1.Antibiotics Application in Liver Transplanting Patients

OBJECTIVE To analyze the application of antibacterials in liver transplanted patients of our hospital in order to improve the rational use of antibacterials in perioperative period of liver transplantation.METHODS According to the criteria of DDD and DUI recommended by WHO,a retrospective study of the application of antibacterials in 82 patients with liver transplantation who discharged hospital during from Jun 2003 to Jun 2005 was made.RESULTS Among the 82 patients,100.0% patients had received antibacterials and 48 patients received combined medication,in which 45.10% used two kinds and 13.41% used three kinds.The longest medication time was 49 days while the shortest was 10 days.Nineteen antibacterials′ DUI were all less than one except meropenem whose DUI was 1.03.CONCLUSIONS This study proved that patients with liver transplantation in our hospital received rational antibacterials.
2.Separation of phospholipid in bear gall by Micellar electrokinetic capillary chromatography

AIM: To develop micellar electrokinetic capilitary chromatography. A method MECC for the separation of major phospholipid from bear gall. METHODS: Following the optimization experiments the MECC was applied to the separation with the 35 m MSDC, 6 mM sodium tetraborate, 10 mM disodium hydrogen phosphate, 30% (v/v) 1-propanol, and pH 8.5 as buffer system. The voltage was 30 kV. the temperature was set at 40 (?C). The capillary length was 570 mm with the effective one of 500 mm. Injection pressure was 50 mbar?4 s and UV detection wavelength was at 200 nm. RESULTS: The separation of phospholipid in bear gall was achieved in the method developed. CONCLUSION: The method is fast, effective and complementary to the HPLC.
3.Effect of RNA silencing of transforming growth factor ?1 on the growth of colon cancer cells by RNA interference

Objectives To observe the effect of inhibition of transforming growth factor (TGF)?1 on the growth and aggression of colon cancer cells by RNA interference. Methods HCT116 colon cancer cells were transfected with small interfering dsRNA (siRNA), the effect of the inhibition of TGF?1 was tested by semi-quantitative RT-PCR, and changes in cell cycle distribution were analyzed by flow cylometry, the ability of aggression was detected by soft agar colony assay. Results The inhibition of the expression of TGF?1 was significantly detected after the transfection of siRNA against TGF?1, which induced the increase of S phase by 59.0%-82. 5% and decrease in G2 phase, colony assay further demonstrated that siRNA against TGF?1 led to a significant reduction in colony formation as compared with the control group. Conclusions The present data suggested that the expression of TGF?1 was conveniently and rapidly blocked by siRNA transfection, and this may lead to the cell cycle arrest and inhibition of the growth and tumorigenesis of colon cells.
4.Role of Th17 Cells and Intestinal Flora in Pathogenesis of Inflammatory Bowel Disease

Inflammatory bowel disease(IBD)is an autoimmune disease and its etiology has not yet been clarified. Dysregulated immune responses resulted from complex interactions among genetic factors,intestinal flora and environmental cues have been considered as the etiology of IBD. Recently,the relationship between Th17 cells and IBD has become a hotspot of study,and more and more studies showed that Th17 cells and their related cytokines regulated by intestinal flora contributed to the pathogenesis of IBD. This article reviewed the role of Th17 cells and intestinal flora in the pathogenesis of IBD.

6.Determination of phthalate esters in physiological saline solution by monolithic silica spin column extraction method

