2.Orthogonal Test for Optimization of Rhubarbs Extraction Process and Pharmacodynamic Studies the Extracts

Objective To optimize the solvent extraction of Rhubarbs. Methods With different concentrations of alcohol as the solvent, the amount of solvent, the extracting time and the extracting frequencies were assayed by orthogonal test to detect their influences on the extraction rate of rhubarb anthraquinone. And the extracts were analyzed and their pharmacodymamic effects were studied. Results Indicated by the content of total anthraquinones, the best conditions was: reflux extraction with 5- fold 50 % alcohol for 3 times, 0.5 h per time. The purgative activity of the extracts was stronger than that in the positive control group. Conclusion Optimization of the extracting conditions can promote the purgative activity of the extracts.
3.Clinical observation of polysomnographic in patients with different subtypes of Parkinson's disease

Objective To evaluate the changes parameters of polysomnographic ( PSG) with different subtypes of primary Parkinson's disease ( PD ) and analyze the clinical characteristics of sleep disorders in PD patients. Methods Ninety patients with primary PD [ tremor-predominant PD group ( n = 40 ) , postural instability gait disorder ( PIGD )-predominant PD group ( n = 50 ) ] and 50 healthy controls were detected by full night PSG monitoring. And the results were compared. Results Compared with tremor-predominant PD group, the total sleep time, bed time, sleep wake-up times, sleep efficiency and rapid eye movement ( REM ) sleep time of PIGD-predominant PD group were significantly lower (all P<0. 05), however, the sleep latency, S1 (min), S1 (%), S2 (min), S2 (%), S3+S4(min), S3+S4 (%) had no statistical significance (all P>0. 05). Compared with the controls, the total sleep time and bed time of tremor-predominant PD group were significantly lower (all P<0. 05), however, the sleep wake-up times, sleep efficiency, REM sleep time, sleep latency, S1 ( min ) , S1 (%) , S2 (min), S2 (%), S3 + S4 (min), S3 + S4 (%) had no statistical significance (all P>0. 05). Combined with video surveillance video observation,the abnormality of REM sleep behavior disorder ( RBD) was 70% with PIGD-predominant PD group, in which the abnormality of tremor-predominant PD group was 2. 5%. Conclusions The PIGD-predominant PD patients are more likely to appear sleep disorders and RBD than tremor-predominant PD patients. It may be related to clinical heterogeneity of different PD subtypes.
4.Influence of anisodamin on guinea pig experimental nasal hypersensitivity

Objective:To observe the influence of anisodamin on guinea pig experimental nasal hypersensitivity.Method:Experimental animals were divided into normal control group, TDI group,reserpine group and anisodamine group.The scores of symptom of nose and the cytologic changes of nasal mucosa were parameters of observing.Result:The nasal symptom of reserpine group was significantly stronger than the TDI group,and the eosinophilia evidently increased (both P<0.01).The nasal symptom of the anisodamine group reduced and the eosinophilia decreased.It is significantly difference between the TDI group and the reserpine group (both P<0.01).Conclusion:Anisodamine may play a role in therapy of the experimental nasal hypersensitivity with hyperfuction of parasympathetic nerve.
5.The value of combined detection of NT-proBNP,CA-125 and Hs-CRP for the diagnosis of chronic heart failure

Objective To investigate the changes of serum N-terminal proBNP (NT-proBNP) ,carbohydrate antigen 125(CA-125) and high sensitive CRP(Hs-CRP) levels in patients with chronic heart failure(CHF) ,to evaluate the severity of CHF using the three markers alone or in combination .Methods A total of 62 patients with CHF(CHF group) from our hospital were select-ed .Among them ,13 cases were in NYHA class Ⅱ ,27 cases were in class Ⅲ and 22 cases were in class Ⅳ .The control group in-cluded 54 patients without CHF .Serum levels of NT-proBNP ,CA-125 and Hs-CRP were detected in CHF group and control group . The left ventricular ejection fraction(LVEF) were detected by echocardiography .Results The serum levels of NT-proBNP ,CA-125 and Hs-CRP of CHF group before treatment were significantly higher than those of control group (P<0 .01) ,and the serum levels increased with the degree of severity of heart failure .There were statistically significant difference among patients in NYHA classⅡ ,class Ⅲ and class Ⅳ (P< 0 .01) .There was a high correlation between NT-proBNP ,CA-125 and LVEF (NT-proBNP vs . LVEF ,r=0 .934 ,P<0 .01 ;CA-125 vs .LVEF ,r=0 .878 ,P<0 .01) ,a low correlation between Hs-CRP and LVEF(r=0 .437 ,P<0 .05) .Compared with the single marker detection ,the combined detection of NT-proBNP ,CA-125 and Hs-CRP increased the AUC .Conclusion The combined detection of NT-proBNP ,CA-125 and Hs-CRP can increase the value of the observed severity of heart failure .
6.Effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials in adolescent patients undergoing scoliosis surgery

