1.Concurrent chemoradiotherapy with low dose weekly paclitaxel in the treatment of stage Ⅲ non-small-cell lung cancer

Background and purpose:The curative effect with radiotherapy or chemotherapy alone for the patients with unresectable stage Ⅲ non-small-cell lung cancer was poor. The 5-year survival was only 5-10 percent. Concurrent chemoradiotherapy could achieve better local control and overall survival of those patients and it had been reported that the 5-year survival was improved to 15.8 percent, but the toxicity were much more severe at the same time. We prospectively evaluated the efficacy and toxicity of concurrent chemoradiotherapy with low dose weekly paclitaxel for unresectable stage Ⅲ non-small-cell lung cancer and tried to make the regime more tolerable without the deterioration of treatment response. Methods:Forty-eight patients with unresectable stage Ⅲ non-small-cell lung cancer were randomized into low dose weekly paclitaxel group and control group.Both groups were treated by the standard fractionation schedule. All patients were irradiated 2.0 Gy/per fraction,five fractions a week,the total radiation dose was 60-64 Gy for tumor. Patients in the low dose weekly paclitaxel group received chemotherapy with 45 mg/m 2 of paclitaxel on every Monday; the patients in control group received 50 mg/m2 of cisplatin on days 2-4 and day 23-25, and 135 mg/m 2 of paclitaxel on days 1 and 22 concomitant with the radiotherapy.Results:The CR (complete response) rates of low dose weekly paclitaxel and control group were 21% and 13% respectively(P0.05).Conclusions:The patients treated by low-dose weekly paclitaxel group showed that both survival fraction and the period of local tumor control were higher than in control group. Additionally, low dose weekly paclitaxel concomitant with radiotherapy was well tolerated and were not statistically different from control group in terms of toxicities.
2.Photoprotective effect of a self-made sunscreen cream: an evaluation in vitro and in vivo by using guinea pigs

[Objective] To determine the sun protection factor (SPF) and protection factor of ultraviolet A (PFA) of a self-made sunscreen cream.[Methods] The sample of a self-made sunscreen cream and a commercial sunscreen cream were painted to an artificial skin (3M film) at a dose of 2 μl/cm2,and a tester was used to evaluate their photoprotective effect.A total of 48 albinism guinea pigs were classified into 8 groups to remain unprotected or be protected by the self-made or commercial sunscreen cream.A solar ultraviolet light simulator (SUV-1000) was used to simulate the ultraviolet rays in sunlight to irradiate the guinea pigs,and the photoprotecfive effect of these sunscreen creams was determined.[Results] The in vitro evaluation revealed that the SPF value of the self-made sun-screen cream and commercial cream was 32.26 ± 2.42 and 30.87 ± 2.57respectively (n =5,t =0.94,P > 0.05),and the PFA value was 24.28 ± 2.44 and 17.53 ± 2.28 respectively (n =5,t =4.52,P< 0.01 ).As the in vivo experiment showed,the SPF value of the self-made sun-screen cream and commercial cream was 30.39 ± 6.65 and 28.79 ± 7.36,respectively (n =12,t =0.38,P> 0.05),and the PFA value was > 8.91 and > 8.93 respectively.[Conclusion] The photoprotective effect of the self-made sunscreen cream is similar to that of the commercial cream.
3.Genetic polymorphism of HIF-1α G1790A associated with radiosensitivity of nasopharyngeal carcinoma

Objective To explore the association between the genetic polymorphism of hypoxia inducible factor 1 α (HIF-1α) G1790A and the radiosensitivity of nasopharyngeal carcinoma.Methods A total of 189 patients with nasopharyngeal carcinoma treated with radical radiotherapy were followed-up for 3 years.The patients were divided into cured group with 135 cases and recurrence group with 54 cases by clinical follow-up results.PCR-RFLP was used to determine the mononucleotide genotypes of HIF-1α G1790A.Results The observed genotype frequencies of HIF-1α gene 1790 (G→A) for GG, GA and AA were 70.04% , 20.74% , 2.22% in cured group and 59.26% , 38.89% , 1.85% in recurrence group, respectively.The allele frequencies for G and A were 87.4% , 13.9% in cured groups and 78.7% ,21.3% in recurrence group, respectively, without significant difference in distribution of allele frequencies between the two groups(x2 =6.919, P =0.077).Conclusions The genetic polymorphisms of HIF-1α G1790A might be related with the radiosensitivity of nasopharyngeal carcinoma.
4.CT-MRI Comparative Observation of Parapharyngeal Space in Nasopharyngeal Carcinoma

