Objective To investigate the effects of Panax notoginseng saponins (PNS) on the cerebral water content and thrombin in the rats with intracerebral hemorrhage (ICH), and explore its mechnism for treating ICH. Methods SD male rats were randomly divided into blank group, sham operation group, model group and treatment group, and time points of 6, 24, 48, 72 h and 7 d were observed. ICH rat model was induced by collagenase and heparin in saline solution injected into caudate nucleus. The treatment group was injected PNS intraperitoneally once a day, and the cerebral water content was assessed by dry-wet weight method. Both content of fibrinogen (FIB) and thrombin clotting time (TT) in 48 h were measured. Results The cerebral water content of model group increased at every time point compared with blank group and sham operation group (P<0.05). The cerebral water content of treatment group was significantly decreased at every time point except for 6 h compared with model group (P<0.05). FIB of treatment group decreased compared with model group (P<0.05), while TT of treatment group was not significantly decreased (P>0.05). Conclusion PNS may play a role in treating ICH by decreasing cerebral water content, FIB and TT of ICH rats.

Objective To investigate the cognitive function of patients with prostate cancer after maximal androgen blockade therapy and its influenced factors,and to provide a new way for early prevention strategy.Methods Montreal cognitive assessment (MoCA),hospital anxiety depression scale (HAD),social support rating scale (SSRS) and self-designed questionnaire were used in 56 cases treated with maximum androgen blockade therapy for more than six months and 37 cases who underwent radical prostatectomy treatment to evaluate their cognitive function and collect the observation indexes between January 2013 and October 2015.Based on MoCA score,all patients were divided into cognitive dysfunction group (n =40) and normal cognitive function group (n =53).The observation indexes in two groups were compared and cognitive function with different treatment in two groups were analyzed.The changes on the influencing factors of cognitive function in patients were filtered using multivariable logistic regression analysis.Results In the cognitive dysfunction group and normal group,the proportion of MAB treatment was 80.0％ (32/40) vs.45.3％ (24/53),the age was 73.7 vs.73.7 years,the proportion of solitary was 32.5％ (13/40) vs.13.2％ (7/53),the proportion of depressive symptoms was 87.5％ (35/40) vs.62.3％ (33/53),the social support level was 32.5 vs.41.1 and the proportion of testosterone decreased was 95.0％ (38/40) vs.45.3％ (24/53).All events showed statistically significant differences (P ＜0.05).Compared MAB treatment group with radical surgical treatment group,the testosterone level was (0.27-±O.15) vs.(12.14 ± 1.86) nmol/L,visual space and executive function score was 4.18 ±0.79 vs.4.54 ±0.56,attention score was 4.73 ±0.99 vs.5.16 ±0.79,delayed memory score was 3.75 ± 1.21 vs.4.30 ± 1.05 and MoCA score was 26.13 ± 1.48 vs.27.27 ± 1.39,which all showed the statistically significant difference (P ＜ 0.05).The results of multiple regression analysis showed that age (OR =1.183,95％ CI 1.135-1.223),depressive symptoms (OR =1.092,95％ CI 1.047-1.149),social support (OR =0.897,95％ CI O.838-0.956),testosterone (OR =2.105,95％ CI 1.369-4.083) were the influenced factors of cognitive dysfunction.Conclusions The incidence of cognitive dysfunction in patients with prostate cancer after maximal androgen blockade therapy more than six months was higher than others.Age,depression,social support level and testosterone levels were related to the occurrence of cognitive dysfunction.

Objective To evaluate the quality of Semen Cassiae from different habitats objectively. Methods To determine the content of chryso-phanic according to ChP and establish HPLC fingerprints with the gradient elution solvent composed of acetonitrile and 1% HAC. A C18 column (250 mm?4.6 mm, 5 ?m) was used, flow rate: 1 mL/min, detecting wavelength: 254 nm, and the column temperature: room temperature. The clustering analysis was carried out by SAS software according to the content of chrysophanic and similarity of HPLC fingerprints obtained by the software of similarity analysis. Results The established HPLC fingerprint has desirable precision, reproducibility, and stability. The content of chrysophanic and HPLC fingerprints of Semen Cassiae from various habitats are different, which differs from the habitats. The content range of chrysophanic in Semen Cassiae is 0.037%-0.170% and the similarity is 0.864-0.962. Conclusion The method indicates the difference of the chemical component in Semen Cassiae from various habitats and can be used as a quality control method for Semen Cassiae.

Objective To observe the morphology of embryonic stem cell-derived neurons after transplanted into A?-injured rat hippocampus. Methods Neural precursor cells (NPCs) were generated from mouse embryonic stem cells (ESCs) expressing EGFP with modified serum-free methods and then transplanted into the hippocampus of A?1-40-injuried rats. The morphology, neurotransmitter phenotypes and receptors of EGFP-positive donor cells were observed with immunofluorescene methods. Results The engrafted NPCs survived and differentiated into glutamatergic and GABAergic neurons and expressed both glutamatergic and GABAergic neurotransmitter receptors. Synaptophysin-positive dots were found surrounding somata and dendrites of transplanted neurons, suggesting the presence of presynaptic terminals adjacent to their membranes. Conclusion NPCs derived from ESCs can differentiate into excitatory and inhibitory neurons after grafted into Alzheimer's disease model rats, and maybe form synapse with the host neurons.

