1.The targeting expression of IL-2 gene driven by alpha fetoprotein promoter and hepatitis B virus envelope in hepatic carcinoma cell

Objective To observe if IL-2 gene can express in hepatic carcinoma cells by double targeting of hepatitis B virus envelope (HBVE) and alpha fetoprotein (AFP) promoter.Methods HepG2 cells,L02 cells,and HepG2.2.15 cells were cultured in vitro.HBVE was obtained by PEG8000 concentration assay,and the acquired HBVE was used to pack recombintional gene.Human AFP promoter-IL-2 recombinational gene was obtained by PCR.Then HepG2 cells and L02 cells were transfected by transient transfection and the expression of IL-2 was detected by RT-PCR and Western blot.Results IL-2 was detected in HepG2 cells by RT-PCR and Western blot but not in L02 cells.Conclusion By using HBVE as a gene transporter,human AFP promoter-IL-2 recombinational gene can express in hepatic carcinoma cells,thereby it can increase the safety of exogenous gene transfection of hepatic carcinoma cells.
2.Clinical observation on hepatocirrhosis of Child-Pugh B treated with Fufangbiejiasan for three years

To observe the therapeutic e ects of Fufangbiejiasan on patients with hepatocirrhosis of Child-Pugh B.Methods:Eighty-eight patients with hepatocirrhosis of Child-Pugh B,Chronic Hepatitis B were enrolled,and divided into two groups,treated with Fufangbiejiasan and Binglian Jianganlin capsule respectively.The therapeutic course was 3 years for both groups.Liver function,HBV.DNA,AFP and CT/MRI were examined at 6th,12th,24th and 36th month after treatment.Results:Levels of blood albumin were markedly increased in Fufangbiejiasan group after treatment at 6th,12th,36th month(38.5?2.1),(39.1 ?3.9),(40.8?3.4)g/L while those in Binglian Jianganlin group were reduced(28.9?1.6),(28.8?1.7),(25.6?1.5)g/L.Level of PTA in Fufangbiejiasan group was signi cantly higher than those in controls;Compared with controls,Fufangbiejiasan treatment was more e ective :34(77.3),41(93.2),42(95.5) and 0(0),0(0),0(0)(P
3.Significance of tamoxifen (TAM) in the combination chemotherapy of human multidrug-resistant bile duet carcinoma cell line

Objective To investigate the significance of tamoxifen in the combination chemotherapy of human multidrug-resistant bile duct carcinoma cell line (QBC939/ADM) and its mechanism. Methods The QBC939/AI)M was established, through exposure to gradually increased and the high and low alternated concentration of ADM persistently. The QBC939 and the QBC939/ADM were effected by single ADM, MMC, VDS or jointly with TAM. Drug sensitivity was measured by MTT. Growth cycle and apoptosis were performed by FCM. The level of their P-gp was detected by IHC. The content of ADM in the human cholangiocarcinoma cell line QBC939 was observed by FCM. Results The inhibitive rate of ADM, MMC, VDS to QBC939/ADM was rather lower than to QBC939. With the use of TAM, their chemotherapy effects were apparently enhanced and the apoptosis ratio increased comparably. The IHC results showed that the level of P-gp on the QBC939/ ADM was overexpressed, and the content of ADM in the QBC939/ADM group was much lower. Effected with the high content of TAM(10 μmol/L), the level of P-gp on the QBC939/ADM was decreased, with the content of ADM in the QBC939/ADM group increased comparably. TAM could both improve the chemotherapy effects of the two types of the cell, but the effect of the QBC939/ADM group was more apparent. Conclusion TAM can enhance the depressant effect of chemotherapy to both the two types of the cell, and increase the content of ADM in the QBC939/ADM group. TAM can be combined with the overexpressed P-gp.
4.The Correlation Between Mood Disorders and Nerve Growth Factor Receptor Expression in Lesions of Psoriasis

Objective To investigate the molecular mechanism of mood disorders in psoriatic patients.Methods The expression of nerve growth factor receptor p75(NGFR p75)and p140trkA were detected by immunohistochemical technique.The patients' mood was measured with self-rating anxiety scale(SAS)and self-rating depression scale(SDS).Results There is a positive correlation of the expressions of NGFR p75and p140trkA with the patients' mood disorders in psoriatic patients.Conclusion NGF and its receptors are the important substance served as a mediator of the mood disorders affecting psoriatic patients' condition.
5.Determination of catechins and caffeine in tea polyphenols

