3.Expression and clinical significance of CD40 protein in human bladder cancer tissue

Objective To investigate the expression of CD40 protein in bladder urothelium cancer and to explore its correlation with clinical stage, pathological grades, cell apoptosis. Methods Expression of CD4o protein was evaluated by immunohistochemistry in paracancerous tissues (20 cases) and bladder urothelium cancer tissues (78 cases). Apoptosis of bladder urothelium cancer tissues was detected by Hoechst to determine the apoptotic rate (AR). Results The expression rate of CD40 protein was 2/20(10 %) in the cases of paracancerous tissues, 55/78(70.5%) in the cases of bladder urothelium cancer tissues, a statistically significant differences of CD40 staining was observed between paracancerous tissues and bladder urothelium cancer tissues (P <0.01). Overexpression of CD40 was strongly inversely related to the tumor grade and clinical stage; the positive group of CD40 protein was (12.60±0.38)%, the negative group of CD40 protein was (6.77±0.53)%, a statistically significant differences of apoptosis were observed between CD40 protein positive group and CD40 protein negative group (P <0.01). It showed CD40 protein might induce cell apoptosis. Conclusion Expression of CD40 protein is closely related with clinical stage, pathological grade and cell apoptotic rate and may offer reliable evidence for diagnosis, treatment and prognosis in bladder urothelium cancer.
4.Determination of icariin,psoralen and isopsoralen in Yishen Buxue Capsule by PR-HPLC

AIM:To determine icatiin,psoralen and isopsoralen in Yishen Buxue Capsule(Herba Epimedii;Fructus Psoraleae;Sanguis Draconis;Cortex Eucommiae,etc)by PR-HPLC.METHODS:The determination of icariin was carried out by HPLC with a Agilent HC-18(150 mm?4.6 mm,5 ?m)column,acetonitrile-water(30∶70)was as a mobile phase,detection wavelength was at 270 nm;With the same Agilent HC-18 column for psoralen and isopsoralen,the mobile phase was methanol-water(52∶48),detection wavelength was at 246 nm.RESULTS:Icariin showed a good linear relationship at the range of 0.110 4-0.883 2 ?g(r2= 0.999 8),the avearge recovery of icariin was 97.49%(RSD=1.47%,n=6).Psoralen also showed a good linear relationship at the range of 0.041 04-0.451 44 ?g(r2= 0.999 0),the average recovery for it was 104.51%(RSD= 1.58%,n=6);Isopsoralen showed a good linear relationship at the range of 0.037 52 ?g-0.412 72 ?g(r2= 0.999 9),the average recovery of isopsoralen was 103.01%(RSD=1.01%,n=6).CONCLUSION:The methods are sensitive,accurate,reproducible,specific and can be used for quality control of Yishen Buxue Capsule.

The nature and preparation of sodium alginate are introduced and its medical applications are reviewed in fields such as medical carrier,gel microspheres,sustained-release materials,surgical wound repairing materials,and so on.Besides,its development prospects are described.It is an inevitable trend that sodium alginate is used in the development of controlled-release reagent.There are also important applications in the partial treatment of ear disease and artificial cartilage for sodium alginate,but the strength and toughness of alginate hydrogel limits its wider application.
6.Effects of the Alkaloid Sinomenine on Expression of TGF-?_1 in Rat Renal Allografts

