1.Phacoemulsification combined with tr abeculectomy glaucoma andcataract after filt rating surgical failure

Objective To evaluate the saf ety and efficiency of phacoemulsificatio n combined with intraocular lens implant and trabeculectomy in patients with glau coma and cataract after filtrating surge ry failure. Methods Thirty-two cases (32 eyes) patients underwent phacoemulsific ation combined with intraocular lens imp lant and trabeculectomy after filtrating surgery failure. Outcome measures inclu ded intraocular pressure (IOP) and visua l acuity. The followed-up periods rang ed from 6 months to 2 years. Results In 21 of the 32 eyes(66%),postoperative vi sual acuity were above 0.5. In 6 of the 32 eyes(18%),postoperative visual acuity were between 0.1 to 0.4. In 27 of 32 e yes(84.3%), postoperative IOP were between 12.23 to 20.55 mmHg. Conclusion phac oemulsification combined with intraocula r lens implant and trabeculectomy in patients with glaucoma and cataract after f iltrating surgery failure is a safe and effective treatment for these patients.
2.Effect of polyene phosphatidyl on synaptic plasticity in region of hippocampus CA3 of young rats

Objective To observe the structure changes of the synapse of the neurons in hippocampus CA3 of young rats and study the basis for the mechanism of polyene phosphatidyl in providing learning and memory and the effect on synaptic plasticity.Methods A total of 20 Wistar rats with 5 months were randomly divided into polyene phosphatidyl group and normal control group.Each group had 10 rats.After 4 weeks feeding,Water maze training was peformed in all the rats for 1 weeks.The immue expressions of synapsin(SYN) of the rats in polyene phosphatidyl group and control groups were observed with immunohistochemical method and analyzed by MOTAC imagine analyzing system.The change of synapse of hippocamal CA3 was observed with electron microscope.And the other 24 to 26 months rats were selected as aged group,and fed in the same condition.Moreover,the ultrastructures of hippocamal CA3 of aged rats were observed by transmission electron microscopy(TEM).Results The SYN in polyene phosphatidyl group(0.430 0?0.022 4) was higher than that in control group(0.3567?0.0209) (P
3.Comparison between the 5/6 nephrectomy SD rat and C57 mouse models of chronic renal failure

Objective To find the similarities and differences between SD rat and C 57 mouse models of chronic renal failure established by 5/6 nephrectomy ,and select an appropriate experimental animal for the study of the treatment and prevention of chronic renal failure .Methods 60 SD rats and C57 mice were randomly and equally divided into 5/6 nephrectomy group and sham operatied group .The mice and rats were killed at 4 weeks , 8 weeks and 12 weeks after the operation, respectively.Serum and renal tissue samples were collected for the detection and analysis of renal function andα-SMA.Results There was no significant difference in the survival rates between the rat and mouse models (P>0.05). The serum creatinine and urea nitrogen in the 5/6NX rat group showed a stable state at 8-12 weeks, which was different from that of the 5/6NX mouse group.Conclusions A 5/6 nephrectomy mouse model of chronic renal failure is successfully established .Although both 5/6 NX mouse and rat groups appear progressive renal failure after the operation , their disease processes are not quite the same .In clinic, appropriate experimental animals should be selected according to the length of the observation period in order to accurately show the disease states .
4.Experimental study of treatment of mitomycin C extravasation injuries

Objective To find an effective method for the treatment of mitomycin C extravasation injuries. Methods The rabbit model of mitomycin C extravasation was made and managed by different methods. The therapeutic efficacy in each group was assessed by using regression index and regression time. Results The therapeutic efficacy in ice compress group and ice compress plus amifostine group was better than that in other groups and the regression time was the shortest. There was no inflammation and necrosis in skin in ice compress group. Conclusion Ice compress, which can prevent inflammation and necrosis in skin, is one of the most effective treatment methods for mitomycin C extravasation injuries.
5.Mechanisms of cell-cell adhesion dependent multicellular drug resistance of pulmonary adenocarcinoma cell line A549

