2.Cloning and functional characterization of a cDNA encoding isopentenyl diphosphate isomerase involved in taxol biosynthesis in Taxus media.

Taxol is one of the most potent anti-cancer agents, which is extracted from the plants of Taxus species. Isopentenyl diphosphate isomerase (IPI) catalyzes the reversible transformation between IPP and DMAPP, both of which are the general 5-carbon precursors for taxol biosynthesis. In the present study, a new gene encoding IPI was cloned from Taxus media (namely TmIPI with the GenBank Accession Number KP970677) for the first time. The full-length cDNA of TmIPI was 1 232 bps encoding a polypeptide with 233 amino acids, in which the conserved domain Nudix was found. Bioinformatic analysis indicated that the sequence of TmIPI was highly similar to those of other plant IPI proteins, and the phylogenetic analysis showed that there were two clades of plant IPI proteins, including IPIs of angiosperm plants and IPIs of gymnosperm plants. TmIPI belonged to the clade of gymnosperm plant IPIs, and this was consistent with the fact that Taxus media is a plant species of gymnosperm. Southern blotting analysis demonstrated that there was a gene family of IPI in Taxus media. Finally, functional verification was applied to identify the function of TmIPI. The results showed that biosynthesis of β-carotenoid was enhanced by overexpressing TmIPI in the engineered E. coli strain, and this suggested that TmIPI might be a key gene involved in isoprenoid/terpenoid biosynthesis.

4.Adrenal tuberculosis:contrast-enhanced CT features with clinical correlation

Objective To investigate the correlation between specific features on contrast-enhanced CT and its clinical findings of adrenal tuberculosis so as to improve the diagnostic accuracy of the disease. Methods Contrast-enhanced CT features in 30 patients with documented adrenal tuberculosis were retrospectively evaluated blindly for the features of location,size,morphology,attenuation and enhancement patterns on CT images,and compared with clinical and pathological materials.Results The common CT manifestations were as follows:enlargement of the adrenal glands in all 30 cases(bilateral 90%,mfilateral 10%)including mass-like enlargement in 13 cases and enlargement but the contours preserved in 17 cases, heterogeneity(28 cases,93.3%),calcification(17 cases,56.7%),and low attenuation in the center with peripheral enhancement(16 cases,53.3%)of the lesions.After antituberculosis chemotherapy, 5 cases of enlarged adrenal glands decreased in size or returned to normal size and configuration,with disappearance of the central low attenuation and new appearance of dot-like calcification in 2 cases.Cochran Armitage trend test showed there was an increasing tendency of calcification rate with clinical duration(X~2= 7.47,P
5.Nasal endoscopic surgical treatment for chondrosarcoma of paranasal sinus and the skull base

Objective To discuss the clinical characteristics and treatments for chondrosarcoma of paranasal sinus and the skull base. Methods The clinical characteristics of chondrosarcoma of paranasal sinus and skull base in 7 patients tmderwent endoscopic surgeries between 2001 and 2008 were analyzed. Of the patients,4 men and 3 women. The patients' age ranged from 18 to 47 years,with a median of 31 years. Clinical symptoms: stuffy, nose bleeding, runny, headache, diplopia, eye outreach limited, blurred vision and even blindness. Surgery methods:under nasal endoscopy,after the attachment sites of the tumors to normal tissues were confirmed,the tumors were peeled off along the clear boundary between the tumors and normal tissues,and the potential residual tumor tissues on bones were cleared by a drill. Results The patients were followed up postoperatively for 24 to 108 months,with a median of 36 months. Five of 7 patients were no recurrence,2 were alive with tumor. Conclusions Chondrosarcoma of paranasal sinus and skull base can be treated by nasal endoscopic surgery, with good clinical outcome.
6.A heterozygous transversion of connexin 50 in a family with congenital nuclear cataract in the northeast of China.

OBJECTIVETo identify the genetic defect causing autosomal dominant congenital cataract (ADCC) in a five-generation family in the northeast of China.
METHODSLinkage analysis was carried out with polymorphic microsatellites on the Human MapPairs marker set, special known loci. Mutation analysis of the candidate gene in the critical region was performed to detect the potential mutation.
RESULTSThe maximum Lod score (2.44 at recombination fraction theta=0) was obtained for markers D1S498,D1S305, and D1S2844. The cataract locus in this family constellation was mapped to 1q21.1 and 21.44 cM interval between D1S2344 and D1S2844, which were known to flank the gene coding Connexin 50 (Cx50) or gap junction protein alpha-8 (GJA8). Sequencing of the coding region of GJA8 gene showed a heterozygous transversion T>G in exon 2, which resulted in the substitution of glycine for valine at amino acid 64, and this position was in the first connexin signature region that characterized this protein.
CONCLUSIONThis is the first report on a mutation in the first connexin signature region of the GJA8 and a different mutation within Cx50 revealed in this family, which might account for the phenotypic differences observed. Furthermore, this study confirmed that GJA8 plays a vital role in the maintenance of human lens transparency.

7.Changes of the mitochondrial DNA copy number and the antioxidant system in the PBMC of hepatocellular carcinoma.

