From the perspective of institute of medical higher education, a set of index system which include three first level indicators has been constructed, for medical academic postgraduate ed-ucation internal evaluation with the method of expert advice as well as in-depth interview, and then through Delphi consulting, the weight of every indicator has been calculated. The weight of three first level indicators are 0.28 (educational condition), 0.42 (educational process), 0.30 (educational results) respectively. Finally, how to carry out internal evaluation and how to apply the results effec-tively have been discussed.

Calcineurin inhibitors, such as cyclosporin A and tacroli mus,are widely-used i mmuno-suppressive drugs clinically. However, renal toxicity, hyperglycae mia and other side-effects can occur over long-term use.Isolation of novel calcineurin inhibitors fro m medicinal plants in recent years has pro-vided a new approach to the development of new immunosuppressive drugs with high efficiency and low toxicity.Recnt studies have shown that medicinal plant extracts, such as isogarcinol, quercetin, kae mpferol, phenylethanoid glycosides, ere mophilane sesquiterpenes A and B,pisiferdiol,as well as extracts fro m Smilax china L and Jasminum humile L,have obvious inhibitory effect on calcineurin.

Calcineurin (CaN) serves as a key enzyme in human immune regulation. The most important target of this enzyme is the transcription factors of nuclear factors of activated T cells (NFATc). The discovery of the immunosuppressive function of CaN inhibitors (CNIs),ciclosporin A (CsA) and tacrolimus (FK506),has helped overcome the immune rejection of organ transplantion and changed organ transplantion fundamentally. Both of these drugs are still widely used in clinical and basic research, but their therapeutic effects are limited by their serious side effects, including renal tox?icity and neurotoxicity. Therefore, the development of new CNIs with higher specificity and fewer side effects in the clinic is a focus of research. In this paper, the newly discovered and synthesized CNIs in recent decades, including the CsA and FK506 derivatives, direct inhibitors of CaN,as well as the inhibitors that specifically interfere with CaN-NFATc interaction, were summarized.

Calcineurin inhibitor screening should benefit from the strengths of each method. This paper reviews three calcineurin inhibitor screening methods:yeast positive screening model, yeast reporter gene high through- put screening model and calcineurin activity detection method. We have compared the range of application, advantages and disadvantages of each method, and combined effect of multiple methods. The yeast positive screening model can prevent the false positiveness caused by bacteriostat by using growth zooms instead of inhibition zooms. The yeast reporter gene high through-put screening model is suitable for pilot screening among a wide range of samples and is more efficient than the other methods. The calcineurin activity detection method can directly report the inhibitory ratio to a specific target. This review is expected provide a model-selection reference for the development of novel low-toxic calcineurin inhibitors.

BACKGROUND:Recently, electrospun materials have been extensively applied in the drug delivery system. OBJECTIVE:To overview the application prospect of electrospun materials in drug delivery systems. METHODS:A computer-based search of PubMed and NCBI databases was performed for literatures about the research progress of electrospinning in tissue engineering and chemotherapy published within the past 10 years using the keywords of“electrospinning, drug delivery system, nanofibers, electrospun materials”.RESULTS AND CONCLUSION:Compared with traditional materials, electrospun stents hold good versatility and control able parameters, thus granting its unique advantage under various physiological conditions. Current drug-loaded materials composed of natural products, synthetic polymers and blended materials;as to drugs, there are antibiotics, chemotherapy medication, DNA and protein. Electrospun materials have been used in tissue engineering, cancer chemotherapy and wound healing. We focus on not only the application progress of electrospun materials in traditional treatments, but also its usage, condition-control ed drug release and living-cel carrying. Electrospun materials combined with various drug-loaded present a broad prospect.
8.Analysis of volatile components of organic solvents in key industry enterprises in Longgang District, Shenzhen City
China Occupational Medicine 2024;51(5):566-569
Objective To analyze the detection situation and variation tendency of volatile components of organic solvents in enterprises in Longgang District, Shenzhen City. Methods A total of 368 enterprises using organic solvents in Longgang District, Shenzhen City from 2019 to 2022 were selected as the research subjects using judgment sampling method. And its 2 086 reports of occupational hazards monitoring on volatile components of organic solvents in workplaces from 2019 to 2022 were collected for analysis. Results Among the 2 086 organic solvent samples, the top 10 chemical substances in terms of detection rates were toluene, methanol, xylene, n-hexane, ethylbenzene, dimethoxymethane, dichloromethane, acetone, ethyl acetate and cyclohexane, with detection rates ranging from 16.73% to 29.72%. The detection rate of benzene in organic solvent samples from 368 enterprises decreased from 13.84% to 5.13% from 2019 to 2022 (P<0.01). The detection rate of trichlorethylene decreased from 3.58% to 0.45% (P<0.01), and the detection rate of dichloroethane decreased from 4.53% to 1.81% (P<0.01). However, the reduction in the detection rate of n-hexane was not significant (from 28.88% to 23.08%, P>0.05). Benzene had relatively high detection rates in yellow glue, super glue, and thinner (10.94%, 10.00%, and 8.45%, respectively). The detection rate of n-hexane was relatively high in super glue, cleaning agent and car wash liquids (90.00%, 61.84%, and 46.81%, respectively). The detection rate of trichlorethylene was relatively high in cleaning agent (3.95%), and the detection rate of dichloroethane was relatively high in super glue (40.00%). Conclusion From 2019 to 2022, the detection rates of traditional occupational hazards such as benzene, trichloroethylene, and dichloroethane in the volatile organic components used in Longgang District enterprises showed a downward trend.

External Guide Sequences (EGSs) represents a novel nucleic acid based gene interference approach to modulate gene expression. They are oligonucleotides that consist of a sequence complementary to a target mRNA and recruit intracellular RNase P for specific degradation of the target RNA. DNA-based EGS1386 with a size of 12 nt was chemically synthesized to target the mRNA coding for the UL49 gene of human cytomegalovirus (HCMV). The DNA-based EGS1386 molecule efficiently directed human RNase P to cleave the target mRNA sequence in vitro. A reduction of more than 50% in the levels of UL49 expression was observed in human cells treated with the DNA-based EGS1386 targeted UL49 assayed by fluorescent quantization PCR and Western blotting. This results showed that the DNA-EGS1386 can effectively guide the RNase P cut the target mRNA. Therefore, DNA-EGS can develop into a new gene silencing technology and potential of the anti-viral reagents.
