1.The expression of Vav1 and its correlation with prognosis in patients with gastric cancer

Objective To investigate the expression of Vav1 in gastric cancer(GC), and analyze its potential relevance to clinicopathological characteristics and prognostic significance in GC patients. Methods The mRNA expression level of Vav1 was detected by real-time quantitative PCR in GC cell lines (HGC-27,SGC7901 and MGC803) and normal gastric mucosa cell line (GES-1). In addition, the protein expression of Vav1 was analyzed by immunohistochemistry in formalin-fixed samples from 105 GC patients. The associations between clinical pathological features and Vav1 protein expression were evaluated in GC patients. Kaplan-Meier method and Cox regression analysis were used to analyze the related factors influencing the prognosis of GC. Results The mRNA expression levels of Vav1 were significantly higher in GC cell lines (HGC-27, SGC7901, MGC803) than those in normal gastric mucosa cell line (GES-1, P<0.05). The positive expression of Vav1 in GC tissues was correlated with diameter of tumor and lymph node metastasis (P<0.05). The univariate analysis showed that size of tumor, degree of differentiation, serosal invasion, lymph node metastasis and Vav1 expression were significantly related with prognosis of GC (P<0.05). Results of Cox regression showed that tumor invasion depth (HR=2.764, 95%CI 1.316-5.817, P=0.007), lymph node metastasis (HR=1.298, 95%CI 1.098-1.534, P=0.002) and Vav1 expression (HR=2.577, 95%CI 1.066-3.946, P=0.006) were the risk factors affecting prognosis of patients with gastric cancer. Conclusion Vav1 performs important role in the aggressiveness of GC, and Vav1 may serve as a novel prognostic factor in GC.

Objective:To identify HLA-A2-restricted CTL epitope of breast cancer differentiation antigen NY-BR-1.Methods:The HLA-A2-restricted CTL epitope of breast cancer differentiation antigen NY-BR-1 is predicted by combination quantitative motif method and the molecular dynamics.The three epitope were assayed their affinity to HLA-A2.Results:The affinity to HLA-A2 of NY-BR-1_ 1043-1051 is a best.Conclusion:NY-BR-1_ 1043-1051 is a HLA-A2 restricted epitope.

5.Relationship between changes of facial soft tissue and hard tissue profile in correction of children's mandible retruding by functional appliance

Objective To evaluate the changes of soft tissue and hard tissue in the correction of Class II division 1 malocclusion by functional appliance, and to investigate the correlation between these changes and the mechanism underlying the soft tissue profile changes. Methods A total of 32 cases of developing class II division 1 malocclusion were selected. They were in the spurt or pre-spurt phase of growth and development and treated with functional appliance, the molar relationship was corrected to either Class I or overcorrected Class I occlusal relationship. Soft tissue and hard tissue variables on the cephalograms were selected and analyzed. Firstly, paired t-tests were performed to compare the changes between before and after-treatment. Secondly, those variables that had statistically significant differences were selected and analyzed with the Pearson coefficient of correlation (r) between soft tissue and hard tissue changes. Results All variables had statistically significant differences between before and after treatment except for soft tissue facial angle and basic upper lip thickness, mandible plane angle and position of basic upper bone; Correlations between soft tissue and hard tissue changes were as follows: height of lower lip to Me' and S-Go (r=0.54); soft tissue chin thick and N-ANS (r=0.55); height of upper lip and ii/OLp (r=0.52); inferior sulcus to H-line and ii/OLp (r=0.60), pg/OLp, saggital relationship between upper-and lower-jaw (ss/OLp-pg/OLp) (r=0.47). Conclusion The changes of soft tissues profile results from the change of lower incisors' position, mandible' moving forward and its subsequent changes of facial heights and saggital relationship between upper-and lower-jaws.
6.Expression of c-myc and MDM2 protein in multiple myeloma and their clinical significance

