1.Dopamine content in the stratum of Parkinson’s disease rats after transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cells

BACKGROUND:Stem cel therapy is superior to drug therapy for recovery of patient’s physiological mode, and cel transplantation therapy is becoming a trend.
OBJECTIVE:To observe the changes in dopamine content in the stratum of Parkinson’s disease rats after transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s.
METHODS:After identification by enzyme digestion, pEGFP-C2-TH plasmid was transfected into the fourth generation of human umbilical cord blood mesenchymal stem cel s by electroporation method, and then transfected cel s were injected into the right cerebral ventricle of Parkinson’s disease rats (experimental group). PBS injection was performed in the control group. Migration of dopamine in the brain tissue of rats was observed, and the content of dopamine was detected by high performance liquid chromatography.
RESULTS AND CONCLUSION:At 8 weeks after cel transplantation, the cel s gradual y migrated to the ventricles;after 12 weeks, the cel s migrated to the cortex, and expressed tyrosine hydroxylase antigen. Meanwhile, the content of dopamine was significantly higher in the experimental group than the control group (P<0.05). These results reveal that the intraventricular transplantation of tyrosine hydroxylase-modified human umbilical cord blood mesenchymal stem cel s has obvious therapeutic effect on Parkinson’s disease rats.
2.Correlation between G919A polymorphism of cystathionine β synthase and breast cancer in Han nationality in Xinjiang

Objective To investigate the relationship between breast cancer and G919A polymorphisms of CBS (cystathionine β synthase) in patients with breast cancer in Han nationality in Xinjiang. Method Three hundred breast cancer patients and 300 normal controls of Han nationality were genotyped by ARMS method, and then the relative biochemical indicators were detected by the automatic biochemical analyzer. Results The Hcy level of breast cancer patients were higher than that of control group (P < 0.05). Genotypes GG, GA, AA of CBS G919A and allele G,A, in breast cancer patients were 63.7%, 24%, 12.3% and 75.7%, 24.3% respectively, which had no statistically significance when compared with those of the controls (P > 0.05). Conclusion The high level of Hcy may be a risk factor for breast cancer in Han nationality in Xinjiang and there was no relationship between CBS G919A polymorphism and breast cancer.
3.Comparison of Immediate Therapeutic Effects Between Endovenous Laser Treatment and Conventional Surgical Therapy for Lower Extremity Varicosity

Objective To compare the immediate therapeutic effects between endovenous laser treatment and conventional surgery for lower extremity varicosity.Methods Data of 350 limbs of 275 patients treated by endovenous laser treatment (endovenous laser treatment group) and 310 limbs of 210 patients by traditional surgery (traditional surgery group) were analyzed and compared in terms of operation time,number of the incision,postoperative pain,complications,postoperative hospitalized duration and 1-year recurrence rate.Results Endovenous laser treatment group had shorter operation time,fewer incisions,less postoperative pain and shorter postoperative hospitalized duration than traditional surgery group (P0.05).Conclusion As a safe and effective new treatment for lower extremity varicosity with minimal invasiveness and no left scars,endovenous laser treatment has the potential to replace conventional surgery and extend the surgical indications for lower extremity varicosity treatment.
4.Diagnosis and treatment of radial nerve injury following internal fixation for humeral shaft fracture

Objective To explore the etiology and treatmen t of radial nerve injury following in ternal fixa-tion for humeral shaft fracture.Methods 15patients were treated in our department from 1997to 2003for radial nerve injury following steal-plate internal fixation.They were selected for this study and followed up from6to 18months.Their injury cause,clinica l manifestation,methods of treatme nt,diagnosis and prognosis were retrospec-tively analyzed.Results The symptoms of nerve injury disappeared in all patients.Their muscle strength of wrist and digit extension returned to grad e V.The electromyogram showed that t heir nerve conduction velocity and l atent period returned to normal.Conclusions Excessive traction,careless manip ulation and steel plate compression are the main causes for iatrogenic radia l nerve injury.The nerve function can be completely recovered with early diag-nosis and early neurolysis.[
5.Allograft cancellous bone combined with autologous red marrow for treatment of periarticular fractures

BACKGROUND:Bone defects often occur after the reduction of periarticuar fractures, and bone grafting is required to fil bone defects, thereby to make the early support of articular surface and prevent the col apse and shift of the articular surface. Al ograft cancellous bone is the transplant material for the treatment of bone defects, but its osteogenesis ability is poor. Autologous red marrow has osteogenesis ability. Therapeutic efficiency of al ograft cancellous bone combined with autologous red marrow for periarticular fractures is stil yet to be
assessed.
OBJECTIVE:To study clinical effect of locking plate fixation and al ograft cancellous bone combined with autologous red marrow in the treatment of periarticular fractures.
METHODS:Forty-three cases of periarticular fractures were incorporated into the Orthopaedics Department of the Third Hospital of Hebei Medical University. After cutting the articular surface anatomical y, composite particles of the red marrow and al ograft cancellous bone were implanted into bone defects, and then, an anatomic locking plate was used. Medial lateral or bilateral locking plate was used for tibial plateau fractures. Dorsal or volar locking plate was used for distal radius fractures, and distal tibial medial or lateral locking plate was used for distal tibial fractures.
RESULTS AND CONCLUSION:Forty-three patients were fol owed up for 12 months to 6 years, an average of 4.3 years. X-ray films and CT scans review showed that 43 patients’ col apse fracture al reached bony union. Fresh fracture healing time was 2-6 months, an average of 4 months;the healing time for old fracture was 3-7 months, an average of 5.5 months. After bone grafting, 43 patients al had no significant immune rejection, two cases showed more wound exudates, and the wound was healed by dressing after 2 weeks. One case had wound infection, the wound was healed after 4 weeks of draining and dressing, and the infection had no recurrence fol owing up for 4 years and 1 month. Forty patients were satisfied with bone grafting, accounting for 93%, and three cases dissatisfied, accounting for 7%, based on Mankin and Komender’s standard assessment. The results confirmed that al ograft cancellous bone combined with autologous red marrow transplantation can play a supporting role in treating periarticular fractures to prevent the col apse of the articular surface and fracture displacement, and to provide reconstruction materials for periarticular defects. Its long-term goal is fracture healing.
6.Anti-apoptosis effect of ligustrazine on the apoptosis of PC12 cells induced by dopamine

