1.Effects of arsenic trioxode and adriamycin on survivin expression in (hepatocellular) carcinoma cell line HepG2

Objective To investigate potential mechanism of arsenic trioxide (As2O3) and/or adriamycin (ADM) induces apoptosis in hepatocellular carcinoma (HCC) cells. Methods Human hepatocellular (carcinoma) cells, HepG2, were treated with As2O3 combined with/without ADM in various concentrations for various time, and then the expression of survivin was detected by immunocytochemistry and RT-PCR (respectively). Results (1)Before the cells were interfered with drugs, survivin was highly expressed in HepG2 cells. (2)Expression of survivin was reduced in cells treated with various concentrations of As2O3 or ADM , and the reduction was dependent on durg concentration and time, these two factors were interrelated. (3)Expression of survivin was reduced more markedly in cells treated with ADM than in those treated with As2O3 and the reduction was additionally increased in cells treated with a combination of the two compounds. Conclusions (1)Either As2O3 or ADM can downregulate the expression of survivin in HepG2 cells in a concentration-or time-dependent manner; (2)At the same concentration, ADM is more effective than As2O3 on the downregulation of survivin expression in HepG2 cells, but the effect can be enhanced by combination of these two drugs. (3)As2O3 and ADM may induce apoptosis in HepG2 cells through the downregulation of survivin expression and thus exert their anti-cancer effect. Their combination may enhance their anti-tumor effect, so that in clinical use, the dosage of each durg may be reduced.
2.Antimicrobials Applying in a Hospital of Traditional Chinese Medicine:Investigations and Analysis

OBJECTIVE To investigate the antimicrobial application in a hospital of traditional Chinese medicine(TCM).METHODS Data of antimicrobial application in patients who were discharged within a month were investigated.RESULTS The usage rate of antimicrobials was 62.62%,the surgical departments were higher in antibacterial use than departments of internal medicine;the pathogenic detection rate was 17.28%;the usage rate of combinations accounted for 34.14%;the duration of preoperative and postoperative antimicrobial use was long,some patients didn′t receive rationally preoperative prophylactic antimicrobials.CONCLUSIONS Management of clinical antimicrobial application in hospital of TCM should be strengthened,especially on the indication of antimicrobial and pathogenic detection.
3.The relationship between renal histology,its mechanism and the treatment of Lupus nephutis

This paper emphasizes the importance of the mechanism of renal tissue damage in conducting treatment of lupus nephritis in addition to the renal histology.There are at least 4 kinds of mechanisms related to the renal tissue damage of LN including circulating immune complex deposition,in situ immune complex formation,vasculitic change and thrombotic mircoangiopathy.Treatment according to the mechanisms of tissue damage will give rise to a much better result as compared with the classic treatment based on morphology alone.Multi-target immune theapy has been recommended for treatment of those severe and complicated LN.

5.Nursing for patients with congenital hypospadia with the application of urethral stent

Objective To discuss effective nursing measures for patients with congenital hypospadia with the application of urethral stent.Methods 142 patients with congenital hypospadia were selected and used soft stent during and after operation.The urethral orifice was kept smooth after operation and the stent was removed until 2 weeks after operation.Results Two patients complicated with urethral fistula,one was because of the poor blood supply of the skin flap and the other because of the edema of the skin flap and the remove of stent.Other patients received good treatment effect. Conclusion Application of urethral stent in patients with congenital hypospadia could reduce the incidence of infection and increase the success rate of operation.
6.Determination of Principal Agents and the Related Substances of Compound Ftibamzone/Dyclonine Liniment by HPLC

OBJECTIVE:To establish a HPLC method for determination of the principal components and the related substances in compound ftibamzone/dyclonine liniment. METHODS: The determination was performed on MACHEREY-NAGEL C18 chromatographic column with the mobile phase consisted of methanol-acetonitrile-0.02 mol?L-1 potassium dihydrogen phosphate solution-triethylamine (25∶25∶50∶0.3,pH value adjusted to 4.0 by the phosphoric acid) with a flow rate of 1.0 mL?min-1. The UV detection wavelength was set at 295 nm. RESULTS: A baseline separation was achieved between foreign materials’ peaks and the principal agents’ peaks. The calibration curve of ftibamzone and dyclonine were linear within the ranges of 20.63~72.21 ?g?mL-1(r=0.999 9) and 20.44~71.54 ?g?mL-1 (r=0.999 9),respectively;the average recovery rate was 98.7% vs. 99.2%;the inter-day RSD was 0.97% vs. 1.10%;the intra-day RSD was 1.52% vs. 1.75%,and the lowest detective limit was 0.4 ng vs. 0.2 ng. The contents of the related substances were ranged from 0.43% to 0.68%. CONCLUSION: This method is simple,rapid,specific,accurate and reproducible,and it can be used for the quality control of the compound ftibamzone/dyclonine liniment.
7.Survey and analysis of special medical services marketing

