1.Effect of baicalin on the expression of receptor activator of nuclear factor-?B ligand and osteoprotegerin in human periodontal ligament cells

Objective To study the effect of baicalin on the expression of receptor activator of nuclear factor-?B ligand (RANKL) and osteoprotegerin (OPG) in cultured human periodontal ligament cells (HPDL cells) as well as its action mechanism. Methods We first constructed small interferring RNA (siRNA) eukaryotic expression vector targeted transforming growth factor ?Ⅱ receptor (TGF-?RⅡ),and then transfected it into T cells. Then HPDL cells together with T cells transfected with siRNA or not were placed in medium that had been added with lipopolysaccharide (LPS) and baicalin. They were divided into six groups and cultured for 48 hours. Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to observe the effect of baicalin on OPG-RANKL expression in HPDL cells. Results The clone sequence correctly identified by RT-PCR was consistent with the designed target sequence. The recombinant vector was constructed successfully and the expression of TGF-?ⅡR of T cells which had been transfected with siRNA1 was inhibited obviously. Objective:To evaluate the immunogenicity of the recombinant plasmid pcDNA 3.1(+)/kgp_ cd.Methods:BALB/c mice were immunized with recombinant plasmid pcDNA 3.1(+)/kgp_ cd,KGP_ cd protein(control) or vector pcDNA 3.1(+)(control) by quadriceps injection or targeted submandibular gland(TSG) injection. Serum IgG and salivary sIgA levels were assayed by indirect ELISA after immunization. Objective ThrouGh the detection of heat shock protein 90(HSP90)Gene expression in the peripheral blood in in patients with hypertensive disorders complicatinG preGnancy( HDCP ),to understand its role in the pathophysioloGy of HDCP. Methods The expression of HSP90 was observed in Groups of normal preGnant women,Gestational hypertension patients,mild preeclampsia patients,severe preeclampsia patients by ELISA. Results The expression of HSP90 in peripheral blood of Gestational hypertension Group,mild preeclampsia Group,severe preeclampsia Group were siGnificantly hiGher than normal preGnant Group( P<0. 0l ) . Conclusion HSP90 may have close relationship with the onset and development of HDCP. It can predict HDCP by detectinG the level of HSP90 in peripheral blood.
4.Investigation on Clinical Teaching of Non-directly Affiliated Hospital

Aim: To enhance the management of clinical teaching hospital and improve the quality of clinical teaching.Methods: The criteria system of evaluation on teaching hospitals were taken to assess the teaching quality of 4 non-directly affiliated hospitals by means of listening to the self-evaluation,checking the file,surveying on the scene and holding student symposium.Prompt feedback were supplied to the 4 hospitals.Result: The 4 hospitals obtained similar scores,which had intelligent teachers,favorable teaching condition,strict management and better teaching quality.Conclusions: Emphasizing teaching,qualified teachers,improving teaching condition and optimizing teaching method are the important measures of improving clinical teaching quality.

6.Distribution of inducible nitric oxide synthase in gingival tissues in elderly patients with periodontitis

Objective: To observe the distribution of inducible nitric oxide synthase (iNOS) in gingival tissues in elderly patients with periodontitis. Methods: Gingival tissue samples were obtained during tooth extraction.The localization of positive expression of iNOS in gingival tissues in 10 patients aged 65-86 years with periodontitis was tested with immunohistochemical method and was compared with that of 10 adult patients(37-54 years old) with periodontitis, 10 juvenile patients (18-29 years old) with periodontitis and 10 healthy elders(65-81 years old),respectively. Results:iNOS expression was mainly found in epithelial cells, inflammatory cells in connective tissues, granuloma tissues, epithelial basement membrane and microvascular endothelial cells in gingiva in the 3 groups of periodontitis. The cases of positive expression of iNOS in microvascular endothelial cells and inflammatory cells in connective tissues in elderly patients with periodontitis were significantly fewer than those in juvenile patients and adult patients ( P

