2.Preliminary study on preparation of Yuanhuzhitong HBS Tablet

Objective: To study the preparation and quality control methods of Yuanhu Zhitong HBS Tablet. Methods: the suitable excipients and formulation process of Yuanhu Zhitong HBS Tablet is sieved by means of testing quantity of tetrahydropalmatine through TLC UV. Results: the beginning floating time of HBS Tablet is 3min; relative dissolution of main component tetrahydropalmatine is 32% within 3hr, 86% within 9 hr. Conclusions: the study is found to be effective in the process of Yuanhu Zhitong HBS Tablet.
3.A methodologic study on the determination of islet cell antibodies

To exclude the interfering factors in islet cell antibodies(ICA) determination some pretreatments on both pancreatic sections and serum samples were made. The results revealed that the islet structures of pretreated pancreatic frozen section appeared clearly. The positive rate of ICA by ABC enzyme-labelled method was higher than that by indirect immunofluorescence method. Comparing the ICA results of exchange sera provided by Immunology and Diabetes Workshop with the reference results,the validity、consistency、sensitivity and specificity was 81%、63%、75%and 88%, respectively. These results suggest that ABC method with the pretreated pancreatic frozen section is a stable method of ICA determination.
4.Variation of dissolution rate of main active ingredient of compatibility of coptis and scutellaria

Objective: To study the scutellaria and coptis in different proportions, the main active ingredient of baicalin, berberine dissolution rate changes and to explore the effect of traditional drug pair of scutellaria and coptis on the dissolution rate of active ingredient. Methods: Based on the clinic common used prescriptions of scutellaria with coptis, ratio of 1:0,1:1,1:2,1:3,2:1,2:3,3:1,3:2,0:1 was selected, after the water boiling reflux extraction, refining separation, various products were obtained for the test, under the optimized conditions of RP-HPLC analysis to compare chromatographic fingerprints and to examine the main component baicalin, berberine in aspects of relative peak area ratio of changes and compatibility relations. Results: Compatibility of Scutellaria baicalensis and Coptis at different proportions can influence the dissolution rate of baicalin, Berberine Hydrochloride. There was a non-linear relationship of dissolution rate of baicalin and Scutellaria baicalensis. The best ratio of scutellaria ratio was 3:1, the best compatibility ratio of coptis was 1:3. Conclusion: Different compatibility proportions of scutellaria and coptis will result in different dissolution rate of baicalin and berberine.
5.Efect of rapid intestinal preparation combined with probiotics in bowel preparation before operation

Objective To explore the effect of rapid intestinal preparation combined with probiotics in bowel preparation before operation.Methods 124 colorectal cancer patients were divided into the probiotic group(65 cases) and control group(59 cases).Control group using traditional 3d bowel preparation,joint probiotic group,to give patients in the 1 d intestinal ready on the basis of probiotic oral.The two groups after patients received isonitrogenous and caloric nutritional support,were observed after the two groups of patients with body temperature and heart rate changes; detection of bacterial DNA ratio of whole blood.Observed in peripheral blood leukocyte count,and systemic inflammatory response syndrome(SIRS) and the occurrence of complications.Results Probiotic group and control group,postoperative fever duration and postoperative heart rate and leukocyte counts return to normal a short time( t =11.52,20.07,P < 0.05 ) ; whole blood PCR detection of bacterial DNA after the positive test group 2 cases (3.30%),the control group was 8 cases ( 26.67% ),the difference was statistically significant ( t =5.07,x2 =34.68,P < 0.05 ).Postoperative SIRS rate and the incidence of complications showed no statistical difference ( P > 0.05 ).Conclusion Probiotics could reduce colorectal cancer patients with postoperative intestinal permeability and reduce the incidence of bacterial translocation and its rapid intestinal preparation method was feasible and effective and knot the protection of the intestinal mucosal barrier function in rectal cancerconductive to knot the early postoperative inflammatory response in patients with rectal cancer recovery.
6.Antitumor mechanism of ursolic acid on human gastric cancer cell lines SGC7901 in vitro