Objective To understand and master the system for assessing the strategies, management and effect of special medical services marketing. Methods Survey sheets on customer satisfaction towards special medical services and survey sheets on the effect of special medical services on the caliber of medical personnel were fandomly distributed to the patients and medical staff in a particular hospital and special topic interviews and statistical analyses of special topic survey data were conducted. Results The satisfaction rate of customers receiving special medical services was 98.9%, and special medical services had a positive effect on the improvement of the quality of medical services, economic benefits and the caliber of the medical personnel. Conclusion Correct orientation is the marketing strategy guiding special medical services. Market-based orientation, marketing-based medical services and resources-based guidance are of important practical values to special medical services while high quality, high caliber and high benefits are the major indexes for assessing special medical services.
8.The study of new targeted-delivery of micro RNAs to bone-metastatic prostate tumors

Objective To construct a new gene delivery system based on atelocollagen (ATE),and explore that modified aptamer (APT),and APT-ATE/miRNA (miRNA-15a and miRNA-16-1) were successfully synthesized to treat bone-metastatic prostatic cancers.Methods Flow cytometry (FCM) analysis was used to characterize APT-ATE complex.The diameter and zeta potential of complexes were measured by Zetasizer Nano-ZS9.The prostatic cancer (PCa) distribution experiments were used to explore its biological characteristics and targeting ability of PCa cells (PC3 and LNCaP).The inhibition of APT-ATE complex on LNCaP cell was determined with the cholecystokinin (CCK)-8 assay.Results FCM results demonstrated the successful synthesis of ATE-APT complex.The cellular uptake of vectors was concentration-dependent.The gene expression in vitro indicated that the modification of APT could increase the efficiency of gene expression and PCa targeting ability of ATE vectors to LNCaP [prostate specific membrane antigen (PSMA) over-expressing prostate cancer cells].The result of biodistribution showed that the bone uptake of APT-ATE was higher than ATE-APT.Conclusions APT-ATE/miRNA might be useful for preclinical and clinical studies on the treatment of bone-metastatic PCa.
9.Investigate and analysis on cognition to the knowledge of peripheral venous indwelling needles of ;clinical nurses in a certain grade A class three hospital

Objective To investigate the cognition level of clinical nurses to peripheral venous indwelling needles, and provide a reference for nursing administrators to carry out targeted intravenous catheter standardized training operation. Methods By convenience sampling, 129 clinical nurses of the certain grade A class three hospital were selected, and their perception about indwelling intravenous catheter were investigated by using self-designed questionnaire in November 2015. Results 125 questionnaires were effectively rewnered. Average score of 125 nurses′cognition to indwelling intravenous catheter was (57.52 ± 10.23) points, pass rate was 65.6%(82/125), and influencing factors were education background, the job tittle, work experience and so on. Conclusions The clinical nurses′assessment, cognition and knowledge master to indwelling intravenous catheter is not ideal, lacking a comprehensive clinical peripheral venous catheter assessment tools. Clinical nurse managers should focus on strengthening vein management, reasonable and better trained clinical nurse intravenous catheter cognitive level, so as to ensure that the effect of catheter using.
10.Expression, purification and characterization of catalytic domain of protein kinase D1 in baculovirus-insect cell expression system.

Protein kinase D (PKD) is a novel family of serine/threonine kinases and diacylglycerol (DAG) receptors and has been documented in a variety of cellular processes. To get high purity catalytic domain of PKD1 (PKD1-cat) for crystallography study, the GST-tagged PKD1-cat gene was cloned into a baculovirus transfer vector pFastBac1 (donor plasmid). When the recombinant plasmid was transformed into DH10Bac competent Escherichia coli, which contains a baculovirus shuttle vector (bacmid), transposition occurs to generate a recombinant bacmid containing the gene of interest (GST-PKD1-cat). The recombinant bacmid DNA was transfected into Spodoptera frugiperda Sf9 insect cells to generate recombinant baculovirus, which was then amplified through multiple rounds of infection in Sf9 cells. After that, Trichoplusia ni insect cells in suspension culture were infected with baculoviral stock at a multiplicity of infection (MOI) of 5 PFU/cell. SDS-PAGE and Western blotting analysis confirmed the detection of a 68 kDa protein by the glutathione S-transferase (GST) monoclonal antibody. The recombination protein was purified by Glutathione sepharose affinity chromatography and cleaved by PreScission Protease to remove GST tag, and a highly pure 42 kDa protein which was consistent with the molecular weight of the expected PKD1-cat protein was detected on SDS-PAGE. The activity of purified PKD1-cat protein was determined by in vitro PKD kinase assay. Our data showed that the kinase activity increased with the concentration of purified PKD1-cat protein. These results showed that the truncated recombinant PKD1-cat protein was highly active and pure, and could potentially be used for solving 3D structure of this protein by Nuclear Magnetic Resonance (NMR) or crystallography.