Objective: To develop a HPLC method for simultaneous determination of catechins (EGC、DC、EGCG、EC、GCG、ECG) and caffeine in tea polyphenols and to quantify seven ingredients in twenty nine batches of tea polyphenols samples from thirteen producers. Methods: The study was achieved using a C 18 column with a methanol water 0.1% formic acid gradient elution system. Results: The peak resolutions of seven ingredients in mixed standards were all above 2; The resolutions in samples were all above 1.5 except EGCG; The average recoveries were 98.56%~100.14%. Conclusion: This method is accurate, stable, reproducible and suitable for quality control of tea polyphenols; the variations of contents are obvious among tea polyphenols samples from different even the same producers.
6.Application of overlapping ratio to fingerprints of green tea and tea polyphenols

AIM: To discuss the application of overlapping ratio to the fingerprints of plant and plant extract. METHODS: We took the HPLC fingerprints of green tea and tea polyphenols for decision procedure. RESULTS: Overlapping ratio could be applied to selecting representative samples for characteristic fingerprints of green tea and tea polyphenols,and could be used to deduce the raw material,process and producer of tea polyphenols.(CONCLUSION:) Overlapping ratio can be used to choose the suitable samples for establishing the characteristic fingerprint spectrum,and can supply references to relationship investigation among products,raw material and process.

9.Bamboo-circled Salt-partitioned Moxibustion for Arthritis of Temporomandibular Joint: A Randomized Controlled Clinical Trial

Objective To observe the clinical efficacy of bamboo-circled salt-partitioned moxibustion in treating arthritis of temporomandibular joint.Method Eighty patients were randomized into two groups. Forty cases in the bamboo-circled salt-partitioned moxibustion group received bamboo-circled salt-partitioned moxibustion at temporomandibular joint; forty cases in the warm needling group were intervened by selecting Xiaguan (ST7), Ashi point, etc. at the affected side. For the two groups, 3-day treatment was taken as a treatment course, and the therapeutic efficacy was analyzed after 2 treatment courses. The improvements in pain and mouth opening were observed before and after the treatment, and the treatment efficacy was evaluated by a 1-month follow-up study.Result The bamboo-circled salt-partitioned moxibustion group was superior to the warm needling group in comparing the real-time analgesic effect (P<0.05) and in the improvement of mouth opening (P<0.05); the comprehensive markedly effective rate was respectively 67.5% and 45.0% in the bamboo-circled salt-partitioned moxibustion group and warm needling group, and the between-group difference was statistically significant (P<0.05), indicating that bamboo-circled salt-partitioned moxibustion is better than warm needling in treating arthritis of temporomandibular joint; the follow-up study revealed satisfactory therapeutic efficacies in both groups: the effective rate was 92.5% in the bamboo-circled salt-partitioned moxibustion group versus 87.5% in the warm needling group, and the difference was statistically insignificant (P>0.05).Conclusion Bamboo-circled salt-partitioned moxibustion can produce a real-time analgesic effect and improve mouth opening; it's especially suitable to treat the patients who are afraid of needling, as it's significantly effective, safe, non-invasive,and easy-to-operate.
10.Hepatic histological changes and its influencing factors of HBeAg negative chronic hepatitis B patients

Objective To investigate hepatic histological features and its influencing factors of HBeAg-negative chronic hepatitis B patients. Methods 134 HBeAg-negative chronic hepatitis B (CHB) patients who underwent percutaneous liver biopsy were recruited in this study. The liver biopsy sections were examined after routine haematoxylin-eosin (HE) staining, and silver staining for assessment of fibrosis. The activity of liver disease was assessed by using a modified Knodell numeric histology activity index (HAI). ALT level, HBV DNA load, HBV serological markers, HBV genotype were assessed with appropriate methods. t test or analysis of variance was used to compare means. Non-parametric was done by Kruskal-Wallis test. The correlation between liver pathological change and clinical factors was analyzed by multivariate linear regression. Results Of 134 HBeAg negative CHB patients, percentages of mild (HAI 4 ~ 8), moderate (HAI 9 ~ 12), and severe hepatitis (HAI 13 ~ 18) were 26.9%, 26.1%, and 47.0%, respectively. As for hepatic fibrosis, 18.7% and 81.3% of the patients had fibrosis score < 3 and ≥3, respectively. Multivariate regression analysis showed that ALT level and hepatic fibrosis were correlated to hepatic inflammation (t = 6.687,P < 0.01; t = 3.478, P < 0.01) while age and hepatic inflammation activity were influencing factors of hepatic fibrosis (t = 3.587, P < 0.01; t =7.136, P < 0.01). However, correlation is not significant between hepatic histological change and other factors, including gender, HBVDNA, HBV genotype and HBeAb status. Conclusions In this study, hepatic histological change tend to become worse in majority of HBeAg-negative chronic hepatitis B , especially in older patients and those with high ALT level.