Objective To discuss the correlation between MTHFR gene polymorphism and unexplained recurrent spontaneous abortion.Methods A case control study was used in this study,140 patients with unexplained recurrent spontaneous abortion(UR-SA) (abortion group)and 143 cases of normal women(control group)were recruited.Genomic DNA was obtained and extracted from the oral mucosa cells.Fluorescence quantitative PCR was used to examine the MTHFR gene polymorphisms,and Taqman-MGB technology was conducted to analysis the relationship between single nucleotide polymorphism and disease.Results There was statistically significant difference in the frequencies of C677T genotype and alleles between the two groups(P<0.05).However,no significant difference in the frequencies of A1298C genotype and alleles between the two groups(P>0.05).Conclusion MTHFR gene C677T polymorphism might be one of the genetic risk factors of URSA.
8.Influencing factors of anxiety symptoms in patients with depression

Objective To investigate the status of depression with anxiety symptoms, and analyze the influencing factors of anxiety symptoms from demographic data and social psychological factors. Methods Hamilton depression rat?ing scale (HAMD), Hamilton anxiety rating scale (HAMA), Eysenck personality questionnaire (EPQ), life event scale (LES), trait coping style questionnaire (TCSQ) and social support scale (SSS) were used to evaluate 729 patients with de?pression. According to HAMA scores, patients were divided into non anxiety symptoms group (HAMA<7) and anxiety symptoms group (HAMA>14). Social psychological factors were compared between two groups, and the influencing fac?tors of anxiety symptoms were analyzed. Results The incidence of anxiety symptoms in depression was 58.85% (429/729), and 119 cases (16.32%) were certainly without anxiety symptoms. Compared with the group without anxiety symp?toms, the anxiety symptoms group had higher scores on neuroticism, psychoticism, negative life events and negative cop?ing style (P<0.001), but lower scores on introversion and extroversion (P=0.010). Degree of depression (OR=9.255, 95%CI:4.726~18.127), neuroticism (OR=1.595, 95%CI:1.197~2.125), negative life events (OR=1.009, 95%CI:1.001~1.017) and negative coping style (OR=1.046, 95%CI:1.013~1.080) were the risk factors of anxiety symptoms (P<0.05). Conclu?sion The incidence of anxiety symptoms in patients with depression is high. Patients with higher degree of depression and typical neurotic personality experiencing more negative life events and those with tendency to adopt negative coping style are more susceptible to anxiety symptoms.
9.Expression of placenta heat shock protein-90 in patients with hypertensive disorders complicating pregnancy

Objective To detect heat shock protein 90 (HSP90) expression in the placenta in pregnancy induced hypertension in order to understand its role in the pathophysiology of hypertensive disorders comphcating pregnancy (HDCP).Methods The gene and protein expression of HSP90 in the placenta of normal pregnant women,gestational hypertension patients,mild preeclampsia patients,severe preeclampsia patients,by RT-PCR and Western blotting.Results The gene and protein expression of HSP90 in placenta of gestational hypertension group,mild preeclampsia group,severe preeclampsia group were significantly higher than that of normal pregnant group (P < 0.01),and it showed the increased progressively with the increasing of disease stage.Conclusion HSP90 may be involved in the onset and development of HDCP.Monitoring HSP90 concentration in the placenta which is the initial theoretical basis for the target as HSP90 for the treatment of HDCP.

Patients' safety is a hot topic of hospital management all over the word. Strengthening the patients' safety education for medical students and nursing students is an effective measure to protect patients' safety. In nursing school of Chongqing Medical University,patients' safety education was con-ducted through the course of undergraduate education. Contents of patients' safety education were com-bined with the professional courses,which are taught step by step in the professional course learning phase for different grades. Patients' safety education took a lot of teaching forms to cultivate the students' patient safety consciousness and preliminary results were achieved.
