1.Experimental Study on the Anaphylactoid Reaction induced by Qingkailing Injections on the Beagle Dogs

Objective To study the sensitization of Qingkailing I njections produc ed in five pharmaceutical factories and the sensitization of each composition an d adminicle of Qingkailing Injections on the Beagle dogs by intravenous injectio n. Methods By intravenous injection with Qingkailing In jections produced by the five pharmaceutical factories and all the compositions and adminicles into the Beagle dogs, the behavior changes of the dogs were observed and the content of serum histamine, I gE, IgG, IgM, ECP and IL-4 were detected. Results The result was determined ac cording to the indices of both behavior changes and the serum histamine, IgE, Ig G, IgM, ECP and IL-4. Qingkailing Injections produced in three pharmaceutical f a ctories and the compositions, extract of gardenia and cholalic acid were positi ve in induc ing the anaphylactoid reaction. Conclusion The Qingkail ing Injections produced in three pharmaceutical factories can induce anaphylactoid reaction. The extrac t of gardenia and cholalic acid may be the relevant substance of anaphylactoid r eaction.
2.Chemical constituents of organic acid part from Camptosorus sibiricus

Objective To study the chemical constituents of the organic acid part from Camptosorus sibiricus.Methods The compounds were isolated by chromatography on silica gel column and identified on the basis of physicochemical constants and spectral analysis.Results Ten compounds were isolated and their structures were identified as 11,12,15-trihydroxy-13-en-octadecenoic acid(Ⅰ),caffeic acid(Ⅱ),courmaric acid(Ⅲ),protocatechuic acid(Ⅳ),4-hydroxybenzoic acid(Ⅴ),isovanillic acid(Ⅵ),2,4-dihydroxybenzoic acid(Ⅶ),cinnamic acid(Ⅷ),succinic acid(Ⅸ),palmitic acid(Ⅹ).Conclusion Compound Ⅰ is a new compound named as camptosoric acid and compounds Ⅲ-Ⅹ are obtained from the plants of Camptosorus Link for the first time.
3.Avian influenza virus surveillance in external environment in Urumqi City during 2013 to 2015

Objective To investigate the distribution of avian influenza A viruses in external environment in Urumqi City in order to provide reference for the prevention and control of human infection with avian influenza virus in Urumqi City.Methods Environmental specimens were collected from 2013 to 2015 to detect avian influenza A virus nucleic acid with real-time fluorescent PCR.The positive specimens were further analyzed to identify the subtypes of avian influenza A viruses.Results A total of 1 043 environmental specimens were collected and tested.Among them,123 specimens were positive for avian influenza A virus nucleic acid with an overall positive rate of 11.79%.H9 was the predominant subtype,accounting for 7.77% of all specimens.Quarterly detection rates of avian influenza A virus in the three years peaked in different quarters of the year.Of all surveillance sites,live poultry markets in urban and rural areas had the highest positive rate (14.23%).Specimens collected from poultry drinking water (18.44%) and other specimens (19.44%) were highly positive for avian influenza A virus.Conclusion Avian influenza A viruses,especially the subtypes of H5,H7 and H9,exist in live poultry markets in Urumqi City.It is necessary to conduct surveillance and health education among people exposed to poultry in live poultry markets.
4.Clinical Observation of Breviscapine Combined with Double Shock in the Treatment of Lupus Encephalopa-thy

OBJECTIVE:To observe the efficacy and safety of breviscapine combined with double shock in the treatment of lu-pus encephalopathy. METHODS:70 patients with lupus encephalopathy were randomly divided into control group and observation group. Control group was treated with Methylprednisolone sodium succinate for injection 500 mg for continuous 3 d,iv,once a day,Cyclophosphamide for injection 1.0 g in the 4th day,iv,and Methylprednisolone sodium succinate for injection 80 mg was used for maintaining treatment in the 5th day for continuous 15 d,once a day. Observation group was treated with Breviscapine for injection 50 mg for continuous 20 d based on the treatment of control group,iv,once a day. The SLE activity(SLEDAI) score, IL-1β,IL-6,IL-8 and IFN-γ were observed,cases of neurological and psychological symptoms,the fatality rate and incidence of adverse reactions were recorded. RESULTS:After treatment,the SLEDAI,IL-1β,IL-6,IL-8 and IFN-γ in 2 groups were signifi-cantly lower than before,and observation group was lower than control group(P<0.05);the cases of neurological symptoms(ex-cept for aseptic meningitis) and psychological symptoms (except for auditory hallucinations) were significantly less than control group(P<0.05). There was no significant difference in the mortality rate and incidence of adverse reactions(P>0.05). CONCLU-SIONS:Breviscapine combined with double shock has good efficacy and safety in the treatment of lupus encephalopathy,it can re-duce the SLEDAI score,improve neurological and psychological symptoms.
5.Dynamic changes in glutamate and γ-aminobutyric acid release in the rostral ventromedial medulla in a rat model of incisional pain

