1.Protective effects of ziprasidone on the damage of hippocampal-derived neural stem cell induced by lipopolysaccharide

Objective To investigate the effects of ziprasidone on the hippocampal-derived neural stem cells (NSCs) viability and apoptosis injured.by LPS.Methods The NSCs were derived from the hippocampus of fetal rats,after the primary neurospheres passaged,the cells were treated with LPS (200 μg/L) and different concentrations of ziprasidone for 48 h or 72 h.The cell viability and the level of LDH were measured by the kit of WST-8 and LDH detected kit,respectively.Furthermore,the apoptosis rate of each dosage group was measured by using Annexin-V-FLUOS Staining Kit.Results (1) Cell viability test showed that the cell viability of LPS group (OD value,0.265± 0.047) was significantly reduced than that of sham (OD value,0.380± 0.029),LPS + 5 μM (OD value,0.316±0.008) and LPS+10 μM (OD value,0.321±0.006) group after ziprasidone treatment for 48 h.There were also significant difference of cell viability between sham,LPS+5 μM,LPS+ 10 μM and LPS group after ziprasidone treatment for 72 h.(2) LDH test showed that the LDH level of LPS group was significantly decreased than that of sham,LPS+5 μM and LPS+10 μM after 48 hour's ziprasidone treatment (P<0.01).The LDH level of LPS group was also significantly reduced than that of sham,LPS+5 μM and LPS+10 μM after 72 hour's ziprasidone treatment (P< 0.01).(3) The apoptosis of LPS + 10 μM ((17.87 ± 2.29) %),LPS + 5 μM ((18.45 ± 3.84) %) treated group or sham group ((9.15 ± 1.54) %) was significantly lower than LPS group ((22.67 ± 2.15)%) by using flow cytometry after 48 h treatment.Furthermore,the apoptosis of LPS+ 10 μM ((10.52± 1.42)%) treated group or sham group((8.45±1.04)%) was significantly lower than LPS group((15.70± 2.97) %) after 72 h treatment.Conclusion The cellular damage of NSCs injured by LPS can be restrained by ziprasidone,and this effect might be one of the cellular mechanism of neural protective effect of ziprasidone.
2.Inhibitory effects of microRNA-375 on biological behaviour of human retinal capillary endothelial cells induced by hypoxia

Background Studies showed that microRNA (miR)-375 suppresses the growth,apoptosis,migration and adhesion of tumor cells,and it plays a regulation to the changes of vascular endothelial growth factor (VEGF) in tumor tissue to arrest neovascularization.However,whether miR-375 intervenes the formation of new blood vessel in eyes is unelucidated.Objective This study was to explore the effects of miR-375 on human retinal capillary endothelial cells (HRCECs) function induced by hypoxia.Methods HRCECs were cultured using IMDM and divided into normal control group,CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2 +miR-375 mimic control group,CoCl2+miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group,and hypoxia cell models were created by adding 200 μmol/L CoCl2.MiR-375 and frizzled 4 (FZD4) small interfering RNA (siRNA) were transfected into the cells by 50 nmol/L miRNA lipidosome for 48 hours.The proliferation of the cells was detected by MTT assay;migrated number of the cells was examined by Transwell chamber assay;ELISA was employed to detect the concentrations of VEGF and VE-cadherin in the medium;the expression of β-catenin,cyclinD1,matrix metalloproteinase-2 (MMP2) and VEGF proteins were analyzed by Western blot;tube length of vessel formation was evaluated by Matrigel assay.Cultured cells were divided into normal control group,CoCl2 model group,CoCl2 +mock group and CoCl2 + FZD4 siRNA group,the relative expression of FZD4,a miR-375 targeted gene,was detected by luciferase reporter.Results The relative expression of miR-375 mRNA was significantly increased in the CoCl2 +miR-375 mimic group compared with the CoCl2 + miR-375 mimic control group and reduced in the CoCo2 + miR-375 inhibitor group compared with the CoCo2 + miR-375 inhibitor control group (t =-19.237,8.764,both at P<0.01),with a higher transfected efficacy for miR-375.The cell proliferative fold,migrated cell number,VEGF and VE-Cadherin contents in the medium and the tube length were significantly different among the CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2 +miR-375 mimic control group,CoCl2 +miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group (F =24.324,26.776,14.113,19.225,15.040,all at P<0.001),and those in the CoCl2 +miR-375 mimic group were evidently reduced in the CoCl2 +miR-375 mimic group compared with the CoCl2 +miR-375 mimic control group,while those in the CoCl2 +miR-375 inhibitor group were considerably elevated in comparison with the CoCl2 +miR-375 inhibitor control group (all at P<0.01).The expressions of β3-catenin,cyclinD1,MMP2 and VEGF protein were significantly different among the normal control group,CoCl2 model group,CoCl2 +miR-375 mimic group,CoCl2+miR-375 mimic control group,CoCl2 +miR-375 inhibitor group and CoCl2 +miR-375 inhibitor control group (F=11.753,13.283,16.770,10.334,all at P<0.001).In addition,the cell proliferative fold,migrated cell number and the tube length were significantly increased in the CoCl2 model group and CoCl2+mock group,and those in the CoCl2+FZD4 siRNA group were decreased in comparison with the CoCl2 +mock group (all at P<0.05).Conclusions MiR-375 inhibits the growth,migration and tube formation ability of HRCECs in hypoxic status probably by regulating the activation of Wnt pathway via directly targeting FZD4.
3.Comparison of Speech-language Abnormalities in Huang Di Nei Jing and Zhong Zang Jing