Objective To explore the protective effect of ligustrazine on the apoptosis of PC12 cells induced by dopamine(DA) and its mechanisms. Methods Apoptosis of PC12 cell was induced by dopamine at the concentrations of 0, 0.30, 0.60 mM. Ligustrazine at the concentrations of 0, 30, 60, 90 ?g/ml was added to defect the effect of ligustrazine on apotosis. The subdiploid peaks showing cell apoptosis rate were detected by flow cytometry. The cell activity was determined by MTT. Concentration of nitric oxide(NO) was determined by the Griess reaction. Results Compared with those of the control, the apoptosis rate and the level of NO induced by 0.30 or 0.60 mmol/L DA were higher, but the activity of PC12 cells was lower. Compared with those of the groups without ligustrazine, the apoptosis rate and the level of NO increased, but the cell activity decreased in a dose dependent manner. Conclusion Ligustrazine can inhibit the apoptosis induced by DA, which might be correlated with the decrease of NO production.

8.Relationship between Hcy and CBS-T833C gene polymorphism and breast cancer in Han of Xinjiang Population

Objective To investigate the correlation between T833C gene polymorphism of cystathionine-βsynthase (CBS) and breast cancer in Xinjiang Han nationality. Methods CBS-T833C gene polymorphism of 200 breast cancer patients in Xinjiang Han nationality (breast cancer group) and 200 health examination people (control group) was analyzed by amplification refractory mutation system (ARMS) assay, Hcy levels and biochemical parameters were detected by automatic biochemical analyzer , the correlation between CBS-T833C gene polymorphism and breast cancer in Xinjiang Han nationality was analyzed. Results 1. Genotype TT,TC,CC of CBS T833C gene in the breast cancer group and the control group were respectively 55.5%, 32.0%, 12.5%and 53.0%, 34.0%, 12.0%. The G, A allele frequency of CBS T833C gen in the two groups were 71.5%,28.5%and 70.0%, 29.0%,the overall frequency distribution between the two groups was not statistically different among the three genotypes (P > 0.05). 2. Hcy levels in plasma of TC, CC genotype were higher than TT genotype in breast cancer group (P <0.05). Conclusion Increase of homocysteine levels may correlates with breast cancer in Xinjiang Han nationality, CBS-T833C gene polymorphism has no correlation with breast cancer in Xinjiang Han nationality.
9.Effects of tumor suppressor NDRG2 gene on proliferation and invasive ability of colon cancer cell line SW620

Objective To observe the influence of N-myc downstream-regulated gene 2 (NDRG2) on the growth and invasive ability of human colon cancer cell line SW620,and to explore its mechanism.Methods pcDNA3.1-NDRG2 and siRNA-NDRG2 were transfected transiently respectively into SW620 by Lipofectamine TM 2000,untreated cells as the control group.Western blotting was used to investigate the expression of NDRG2 and matrix metalloproteinase-2 (MMP-2).Matrigel invasion assay was used to study the invasive abilities of SW620 cells in all groups.The growth curve was determined through 3-(4,5-dimethyl-2-thiazoly)-2,5-diphenyl-2H-tetrazolium bromide method.Result After transfecting pcDNA3.1-NDRG2 into the SW620 cells,the protein level of NDRG2 increased and the expression of MMP-2 declined markedly.After transfecting siRNA-NDRG2 into the SW620 cells,the protein level of NDRG2 declined and the expression of MMP-2 increased markedly.In addition,compared with the control group (75.80 ± 4.82),the numbers of transmembrane cells in pcDNA3.1 group (56.20 ± 7.40) and in siRNA group (94.20 ± 9.23) were significantly different (t =13.102,P =0.000;t =11.820,P =0.000).The growth curve showed that:compared with the control group (0.67 ±0.01),the absorbance of the fifth day after transfection in pcDNA3.1 group (0.46 ±0.01) and in siRNA group (0.91 ± 0.02) were different significantly (t =9.561,P =0.000;t =10.922,P =0.000).Conclusion NDRG2 can reduce the invasion and proliferation ability of colon cancer cell SW620,and its mechanism may be related to the down-regulation of MMP-2 expression.
10.MRI of Acute Pancreatitis

Objective To assess the value of magnetic resonance imaging for acute pancreatitis.Methods 77 patients of acute pancreatitis were examined with holding breath MRI sequence.Transverse scan:2D Flash T 1WI/T 1W+FS,TSE T 2WI+FS or HASTE+FS,Trufisp.Coronals scan:2D Flash T 1W+FS.MRCP was used in 34 patients.2D Flash dynamic contrast enhanced were used in 77 patients.Results 59 cases of acute edematous pancreatitis showed swelling and low signal intensity on T 1WI and higher signal intensity on T 2WI.Excepted the above findings,18 cases of hemorrhagic necrosis pancreatitis alos showed the higher signal intensity on T 1WI and peripancreatic exudative hydrops.Conclusion MRI is a reliable method for demonstrating acute pancreatitis.
