Obesity is a rapidly growing international health problem. The association of fat mass and obesity associated(FTO) gene and obesity is becoming the hot topic in the gene research. FTO is a member of the nonheme dioxygenase [Fe(Ⅱ) and 2-oxoglutarate-dependent dioxygenases]superfamily. FTO levels in the brain,especially in the hypothalamus,participate in the central control of food intake. The mutation of FTO gene accentuates body mass index,hip circumference and total weight,leading to obesity.
3.Effects of Extract 5F from Pteris Semipinnata L. on the Apoptosis of Skin Fibroblasts in Patients with Systemic Scleroderma

Objective To study the effects of extract 5F from Pteris semipinnata L. on the apoptosis of skin fibroblasts in patients with systemic sc leroderma (SSc) and the possible mechanism of its inhibition on the proliferatio n of skin fibroblasts. Methods The cultured SSc skin fibroblasts were used as an experimental model, and the apoptosis of SSc skin fibroblasts before and afte r the extract 5F stimulation was detected by TUNEL assay and fluorescent microsc opy. Results The apoptosis of SSc skin fibroblasts was induced by the extract 5F, and the apoptosis was enhanced along with the increase of the concentration and incubation time of this compound within definite limits. Conclusions These results indicate that the extract 5F can induce the apoptosis of SSc skin fibro blasts, and significantly inhibit the proliferation and collagen synthesis in th ese cells.
4.Determination of thiodiglycolic acid in urine with gas chromatography-mass spectrometer.

OBJECTIVETo establish a rapid determination method with gas chromatography-mass spectrometer (GC-MS) for thiodiglycolic acid (TDGA), a vinyl chloride (VCM) biomarker.
METHODSA high- sensitivity determination method was established using a moderate methyl esterification instead of methyl esterification of highly toxic diazo reaction.
RESULTSThe standard curve regression linear equation of the method was: y=8460.5x-4758.2, the linear coefficient was 0.999 7, the minimum quantity concentration was 2.0 µg/L, the range of precision value was 0.81%-2.38%, and the average recovery of standard addition was 99.0%-102.9%.
CONCLUSIONThis method reduces the risk of traditional methyl esterification, improves the determination sensitivity compared with the GC-FPD method, and meets the determination requirement of TDGA.

5.Concordance of endoscopic grading and scoring systems for inflammatory bowel diseases

Objective To assess the agreement of different endoscopy grading or scoring systems for inflammatory bowel diseases(IBD)including ulcerative colitis(UC)and Crohn's disease(CD).Methods A standardized table was prepared based on the searches for endoscopic grading or scoring systems on Medline and Chinese Biomedical Database,the data of 80 patients with UC and 31 with CD.who underwent colonoscopy in Shanghai Renji hospital from June 2006 to February 2007,were evaluated with each system by two physicians independently.Data were analyzed with SPSS 13.0.Results Six endoscopic grading and scoring systems of UC and three of CD were included for evaluation.For the systems of UC and CD,Kendall's coefficients of concordance were 0.71(P<0.01)and 0.34(P<0.01),respectively.There was no significant differenee between every two systems for UC.Nonetheless.Spearman's correlation coefficient between Chinese Grading System of Crohn's Disease(CGSCD)and Crohn's Disease Endoscopic Index of Severity(CDEIS)was 0.32(P=0.08).Significant differences in frequencies were detected in endoscopic systems for UC by Kruskal Wallis test(P<0.01).Conclusion There is satisfactory concordance among the endoscopic grading and scoring systems of UC,while CGSCD needs further improvement.Furthermore,Jeroen elassifieation inclines to severe category,while modified Baron scale tends to be a mild one.
6.Cloning and expression of the key enzyme hyoscyamine 6 beta-hydroxylase gene (DaH6H) in scopolamine biosynthesis of Datura arborea.

Hyoscyamine 6 beta-hydroxylase (H6H) is the last rate-limiting enzyme directly catalyzing the formation of scopolamine in tropane alkaloids (TAs) biosynthesis pathway. It is the primary target gene in the genetic modification of TAs metabolic pathway. Full-length cDNA and gDNA sequences of a novel H6H gene were cloned from Datura arborea (DaH6H, GenBank accession numbers for cDNA and gDNA are KR006981 and KR006983, respectively). Nucleotide sequence analysis reveals an open reading frame of 1375 bp encoding 347 amino acids in the cDNA of DaH6H, while the gDNA of DaH6H contains four exons and three introns, with the highest similarity to the gDNA of H6H from D. stramonium. DaH6H also exhibited the most identity of 90.5% with DsH6H in amino acids and harbored conserved 2-oxoglutarate binding motif and two iron binding motifs. The expression level of DaH6H was highest in the mature leaf, followed by the secondary root, and with no expression in the primary root based on qPCR analysis. Its expression was inhibited by MeJA. DaH6H was expressed in E. coli and a 39 kD recombinant protein was detected in SDS-PAGE. Comparison of the contents of scopolamine and hyoscyamine in various TAs-producing plants revealed that D. arborea was one of the rare scopolamine predominant plants. Cloning of DaH6H gene will allow more research in the molecular regulatory mechanism of TAs biosynthesis in distinct plants and provide a new candidate gene for scopolamine metabolic engineering.
7.The differential diagnostic value of high-frequency ultrasonography in infantile cholestatic hepatopathy

