1.Establishing a rabbit model of peripheral partial growth plate closure by immediate filling after partial growth plate resection

BACKGROUND: Animal models of peripheral partial epiphyseal plate closure are commonly used in related trials.OBJECTIVE: To modify the original animal models of distal femoral partial epiphyseal plate closure.DESIGN, TIME AND SETTING: A self-controlled trial was performed at the Experimental Animal Center of General Hospital of Chinese PLA from March to October 2007.MATERIALS: Twenty healthy New Zealand rabbits of 4-6 weeks old weighing 1.0-1.5 kg were selected.METHODS: A straight incision about 2 cm was made in lateral knee of 20 rabbits. The subcutaneous tissues and knee capsule were cut open to expose distal femoral condyle. The condylar plate line was clear. Two holes were drilled in the fight lateral condyle, about 3.0-4.0 mm deep, and partial normal epiphyseal plate (about 33%-44% of the total growth plate) was removed. An incision deep to periosteum was created in the left side and regarded as self-control.MAIN OUTCOME MEASURES: Femoral length, valgus angle of the distal femur, and femoral deformity were detected by X-ray photograph, and gross observation. Bone bridge formation of bilateral femora was observed by histological examination.RESULTS: One rabbit died of diarrhea 31 days after surgery, and 19 were included in final analysis. ①Radiographs showed that the mean length of left femur (control side) was significantly longer but valgus angle was significantly smaller than the right side (experimental side) 4 months after surgery (P < 0.01). The left distal femur averagely grew for 2.27 cm,accounted for 57.6% of the overall femoral growth, and the right side averagely grew for 0.45 cm, accounted for 21.4% of the overall femoral growth.②Four months after surgery, femoral appearance of the control side was normal, with smooth and complete condylar and facies articularis patellae. While, the experimental side femur was significantly shortened, and severe vaigus deformity was observed in the distal femur. ③Five weeks after surgery, layer cells of the epiphyseal plate arranged regularly in the control side. Bone trabecula with many fibers was found in bone defect cavity of the experimental side, and the cell layer of the remnant epiphyseal plate was decreased. The epiphyseal plate was closed in the control side,and bone trabecula of the experimental side was thickened 10 weeks postoperatively.CONCLUSION: This modeling method is simple, efficient, and easy to manipulate; in addition, the bone bridge area is controllable. The models can meet the study demands for peripheral partial epiphyseal plate closure, and are significant for filling material selection during treatment.
2.The diagnostic value of ultrasound for the residual gallbladder

Objective To explore the diagnostic value of ultrasonography for the residual gallbladder after cholecystectomy.Methods Application of the ultrasound diagnostic apparatus in cholecystectomy patients was to be screened to find the residual gallbladder.Results Ultrasound resuts showed 18 cases of residual gallbladder,which combined stones in 12 cases(66.7% ),polyps in 1 case(5.6% ) ;Atrophic cholecystitis,acute cholecystitis,postoperative residual gallbladder probability were high,accounting for 83.0%.Conclusion Ultrasound for the preoperative prevention and postoperative diagnosis of the residual gallbladder has better value,and the cholecystectomy patients should be routinely review the ultrasound.
3.Influence of arterial hemostasis with three tourniquets on venous blood gas in dogs

Objective To observe changes of venous blood gas when applying tourniquets to arterial bleeding dog models and compare the injury of the bundled tissues caused by three different tourniquets. Methods A total of 18 dogs were randomly divided to three groups and made into femoral artery bleeding model of the hind limbs. Then, rubber tubing, half-hitch tourniquet and multi-function pressure-control bandage (hemostatic bandage group) were employed respectively for femoral artery hemostasis in three groups. The pressure produced by the bandage and femoral vein blood gas indices were measured at 0.5, 1, 3 and 6 hours respectively after tourniquet application. Results The pressure produced by the hemostatic bandage group was 0.74 kg, which was obviously lower than 1.46 kg in the rubber tubing group and 2.09 kg in the half-hitch tourniquet group ( P < 0.05 ). At one hour after hemostasis, PvO2 was (36.45 ± 10.45 )mm Hg in the hemostatic bandage group and (28.73 ± 9.07 )mm Hg in the rubber tubing group, which was significantly higher than (21.87 ±4.30) mm Hg in the half-hitch tourniquet group ( P < 0.05 ). At 3 and 6 hours after hemostasis, PvCO2 was (50.67 ± 9.31 ) mm Hg and (60.02 ± 10.85)mm Hg respectively in the hemostatic bandage group, while (16.85 ± 16.51 )mm Hg and ( 60.02 ± 10.85) mm Hg respectively in the rubber tubing group, which were lower than that in the half-hitch tourniquet group ( P < 0.05 or 0.01 ) . Conclusion Compared with rubber tubing and Halfhitch tourniquet, the hemostatic bandage can efficiently stop bleeding under slight pressure, with the least injury to the bundled tissues.
4.Expression of fusion protein EGFP-HBVP22~e in HepG2 cells

