BACKGROUND: Some researches indicated that the acupuncture treatment, especially at Sanjiao meridian might be effective for otopathy. However, the mechanism is not clear yet.OBJECTIVE: To observe the hearing improvement of the cavies induced by gentamicin after the animals received the acupuncture at Sanjiao meridian acupoints: Yifeng (SJ 17), Ermen (SJ 21) and Zhongzhu (SJ 3).DESIGN: Complete randomization.SETTING: Department of Neurology, Xuanwu Hospital, Capital University of Medical Sciences.MATERIALS: Sixty healthy white male cavies of 3-4 months old, common grade, 250-300 g weight and normal auricle reflex were selected and randomly arranged into 5 groups: normal group, model group, acupuncture groups of Yifeng (SJ 17), Ermen (SJ 21) and Zhongzhu (SJ 3), with 12 ones in each.METHODS: The experiment was conducted at the Laboratory of Acupuncture and Moxibustion, Beijing University of Chinese Medicine from January to July in 2001. ①The normal cavies were bred at normal conditions, while other cavies were injected intramuscularly with 80 mg/kg gentamicin every day, continuously for 20 days. At the same time of gentamicin injection, Yifeng (SJ 17), Ermen (SJ 21) and Zhongzhu (SJ 3) were selected for acupuncture treatment in the corresponding groups. The fixed cavies were pricked subcutaneously with 0.35 mm×25 mm stainless steel acupuncture needles for 10 mm straight depth. The needles were twisted with mild reinforcing-reducing method once in half a minute, repeated every 10 minutes, and remained for 30 minutes one day for 20 days.②Before and after the intervention, auditory brain-stem response (ABR) was detected with 7S11A evoked potential apparatus, in which the sound source was input by earphone for measuring the threshold value of ABR in cavies. ③The difference of measurement data were compared with t test between groups.MAIN OUTCOME MEASURES: The effect of acupuncture treatment at Yifeng (SJ 17), Ermen (SJ 21) and Zhongzhu (SJ 3) on the ABR threshold in cavies injured by gentamicin.RESULTS: Totally 60 cavies were involved in the result analysis. There was insignificant difference in the ABR threshold of each group before the intervention (P > 0.05); After the intervention, the ABR threshold was obviously lower in normal group than in other 4 groups (P < 0.01), and also obviously lower in 3 acupuncture groups than in model group (P< 0.01),and especially in Yifeng group than in model group.CONCLUSION: The acupuncture treatment at Ermen (SJ 21), Zhongzhu (SJ 3), and especially at Yifeng (SJ 17) acupoints can relieve the hearing impairment in the cavies due to drug toxicity.

Objective To investigate the effect of astaxanthin( ASX)on endothelial progenitor cells( EPCs)injury induced by oxidative stress in vitro and to explore its underlying mechanism. Methods Cultured EPCs isolated from peripheral blood were randomly divided into 5 groups:normal control, model group[ tert-butyl hydroperoxide( tBHP)100μmol·L-1 ],and ASX+tBHPgroups(thecellswerepreconditionedwithASX0.1,1.0,and10.0nmol·L-1,respectively).Thecellviabilitywas measured by MTT method. The level of reactive oxygen species( ROS)was determined by DCFH-DA method. The changes of mitochondrial membrane potential( MMP)and apoptosis ratio were detected by JC-1 method and DAPI method, respectively. caspase-3 activity changes of EPCs were detected. Results The cell viability of EPCs was improved with the increasing concentration of ASX. Compared with the model group[(48. 5±4. 3)%],0. 1,1. 0,10. 0 nmol·L-1 ASX significantly increased the cell viabilities[(57. 6±8. 2)%,(77. 6±7. 5)%,and(85. 3±6. 1)%,P﹤0. 05]. The results of DAPI staining revealed that ASX pretreatment could significantly reduce the apoptotic rate of EPCs. The apoptotic rate of the model group was( 27. 8 ± 3. 2)%,while that of ASX+tBHP groups was[(20. 4±2. 9)%,(14. 9±1. 7)%,and(7. 8±0. 7)%,P﹤0. 05],respectively. The data from caspase-3 activity assay indicated that ASX precondition could also remarkably decrease the caspase-3 activity for EPCs. The caspase-3 activity of the model group was(0. 345±0. 018),while that of the ASX+tBHP group were[(0. 291± 0. 013),(0. 209±0. 004),and(0. 169±0. 013),P﹤0. 05],respectively. In addition, treatment with tBHP resulted in an increase of DCF fluorescence, while ASX precondition could decrease the DCF fluorescence, which suggested the accumulation of intercellular ROS for EPCs. Injury of michondrial membrane resulted in the loss of mitochondrial membrane potential( MMP). The MMP detected by JC-1 method revealed that compared with model group, pretreatment of ASX inversed the reduction of MMP. Conclusion Astaxanthin inhibits endothelial progenitor cell apoptosis induced by oxidative stress through inhibiting ROS production, improving the mitochondrial function and down-regulating caspase-3 activity.