Purpose To investigate the expression of c-myc and MDM2 protein in multiple myeloma (MM) and their correlation with clinical stage.Methods Immunohistochemical SP three-step method was used to detect the expression of MDM2 and c-myc protein in 60 cases of MM with different clinical stages and 10 cases of nontumorous bone marrow tissue.Results The positive rate of c-myc protein in MM and nontumorous bone marrow tissue were 71.7% and 10.0%;the positive rate of MDM2 protein in two tissues were 80.0% and 20.0%,respectively.The expression difference of two proteins between MM and nontumorous bone marrow tissue was statistically significant (P < 0.05).The expression of c-myc and MDM2 protein was positively correlated with MM clinical ISS stage (P < 0.05),but irrelevant with the age and gender.The expression of c-myc and MDM2 protein in MM was positively correlated (P < 0.05).Conclusion The c-myc and MDM2 protein have a highly expression in MM,and it may be relative to the occurrence of MM and clinical stage.In the MM,c-myc and MDM2 protein may have synergetic functions and promote the development of tumor.
9.Vacuum sealing drainage facilitates the survival of slender narrow pedicle flap

Objective To probe into the application of vacuum sealing drainage (VSD) in flap with slender narrow pedicle and to observe the survival,the shaping of blood vessel,congested edema and the expression of CD34 in the flap,and to analyze the underlying mechanism of VSD on the flap survival.Methods 64 SD rats were randomly divided into 8 groups:4-hour group,8-hour group,1-day group,2-day group,3-day group,4-day group,and 6-day group.A flap was designed and put on either side of a rat's back.The both width and length of the flap were 0.8 cm and the pedicle sized in the diameter of 3.5 cm.Both sides of the flaps on the rats' backs were randomly arranged into control side and experimental side.VSD was fixed sideways to each group after the shaping of flaps.Each of the equipments was dismantled respectively in the corresponding time after the operation.Afterwards the color and swelling degree of the flaps were observed,a sample was cut and the flap survival area was measured altogether on the 7th day.Flap histology and the expression of CD34 were observed mainly through HE dyeing and immunohistochemistry.The statistical data were treated to figure out the flap survival rate and to determine whether statistical differences existed in the microvessel density (MVD).Results As the time of the negative pressure suction went on,the survival area of the experimental side flap was increasing day by day and came to the maximum on the third day (95±4)%,but no statistical difference was discerned between on the third day and on the sixth day (P>0.05).As the time of the negative pressure suction went on,the MVD in the flap was also increasing.Statistical differences (P<0.05) existed between the experimental side of the instant group,4-hour group and the 8-hour group and those of 3-day group,4-day group,and 6-day group.Conclusions The application of VSD can advance the survival of the flap,whose survival area can reach its maximum within 3 days under application.Besides,it can promote the increase of MVD in the flap.
10.Balloon combined with stent-assisted steel-coil embolization for the treatment of intracranial wide-necked aneurysms located at artery bifurcation sites

Objective To discuss the advantages of balloon combined with stent-assisted steel-coil embolization in treating intracranial wide-necked aneurysms located at artery bifurcation sites. Methods The clinical data of 20 patients with intracranial wide-necked aneurysms located at artery bifurcation sites (25 aneurysms in total), who were admitted to the First Affiliated Hospital of Kunming Medical University during the period from January 2014 to May 2014 to receive balloon combined with stent-assisted steel-coil embolization treatment, were retrospectively analyzed. Among the 20 patients, 16 had a history of subarachnoid hemorrhage and 4 had no history of subarachnoid hemorrhage. A total of 25 intracranial aneurysms located at artery bifurcation were detected; the locations included basilar terminal bifurcation (n=14), middle cerebral artery (MCA) bifurcation (n=8) and internal carotid artery (ICA) bifurcation (n=3). Three dimensional DSA was performed immediately and three months after the treatment , and the manifestations were evaluated according to Raymond classification. The clinical efficacy at three months after the treatment was assessed with the modified Rankin scale (mRS). Results Three dimensional DSA performed immediately after the balloon combined with stent-assisted steel-coil embolization treatment showed that among the 25 intracranial wide-necked aneurysms located at artery bifurcation 21 belonged to Raymond gradeⅠ, 2 belonged to Raymond grade Ⅱand 2 belonged to Raymond grade Ⅲ. Follow-up DSA performed three months after the treatment revealed that 20 aneurysms were Raymond grade Ⅰ, 3 aneurysms were Raymond gradeⅡand 2 aneurysms were Raymond grade Ⅲ. Three months after the treatment the mRS score was 0 point in 17 patients and one point in one patient, and the prognosis of these patients was good; the mRS score was 4-6 points in 2 patients, and the prognosis of the two patients was poor. Conclusion For the treatment of intracranial wide-necked aneurysms located at artery bifurcation sites, balloon combined with stent-assisted steel-coil embolization has obvious advantages.