Objective To detect the interaction domains' gene mutations of KEAP1 and NRF2 in non-small-cell lung cancer cell lines,and to analyze the significance of these mutations on the study of lung cancer cell lines.Methods Six non-small-cell lung cancer cell lines were used as the research materials.The 2nd to 6th exons of KEAP1 and the coding sequences of DLG and ETGE motif were amplified by PCR,and the products were used for gene sequencing analysis.Obtained gene sequences were analyzed using NCBI databases to get the base locations of gene mutations,as well as the subsequent amino acid sequence changes.Meanwhile,the relative intracellular reactive oxygen species (ROS) concentrations in the tested cell lines were detected and used in the analysis of abnormal NRF2 transcriptional activity in lung cancer cell.Results Four mutations were detected in the 4th exon of KEAP1 gene from all the 6 cancer cell lines,several other missense mutations were also investigated in the 3rd exon of KEAP1 from some cancer cell lines.Multiple genetic variations were found in all the NRF2-ETGE motif-encoding sequences of the 6 cancer cell lines.All 6 cancer cell lines were found to have higher ROS concentration than the lung germ cell line.Conclusion Lung cancer cells generally contain high levels of ROS as well as gene mutations in KEAP1 and NRF2 genes which lead to abnormal transcriptional activity of NRF2 in lung cancer cell lines.

Objective To explore the correlation between humanistic care ability of nursing students and their family environmental factors. Methods A sample of 198 nursing students was recruited with cluster sampling method. Demographic data, humanistic caring capacity, and family environmental factors were collected using a self-designed questionnaire, Caring Ability- Inventory (CAI) and Family Environment Scale (FES- CV) and then an analysis was conducted. Results Han nationality students accounted for 32.83%(65/198), minority students accounted for 67.17%(133/198) . The total average score of humanistic care ability was 190.79±18.84, the average score of items was 5.16±0.99. There were significant differences between our participants and those regarded as international norms on the three dimensions of humanistic care ability (t=-16.27--8.12, P < 0.01). The total average score of family environment was 42.57±8.88. Significant differences were also presented between our participants and those of domestic norms on the ten factors of the family environment (t=-10.94-30.04, P < 0.01). The significant correlation was not documented between the total scores of humanistic care ability and family environment. However, the correlations were documented between the six domains of family environment (intimacy, emotional expression, knowledge, entertainment, contradiction, sense of organization) and the total score and each dimension of humanistic caring capacity (P < 0.05 or 0.01). Conclusions The humanistic care ability of nursing students in minority area is relatively low. The humanistic care ability and family environment are correlated on the multi- dimen-sional facets. School educational strategies should be combined with family environment factors of nursing students to jointly promote the humanistic care ability of nursing students.

Objective To identify the features of the NCAM+ c-Kit+ subset of hepatic progenitor cells in the intrahepatic cholangiocarcinoma (ICC) cell line RBE.Method Magnetic activated cell sorting was used to isolate NCAM+ c-Kit+/NCAM-c-Kit-subset cells,which were tested for hepatic progenitor cell properties and proliferation,colony formation,and invasive abilities in nude mice.Resuits The cell proliferation ability of NCAM+c-Kit+ subset cells was stronger than that of NCAMc-Kit-subset cells (P＜0.01).In serum-free condition,the number of colonies formed by NCAM+c-Kit+ subset cells was more than that of NCAM-c-Kit-cells (P＜0.01).1 × 104 NCAM+c-Kit+ cells were enough to form tumors in nude mice after subcutaneous inoculation for two weeks,while 1 × 106 NCAM-c-Kit-cells were necessary to form tumors for three weeks.The tumor formation rate of NCAM+c-Kit+ cells was higher than that of NCAM-c-Kit-cells (P=0.04).Conclusions It is possible that NCAM+c-Kit+ subset cells in RBE have the properties of hepatic progenitor cells,and NCAM combined with c-Kit might be a valuable marker for isolating and purifying ICC stem/progenitor cells.

Objective To evaluate the application value of virtual touch tissue quantification (VTQ)for evaluating small focal liver lesions.Methods Seventy-seven patients with 89 small focal liver lesions,whose largest diameter were less than or equal to 3 cm were evaluated with VTQ.The shcar wave velocity (SWV) of the tumor and background liver parenchyma were calculated,and their results were compared with those of 40 healthy subjects.Statistical analysis was performed on the SWV for the differentiation of normal liver,background liver parenchyma and lesions.The cut-off point for SWV was obtained by using ROC curve analysis which was used to predict malignancy lesions.Results The SWV of malignant lesions was significantly higher than that of benign ones [(2.56-± 0.67) m/s vs (1.70 ± 0.55) m/s,P =0.000].The SWV of metastatic liver carcinoma which came from lung cancer and breast cancer were higher than that of gastrointestinal tract cancer transfers (P =0.000).Based on 1.93 m/s as the cut-off point of SWV value,the sensitivity,specificity,accuracy,positive predictive value and negative predictive value for diagnosing malignancies were 82.0 ％,84.6％,83.15 ％,87.23 ％ and 78.57 ％ respectively.Conclusions VTQ technique can help the differential diagnosis of small focal liver lesions.