Objective To study the mechanism of multicellular drug resistance of pulmonary adenocarcinoma cell line A549 mediated by cell-cell adhesion. Methods We compared the sensitivity of monolayer cells (MCs) to adriamycin (ADM) with that of multicellular spheroids (MCSs) which was employed as a three dimensional cell culture model. Transmission electron microscopy was applied to observe the cellular ultrastructure. Bcl-2, Bcl-xL, and Bax expression levels were detected by flow cytometry and indirect immunofluorescent staining. Results Compared with MCs, MCSs had more than two layers of cells, more extensive and compact cell adhesion, and inlay junctions were found within them. MCSs were more resistant to ADM. At the same time, Bcl-2 and Bcl-xL expressions in MCSs were much higher. After treatment with ADM, expressions Bcl-2 and Bcl-xL increased markedly in MCSs. Conclusion MCSs could simulate the solid tumors in vivo and has multicellular drug resistance mediated by cell-cell adhesion. The possible mechanisms may be associated with the upregulation of Bcl-2 and Bcl-xL.
6.Construction of three dimensional in vitro culture model of pulmonary adenocarcinoma and detection of its drug resistance

Objective To explore an optimal model of three dimensional in vitro cell culture for simulating solid tumors in vivo . Methods The model of three dimensional cell culture was constructed under the conditions of inhibiting the cell wall attachment and stirring the medium. Multicellular spheroids (MCS) were cultured using microcarriers (CutiSpher). Drug sensitivity of monolayer cells (MC) and MCS was tested by MTT staining and cytometry, respectively. Ultrastructures of the MC and MCS were observed by transmission electron microscopy. Results Cells in three dimensional cell culture model without microcarriers were compacted into mass at 4 d while cells in MCS were found to attach to the microcarriers at 0.5 h. MCS had more than two layers of cells growing within it at 5 d. Compared with MC, MCS was more resistant to the anticancer drug, and had more plenty of organell and microvilli with more extensive and compact cell adhesion. Conclusion MCS has strong developmental properties and can simulate the cell cell interactions in vivo , especially cell adhesion, which may contribute to the drug resistance of MCS.
7.Waist-to-height ratio as a predictor of dyslipidemia for Chinese adults

Objective To explore the association between waist-to-height ratio (WHtR) and dyslipidemia in Chinese adults and to find out an optimal threshold of WHtR for predicting dyslipidemia.Methods A total of 221 270 adults from 4 health checkup centers nationwide were selected by using cluster random sampling method.Height,body weight,waist circumference (WC),total cholesterol (TC),triglyceride (TG),high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol(LDL-C) were measured.Body mass index (BMI) and WHtR were then calculated.The area under the receiver operating characteristic curve(AUC) was analyzed and optimal cutoffs were estimated by maximizing the sums of sensitivity and specificity.Results WHtR showed the largest AUCs in the participants with higher TC,TG and LDL-C,followed by increased WC and BMI.In male and female,the optimal thresholds of WHtR,WC and BMI for predicting dyslipidemia were 0.49-0.50 and 0.47-0.49,83-85and 73-76 cm,and 24-25 and 22-23 kg/m2,respectively.Conclusion WHtR may be superior to WC and BMI for predicting dyslipidemia,and an unisex cutoff of 0.5 should be recommended.
8.Change of Na~+/dicarboxylate cotransporter-1 expression in rats with nephrolithiasis induced by ethylene glycoi and mechanism of potassium citrate prevention

Objective To investigate the change of Na+ /dicarboxylate cotransporter (SDCT) 1 expression in renal tissues of rats with nephrolithiasis induced by ethylene glycol (EG) and the mechanism of potassium citrate prevention. Methods Male Wistar rats were divided into control, nephrolithiasis and potassium citrate treated groups. Calcium oxalate crystal deposition and histological changes in kidney were examined by anatomical and light microscope. The plasma and urinary biochemical parameters, such as citrate, oxalate etc., were analyzed by routine biochemical method. The expression of SDCT1 mRNA in kidneys was determined by Northern blot, and the change of SDCT1 protein abundance was detected by immunohistochemistry. Results On day 3, the animals in the nephrolithiasis group had a higher level of SDCT1 mRNA and protein abundance in kidneys, as well as a lower level of citrate in the urine when compared with the control group. However none of the rats in this group had obviously calcium oxalate crystal deposition in kidneys. On day 7 and 14, the expression of SDCT1 mRNA and protein abundance were shown further increase, when the urinary citrate concentration was decreased progressively, and 87. 5% to 100% of the rats in this group displayed a large quantity of calcium crystal deposition in the kidney. In the potassium citrate treated group, both the expression of SDCT1 mRNA and protein abundance were shown almost complete inhibited during the whole experiment time, meanwhile the urinary citrate level was significantly elevated with time; furthermore, the occurrence of the renal crystal deposition decreased to 37. 5% on day 14, and the pathologic changes such as tubular dilation and inflammatory cells infiltration were shown to be alleviated. Conclusions The upregulation of SDCT1 mRNA and protein abundance in kidney has a close relationship with hypocitraturia, which may play an important role in the development of nephroliathisis. The treatment with potassium citrate has a beneficial effect on the experimental nephrolithiasis rats through inhibiting the expression of SDCT1 in the renal tissue.
9.Protective Effects of Dexmedetomidine on Systemic Inflammatory Response Syndrome during Cardiopulmonary Bypass