The record of speech-language abnormalities scattered in some chapters of two classics of traditional Chinese medicine, Huang Di Nei Jing and Zhong Zang Jing. The descriptive modes of these abnormalities in Huang Di Nei Jing were more diversified than in Zhong Zang Jing. In aspect of clinical values, speech-language abnormalities in Huang Di Nei Jing were used to guide clinical diagnosis and treatment, such as describing body constitution and clinical mistreatments, but rare in Zhong Zang Jing.
4.Selection of SPE column for trace pesticides in large volume water

Objective The selection of SPE column to extract trace pesticides in the water. Methods Investigated the recovery of 6 kinds SPE column (including HLB,GDX403,GDX103,C18,C8,C6) to extract phorate, sulfotep, propoxur, carbofuran, cypermethrin, fenvalerate in the water. Results By using oasis HLB cartridge to extract 21 kinds of pesticides in water, the recoveries were over 60%for the most pesticides. Conclusion The method could be applied to the cases which extract and analyse the trace pesticides in large volume water.
5.Protective effects of transfecting bone morphogenetic protein-7 on rat cardiomyocytes against hypoxia-reoxygenation injury

Objective To investigate the protective effect of transfecting bone morphogenetic protein-7 (BMP-7) on cultured neonatal rat cardiomyocytes against hypoxia-reoxygenation injury. Methods Neonatal rat cardiomyocytes were cultured. The pcDNA-rrBMP7 was introduced into cardiomyocytes by Fugene 6.0 transfection method. The cardiomyocytes were divided into three groups: control group (group C), hypoxia-reoxygenation group (group HR) and gene transfecting group (group BT). Trypan blue exclusion test was performed to detect cell viability. The activity of lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) was assayed to evaluate cell injury. For evaluating the cell antioxidant ability, the activity of superoxide dismutase (SOD) and the level of malondialdehyde (MDA) were determined by colorimetric assay. Fluo-3 labeling method and confocal laser scanning microscopy were used to observe the change in intracellular calcium. Results The results showed that after 120 min of simulated ischemia followed by 240 min of reperfusion, cell pulsation rate was decreased, the activity of LDH, CPK and the trypan blue uptake rate were increased. As compared with the group C, SOD activity decreased and the content of MDA increased in Group HR. Compared with Group HR, the SOD activity increased and the content of MDA decreased in group BT. 6.Microtensile bond strength and SEM evaluation of total-etch and self-etching adhesives to caries-affected dentine

Objective:To evaluate the microtensile bond strength of total-etch or self-etch adhesives to caries-affected dentine versus normal dentine. Methods:Twenty-four extracted human molars with mesial coronal carious lesions extending into mid-dentin were selected. The softened carious dentine was removed with a slow-speed, carbon-steel bur and Caries Marker was used as guides. Prepared cavity in sound dentine in the distal coronal part of the identical teeth was used as the control. Surfaces were bonded with either Prime&Bond NT, Adper Single Bond 2, Adper Prompt or XenoⅢ,according to manufactures' recommendations. A crown was built up using resin composite(Filtek P60).After storage in water (37 ℃,24 h),teeth were vertically serially sectioned into 1.0 mm thick slabs and trimmed to yield 1 mm2 test area that contained either caries-affected or normal dentine. Then the specimen were tested in tension in an Instron machine at 0.8 mm/min.The bonding interface between the dentine and adhesives was qualitatively evaluated under the observation of scanning electron microscope(SEM). 7.Total hip replacement in patients with ankylosing spondylitis: problems and their solutions

Objective To evaluate the efficacy of total hip replacement in patients with ankylosing spondylitis(AS) and to solve the problem often encountered during surgery. Methods One hundred and twelve hips of 98 patients with AS undergone total hip replacement were reviewed. The mean duration of follow up was 4.2 years(range, 1.5 to 8 years). The pre and postoperatively comparative study on pain, range of motion, correction of deformity and total function evaluation were conducted. Results Pain relief obtained in all but 4 hips. Mean range of motion was improved from 37.5? to 69.5? and the flexion deformity of the involved hip was corrected from mean 38.2? to 5.7?, all of the patients could take care of daily living by themselves. Complications included penetrating fracture of proximal femur in 4, sciatic nerve injury in 3, early dislocation of the hip in 2, disassociation of the acetabular component in 2 and sinking of the femoral stem in 23. Conclusion Total hip replacement is a very important and effective treatment of choice for patients with AS. The modified combination of anterolateral and lateral approach used by the authors facilitated release of contracture of the soft tissue anterior to the hip joint and correction of flexsion deformity of the hip. In case of multiple joint involvement, the surgery should be performed on the most severely involved one and the next surgery, if required, should be scheduled later within 3 to 6 months.
8.Video-Assisted Thoracoscopic Surgery for Open Thoracic Traumas

