1.Antibody array analysis of atherosclerosis cytokine levels in rats

Objective To investigate the changes of the cytokines on the aortas of atherosclerosis (AS) rats. Methods Zymosan-liquid medical paraffin suspension (contain Zymosan 20 mg/kg) was injected intra- peritoneally once a week for 10 times and given intraperitoneal injections of ovalbumin, 2.5 mg/kg, once a week for 7 times after initial subcutaneous sensitization. All animals were fed cholesterol-rich diet to induce AS. Electron microscopy was used to observe the foam cells, and RayBio Rat Cytokine Antibody Array 1.1 was employed to detect the cytokines in the aorta wall. Results Electron microscopy showed all models had monocytes migration from lumen to subendothelium in the aorta and formed to foam cell for phagocytosis lipid, indicating typical characteristic of AS in the rats. Most pro-inflammatory cytokines were increased except Fractalkine and IL-6, but anti-inflammatory cytokines were all slightly decreased. Conclusion Inflammation and immune can induce the formation of foam cells. The balance of pro-inflammatory cytokines and anti-inflammatory cytokines is broken is the important reasons of atherogenesis。
2.Characteristics of multiple evanescent white dot syndrome with multimodal imaging

Objective To observe the characteristics of multiple evanescent white dot syndrome (MEWDS) with modern multimodal imaging modalities.Methods This was a retrospective case study.Eleven patients (11 eyes) diagnosed with MEWDS were enrolled.There were 10 females and 1 male,mean age was 27.6 years (range 15-41 years).The period between disease onset and visiting to the hospital was between 2 to 13 days,the average time was 4.7 days.All the patients underwent examinations of best corrected visual acuity,slit-lamp biomicroscope,indirect ophthalmoscope,fundus color photography,fundus autofluorescence (FAF),fundus fluorescein angiography (FFA),indocyanine green angiography (ICGA) and spectral domain optical coherence tomography (SD-OCT).The mean follow up duration was 3.2 months.The imaging characteristics were compared.Results Fundus color photography showed foveal orange-red granularity in all eyes.FAF showed strong autofluorescence with a vague boundary.FFA showed a variable number of highly fluorescent fine needle-like dots arranged in a ring in the early stage,and fluorescence remained in the late stage.ICGA showed advanced lesions of vague boundary merged into a large plaque of deep retinal hypofluorescence.SD-OCT showed the hyperreflectant material deposit over the retinal pigment epithelium and extending anteriorly through the interdigitation zone,ellipsoid layer,and toward the external limiting membrane.At the site of extrafoveal lesions,SD-OCT revealed the presence of discontinuities or disruptions centered on the ellipsoid zone to include the interdigitation.Conclusions In MEWDS patients,fundus photography showed foveal orange-red granularity;FFA showed early fluorescent dots distributed in a ring pattern;ICGA showed hypofluorescent lesions in the later stage;SD-OCT showed disruption of the interdigitation zone and ellipsoid zone and accumulations of hyperreflective material that was of variable size and shape;FAF showed strong autofluoresce areas that correlated to spots observed with FFA and ICGA.
3.Effects of procainamide on a canine model of electropharmacology as assessed by programmed electrical stimulation

A canine model of electro-physiologic-electropharmacologic testing on ischemic ventricular tachyarrhythmias were established in the open-chest dogs subjected to programmed electrical stimulation (PES) on 5~8days after acute my-ocardial infarction produced with two -stage occlusion of left anteriol decending coronary artery (LAD) followed by partly reperfusion. The electrophysiologic -electropharmacologic effects of pro-cainamide (PA ) were observed in this canine model. pA distinctly lengthened the QTc interval and the effective refractory period ( ERP) of normal and infarct myocardium in both ventricles and decreasedthe dispersion of ERP in infarct myocardium (IDR) as well as the dispersion of ERP in left ventricle (VDR). The PES - induced ventricular tachycardia (VT) or ventricular fibrillation (VF) was prevented in 5 out of 6 PA treated dogs (n = 6),Normal saline (NS) did not prevented PES- induced VT/VF. The results suggest that PA may be effective in preventing the onset of reentrant ventricular tachyarrhythmias after myocardium ischemic damage.
4.Endovascular therapy for dysfunctional native arteriovenous fistulas