Objective: To investigate the effects of dexmedetomidine (DEX) on oxidative stress and systemic inflammatory response syndrome(SIRS) during cardiopulmonary bypass (CPB).Methods: Totally 62 patients scheduled for mitral valve replacement surgery were enrolled in this study and randomly assigned to DEX group with constant infusion of DEX (0.5 μg·kg-1 ·h-1 , n =31) and the control group with the same amount of 0.9% sodium chloride injection(n =31).A volume of 4 ml of jugular vein blood was respectively withdrawn before the induction of anesthesia (T0), at 10 min after aortic-off clamping(T1), the end of bypass(T2), 4h after bypass(T3),12h after bypass(T4) and 24h after bypass(T5).The serum C-reactive protein (CRP), tumor necrosis factor-α (TNF-α), interleukin-6(IL-6), interleukin-8(IL-8) and interleukin-10(IL-10) were determined.Results: The serum CRP in both groups increased remarkably at T1-T5 , while the serum CRP in DEX group was significantly lower than those in the control group at T3-T5.The serum TNF-α increased remarkably in control group at T2-T4, while the serum TNF-α increased remarkably in DEX group at T2-T3;the serum TNF-α in DEX group was significantly lower than those in the control group at T3-T4.The serum IL-6, IL-8, IL-10 in both groups were increased remarkably at T2-T5, T1-T5,T1-T3, respectively.The serum IL-6 and IL-8 in the DEX group were significantly lower than those in the control group at T2-T4 while the serum IL-10 in the DEX group was significantly higher than that in the control group at T1-T3.The serum WBC in both groups at T3-T5 were higher than those at T0, while the serum WBC in the control group was significantly higher than that in the DEX group at T3-T5.However, the differences between the two groups at mechanical ventilation time, ICU observation time, the incidence of postoperative SIRS after 24h, hospitalization days, and the adverse reactions were not statistically significant.Conclusion: DEX has notable protective effects on systemic inflammatory response syndrome during extracorporeal circulation.
10.Prediction of antigenic epitopes on HA, NA amino acid sequences of novel influenza A (H7N9) virus and analysis association between susceptibility and HLA-Ⅱalleles

Objective:To compare the amino acid sequences difference of HA,NA novel influenza virus A/H7N9 isolates, decipher possible B cell epitopes and T cell epitopes of HA,NA protein,and analyze the association between susceptibility and HLA polymorphisms.Methods:The amino acid sequences of novel influenza A ( H7N9) virus were downloaded from Genbank.Phylogenetic trees were constructed based on the amino acid sequences of HA and NA by using software Clustal X and MEGA 4.0.B cell and T cell epitopes were respectively predicted with Protean software and NetMHCⅡ2.2 Server online server.Results:The homology of HA and NA proteins of H7N9 virus was high.10 B cell epitopes and 15 T cell epitopes were randomly distributed throughout HA sequence and 12 B cell epitopes and 9 T cell epitopes were randomly distributed throughout NA sequence.HLA-DRB1*0701 allele which was commonly observed in Northern Chinese population have a high binding affinity for 9-mer peptides of HA and NA proteins.Conclusion:The prediction of B and T cell epitopes of HA and NA proteins with multiple methods benefits the research and development of vaccine against human infection with avian influenza A H7N9 virus.HLA-DRB1*0701 allele may contribute to susceptibility to novel influenza A (H7N9) virus.H7N9 influenza virus is more easily spread in Urumqi,Harbin,Shandong Province,Liaoning Province,Beijing, Shijiazhuang and Tianjin of China.
