Objective To screen MUC1 antigen mimic epitope by phage display peptide library technology and to construct a recombinant plasmid expressing MUC1 antigen mimic epitope.Methods MUC1 mimic epitope was screened by Biopanning,DNA sequencing and amino acid sequence comparison.The gene was constructed into PET-31b(+) expression vector and expressed in Escherichia coli BL21(DE3) after transformation and induction by IPTG.The complete protein of the host bacteria was extracted for SDS-PAGE.The recombinant protein was purified by affinity chromatography on a Ni~(2+)-sepharose column and detected by Western blotting.Results The selected MUC1 mimic epitope could specifically combine with MUC1 monoclonal antibody.The recombinant expression vector PET-31b(+)-MUC1 was constructed successfully after the fusion protein was induced with IPTG.A specific protein band was shown on SDS-PAGE profile and detected by Western blotting.Conclusion An MUC1 mimic epitope was screened out.The epitope fusion protein was successfully expressed for the development of tumor vaccines targeting MUC1.

Elbow stiffness is a common complication after trauma or surgery to this joint. The soft tissue is a primary factor resulting in stiffness of the elbow. Splints increasing passive range of motion (PROM) with creep or stress relaxation principle play a important role in reconstruction for post-traumatic elbow stiffness. The clinician should follow the basic principle in designing and application of splints. With reasonable resign and appropriate regimen, clinical experience and related research show that splints can improve elbow ROM of post-traumatic elbow stiffness efficiently.

ObjectiveTo summarize the recent therapeutic effects of mesh-plug tension-free hernioplasty in elderly inguinal hernia. MethodsAll cases were performed with mesh-plug case-hardened products, which are manufactured by American Bard Company. ResultsAll the operative procedures were performed smoothly. Postoperative complications were:6 cases of urinary retention,4 scrotal hydroceles,2 persistent wound pain,2 local lightly projections with foreign body sensation,2 hypoincisional haematoma. 1～24 months of follow-up were taken,2 recent recurrences were reported. ConclusionMesh-plug tension-free hernioplasty is a perfect surgical operation. Its main characteristics are:simple performance, less trauma, tension-free, time-saving, good recent therapeutic effect, and fewer recent recurrence. It is especially suitable to the elderly patients and/or Patients with other diseases.

Objective To study the curative effect of continuous positive airway pressure (CPAP) on elderly patients with obstructive sleep apnea-hypopnea syndrome (OSAHS) complicated with diabetes mellitus. Methods Eighty patients with OSAHS and diabetes mellitus were included and randomly divided into treatment group (n=40) and control group (n=40). The control group was given conventional treatment including diabetes diet and hypoglycemic drugs. The treatment group was given Futong ST-25 continuous positive airway pressure (CPAP) ventilation besides the conventional treatment. The serum levels of fasting blood glucose (FBG), two-hour postprandial blood glucose (2 hPG), insulin, sleep apnea hypop-nea index (AHI), low oxygen saturation (LSpO2) and the longest apnea time were monitored before and after treatment in two groups. Results After four-week treatment, values of FPG, 2 hPG, AHI and the longest apnea time were significantly lower in treatment group than those in control group (P < 0.01), but values of insulin, two-hour postprandial insulin and LSpO2 were significantly higher in treatment group than those in control group (P<0.01). Conclusion CPAP therapy can effec-tively decrease blood sugar level and improve AHI, LSpO2 and the longest apnea time in elderly patients with OSAHS compli-cated with type 2 diabetes.

Objective To investigate the effects of RDP1258 on survival of rat cardiac allograft. Methods RDP1258 was synthesized and the model of rat heart abdominal transplantation was established. Animals were divided into four groups. Group 1 received no immunosuppression. Group 2 received CsA alone. Group 3 received RDP1258 alone. Group 4 received RDP1258 and subtherapeutic CsA. In all cases RDP1258 was administrated intraperitoneally and CsA was gavaged. Light and electron microscopic examinations were taken . Transplanted hearts were monitored daily by direct palpation. Results The purity of synthesized RDP1258 was over 95 % and the molecular weight was in accord with theoretical value. The histology and the ultrastructure changed little in grafts in group 3 and group 4. Survival of rat cardiac allograft was significantly prolonged in group 4. Conclusions RDP1258 can suppress acute rejection. Perioperative administration of RDP1258 in combination with CsA can significantly prolong survival of rat cardiac allograft.

Objective To research an ultrasound imaging workstation based on DICOM so as to construct a patient-centered effective digital hospital.Methods Several interfaces of ultrasound imaging workstation based on DICOM were showed and their advantages and disadvantages were analyzed.Ultrasound imaging workstation based on DICOM was also compared with that based on video capture card.Results DICOM-based ultrasound imaging workstation gained advantages over the one based on video capture card in availability,resolution and management.Conclusion The workstation may contribute to enhancing clinical efficiency,promoting digital hospital and regional medical image sharing,and thus is worthy popularizing practically.

Information literacy is the essential literacy in medical education and training of information analysis ability is the difficulty in information literacy education .Described in this paper is the trial to help postgraduates and long-term schooling undergraduates select their study orientation and topics in ChinaMedical University , including their course targets, contents and teaching outcomes.

Objective To compare the efficacy and complications among three surgical treatments of femoral intertrochanteric fractures in elderly patients.Methods Totally 209 elderly patients with femoral intertrochanteric fracture were divided into 3 groups:DHS group[n =115,patients treated with dynamic hip screws (DHS)],PFNA group [n=57,patients treated with proximal femoral nails anti-rotation (PFNA)] and SEFT group [n=37,patients treated with singlearm external fixator technique (SEFT)].The length of hospitalization,operative time,bleeding volume,hip function scores and complications were compared among the three groups.Results All patients were followed up for 5-12 months (10.6 months in average).The length of hospitalization,operative time,bleeding volume,weight-bearing time and Harris hip score in postoperative 3 months were (14.3±2.7) d,(102.4±22.7) min,(240.8±38.4) ml,(50.2±7.7) d,(88.7±12.8) min in DHS group,(11.2±3.4) d,(50.3±15.6) min,(80.5±18.7) ml,(22.6±4.8) d,(92.6±15.4)min in PFNA group,(15.1±3.3) d,(35.8±10.9) min,(52.6±12.9) ml,(55.4±10.1) d,(87.3 ± 12.6) min in SEFT group,respectively.There were significant differences in the length of hospitalization,operative time,bleeding volume,weight-bearing time and Harris hip score in postoperative 3 months among the three groups (all P ＜ 0.05),while there was no significant difference in healing time among the three groups (all P＞0.05).The incidence of complications was highest in DHS group,higher in SEFT group,and lowest in PFNA group (all P＜0.05).Conclusions DHS is more suitable for patients with good physical condition and stable fractures.SEFT is suitable for patients with poor condition,who can not tolerate trauma,anesthesia and bleeding.PFNA has more advantages including stable fixation,less invasive,less complications and wide indications,which is more suitable for femoral intertrochanteric fractures than the other two treatments.