Objective To investigate the potential mechanism of inhibition of ursolic acid(UA) on growth of human gastric cancer cell lines SGC7901 in vitro.Methods SGC7901 cells were cultured in vitro,MTT assay was used to observe the effect of UA on growth of SGC7901 cells in various concentrations for different times.After SGC7901 cells were treated by 0—40 ?mol/L UA for 24 h,morphological changes were observed by inverted microscope.Apoptotic changes were detected by fluorescence microscopy and flow cytometry (FCM).Protein expressions of Bcl-2 and Bax were determined by Western blotting.Results UA(20—40 ?mol/L) could significantly inhibit the growth of SGC7901 cells in a(dose-and) time-dependent manner,the IC_(50) value of UA for SGC7901 cells for 12,24,36,and 48 h were((57.50?)1.18),(34.28?2.05),(27.54?1.11),and(24.83?1.02) ?mol/L,respectively.After UA((20—)40 ?mol/L) treatment for 24 h,SGC7901 cells turned round and floated at different levels;SGC7901 cells were arrested at G_0/G_1 phase and apoptosis was induced,and the apoptotic rate was increased along with the increase of UA concentration.Meanwhile Bcl-2 protein expression decreased,whereas Bax protein expression unchanged.Conclusion UA has a stronger antitumor effect on SGC7901 cells.Cytotoxic effect,proliferation inhibition,and apoptosis may be involoved in the mechanism of UA,and the apoptosis caused by UA may be enhanced by decrease of Bcl-2 protein expression.
7.Quality evaluation of Semen Cassiae by both indicated component determination and HPLC fingerprint

Objective To evaluate the quality of Semen Cassiae from different habitats objectively. Methods To determine the content of chryso-phanic according to ChP and establish HPLC fingerprints with the gradient elution solvent composed of acetonitrile and 1% HAC. A C18 column (250 mm?4.6 mm, 5 ?m) was used, flow rate: 1 mL/min, detecting wavelength: 254 nm, and the column temperature: room temperature. The clustering analysis was carried out by SAS software according to the content of chrysophanic and similarity of HPLC fingerprints obtained by the software of similarity analysis. Results The established HPLC fingerprint has desirable precision, reproducibility, and stability. The content of chrysophanic and HPLC fingerprints of Semen Cassiae from various habitats are different, which differs from the habitats. The content range of chrysophanic in Semen Cassiae is 0.037%-0.170% and the similarity is 0.864-0.962. Conclusion The method indicates the difference of the chemical component in Semen Cassiae from various habitats and can be used as a quality control method for Semen Cassiae.
8.LABORATORY DIAGNOSIS OF BENIGN MONOCLONAL GAMMOPATHY: ANALYSIS OF 38 CASES

38 cases of Benign Monoclonal Gammopathy (BMG) representing 17.9% were screened from 212 cases of M-proteinemia by laboratory examinations. Among the 38 cases, 17 cases had increase of IgG, another 17 cases of IgA and 4 cases of IgM. Their average M-proteins were 1.42, 0.88 and 1.84 g/dl respectively.The salient points of differential of benign or malignant M-proteinemia were also discussed. The recommended sequential laboratory diagnostic procedure of BMG were as follows: tatol serum protein determination, plasma protein zone electrophresis, urine protein analysis, immunoglobulin quantitation and immunoelectrophresis or selective electrophresis.

10.Effect on cGMP of Meridian Skin with Electroacupuncture on Myocardial Ischemia Rats

Objective To observe the specific changes of content of cyclic guanosine monophosphate (cGMP) around meridian skin and specific influences of electroacupunture Neiguan (PC6) on cGMP of rats with acute myocardial ischemia (AMI). Methods With 6 male Wistar rats as normal group, 18 rats were made AMI model and divided into model group, high-frequency group and low-frequency group (n=6). The high-frequency and low-frequency group were given electroacupuncture in bilateral PC6 with 100 and 2 Hz for 20 min respectively. The content of cGMP in skin around PC6, Ximen (PC4), Tianquan (PC2), Waiguan (TE5) and Zusanli (ST36) was detected by using enzyme-linked immunosorbent method after treatment. Results In model group, cGMP content increased significantly around PC6, PC2, TE5 and ST36 compared with normal group (P <0.05,P <0.001). After the treatment with high-frequency or low-frequency electroacupunture in PC6, cGMP content decreased significantly in skin around PC6, PC2, TE5 and ST36 compared with model group (P <0.05, P <0.001). Conclusion High-frequency electroacupunture or high-frequency electroacupunture used in PC6 can protect rats from the injury of AMI. The mechanism may be related to the decrease of cGMP content around the acupoint areas in heart-related meridian.
