BACKGROUND:Stem cel therapy is superior to drug therapy for recovery of patient’s physiological mode, and cel transplantation therapy is becoming a trend.
OBJECTIVE:To observe the changes in dopamine content in the stratum of Parkinson’s disease rats after transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s.
METHODS:After identification by enzyme digestion, pEGFP-C2-TH plasmid was transfected into the fourth generation of human umbilical cord blood mesenchymal stem cel s by electroporation method, and then transfected cel s were injected into the right cerebral ventricle of Parkinson’s disease rats (experimental group). PBS injection was performed in the control group. Migration of dopamine in the brain tissue of rats was observed, and the content of dopamine was detected by high performance liquid chromatography.
RESULTS AND CONCLUSION:At 8 weeks after cel transplantation, the cel s gradual y migrated to the ventricles;after 12 weeks, the cel s migrated to the cortex, and expressed tyrosine hydroxylase antigen. Meanwhile, the content of dopamine was significantly higher in the experimental group than the control group (P<0.05). These results reveal that the intraventricular transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s has obvious therapeutic effect on Parkinson’s disease rats.

Objective: To evaluate the effectiveness that gelatin lines the injured vasuclar wall to prevent from restenosis.Methods: The animal model of restenosis was established by balloon dilation in femoral artery,and 30% gelatin was lined on injured vascular wall in experimental group.The samples were collected at the 2nd week and 4th week after operation.The neointimal area(NEA),neointimal area/media area(NEA/MA)and cell apoptosis were observed by image analysis system and TUNEL technique. Results: The intima of the control group was thicker than that of the experimental group,and more cells appeared in the control group.Compared with the control group,the NEA was reduced significantly in the experimental group at the 2nd and 4th week (0.67?0.25cm 2 vs 1.02?0.41cm 2,and 1.18?0.36cm 2 vs 1.46?0.15cm 2).The NEA/MA ratio in the experimental group was lower than the control group at the 2nd and 4th week(0.35?0.05% vs 0.54?0.14%,and 0.62?0.25% vs 0.75?0.10%).The apoptosis rate of vascular smooth muscle in the experimental group was higher than the control group at the 2nd and 4th week(18.2?5.8% vs 13.6?2.4%,and 15.1?2.1% vs 13.0?1.5%).These diffences were all significant (P

Objective To compare the retromolar space (RMS) between different antero-posterior (A-P) skeletal patterns, and explore the relationship between the status of third molar eruption/impaction with the different A-P skeletal patterns. Methods A total of 90 orthodontic patients (25-35 years old) were investigated from dental pantomograms (DPTs) and lateral cephalograms (LC). The subjects were divided into three groups according to their ANB angle:skeletal classⅠ, skeletal classⅡ, and skeletal classⅢ. Each group was subdivided into impacted and erupted subgroups. Values of DPT and LC were traced, and the following variables were measured including mandibular body length, third molar angulation and ret-romolar space width. Results The retromolar space width was significantly smaller in classⅡsubjects than that in classⅢsubjects, and RMS was significantly smaller in impacted group than that in erupted group, respectively (P<0.05). There was no significant difference in the mandibular angle in erupted subgroup (P<0.05). There was a smaller mandibular body length in impacted subjects (P<0.05). Conclusion There is a relationship between different morphological characteristics of sagittal skeletal pattern and the impacted status. Patients with shorter mandibular body length and decreased RMS width are more inclined to impaction.

Objective To investigate the irradiating effect of low intensive microwave(LIM) on pathological process of blood vessel restenosis(RS) and assess the probability of LIM irradiation to prevent was used RS.(Methods Forty-four) male healthy New Zealand rabbits were randomly divided into 2 groups.Fogarty catheter traumatize to the tunica intima of iliac artery so as to establish RS models.Two thousand four hundred and fifty MHz microwave with different power of 2,5 and 10 mW/cm~2 was used,locally to irradiate EIA in irradiating group(1 h/d).Specimens were obtained at different time of 3,7,14 and 28 d after operation.Morphological changes of tissues were observed with HE and EF staining and the area of tunica intima,tunica media and the rate of cavity stenosis were analyzed with image analysis system;apoptosis was detected with TUNEL;phenotype and microstructure of VSMC were observed with TEM.Results After microwave irradiating,inflammatory reaction in early period was suppressed,mural thrombus decreased,the proliferation and migration of VSMC depressed,the area of tunica intima and the rate of cavity stenosis obviously reduced comparing with the control group(P0.05).At other different time,however,the rate of apoptosis cells in irradiating groups obviously increased than that of the control group(P