OBJECTIVETo investigate the relationship between the changes of the copy numbers of mtDNA in peripheral blood mono-nucle- ar cell(PBMC) and the disordered of antioxidant capacity of hepatocellular carcinoma (HCC) patients.
METHODSThe Ficoll Hypaque method was used to isolate the PBMC from blood specimens. The ND1 gene of the mitochondrial was amplified by real-time PCR; meantime β-actin was served as a quantitative standard marker; the difference of mtDNA copy number in PBMC was compared between HCC and healthy control group. The level of reactive oxygen species (ROS) in PBMC was determined by flow cytometry. The change of total antioxidant capacity (T- AOC) of plasma was detected by the biochemistry examination.
RESULTSThe copy numbers of ND1 gene in PBMC of HCC was 73% that of the healthy control group,which suggested a decrease of the copy numbers of mtDNA in HCC. The levels of ROS of PBMC in HCC was (417. 82 ± 110.62) and (301.82 ± 75.54) in control group, which showed that the levels of ROS of PBMC in HCC were significant higher than that in control group (P < 0.01).Plasma T-AOC in HCC was (1.30 ± 0.85), and (3.20 ± 1.62) in control. The T-AOC of plasma of HCC was significantly lower than in control group (P < 0.01).
CONCLUSIONThere was a certain relationship between the decrease of the copy numbers of mtDNA and the disordered antioxidant capacity in hepatocellular carcinoma, which may be associated with the development of hepatocellular carcinoma.

9.Analysis of iodine nutritional status of high-risk population in coastal salt-producing areas and coastal nonsalt-producing areas in Xiamen city of Fujian province

Objective To assess the iodine nutritional status of special target population in coastal saltproducing areas and coastal non-salt-producing areas in Xiamen city,and to provide a basis for take appropriate measures for prevention of iodine deficiency disorders.Methods The Xiang-An salt-producing areas and the JiMei non-salt-producing areas were chosen as research spots in 2009.One sample of produced water and 2 samples of tap water were collected to test iodine level; 600 children aged 8 to 10 were selected and thyroid palpation was performed,besides,the urine sample and household salt sample were also collected for iodine determination.Sixty pregnant women,breasffeeding women,and 0 - 2 year old infants were recruited,respectively,and urine samples and household salt samples were collected to perform the determination of iodine level.Results The iodine levels in drinking water of Xiang-An district and Ji-Mei district were 3.23 and 6.05 mg/L,respectively.The consumption rates of edible qualified iodinated salt were 84.4% (438/519) and 98.3% (392/399),respectively.The goiter rates of children aged 8 - 10 were 3.03%(19/628) and 0.67%(4/600),respectively.The medians of urinary iodine were 202.80 and 238.40 μg/L,respectively.The proportions of urinary iodine level < 50 μg/L were 3.5% (14/405) and 1.0%(2/202),respectively.The medians of urinary iodine of the pregnant women were 120.55 and 153.35 μg/L,respectively,and the proportions of urinary iodine level < 150 μg/L were 62.1% (46/74) and 46.8% (29/62),respectively.The medians of urinary iodine in three trimester were 173.10,144.75 and 101.90 μg/L,respectively,early trimester of pregnancy > second trimester and third trimester (Z =6.151,3.052,all P < 0.05),second trimester > third trimester (Z =2.016,P < 0.05 ).The medians of urinary iodine of the breastfeeding women were 131.20 and 104.35 μg/L,respectively.The proportions of urinary iodine level < 100 μg/L were 35.3% (24/68) and 46.7%(28/60),respectively.The medians of urinary iodine of the infants were 81.95 and 80.20 μg/L,respectively,the proportions of urinary iodine level < 100 μg/L were 59.7%(37/62) and 61.6%(40/65),respectively,< 50 μg/L were 32.3% (20/62) and 30.8% (20/65),respectively.Conclusions The levels of iodine nutrition in pregnant women,breastfeeding women,and 0 - 2 year old infants from Xiang-An district and Ji-Mei district in Xiamen city are still below the desired level of iodine nutrition,and the infants and pregnant women in coastal salt-producing areas are poor in iodine nutrition,we should pay close attention.We should strengthen market supervision on iodized salt,carried out iodine nutrition monitoring on pregnant women,breasffeeding women,and infants,and disseminate knowledge of iodine nutrition among high-risk population should be carried out immediately.

Tetramethylpyrazine (TMP), an effective component of traditional Chinese medicine Chuanxiong, is commonly used to resolve embolism. Its possible therapeutic effect against atherosclerosis has received considerable attention recently. Angiotensin II (Ang II) is highly implicated in the proliferation of vascular smooth muscle cells (VSMCs), resulting in atherosclerosis. The mechanisms of TMP in the proliferation of VSMCs induced by Ang II remain to be defined. The present study was aimed to study the effect of TMP on Ang II-induced VSMC proliferation through detection of nuclear factor-kappaB (NF-kappaB) activity and bone morphogenetic protein-2 (BMP-2) expression. Primary cultured rat aortic smooth muscle cells were divided into the control group, Ang II group, Ang II + TMP group and TMP group. Cells in each group were harvested at different time points (15, 30 and 60 min for detection of NF-kappaB activity; 6, 12 and 24 h for measurement of BMP-2 expression). NF-kappaB activation was identified as nuclear staining by immunohistochemistry. BMP-2 expression was observed through Western blot, immunohistochemistry and in situ hybridization. The results showed that: (1) Ang II stimulated the activation of NF-kappaB. Translocation of NF-kappaB p65 subunit from cytoplasm to nucleus appeared as early as 15 min, peaked at 30 min (P<0.01) and declined after 1 h. (2) TMP inhibited Ang II-induced NF-kappaB activation (P<0.01). (3) Ang II increased BMP-2 expression at 6 h but declined it significantly at 12 and 24 h (P<0.01). (4) BMP-2 expression was also kept at high level at 6 h in Ang II + TMP group but maintained at the normal level at 12 and 24 h. (5) There was no significant difference in NF-kappaB activation and BMP-2 expression between the control group and TMP group. These results indicate that TMP inhibits Ang II-induced VSMC proliferation through repression of NF-kappaB activation and BMP-2 reduction, and BMP-2 expression is independent of the NF-kappaB pathway. In conclusion, TMP has therapeutic potential for the treatment of atherosclerosis.