Objective To afford the basis for the application of Sinomenine(SIN) in the proposal for organ transplantation, the expression of TGF-? 1 was explored in inborn WistarSK kidney transplantation model. Methods Forty-eight recipients were divided into 4 groups at random:group 1 being used as control and treated with 0 9% natrii chloride(3ml?kg -1 ?d -1 ,ip), group 2 treated with SIN(30mg?kg -1 ?d -1 ,ip),group 3 treated with CsA(2 5mg?kg -1 ?d -1 ,ip),group 4 treated with SIN(30mg?kg -1 ?d -1 ,ip) combined with CsA(2 5mg?kg -1 ?d -1 ,ip).Allografts of 6 recipients from each group were harvested at day 6 before end stage rejection for histological studies. The synergistic effects of SIN combined with CsA were assessed by COX proportional hazards model. The quantitative methods of immunohistochemistry were used for analysis on expression of TGF-? 1.Results Control recipients rejected their allografs at day 9 post-operation, the average life time was 7 83?9 75 days. Treated with either SIN(30mg?kg -1 ?d -1 ,ip) or subtherapeutic doses of CsA(2 5mg?kg -1 ?d -1 ,ip) allografts survival was mininally prolonged. Combined treatment with SIN and subtherapeutic doses of CsA led to a significant prolongation of graft survival(group 4), more than 18 days. The expression of TGF-? 1 was enhanced significantly in group 2 and group 3.No synergistic effect of SIN combined with CsA on expression of TGF-? 1 was observed. Conclusion The therapeutic value of SIN in rat kidney allograft model suggests that this safe chinese medicine has remarkable efficacy of anti-acute rejection in rat kidney transplantation. It shows significant synergistic effect with sub-therapeutic CsA. The immunosuppressive efficacy of SIN is probably based on enhancing express of TGF-? 1. No synergistic effect of SIN and CsA on expression of TGF-? 1 indicates low danger of fibrosis of allografts. SIN will be a hopeful chinese medicine for immunosuppressive treatment in organ transplantation.
7.Determination of Polysaccharides in Hedyotic Diffusa from Differents Habitat

Objective To establish a method to determinate polysaccharides in Hedyotic diffusa, then compare the polysaccharides content in Hedyotic diffusa form Jiangxi, Guangdong, Guangxi. Methods Sulfuric acid -phenol coloration were applied to analysis polysaccharides content. The detective wave length was 486 nm. Results Polysaccharides in Hedyotic diffusa from Jiangxi province was a little higher than the others. Conclusion Polysaccharides content in Hedyotic diffusa form different habitat are almost alike.
8.Percutaneous nephrolithotomy (PCNL) with the single channel double sets of catheter sheath method in the management of complicated ureteric calculus

Objective To evaluate the effect of the percutaneous nephrolithotomy (PCNL) with the single channel double sets of catheter sheath method in the management of complicated ureteric calculus. Methods The clinical data of forty-eight cases with complicated ureteric calculus were retrospectively investigated. Among them, 18 cases with ipsilateral kidney calculus, 1 case with repeat renal deformity, within the ureteral calculus in upper kidney. Results 48 patients smoothly under ulime from ultrasound guided puncture percutaneous renal channel is established successfully, The establishing the channel to setting the double sheath to the ureteropelvic junction was (18.0 ± 5.0) min, the operation time was (59.0 ± 6.0) min, the stone free rate was 96.6% and no significant complications, such as septic shock, intraoperative or postoperative bleeding and damage to surrounding organs, were detected. The patients were followed up for 3 to 6 months, the degree of hydronephrosis significantly reduced and no ureteral stricture were developed during the period. Conclusion The percutaneous nephrolithotomy (PCNL) with the single channel double sets of catheter sheath method is safe and effective for complicated ureteral calculus.

10.Experimental study on preparation and targeting ability in vitro of HCC-targeted lipid microbubble containing 10-HCPT

Objective To prepare HCC-targeted lipid ultrasound microbubble containing 10-hydroxycamptothecin (10-HCPT), and to assess its targeting function in vitro. Methods After the biotinylated monoclonal antibody Hab18 was prepared, the biotinylated degree was determined. Microbubbles containing 10-HCPT were prepared by mechanical vibration. Then the biotinylated antibody was attached to the surface of the microbubbles by avidin-biotin interaction. The physical property, entrapment efficiency and the drug-loading amounts of 10-HCPT lipid microbubbles were determined. The combination of biotinylated Hab18 with microbubbles containing 10-HCPT was proved by immunofluorescent assay, and the targeting function of the targeted microbubbles containing 10-HCPT was observed with light microscope, served non-targeted microbubbles as control group. Results About 13 biotin molecules were coupled to each antibody in average. The disposition of the prepared microbubbles was steady and the mean diameter was 1.52 μm. The drug entrapment efficiency was 76.32% and the drug-loading amounts was 21.81%. Red fluorescence was observed at the edge of the microbubbles in immunofluorescent assay, and the conjugation of the targeted microbubbles containing 10-HCPT with 7721 cells was tight while control group was negative. Conclusion HCC-targeted lipid ultrasound microbubbles containing 10-HPTC can be prepared successfully, with higher entrapment efficiency and drug-loading amounts and strong targeting function in vitro.