Objective To investigate the time-dependent changes in glutamate (Glu) and 7-aminobutyric acid ( GABA) release in the rostral ventromedial medulla in a rat model of incisional pain. Methods Healthy male SD rats weighing 250-300 g were used in this study. Twelve rats in which microdialysis cannulae were implanted in the right rostral ventromedial medulla without neurological deficits were randomly divided into 2 groups ( n = 6 each): group A control and group B incisional pain. In group B an 1 cm long incision was made in the plantar surface of right hindpaw under 1.2% isoflurane anesthesia which was maintained for 5 min. Samples of dialysate were collected before incision (T_0 baseline) and at 3 h, 1 d, 2 d and 3 d after incision was made (T_(1-4)) in both groups for determination of Clu and GABA concentrations (by HPLC). Results In group B Glu and GABA concentrations in the dialysate were significantly increased at 1 d (T_2) and 3 h-3 d (T_(1-4)) respectively as compared with the baseline value at T_0 and were significantly higher than those in group A (control group). Conclusion Incisional pain increases the release of Glu and GABA in the rostral ventromedial medulla which might influence the function of descending pain modulation pathway.
6.The impact of auto-mA low-dose colonic CT scanning on image quality and lesion visualization

Objective To evaluate the effect of low-dose colonic CT scanning in image quality and lesion revealing with different noise idex.Methods Model group:in order to find the optimal scanning parameter range,small simulated polyps in pig colon was prepared and mA was modulated by regulating noise index.Images were reprocessed with CT colonography and Raysum reconstructions.Image quality and the lesion revealing situation were evaluated and scored based on both reconstructive and axial images.The study of patient group was prospective and continuous.All positive cases were comfirmed by biopsy or surgery pathology.Two groups of patients were enrolled.First group(39 cases from January 2007 to June 2007):patients were divided into three sub-groups randomly(15 cages with NI=10,13 cases with NI:13,11 cases with NI=16),while NI was not changed with position.Second group(32 cases from Jury 2007 to January 2008):patients were scanned and NI was changed with different position(NI=10,20).The average X-ray radiation values of two patient groups were compared with standard values.All data were processed with rank sum tesL Results First group:when NI=10,the average scores of axial and constructive images were 4.2 and 2.4 respectively,when NI=13,those were 3.2 and 2.5 respectively,When NI=16.those were 2.9 and 2.7 respectively.and the average CTDlw were 17.51,12.90 and 5.94 mGy respectively.When NI increased,the average scores of axial decreased(H=20.01,P<0.01),the average scores of constructive images did not changed(H=0.81,P>0.05).Second group:when NI=10,the average scores of axial and constructive images were respectively 3.6 and 2.3,when NI=20,those were 2.2 and 2.3 respectively,and the average CTDIw was 11.63 mGy.When NI increased from 10 to 20,the average scores of axial decreased(H=20.84,P<0.01),the average scores of constructive images did not decreased(H=0.29,P>0.05).In the first group,the average CTDIw wag reduced by 35.0%,52.1%and 77.9%respectively when NI was 10,13 and 16.In the second group,the average CTDIw Was reduced by 56.8%.Conclusions Auto-mA low-dose CT scanning could reduce X-ray radiation obviously,while not affecting image quality.
7.Establishment of a severe aplastic anemia mouse model by using recombinant human interferon-gamma plus busulfan