Objective To evaluate the video-assisted thoracoscopic surgery(VATS) in the diagnosis and treatment of open thoracic traumas. Methods From July 2003 to July 2008,video-assisted thoracoscopic surgeries,including thoracic exploration,repair of the pulmonary laceration,parietal pericardium and diaphragm,hemostasis,evacuation of the clotted hemothorax,and mini-incision thoracotomy,were carried out in 64 selected patients with open thoracic traumas. Results Via VATS,all the 64 patients were diagnosed accurately: pulmonary laceration in 43(56 lesions),pericardiac rupture in 7(one of them showed right ventricular rupture),diaphragmatic rupture in 9(5 of them were complicated with spleen rupture,1 with hepatic rupture,and 1 with gastric rupture perforation),single intercostal vessel injury in 5,and combined intercoastal vessel injury in 3.The procedures involved VATS pulmonary laceration repair in 24(combined with thoracic wall hemostasis in 2),VATS combined mini-incision thoracotomy pulmonary laceration repair with Endo-GIA in 11(silk suture was employed in 6,and pulmonary wedge resection in 2).Through VATS procedure,hemostasis and repair were completed for pericardiac repair in 3,and diaphragmatic repair in 5.Thirteen cases were converted to open thoracic surgery.The duration of VATS procedure ranged from 32 to 124 minutes with a mean of 65.8 minutes.After the procedure,6 patients developed pneumothorax,4 showed mild subcutaneous emphysema,and 9 had slight hemoptysis in 1 to 12 days after the treatment.The patients were followed up for 2 to 60 months(mean,28.5 months).Chest X-ray performed during the period showed no pleural effusion,pneumothorax or other trauma-related complications.Conclusion VATS is effective for the diagnosis and treatment of open thoracic traumas with less surgical injuries and good outcomes.
9.Chemotherapy plus pirarubicin in treatment of malignant lymphoma in the elderly:an analysis of 50 cases

Objective To investigate the effects and toxicity of pirarubicin and chemotherapy regimen in treatment of malignant lymphoma in the elderly. Methods 50 cases(over 60 years of age) with non Hodgkin's lymphoma collected in Lishui Central Hospital of Zhejiang province from January 2011 to January 2013 were randomly divided into control group(n=25)and experimental group(n=25). Experimental group were received CTOP chemotherapy, control group were received the standard CHOP chemotherapy, vomiting, stomach, blood pressure, blood glucose control and other appropriate symptomatic treatment were adopted according to the speciifc circumstances of each patient in both two groups. Regular inspection were given to all patients, and bone marrow suppression, treatment of lymphoma, complications and other indicators were observed. Results Complete remission in experimental group after treatment was 13 cases, accounted for 52%within group, partial remission were 9 cases, accounted for 36%. The total effective number was 22, and the rate was 88%. Complete remission in control group after treatment was 8 cases, accounted for 32%within group, partial remission were 5 cases, accounted for 20%. The total effective number was 13 cases, and the rate was 52%. Bone marrow suppression in experimental group was signiifcantly less than control group (P<0.05). Conclusion Elderly patients with malignant lymphoma in chemotherapy plus pirarubicin can improve the therapeutic effect, reduce bone marrow suppression and complications.
10.Dendranthema morifolium attenuated the reduction of contraction of isolated heart and cardiomyocytes induced by ischemia/reperfusion

AIM: To investigate the effect of Dendranthema morifolium (Ramat) Tzvel (DM) on isolated rat heart and ventricular myocytes during ischemia/anoxia and reperfusion/reoxygenation.METHODS: The Langendorff perfused rat hearts were used to measure intraventricular pressure and coronary flow. The cell contraction and intracellular calcium transient in enzymatically isolated ventricular myocytes were determined. RESULTS: (1) DM (0.5 g/L) significantly attenuated the inhibitory effects induced by ischemia/reperfusion on left ventricular developed pressure (LVDP), ?dp/dt max, coronary flow and LVDP?HR, meanwhile increased the content of SOD and decreased the content of MDA in the myocardium; (2) DM (0.5 g/L) attenuated the inhibitory effects of anoxia and reoxygenation on [Ca 2+]i transient and cell contraction in isolated ventricular myocytes. CONCLUSION: DM attenuated the effects on contractility and intracellular calcium induced by ischemia/anoxia and reperfusion/reoxygenation in the isolated rat heart and the ventricular myocytes. The mechanism might be related to increase in SOD activity and maintaining [Ca 2+]i homeostasis.