Objective To assess the feasibility and effectiveness of percutaneous transluminal angioplasty (PTA)for the salvage of immature arteriovenous fistula (AVF) and to identify the incidence of arterial and venous puncture site spasm.Methods The medical records and radiological data of 88 patients with 112 interventional procedures for immature AVFs were retrospectively reviewed.Results The stenosis lesions were (2.0 ± 1.4) cm long.Technical success rate and clinical success rate were 80.4% (78/97) and 92.8% (90/97) for PTA via brachial artery,85.7% (6/7) and 100% (7/7) for PTA via vein,25% (2/8) and 50% (4/8) for PTA via both brachial artery and vein,respectively.Spasm of pure arterial PTA occurred in 2 patients (2.1%) and was mild and moderate.Spasm of pure venous PTA occurred in 2 patients (28.6%) and was both moderate.Spasm of combined arterial and venous PTA occurred in 3 patients (37.5%) and from being severe to completely occluded.By comparison,there were statistical differences of technical and clinical success rate (P =0.000,0.019 ; P =0.000,0.029),fistulas spasm rate was statistically significant different (P =0.000).Conclusions Endovascular therapy was effective in restoring the dysfunctional native AVFs,it was safer and more effective and with less sideeffects especially in selecting coronary balloon to treat patients without large phlebangioma and round fistulas.
5.Anatomical MRI study for rotational alignment of tibial component in total knee arthroplasty

Objective To compare the difference between a vertical line (AA) drawn to the line connecting the inner edge of the patellar tendon with the mid-point of the ending point in the posterior cruciate ligament, tibial posterior condylar line (PC), tibial plateau anterior line (AC), the maximal mediolateral distance (MMLD) and a vertical line (BB) drawn to aligning the mid-point of ending point in the posterior cruciate ligament with the medial 1 / 3 of the patellar tendon relative to the surigical transepicondylar axis (STEA) by MRI, and to explore a reliable reference to determine tibial component rotation in total knee arthroplasty , and whether it will change in knees with varus deformity. Methods Thirty healthy volunteers (Group1) and thirty osteoarthritis patients (Group2) were enrolled in this study. The angles were measured among the five tibial rotation axes and STEA after MRI. Results The angles were (-1.48 ± 2.38)°, (6.16 ± 4.53)°, (6.45 ± 5.24)° ,(5.08 ± 4.99)° and (3.24 ± 2.68)° respectively in group 1 and (-1.88 ± 2.21)°, (-3.13 ± 4.66)°, (11.13 ± 5.72)°, (4.11 ± 4.15)° and (5.12 ± 4.87)° respectively in group 2. The angle between AA and STEA was not affected by varus deformity (P > 0.05), but the others were (P < 0.05). Conclusion The angle between AA and STEA is the smallest which is used to determine tibial component rotation in knees with varus deformity is the most reliable one.
6.Relationship between absolute monocyte/lymphocyte count and prognosis in low-risk patients with aggressive B cell lymphoma

Objective To investigate the prognostic significance of peripheral blood absolute monocyte/lymphocyte count in low‐risk patients with aggressive B cell lymphoma .Methods Retrospective study was performed in 42 low‐risk patients with ag‐gressive B cell lymphoma approved by histology between 2003 to 2013 .Peripheral blood absolute monocyte count (AMC) ,absolute lymphocyte count (ALC) ,lymphocyte/monocyte(LMR) and the some other clinical characters were estimated .Results The best absolute monocyte/lymphocyte count cut‐offs respectively were 475/mm3 and 1 005/mm3 ,analyzed by receiver operating character‐istic curve .AMC≥475/mm3 and ALC<1 005/mm3 were found to be independent predictors of progress free survival (RR=4 .271 , 3 .023 ;P= 0 .01 ,0 .39);AMC≥475/mm3 was found to be independent predictors of 5‐year overall survival (RR= 4 .680;P<0 .0001) in low‐risk patients with aggressive B cell lymphoma .Conclusion Higher AMC and/or lower ALC are poor prognosis fac‐tors .AMC and ALC may be important prognostic factors of low‐risk patients with aggressive B cell lymphoma .
7.Expressions of nitric oxide synthase in neonatal rat after hypoxia-ischemia and their changes while ganglioside GM_1 administrated

