Objectives: The incidence of cervical carcinoma is increasing in young females.More and more clinical researches focus on the choice of therapeutic methods and improvement of the patients' survival.This study aimed to evaluate the efficacy of preoperative concurrent chemoradiation for stage Ⅰb-Ⅱb high-risk cervical carcinoma in young females.Methods: We retrospectively analyzed the clinical data of 118 female patients(≤35 years) with stage Ⅰb-Ⅱb high-risk cervical carcinoma,bulky,low-differentiation or non-squamous.According to preoperative treatments,the patients were divided into three groups,Group 1 preoperatively treated by concurrent chemoradiation,Group 2 by neoadjuvant chemotherapy and Group 3 by radiotherapy(intracavitary irradiation),all followed by radical hysterectomy.And those with pathologically reported pelvic lymph node metastasis,deep cervical stromal invasion,vascular tumor thrombosis,or surgical margin positivity received postoperative chemotherapy and/or radiotherapy.Results: The disappearance rate of cancer cells and the rate of superficial myometrial invasion were significantly higher in Group 1 than in the other two groups(P
2.Pacilitaxel in association with gemcitabine for anthracycine-resistant advanced breast cancer

Objective To evaluate the effect and safety of combined therapy of paclitaxel and gemcitabine for anthracycine(ANT)-resistant advanced breast cancer(ABC).MethodsFrom May 2000 to Aug 2004,twenty six patients with ANT-resistant ABC were treated with this combined regime.The median chemotherapy cycles were 2.5(range from 2 to 3 cycles).ResultsOf 26 patients, there were 3 complete(11.5%) and 11 partial(42.3%) responses for an overall response rate of 53.8%. Eight cases remained stable (30.8%) and 4 progressive (15.4%). The median survival time was 18 months. The median time to progression was 7 months. The main toxic reaction included bone marrow depression, liver function damage, alopecia, mucositis and peripheral neurotoxicity. ConclusionsCombined medication of gemcitabine and paclitaxel is effective in therapy of ANT-resistant advanced breast cancer with acceptable toxicity.
3.Effects of Qingfeitang on ET-1 and PGE2 in exhaled breath condensate and serum of patients with ARDS

Objective To observe the changes of endothelin-1 (ET-1) and prostaglandin E2 (PGE2) in exhaled breath condensate (EBC) and serum of patients with acute respiratory distress syndrome (ARDS) after treated by Qingfeitang and investigate its clinical value. Methods 52 ARDS patients receiving mechanical ventilation at intensive care unit (ICU) were divided into the Qingfeitang treatment group and the control group, with 26 cases in each group. The EBC were collected by Ecoscreen condenser within 24 h after diagnosis of ARDS and on the 5th day of medication, and the venous blood were collected at the same time. The levels of ET-1 and PGE2 in the EBC and serum of different period were measured by EIA. Results (1) After treatment, The levels of ET-1 in EBC and serum of the Qingfeitang treatment group were significantly lower than those of the control group. (2) After treatment, the levels of PGE2 in serum of the Qingfeitang treatment group were significantly lower than that of the control group. (3) The oxygenation index difference before and after treatment of Qingfeitang in the treatment group was higher than in the control group. (4) The duration of mechanical ventilation of the Qingfeitang treatment group was significantly less than that of the control group. Conclusions Qingfeitang could be an effective method for alleviating acute respiratory distress syndrome.
4.Construction, expression and in vitro biological behaviors of Ig scFv fragment in patients with chronic B cell leukemia.

The expression vector of SmIg scFv fragment was constructed in patient with B cell chronic lymphocyte leukemia (B-CLL) and expressed in E. coli to obtain scFv fragment, and the effect of the protein on the proliferation of stimulated peripheral blood mononuclear cells (PBMC) was investigated in vitro. Two pairs of primers were designed, and variable region genes of light chain and heavy chain were amplified by PCR respectively from the pGEM-T vectors previously constructed in our laboratory which containing light chain gene or Fd fragment of heavy chain gene. The PCR product was digested, purified and inserted into pHEN2 vector to construct the soluble expression vector pHEN2-scFv. After the induction by IPTG, the scFv protein was identified by SDS-PAGE electrophoresis and purified by Ni-NTA-Chromatography. MTT was used to determine the effect of purified protein on the proliferation of stimulated PBMC in vitro. Plasmid PCR and restriction enzyme digestion of pHEN2-scFv revealed the pHEN2-scFv vector was constructed successfully. Id-scFv protein was expressed in positive clone after induced by IPTG. SDS-PAGE analysis showed that the relative molecular weight of fusion protein was about 30 kD (1 kD= 0.9921 ku), which was consistent with the theoretically predicted value. Proliferation of PBMC could be induced by purified Id-scFv. It was suggested that the expression vector of SmIg scFv fragment was constructed successfully, and scFv protein was expressed and secreted from E. coli, which could induce proliferation of PBMC. This may lay an experimental foundation for further research of Id-HSP complex vaccine for B-CLL.