Monolithic silica spin column extraction (MonoSpin-SPE) was developed as a simple,sensitive,and eco-friendly pretreatment method which combined with ultra-fast liquid chromatography-mass spectrometry (UFLC-MS) to determine the levels of six phthalate esters,dimethyl-(DMP),diethyl-(DEP),dipropyl-[DPrP],butyl-benzyl-(BBP),dicyclohexyl(DcHP),and di-n-octyl-(DOP) phthalate in physiological saline samples.Under optimized experimental conditions,the method was linear in the following ranges:0.2 - 50 μg/L for DMP,DEP,DPrP,DcHP and DOP; 5- 100,μg/L for BBP.The correlation coefficients (R2 ) were in the range of 0.9951- 0.9995 for all the analytes and the limits of detection (LODs) and limits of quantification (LOQs) were in the ranges of 0.02- 0.9 μg/L and 0.08- 2.7 μg/L,respectively.The pretreatment process showed good reproducibility with inter-day and intra-day relative standard deviations (RSDs) below 8.5% and 11.2%,respectively.This method was used to determine the levels of six phthalate esters in physiological saline samples and the recoveries ranged from 71.2% to 107.3%.DMP and DEP were found in actual physical saline samples (brand A and brand B).

8.Improvement of determination method of total anthraquinone in Rhei Radix et Rhizoma in Pharmacopoeia of the People’s Republic of China

Objective:To improve the determination method for the total anthraquinone of the Rhei Radix et Rhizoma in the Pharmacopoeia of the People’s Republic of China, and compare this method with the method in the pharmacopoeia to determine the feasibility of such method. Methods:By changing the determination of total anthraquinone from biphasic hydrolysis to monophase hydrolysis, the method included in the pharmacopoeia was improved to determine the total anthraquinone content in Rhei Radix et Rhizoma. Chromatographic conditions were Symmetry C18 (4.6 mm × 250 mm, 5 μm) chromatographic column; the mobile phase is methanol-0.1% phosphoric acid water (85:15); the flow rate was 1 ml/min; the column temperature is 30 ℃; the detection wavelength is 254 nm. Results:The concentrations of aloe-emodin, rhein, emodin, chrysophanol, physcion in the range of 0.003 3-0.332 0 μg, 0.006 9-0.668 0 μg, 0.002 3-0.232 0 μg, 0.010 4-1.040 0 μg, 0.008 4-0.836 0 μg have good linear relationship with the peak area; RSDs of precision, stability and repeatability were less than 2%; the recovery rates of aloe-emodin, rhein, emodin, chrysophanol and physcion were 101.50%, 99.30%, 99.62%, 101.57%, and 103.11%, and the RSDs were less than 2%. Conclusion:The improvement method is simple, accurate, reliable and reproducible, which could be used for the quality control of Rhei Radix et Rhizoma.
9.Pathogen Distribution and Antimicrobial Resistance after Liver Transplantation

OBJECTIVE To evaluate the distribution of pathogens causing infection and current status of antimicrobial resistance in order to offer experimental data for clinical administration.METHODS The resistance tests were performed using K-B method.WHONET5.2 and EXCEL were used to analyze bacteria distribution and resistance.RESULTS From 119 cases of patients were earned the routine bacteria culture results,the tested rate was 96.75%,the positive specimens were 379 and the postitive rate was 83.85%.The strains of Pseudomonas aeruginosa were the most frequent organisms isolated then were the strains of Streptococcus pyogenes,coagulase-negative Staphylococcus,Nesseria spp,and Stenotrophomonas maltophilia.CONCLUSIONS It is important to strengthen antibiotics administration,bacteria isolation and resistance surveillance.
10.Inhibitory Effect of Acupuncture Combined with Herbal Medicine on Hyperplasia of Mammary Glands in Rats

[Objective] To observe the inhibitory effect of acupuncture combined with herbal medicine on hyperplasia of mammary glands in rats. [Methods] Rats were randomized into 6 groups: A (normal) , B (model) , C (acupuncture) , D ( herbal medicine), E (acupuncture combined with herbal medicine) and F (triphenyloxamine) . Rat models of hyperplasia of mammary glands in Group B, C, D, E and F were established by intramuscular injection of estradial benzoate and progesterone. Nipple height in rats were detected before and after treatment. The number of acini in each lobule, the diameter of acinar cavity and duct in the second pair of mammary glands were calculated under light microscope. The expression of estrogen receptor (ER) and progesterone receptor (PR) in the tissues of mammary gland were examined by immunohistochemical method after treatment.