Objective Finding out the kinds and activities in protease of brain,heart,lung,kidney,eyes,skin and muscle of Andrias davidianus. Methods Using protease returned electrophoresis technique(G-PAGE). Results 1.Activity of proteases in brain was weak at pH 4.5,and showed no activity at pH 7.0 and pH 9.5;2.Proteases of heart and lung had activities at pH 4.5 and pH 7.0;3.Activity of kidney proteases was strong at pH 4.5 and stronger at pH 7.0,and kinds of proteases were more at pH 7.0 than at pH 4.5;4.Proteases in eyes had no activity at pH 4.5 and pH 9.5;5.Activity of proteases in muscle was very strong at pH 7.0 and pH 9.5,and kinds of proteases were more at pH 7.0 than at pH 9.5;6.Activity of skin proteases was similar to that of muscle,but the former was weaker than the later. Conclusions Among the seven kinds of tissues and organs of Andrias davidianus,kinds and activities of kidney and muscle proteases were stronger and more than others;and the eyes showed almost no protease activity.The proteases-activity optimal pH of brain was acidic;The proteases-activity optimal pH of heart,lung and kidney was acid of neutral;and the proteases-activity optimal pH of skin and muscle was neutral.

Objective To analyse the experience and treatment of early phase severe acute pancreatitis (SAP) in intensive care units (ICU).Methods A multicenter retrospective study was done on patients with SAP treated in three major teaching hospitals (Beijing Hospital,Peking University First Hospital and Peking University Shenzhen Hospital) in China from Jan.2001 to Dec.2011.Results There were 188 patients who were enrolled in the study,including 121 males and 67 females.The age ranged from 19 to 104 (51.0±18.2) years.The mean APACHE Ⅱ score was (22.2±4.6).84.0％ of patients survived,the mortality was 10.1％ in the early phase and 5.9％ in the late phase.The most common systemic complications were acute renal injury (46.3 ％),acute respiratory distress syndrome (35.6％),and septic shock (17.6％).The local complication rate was 47.3％,which included acute peripancreatic fluid collections (32.8％),acute necrotic collection and walled-off necrosis (48.4 ％) and pseudocyst (18.8 ％).The conservative treatments included intensive care,fluid resuscitation,mechanical ventilation,continuous renal replacement therapy,antibiotics,glucose control,inhibition of pancreatic enzyme activity and secretion,and nutritional support.Surgical intervention included endoscopic retrospective cholangio-pancreatography and endoscopic sphincterectomy,B ultrasound or CT guided puncture and drainage,and surgical drainage and debridement of necrosis.Conclusions The early phase of SAP was characterized by systemic inflammatory response syndrome and multiple organ dysfunction syndrome which accounted for the first peak in mortality.Intensive care therapy and multi disciplinary comprehensive combined strategy were very important for these patients with systemic and local complications.ICU treatment in the early phase was preferred for patients with SAP.

Subjective To acquire and analyze adult stage Schistosoma japonicum (Chinese strain) expressed sequence tags and new genes from an adult S. japonicum cDNA library, and to search new vaccine candidates and drug targets. Methods A cDNA library was constructed from adult stage S. japonicum. Clones were selected randomly from the cDNA library and were se-quenced. ESTs and new genes were acquired after analysis in GenBank databases by BLAST and other programs. All ESTs and new genes were submitted to GenBank and received accession numbers. Results 149 ESTs were acquired from a total 382 clones that were randomly selected from the adult S. japonicum cDNA library. All ESTs were successfully submitted to the dbEST at Genbank. Some of them were homologous with sequences of male, female, egg, schistosomula, cercaria and miracidia of S. japonicum. 18 new genes of adult S. japonicum were acquired. Some genes were housekeeping genes and some genes might be interesting as vaccine candidates or drugs targets. Conclusions The EST straltegy is a rapid, efficient and economical method to acquire ESTs and to discover new genes of adult stage S. japonicum from cDNA libraries.[

Objective To observe the changes of postprandial blood pressure and heart rate with two types of nasal feeding in the very elderly and explore the method for decreasing the postprandial hypotension (PPH).Methods Totally 49 patients (aged＞80 years) were randomly divided into two groups of continuous versus intermittent nasal feeding.The clinical symptoms and changes in blood pressure and heart rate before and after meals were observed.Results The incidence of PPH was 100.0％(26 cases)in the group of intermittent nasal feeding,and 47.8％ (11 cases) in the group of continuous nasal feeding (P＜0.01).Postprandial blood pressure in the group of intermittent nasal feeding was decreased significantly as compared with the group of continuous nasal feeding (P ＜0.05); and the heart rate was accelerated in the group of intermittent nasal feeding compared with continuous nasal feeding (P＜0.05).The incidence of serious cardio-cerebral ischemia was 7.7％ (2cases)in the group of intermittent nasal feeding.The patients had no clinical symptoms in the group of continuous nasal feeding.Conclusions Continuous nasal feeding for the very elderly with gastrointestinal nutrition can reduce the extent of the falling blood pressure and the incidence of PPH,and avoide cardiocerebrovascular events.