Objective With eukaryote expression vector pEGFP-C2, to establish the HepG2 cell line which may stably express the P22e of hepatitis B virus (HBV), and may be used to study the existed relationship between HBVP22e and hepatitis B. Methods HBVP22e cDNA obtained by PCR from HBV adr subtype 1.2 copies genome plasmid p3.8II was inserted into universal vector pMD18-T for identification. The plasmids were transfected into HepG2 cells via liposome, while EGFP-HBVP22e was analyzed by Western blot, and observed with fluorescence microscope in HepG2 cells. Results Expression vectors of recombinant pEGFP-C2HBV P22e were constructed and expressed steadily in HepG2 cells. Conclusion The results can be used to explore biological activity of HBVP22e in hepatitis B.

Both receiving economic support and receiving emotional support from adult-children were significant positive predictors of the elderly's SWB (R2=8.3%). Conclusion: The receiving economic support and receiving emotional support from adult-children significantly affected the elderly's SWB.

Objective To investigate the effect of rhein on endothelial plasminogen activator inhibitor-1 (PAI-1) mRNA expression and protein synthesis induced by transforming growth factor ?1 (TGF-?1), and explore the protective mechanism of rhein on human endothelial cells. Methods A human umbilical endothelium derived cell line (ECV-304) from ATCC was used in this study. The endothelial PAI-1 mRNA expression and protein synthesis were detected by Northern blot and flowcytometry analysis respectively. The activity of phospho-p44/p42 MAPK induced by TGF-?1 was determined by immunoprecipitation analysis and Western blot. Results TGF-?1 (2?g/L) could increase rapidly endothelial PAI-1 mRNA expression and persist for 24 hours. The PAI-1 mRNA expression induced by TGF-?1 in endothelial cells was inhibited by incubating with rhein. Rhein could also inhibit the endothelial PAI-1 protein synthesis. Further study revealed rhein showed a significantly inhibitory effect on the activity of phospho-p44/p42 MAPK induced by TGF-?1 in human endothelial cells. Conclusions Rhein shows a protective action on endothelial cells by inhibiting PAI-1 overexpression, and this may provide beneficial effects on the treatment of diabetes mellitus and its complications.

Objective To investigate the role of hexosamine biosynthesis pathway (HBP) in the functionalalterationofmesangialcellstransinfectedwith glucose transporter 1 (GLUT1) gene. Methods Rat mesangial cells were transinfected with the human GLUT1 gene (MCGT1) by retrovirus vector. Mesangial cells transinfected with bacterial ?-galactosidase (MCLacZ) were used as control. Glucose uptake was detected with 2-deoxy-〔 3H〕-D-glucose (2-DG). Cell size, RNA/DNA ratio, protein/DNA ratio and the synthesis of fibronectin were evaluated by flow cytometry. The activity of glutamine: fructose-6-P aminotransferase (GFAT), which is the key enzyme of HBP, was assayed by spectrophotometry. The expression of GFAT gene was analyzed by RT-PCR. Objective To explore the methods of large sample research for traditional Chinese medicine(TCM) symptoms of exercise-induced diseases and to define the main symptoms and the main symptom compositon of exercise-induced diseases.Methods The methods of TCM theory of differentiation of syndrome, epidemiology study and multivariable statistical analysis were properly combined. We investigated 518 cases of exercise-induced insomnia(EII) by using the clinical epidemiology program with the uniform diagnostic standard and data processing method.Results The asthenia of both the heart and the spleen(56 cases, 10 8%) was one of the primary syndrome of EII.Conclusions This study showed that main symptoms in differential diagnosis of asthenia of both the heart and the spleen might be sleeplessness, poor appetite, excessive dream, dizziness and weakness of limb.
10.Effects of mild hypothermia on the nitric oxide and water content of brain tissue in rats with traumatic brain edema

Objective To explore the effects of mild hypothermia (MH) on the nitric oxide (NO) and water content of brain tissues (WBT) in rats with traumatic brain edema (TBE). Methods Fifty-four Wistar rats were divided into a control group (group C), a normithermal traumatic group (NT group) and a mild hypothermia traumatic group (MHT group). The NT and MHT groups were then divided into 4 subgroups for study at 30 min, 2 h, 4 h and 8 h post-trauma. TBE models were established according to Yuan Shaoji′s method. The concentration of NO in the jugular vein was measured using chemical luminescence, and water in the brain tissues was calculated with Elliot′s formula. Results Compared with those in the group C, the concentrations of WBT and NO were significantly increased 30 min post-trauma in the NT group, and reached a peak 8h after trauma. These levels were markedly decreased in the MHT group in comparison with the NT group. Conclusions NO levels might play an important role in the development of TBE, and change synchronously with WBT. TBE could be mitigated by MH, which might promote early rehabilitation of TBE by reducing NO.