Objective To recognize CT signs of parapharyngeal space (PPS) changing in nasopharyngeal carcinoma (NPC) further.Methods 30 patients with proven NPC by biopsy were performed CT and MR scanning before radiotherapy. Results According to CT findings,four types of PPS display in 30 cases were found,each of which was observed and compared with MR findings and were as follows: (1)PPS ,which had no changing on CT, showed fat-filled triangular space continuously on MRI. (2) PPS, which became narrow and linear hypodense on CT,showed that fat intensity was pressed, but continuously on MRI; (3) PPS, which became narrow and linear hyperdense on CT,showed fat intensity,was pressed exceedingly but continuously either on MRI. (4)PPS,which was diminished on CT, showed the mass occupied PPS and lateral pterygoid muscles was pushed, medial fatty space of which was diminished, but intensity of medial or lateral pterygoid muscles was normal or abnormal on MRI. Conclusion (1)When PPS becomes narrow on CT,whether PPS shows hyperdense or hypodense,the mass makes a breakthrough PPS. (2) When PPS is diminished completely on CT,the mass steps PPS to the infratemporal fossa.
5.Research progress of sequential immunotherapy for neuromyelitis optica spectrum disorders in children

Pediatric neuromyelitis optica spectrum disorders (NMOSD) is an inflammatory demyelinating di-sease spectrum of the central nervous system, mainly involving the optic nerve and spinal cord.Due to the high disability and recurrence rate, sequential immunotherapy is usually needed to prevent the recurrence of NMOSD after acute treatment.Among the commonly used immune suppressants, Rituximab and Mycophenolate mofetil are the preferred drugs due to the best efficacy.Drug tolerance of Mycophenolate mofetil is better than that of Rituximab, and that of Cyclophosphamide remains the worst.Azathioprine is not recommended as the first-line drug of NMOSD because of frequent adverse events and the slow onset.This study aims to review the sequential immunotherapy of NMOSD in children.
6.Design of personal dosimeter system based on ZigBee wireless network

Objective To research and design-an personal dosimeter system to provide data for nuclear radiation injury protection.Methods The overall architecture,hardware module and software of the system were designed with ZigBee wireless network technology and the principle.The system was composed of a terminal node,a router and a coordinator.Results The system could collect the information on nuclear radiation dosage of the serviceman within 1 km,and then the data were uploaded with the wireless network.Conclusion The system gains advantages in low power consumption,low cost,low interference and etc,and plays a very important role for commander to hold the combatants' nuclear radiation dose in the battlefield.In addition the system can also be applied in civilian field to enhance the personal dose management.
7.Methodology and clinical application of left heart contrast echocardiagraphy with perfluoropropane-albumin microsphere

0.05 ). The average enhancement rate of LV endocardial border was 96.6 %. The myocardial contrast agent perfusions of left ventricular walls were clearly visualized in 30 patients. CONCLUSION: Clinical application of intravenous left heart contrast echocardiagraphy with perfluoropropane-albumin microsphere is feasibility and effective.
8.Analysis of clinical and biological characteristics of relapsed childhood low-risk acute B lymphoblastic leukemia

Objective:To investigate the clinical and biological characteristics of relapsed childhood low-risk acute B lymphoblastic leukemia (B-ALL).Methods:The clinical and laboratory data of 34 children who admitted in Beijing Boren Hospital from July 2017 to July 2018 were retrospectively analyzed, and 127-339 mutations of hematological malignancy related genes were analyzed.Results:The median time from the diagnosis to the recurrence was 871 d (87-1 446 d). The recurrence at early stage and late stage had 26 cases (76%) and 8 cases (24%), respectively. The recurrence before maintenance treatment, during maintenance therapy and after withdrawal of chemotherapy had 3 cases (9%), 12 cases (35%) and 19 cases (56%) (13 cases relapsed within 1 year after withdrawal, 6 cases relapsed after withdrawal 1-2 years and no one relapsed after withdrawal 2 years). The sites of recurrence included bone marrow alone accounting for 26 cases (76%), both intramedullary and extramedullary disease (EMD) accounting for 6 cases (18%), EMD alone accounting for 2 cases (6%). Flow cytometry showed that 9 patients presented minimal residual disease (MRD)-positive (6 cases with one positive, 2 cases with twice positive and 1 case with 3 times positive), including 8 cases occurred at early stage and 1 case occurred at late stage; and the level of MRD was 0.02%-3.82%. Complex chromosomal karyotype appeared in 6 relapsed children with normal or hyperdiploid karyotype at first diagnosis. Hematological malignancy related gene mutation detection was made in 28 cases, and the results showed that each patient had at least one gene mutation, and 2 or more gene mutations were detected in 25 cases (89%). The high frequency of gene mutations were as follows: CREBBP (7 cases, 25%), NRAS (7 cases, 25%), KRAS(7 cases, 25%), TP53 (4 cases, 14%), and NT5C2 (4 cases, 14%).Conclusions:The recurrence of childhood low-risk B-ALL occurs mostly in the maintenance treatment or in two years of withdrawal of chemotherapy. Positive MRD after complete remission is likely to show the risk of early recurrence. The gene mutations after the poor prognosis in cancer cells may be related to the recurrence of childhood low-risk B-ALL, and the common gene mutations include CREBBP, RAS signaling pathways genes and TP53, NT5C2.
9.Effects of Qidi Tangshen granules and their separate prescriptions on podocytes in mice with diabetic nephropathy