6.Study on glycosaminoglycan from Apostichopus Japonius on cellular immuno-regulation function of peripheral blood cells from pulmonary tuberculosis patients in vitro

Objective To investigate the effects of glycosaminoglycans (HGAG) on the immune function of peripheral blood cells from patients with pulmonary tuberculosis.Methods Peripheral blood monouclear cells (PBMC) were isolated from peripheral blood of 40 healthy people (healthy group) and 30 tuberculosis patients (tuberculosis group) and cocultured with HGAG in vitro for 24 hours.Flow cytometry was used to detect the expression of CD45RA and CD45RO,as well as the expression of CD1a and CD83.Results The results showed that the expression of CD45RA and CD45RO in the tuberculosis group was the most significant (P < 0.05) at the concentration of 50 μg/m coculturing with HGAG.The expression of CD45RA and CD45RO were most obvious in the healthy group at the concentration of 10 μg/ml and 50 μg/ml respectively (P <0.001).The difference of CD45RA between the two groups was no significant (P >0.05),while the difference of CD45RO was statistically significant (P < 0.01) before co-culturing.The expression of CD45RA and CD45RO at 10 μg/ml and 50 μg/ml after co-culturing with HGAG were statistically significant (P < 0.05).There was no statistical difference in CD1a and CD83 in healthy group before and after co-culturing (P > 0.05),while there was statistically difference (P < 0.05) before and after culturing in tuberculosis group.Before co-culturing,there was no significant difference in the expression of CD1a between the healthy group and the tuberculosis group (P > 0.05),but CD83 expression was statistically different (P < 0.001).After co-culturing,there were no significant differences in CD1a and CD83 expression between healthy and healthy groups (P > 0.05).Conclusions HGAG can down-regulate the expression of CD45RA and up-regulate the expression of CD45RO in a certain concentration range,and promote the maturation of dendritic cells (DC) in tuberculosis patients and regulate the cellular immunity of patients with pulmonary tuberculosis in vitro.
7. Expression and significance of tight junction protein 3 in colorectal cancer

Objective:
To analyze the expression of tight junction protein 3(claudin-3) in colorectal cancer and its relationship with the development, metastasis and prognosis of colorectal cancer.
Methods:
From February 2013 to February 2015, 78 patients with colorectal cancer operated in the People's Hospital of Sanmen County were selected in this study.The tissues of colorectal cancer and adjacent tissues were collected and claudin-3 expression was detected.The relationship between claudin-3 and clinicopathological features of colorectal cancer was analyzed.
Results:

The expression vector of SmIg scFv fragment was constructed in patient with B cell chronic lymphocyte leukemia (B-CLL) and expressed in E. coli to obtain scFv fragment, and the effect of the protein on the proliferation of stimulated peripheral blood mononuclear cells (PBMC) was investigated in vitro. 9.Methylation of RAR-β2, RASSF1A, and CDKN2A genes induced by nickel subsulfide and nickel-carcinogenesis in rats.

OBJECTIVETo investigate the expression variation of RAR-β2, RASSF1A, and CDKN2A gene in the process of nickel-induced carcinogenesis.
METHODSNickel subsulfide (Ni(3)S(2)) at dose of 10 mg was given to Wistar rats by intramuscular injection. The mRNA expression of the three genes in induced tumors and their lung metastasis were examined by Real-time PCR. The methylation status of the 5' region of these genes were detected by Quantitative Real-time methylation specific PCR.
RESULTSThe mRNA expressions of the three genes both in muscle and lung tumor were decreased distinctly in comparison with normal tissue. But hypermethylation was found only in muscle tumor.
CONCLUSIONThese findings suggest that loss of function or decrease of RAR-β2, RASSF1A, and CDKN2A, as well as the hypermethylation of 5' region of these genes, are related with nickel exposure.

10.Epidemiological investigation on chronic kidney disease and its main risk factors among the elderly in Tianmen, Hubei

Objective To investigate the epidemiological characteristics and main risk factors of chronic kidney disease in the elderly in Tianmen, Hubei Province. Methods A descriptive method was adopted in this study. Using stratified sampling, a total of 720 local residents over 65 years old in Tianmen of Hubei Province were selected from January 2017 to January 2020 as the research subjects. The general information of patients with chronic kidney disease was compared, and the difference in the characteristics of patients with different disease progression was compared. The risk factors for chronic kidney disease and disease progression were analyzed. Results Through the diagnosis of chronic kidney disease, a total of 82 patients with chronic kidney disease were found, including 33 patients in stage I, 26 patients in stage II and 23 patients in stage III. As the disease progressed, the patient's age, serum uric acid, triglyceride, total cholesterol, fasting blood glucose, serum creatinine, and the incidence of chronic diseases all increased significantly. The results of correlation analysis showed that the incidence of chronic kidney disease and the progression of the disease were positively correlated with age, body mass index, serum uric acid, triglyceride, total cholesterol, fasting blood glucose, serum creatinine level, diabetes, hypertension, and dyslipidemia, which were independent risk factors for chronic kidney disease. Conclusion The prevalence of chronic kidney disease in Tianmen was 11.39%. Higher age, serum uric acid, blood lipid, blood pressure and blood glucose levels are all important factors for the progression of chronic kidney disease.