Objective To evaluate the differential diagnostic value and superior of biliary atresia(BA)in the infants with cholestatic hepatopathy by high-frequency ultrasonography (HUS).Methods After 4 hours fasting,124 infants with cholestatic hepatopathy were scanned with high-frequency US.The data of hepatic size and parenchyma,gallbladder,triangular cord (TC) sign,bile duct,right hepatic artery (RHA)and portal vein (PV) were observed and measured.Meanwhile,the other data were collected,which included the clinical diagnosis,blood biochemical tests,the MRCP and dynamic duodenal liquid color check finding,the pathological results after liver puncture biopsy and so on.Results In 124 infants with cholestatic hepatopathy,BA was found in 61 infants and ruled out in 63.TC thickness,RHA diameter,and gallbladder length and width exhibited significant differences between the group with BA and the group non-BA(all P <0.001).The correctness for the diagnosis of BA was 90.3% by the combination of TC sign and abnormal gallbladder morphology,and 83.1 % by stool color,81.5 % by γ-GT,47.5 % by MRCP,83.3 % by dynamic duodenal liquid color check,95.2% by the pathology after liver puncture biopsy,respectively.Conclusions HUS is superior to other diagnostic methods in BA with higher accuracy rate,noninvasion,simplicity and economy.
8.Evaluation of non-invasive ventilation in the treatment of chronic obstructive pulmonary disease complicated with typeⅡ respiratory failure

Objective To investigate the clinical significance of BiPAP mechanical ventilation in the treatment of chronic obstructive pulmonary disease(COPD)with type Ⅱ respiratory failure.Methods A total 56 patients suffering from respiratory failure due to exacerbation of COPD were randomly divided into treatment group (BiPAP plus routine therapy)and control group(routine comprehensive therapy).Blood gases,respiratory rates,heart rates,scores of the activity of accessory respiratory muscles,incubation rate,fatality rate were observed.Results In the treatment group,PO2 greatly increased,PCO2 greatly decreased,pH improved significantly,and respiratory rate,heart rate and scores of the activity of accessory respiratory muscles greatly decreased.The incubation rate and the fatality rate were significantly lower in the treatment group than those in the control group(P<0.05).Conclusion BiPAP mechanical ventilation plus routine therapy on COPD with type Ⅱ respiratory failure can improve the curative effects,lessen respiratory muscle fatigue,and decrease the need for incubation and fatality rate.
9.Drug-induced Edema:Literature Analysis of 173 Cases

OBJECTIVE:To discuss the general regularity and characteristics of the drug-induced edema so as to provide reference for clinical safe medication.METHODS:173 drug-induced edema cases publicly reported in domestic journal from 2001 to 2006 were analyzed statistically.RESULTS:The drug-induced edema was more often seen in males than in females,but not linked to age.It was characterized by early onset,with 76(43.9%)showing edema within 1h.It was chiefly induced vial oral and intravenous routes.The drug-induced edema was caused by 17 drug categories(or 115 kinds),with antibacterials showing the highest percentage(31.2%,54 cases).Laryngeal edema,which occurred in 57 cases,was the chief type among all the edema cases.CONCLUSION:Drug-induced edema is a common adverse drug reaction,which should be given great attention in the clinic,much as in the case of acute laryngeal edema.Clinicians should give close monitoring on patients after medication to guard against possible events.
10.Inhibitory Effect of Acupuncture Combined with Herbal Medicine on Hyperplasia of Mammary Glands in Rats

[Objective] To observe the inhibitory effect of acupuncture combined with herbal medicine on hyperplasia of mammary glands in rats. [Methods] Rats were randomized into 6 groups: A (normal) , B (model) , C (acupuncture) , D ( herbal medicine), E (acupuncture combined with herbal medicine) and F (triphenyloxamine) . Rat models of hyperplasia of mammary glands in Group B, C, D, E and F were established by intramuscular injection of estradial benzoate and progesterone. Nipple height in rats were detected before and after treatment. The number of acini in each lobule, the diameter of acinar cavity and duct in the second pair of mammary glands were calculated under light microscope. The expression of estrogen receptor (ER) and progesterone receptor (PR) in the tissues of mammary gland were examined by immunohistochemical method after treatment.