Objective:Previous studies have found that Qidi Tangshen granules (QDTS),a combination therapy of supplementing essence (Tianjing,TJ) and unblocking the collaterals (Tongluo,TL),can reduce kidney damage in db/db mice.This study aimed to explore the effect of QDTS and their separate prescriptions on podocytes in mice with diabetic nephropathy.Methods:The db/db mice were used in this experiment as an animal model,while wild-type C57BL/6J mice were used as normal controls.At the age of 12 weeks,the db/db mice were randomly divided into 5 groups (db/db,db/db + valsartan,db/db + QDTS,db/db + TJ and db/db + TL).The urine albumin excretion ratio (UAE) was measured by enzyme-linked immunosorbent assay before and after the intervention.The ultrastructure of the kidney podocytes was observed by transmission electron mi-croscopy.The protein expression levels of nephrin and desmin were detected by immunohistochemistry.Results:QDTS and their separate prescriptions significantly decreased the UAE and attenuated the renal pathological injury.QDTS and their separate prescriptions also reduced the fusion rate of the foot pro-cesses and increased the expression of nephrin protein.In contrast,QDTS and their separate pre-scriptions (TJ and TL) reduced the expression level of desmin protein.Conclusion:QDTS and their separate prescriptions might reduce diabetes-induced renal injury by reducing podocyte damage.The therapeutic effect of QDTS was more pronounced than TJ and TL.
10.Mesenchymal stem cells inhibit hypoxic damage to rat pulmonary microvascular endothelial cells by regulating oxidative stress

OBJECTIVE To explore the role and mechanism of dental pulp stem cells(DPSCs)in repairing hypoxic injury to rats pulmonary microvascular endothelial cells(PMVECs).METHODS ①PMVECs were treated with cobalt chloride at 0,10,25,50 and 100 μmol·L-1 for 72 h.CCK-8 was used to detect the cell viability,and the protein levels of hypoxia-inducible factor 1α(HIF-1α),zona occludens small-band protein 1(ZO-1),and occludin(OCLN)were detected by Western blotting.②There was a cell control group,model group,and model+DPSCs group,and the levels of reactive oxygen species(ROS)was detected by immunofluorescence staining after at 24 and 48 h of action.The levels of ZO-1 and OCLN proteins were detected by Western blotting.③ A cell control group,model group,model+DPSC group and model+DPSC cell knockdown superoxide dismutase 1(SOD1)group were set up.The mRNA level of SOD1 was detected by real-time fluorescence quantitative PCR 24 and 48 h later,while the protein levels of ZO-1 and OCLN were detected by Western blotting.RESULTS ① Com-pared with the cell control group,72 h of cobalt chloride 100 μmol·L-1 treatment of PMVECs resulted in a cell survival rate above 80%,a significant increase in the level of HIF-1α protein(P<0.05),a signifi-cant decrease in the levels of ZO-1 and OCLN proteins(P<0.01),and establishment of a model of hypoxic injury in PMVECs.② Compared with the cell control group,the ROS level was significantly higher in the model group(P<0.01).Compared with the model group,the ROS level was significantly lower in the model+DPSCs group(P<0.01),while the levels of ZO-1 and OCLN proteins were signifi-cantly higher in the model+DPSCs group(P<0.05).③ Compared with the DPSC group,ZO-1 and OCLN expressions were significantly decreased after knockdown of SOD1 in DPSCs(P<0.05,P<0.01).CONCLUSIONS DPSCs can repair hypoxic injury to PMVECs,and the anti-oxidative stress capacity of DPSCs plays an important role in hypoxic injury repair of PMVECs.