AIM: To study the neuroprotective effect and possible mechanism of ganglioside GM 1 on neonatal hypoxic-ischemic-encephalopathy(HIE). METHODS: A rat model of neonatal HIE was established, then the pathological changes and expressions of nitric oxide synthase (NOS) in the brain tissues were investigated in different periods after hypoxia-ischemia (HI) and the subseqent changes of the above results after GM 1 administrated. RESULTS: The damage of the brain exposed to HI were alleviated significantly after GM 1 administrated. The levels of NOS expressions in the brain tissue increased after HI. GM 1 could inhibit NOS expressions induced by HI. CONCLUSION: GM 1 may have some protective effects on neonatal HIE, and the possible mechanism is related to the partial inhibition of NOS expression.
8.Effects of lentivirus-mediated siRNA interference of USP39 on proliferation and migration of mice vascular smooth muscle cell

Objective To investigate the effect of lentivirus-mediated siRNA interference of USP39 on proliferation and migration of mice vascular smooth muscle cell in vitro.Methods Five siRNAs of siControl, siRNAUSP39-70, siRNAUSP39-71, siRNAUSP39-72 and siRNAUSP39-73 were designed and sythezied,mice VSMCs were infected with the lentivirus for delivering siRNAUSP39-73, and the stably transfected cells were selected by puromycin.The interference efficiency of siRNAUSP39-73 was assessed with Western blot.The effect of USP39 interference on the proliferation of VSMCs was determined by cells counting and MTT assay.Transwell assay was used to detect the migration of VSMCs.Results Recombinant lentiviral vector siRNAUSP39-73 was successfully transfected into mice VSMCs.Comparing with siControl group and Normal group, USP39 protein level of siRNAUSP39-73 VSMCs were decreased(P<0.05), and the proliferation and migration ability were all inhibited(P<0.05).Conclusion Targeted down-regulation of USP39 expression can inhibit the proliferation and migration of mice VSMCs in vitro.
9.Effect of EGF on angiogenesis induced by adipose-derived stem cells

Objective To explore the effect of epidermal growth factor (EGF) on tube formation of HUVEC induced by the secretion of angiogenesis factors of adipose-derived stem cells (ADSCs).Methods ADSCs were primarily cultured by enzyme digestion method.The flow cytomertry was performed to detect the expression of cell surface marker.ELISA was used to detect the expression of VEGF,HGF,and SDF-1 after given different doses of EGF.Tube formation assay was used to examine the effect of EGF on the tube formation induced by ADSCs.Results ADSCs were successfully isolated and cultured from human liposuction tissue and specific markers were expressed on ADSCs.EGF promoted the secretion of angiogenesis factors VEGF,HGF,and SDF-1,which were secreted by ADSCs.EGF pretreatment increased the ability of tube formation of HUVECs induced by ADSCs.Conclusions ADSCs induce the secretion of angiogenesis factors in vitro,and thus increase the ability of tube formation of HUVECs.EGF promotes the secretion ability of ADSCs,and the best concentration is 15 mg/L.
10.Dermal-fat graft combined with secondary autologous fat transplantation to reconstruct severe facial depression

Objective To investigate the method of dermal-fat graft combined with secondary autologous fat transplantation in repairing severe facial depression and to evaluate the clinical effects.Methods Twelve cases of facial depression had been repaired by the transplantation of dermal-fat flap which was removed from the abdomen at the first stage.They were given fat granules injection 1 to 3 times postoperatively,and 3,6 and 3 patients were given fat granules injection three times,twice and once,respectively,with the interval period of 3 to 6 months.The result was based on comparison of the photos taken from preoperation and postoperation.Results All patients were healed primarily except one of which was formed hematoma after operation and scavenged thereafter.After 6 months to 2 years follow-up,all the patients had satisfactory facial contour.Conclusions Combined autologous fat granules with free dermal-fat graft to reconstruct severe facial depress is an easy,safe and effective technique and deserves to be recommended.