Objective To investigate the effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials (SEPs) in the adolescent patients undergoing scoliosis surgery.Methods Forty-five adolescent patients of both sexes,aged 12-17 yr,with body mass index of 15-23 kg/m2,undergoing scoliosis surgery under general anesthesia,were randomly divided into 3 groups ( n =15 each):low concentration sevoflurane group ( group S1 ),moderate concentration sevoflurane group ( group S2 ) and high concentration sevoflurane group (group S3 ).Anesthesia was induced with fentanyl 4 μg/kg,midazolam 0.03 mg/kg,propofol 2.0 mg/kg,and rocuronium 0.6 mg/kg.The patients were endotracheal intubated and mechanically ventilated.Anesthesia was maintained with propofol infusion at 8 mg· kg-1 · h-1 and remifentanil infusion at 0.25 mg· kg-1 ·min-1.After beginning of the surgery,1.8%,4.0% and 6.0% sevoflurane were inhaled for 10 min in groups S1-3 respectively,SEPs were measured by stimulating the posterior tibial nerve and significant SEP suppression was detected and recorded.The wave P40 latency,wave P40-N50 amplitude,and time for P40 waveform changes were recorded before sevoflurane inhalation (baseline) and at 10 min of sevoflurane inhalation,and the time for recovery of P40 waveform was recorded after sevoflurane inhalation was stopped.The prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were calculated.Results The rates of significant SEP suppression were 80%,100% and 100% in groups S1,S2 and S3 respectively ( P > 0.05).Compared with group S1,the prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in groups S2 and group S3 ( P < 0.05).Compared with group S2,the decreased percentage of P40-N50 amplitude was significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in group S3 ( P < 0.05).Conclusion 1.8%,4.0% and 6.0% sevofiurane combined anesthesia can suppress SEPs in the adolescent patients undergoing scoliosis surgery,and they are not suitable for the surgeries requiring SEP monitoring.
7.The clinic and imaging features of Klebsiella pneumoniae liver abscess in diabetes mellitus patients with different levels of HbA1c

Objective To analyze the clinic and imaging features of Klebsiella pneumoniae liver abscess in diabetes mellitus patients with different levels of HbA1c.Methods One hundred and forty-six diabetes patients with KPLA were divided on the basis of their levels of HbA1c into three groups:complete glycemic controlled (HbA1c ≤ 7%);good glycemic controlled (7% <HbA1c≤9 %) ≤ bad glycemic controlled (HbA1c> 9 %).Compared the patients' characteristics,clinic features,imaging features and complications among each group.Results Compared with patients in groups of complete and good glycemic controlled,patients in group of bad glycemic controlled tend to have younger age at onset[(with an average age of (59.1 ± 13.8)years],longer hospital stay[(with an average stay of (23.1 ± 7.6) day] and more experience complications such as:hyperlipoidemia(49 cases,77.8%),chronic renal failure(12 cases,19.2 %),life-threatening clinical crisis,and higher infection rate.Patients in bad glycemic controlled also had high risk of biliary pneumatosis(18 cases,28.6 %),hepatic venous thrombosis (20 cases,31.8 %) and gas-forming pyogenic liver abscess(26 cases,28.6 %)(P<0.05).Conclusion The complications of hepatic venous thrombosis,gas-forming and infection in diabetes patients with KPLA were associated with glycemic uncontrolled.
9.Expression of matrix metalloproteinase-11 in gastric cancer and its relation with transcription factor Sp1

Objective To explore the expression of matrix metalloproteinase-11 (MMP-11) gene in gastric cancer and its correlation with transcription factor Sp1.Methods The expressions of the MMP-11 and Sp1 in level of RNA and protein in 11 specimens from the gastric cancer patients s were detected by RT-PCR,differential PCR and Western-blot,respectively.Results Electrophoresis illustration showed MMP-11 mRNA expression had significant differences in eight pairs of gastric cancer tissues and normal tissues,including 7 pairs of over expressed in gastric cancer tissues and low or no expression in normal tissues,and 1 pair of low expression in cancer tissues but high in normal tissues.The relative molecular weight of Sp1 protein was mainly 95×103 in cancer tissue and 106×103 in normal tissues.Activated MMP-11 protein mainly was over expressed in gastric cancer tissues,and low or no expression in normal tissues,but degradable MMP-1 1 protein was not observed.The enzyme prototype of and activated type of MMP-11 protein were also associated with Sp1 protein.Conclusions The MMP-11 mRNA expression in gastric tissue is higher than that in the adjacent tissues.Sp1 protein and MMP-11 protein are expressed in gastric cancer,but are low or no expression in normal tissue.There is relationship between Spl and MMP-11.
10.The study of influence on cyclin and cyclin-dependent kinase inhibitor of Kasumi-1 cell treated with VPA

Objective To explore the influence on cyclin and cyclin-dependent kinase inhibitor of cell cycle of Kasumi-1 cell-derived from t(8;21)/AML treated with histone deacetylase(HDAC)inhibitor valproic acid(VPA).Methods RT-PCR assay was applied to detect mRNA expression of regulation factor of cell cycle of Kasumi-1 cell with VPA at different concentration and different time. Results VPA could down-regulate mRNA expression of cyclinD1,cyclinE1 and cyclinB1,and up-regulate mRNA expression of p21 WAF1/CIP1 with no obvious variation of mRNA level of p27KIP1. Condusion VPA could regulate cell cycle through the regulation of cyclins and cyclin-dependent kinase,arresting Kasumi-1 cell at G0/G1 phase.
