2 years after the initial curettage and was treated with curettage again. No recurrence occurred at the last follow-up 6 months after the second curettage. The function of the involved joints was nearly normal in all cases except for one case with slight limitation of knee flexion. Conclusions Mastering the image and clinical characteristics of chondroblastoma can avoid misdiagnosis and mistreatment Curettage can control the tumor satisfactorily. Postoperative rehabilitation can improve the function of the involved limb.

AIM As one of the reactive oxygen species, toxic dose of H2O2 leads to the necrosis or apoptosis of many kinds of cells. It is nuclear whether these processes are related with the changes of nuclear Ca2+ elicited by H2O2. This study on the effect of taurine on changes of nuclear and cytosolic (Ca2+) elicited by H2O2 in rat vascular smooth muscle cells. METHODS The techniques of Fluo-3 and laser scanning confocal microscopy were used. RESULTS Rapid and sustaining rise of nuclear [Ca2+] ([Ca2+]n) and cytosolic [Ca2+]([Ca2+]c) were found in cultured rat vascular smooth muscle cells upon adding of 0.5% H2O2. Pretreatment of the cells with 20 mmol*L-1 taurine, a regulator of cellular Ca2+ homestasis, significantly decreased the rise amplitude of [Ca2+]n (P<0.05), but failed to affect that of [Ca2+]c (P>0.05) elicited by H2O2. It was suggested that the changes of [Ca2+]n and [Ca2+]c have different respond to taurine. CONCLUSION It was concluded that there exist the independent regulating mechanisms of Ca2+ in the nucleus from differences between the nucleus and cytosol Ca2+ in sensitivity to action of H2O2 and taurine.

BACKGROUND:Plate fixation or intramedulary fixation for clavicle fractures has remained a matter of debate. At present, there is a need for a higher level of evidence to evaluate the advantages and disadvantages of two different surgical methods.
OBJECTIVE:To assess the clinical efficacy of intramedulary fixationversusplate fixation in the treatment of clavicle fractures.
METHODS:We conducted a computerized search of the electronic databases like Cochrane Library, PubMed, Embase, OVID, CBM, CNKI, Wanfang and VIP for relevant randomized controled trials on plate fixation or intramedulary fixation for clavicle fractures. The quality of the included studies was independently assessed with RevMan 5.3 software. The meta-analysis was performed in Stata 13.0.
RESULTS AND CONCLUSION:Eight randomized controlled trials with 527 patients were included in this meta-analysis. The meta-analysis results showed union time, surgery time, incision length, hospital stay and infection rate were less in the intramedullary fixation group than in the plate fixation group (P< 0.05). No significant difference in shoulder function score, upper limb function score, fracture nonunion and refracture was determined between the two groups (P< 0.05). The current evidences suggested that intramedullary fixation is the optimum choice in the treatment of clavicle fractures.

Objective To observe the clinical effects of tibial locking multidirectional interlocking intramedullary nail (TLMIIN) for tibial plateau fractures. Methods 38 cases with closed tibial plateau fractures treated by TLMIIN from October 2008 to May 2012 were retrospectively analyzed. There were 22 males and 16 females, with an average age of 48.7 years (range, 28-67 years). There were 24 fractures on the left side while the other 14 factures were on the right side, which were all fresh frac?tures. According to AO/OTA classification of tibial plateau fractures, there were 4 cases of B1, 1 case of B2, 14 cases of B3, 8 cas?es of C1, 5 cases of C2 and 6 cases of C3. Close reduction were performed on 4 cases (10.5%of all cases). Open reduction were performed on the other 34 cases. The limited incision was decided by the distribution of the fragments and fracture line. Hohl?Luck evaluation system was applied for the follow?up. Results The mean follow?up period was 18.1 months (range, 11-23 months). All fractures were healed at an average period of 87.4 days (range, 48-131 days). The average time from operation to full weight?bearing was 108.9 days (range, 80-128 days). Hohl?Luck evaluation system was used in the final follow?up. The excellent and good rate of functional score was 94.7%(36/38), including 28 cases excellent, 8 cases good and 2 cases fair from functional as?pect. The excellent and good rate of radiological score was 84.2%(32/38), including 23 cases excellent, 9 cases good and 6 cases fair from radiological aspect. No complications such as infection, breakage and loosening of the screw, malunion, nonunion oc?curred at the time of the latest follow?up. 6 cases with serious swelling of the knee joint and the soft tissue of crus were cured by an?ticoagulation, dehydration and physiotherapy treatments after operation. The 2 cases with a little exudates and incrustation was bacterial cultured negative and healed after 16 days and 18 days on the incision. 1 case had lost of reduction due to weight?bearing 1 week after operation, who had 25 degree of varus deformity, which was left dispose, and the bone healing and joint function were unaffected. Conclusion The intramedullary support, restrictive and non?restrictive multidirectional tridimensional fixation of TLMIIN technology had satisfactory effects in treating plateau fractures. It can supply a new therapeutic method, with little dam?age of soft tissues when taking out the internal fixation and reduce some postoperative complications.