BACKGROUND:It is important to establish an ideal mouse model of severe aplastic anemia for investigating the mechanism and finding new therapies for aplastic anemia. OBJECTIVE:To establish a severe aplastic anemia mouse model by using recombinant human interferon-γand busulfan. METHODS:Sixty healthy Kunming female mice were randomly divided into two groups:model group (n=50) and control group (n=10). The model group was given recombinant human interferon-γat a dose of 1×104 U/d by intraperitoneal injection and busulfan at a dose of 18 mg/(kg·d) through stomach feeding for 7 days. The same volume of physiological saline was given to control group. Multi-parameters, including general condition, body weight, blood cellcount, morphology and biopsy of bone marrow were analyzed in two groups. RESULTS AND CONCLUSION:At day 7 after treatment, the weight, white blood cellcount, hemoglobin, blood platelet, reticulocyte count in model group were significantly lower than control group (P<0.05). Bone marrow smears and biopsy of model group showed marked reduction of bone marrow proliferation and increases of percentages of non-hematopoietic cellclusters and adipose tissue. The oil drop and fat vacuole were apparently seen in the model group. Severe aplastic anemia mouse model can be established by using recombinant human interferon-γand busulfan successful y, which is economic, stable and easy to operate.
8.The platelet activation and the expression of interleukin-1β in patients with rheumatoid arthritis

Objective To evaluate the significance of platelet activation and the expression of inter-leukin (IL)-1β in patients with RA. Methods The activation of platelets and the expression of IL-1β in pla-telets in 50 RA patients(22 high-active, 28 mediate/low active ) and 30 normal controls were determined us-ing flow cytometry. Meanwhile, inflammatory indicators such as erythrocyte sedimentation(ESR), C-reactive protein (CRP) and DAS28 were also recorded. T test and correlation analysis were performed. Results The platelet activation in RA group(19.2±4.8) was higher than the control group(9.0±2.9)(t=10.5, P=0.001). The expression of IL-1β in platelets in RA group(41±11) was higher than control group(21±8)(t=9.01, P =0.000) .The platelet activation in high-active RA group(22 ±4) was higher than mediate/low active RA group(17 ±4)(t =3.96,P =0.001). The expression of IL-1β in platelets in high-active RA group(45 ±10) was higher than mediate/low active RA group (38 ±10)(t =2.329,P =0.024). The expression of IL-1β in platelets in RA group was positively correlated with the level of ESR、CRP and DAS28 (r value and P value were 0.576, 0.578, 0.618 and 0.000, 0.000, 0.000 respectively). Conclusion The platelets of patients with RA are activated and may suggest that IL-1β, which may associate with disease activity. Our research suggest that platelet may play a role in the inflammatory process of RA by secreting IL-1β.
9.The photodynamic effects mediated by upconversion nanoparticles on rat astrocytes in vitro

Objective To investigate the phototoxicity effects of the nanocompound of upconversion nanoparticles (UCNPs) on rat astrocytes in vitro.Methods The spinal astrocytes cells were cultured successfully in vitro and then incubated with the UCNPs-MC540 of various concentrations and exposured 980 nm infrared laser irradiation of different energy densities.The cell survival rates of each group were detected by MTT assay.The cellular morphology was observed via transmission electron microscope after photodynamic therapy.Results UCNPs-MC540 of different concentrations without laser irradiation or laser of different energy had no significant effects on cell survival rates.when cells incubated with 100 μg/ml UCNPs-MC540 for 12 h underwent laser irradiation of different energy,the cellular survival rates significantly decreased with the increased energy densities.when the cells incubated with UCNPs-MC540 of various concentrations for 12 h underwent laser irradiation of 2 000 J/cm2,the cellular survival rates significantly decreased with the increased concentrations.Compared with controls,the TEM show the apoptosis sign in the cells incubated with 200 μg/ml UCNPs-MC540 after laser irradiation of 2 000 J/cm2.Conclusion The UCNPs-MC540 mediated photodynamic therapy have effective killing effect on astrocytes by the mechanism of induction the apoptosis.
10.Synthesis method optimization and biodistribution study of 18F-T807 on TRACERlab FXFN synthesizer

Objective To optimize the synthesis method of 18F-T807 and study preliminary biodistribution. Methods 18F-T807 was synthesized using an optimized method in TRACERlab FXFN synthesizer with a t-BOC(t-Butyloxy carbonyl)-protected 18F-T807 precursor NPPI-9 as starting material, improving experimental conditions for synthesis, then QC and biodistribution study in Wistar rats conducted. Results The improved synthesis conditions increased the synthesis yield from 20.5%±6.1% to 25.7%±5.8%. QC met the standard. Wistar rats had higher intake in kidney, liver, blood and lowest intake in brain, heart, lung. Conclusion The optimized synthesis method to synthesize 18F-T807 is simple and easy, and high yield, which can meet the needs of scientific research and clinical practice.