Objective To evaluate the modification of C_(60) on the radiation effects of ~(60)Co-γ irradiation on zebrafish. Methods The adult and embryonic zebrafish were used as model organisms to examine the potential of C_(60) to elicit oxidative stress responses on the surviving rate, hatching rate and malformation occurrence, both upon exposure to light or in the dark.Reactive oxygen species (ROS) production and DNA damage were examined as the possible underlying mechanism. Results 500 × 10~(-9) nano-C_(60) waterborne exposure could enhance the γ-irradiation effects by decreasing adult fish survival upon light exposure, which resulted in ROS and DNA damage increasing. The hatching rates were also inhibited with higher malformation, though dark exposure did not make any enhancement, except that the 5000× 10~(-9) C_(60) would inhibit larvae hatching and induced more malformation. Conclusions Waterborne nano-C_(60) exposure may enhance the radiation effects on zebrafish,ROS production and DNA damage increasing may be the underlying mechanism.

Journal of Preventive Medicine 2024;36(2):119-122
Objective:
To investigate the awareness of hepatitis C prevention and control knowledge and its influencing factors among female sex workers (FSWs) in Jiaxing City, Zhejiang Province.
Methods:
According to the HIV/AIDS Sentinel Surveillance Plan, FSWs at ages of 15 to 65 years monitored by the national AIDS surveillance sentinel in Jiaxing City were recruited, and demographic information, awareness of hepatitis C prevention and control knowledge and related behaviors were collected by questionnaire surveys. Factors affecting the awareness of hepatitis C prevention and control knowledge were identified by a multivariable logistic regression model.
Results:
A total of 430 questionnaires were allocated, and 412 were valid, with an effective rate of 95.81%. The respondents had a mean age of (28.58±4.93) years, and included 258 unmarried FSWs (62.62%), 344 with registered residence outside Zhejiang Province (83.50%), 212 with junior high school education or below (51.46%) and 243 from high-end entertainment places (58.98%). The overall awareness of hepatitis C prevention and control knowledge among FSWs was 20.39%, and the awareness of "Transfusion of blood containing hepatitis C virus may acquire hepatitis C" (38.83%) and the awareness of "tattooing, eyebrow tattooing and ear piercing in streets or small shops may infect hepatitis C" (38.11%) were relatively low. Multivariable logistic regression analysis identified marital status (divorced or widowed, OR=0.161, 95%CI: 0.054-0.482), educational level (high school or technical secondary school, OR=2.568, 95%CI: 1.446-4.560; junior college or above, OR=6.110, 95%CI: 2.658 -14.045) and grade of entertainment places (high-end entertainment places, OR=2.756, 95%CI: 1.525-4.982) as factors affecting the awareness of hepatitis C prevention and control knowledge among FSWs.
Conclusion
FSWs in Jiaxing City have a low awareness of hepatitis C prevention and control knowledge, especially lacking of knowledge about the transmission routes and prognosis of hepatitis C.

Objective:
To evaluate the protective effect of influenza vaccine among people aged 70 years and older in Jiaxing City, Zhejiang Province, so as to provide a basis for formulating immunization strategies.
Methods:
The influenza-like illness (ILI) cases aged 70 years and older treated in influenza surveillance sentinel hospital in Jiaxing City from November 2022 to May 2023 were selected. The medical information and influenza vaccination information were collected by a questionnaire survey, and influenza virus was detected using the quantitative fluorescent real-time PCR assay. The test-negative design case-control study was used to analyze the influencing factors of influenza virus positive and evaluate vaccine effect (VE).
Results:
Totally 1 084 ILI cases were enrolled, including 535 males (49.35%) and 549 females (50.64%). There were 732 cases (67.53%) aged 70 to 79 years, and 352 cases (32.47%) aged 80 years and older. There were 689 cases with underlying diseases, accounting for 63.56%. A total of 224 influenza virus positive samples were detected, with a positive rate of 20.66%. Multivariable logistic regression analysis showed that a lower possibility of influenza virus positive was seen in ILI cases aged 80 years and older, with underlying diseases and with influenza vaccination in the current season (all P<0.05). A total of 345 cases were vaccinated against influenza in the current season, with a vaccination rate of 31.83%. The VE of influenza vaccine was 37.40% (95%CI: 12.40%-55.40%), of which the VE to A (H1N1) was 36.00% (95%CI: 7.50%-55.70%) and to A (H3N2) was 40.90% (95%CI: -26.00%-72.30%). The VE for ILI cases aged 70 to 79 years was 41.00% (95%CI: 13.90%-59.60%), and for ILI cases aged 80 years and older was 20.60% (95%CI: -64.60%-61.70%).
Conclusions
Influenza vaccine has a certain protective effect on cases aged 70 years and older. Free influenza vaccination for the elderly should be continuously promoted and the vaccination coverage should be increased.
