1.Lenalidomide combined with anti-B-cell maturation antigen chimeric antigen receptor T cell for relapsed/refractory multiple myeloma: report of one case and review of literature

Objective:To explore the clinical effect of lenalidomide combined with anti-B-cell maturation antigen chimeric antigen receptor T cell (anti-BCMA CAR-T) in the treatment of relapsed/refractory multiple myeloma (RRMM).Methods:The clinical data of a patient with RRMM who underwent lenalidomide combined with anti-BCMA CAR-T therapy in Henan Province Hospital of Traditional Chinese Medicine in January 2020 were analyzed. Clinical manifestations, diagnosis and treatment were also analyzed, and the related literature was reviewed.Results:The patient was a 51-year-old man who was diagnosed as IgD-λ multiple myeloma (MM) in October 2015. The patient achieved remission after 10 courses of chemotherapy regimens including immunomodulators and proteasome inhibitors, followed by autologous hematopoietic stem cell transplantation. MM relapsed after 14 months of transplantation. His disease continued to progress after multiple chemotherapy regimens and mouse or human-derived anti-BCMA CAR-T therapy. After a conditioning chemotherapy regimen of fludarabine and cyclophosphamide, the patient took lenalidomide on day 1 and was infused human-derived anti-BCMA CAR-T cells on the next day. Grade 3 cytokine releasing syndrome (CRS) appeared after infusion, and was resolved after symptomatic treatment. Very good partial response (VGPR) was achieved on day 14 after anti-BCMA CAR-T treatment. VGPR had been maintained for more than 3 months by press time.Conclusion:Lenalidomide combined with anti-BCMA CAR-T therapy is feasible and effective in the treatment of RRMM.
2.Effect of jiangu erxian pill on proliferation and cell cycle of human osteoblast.

OBJECTIVETo further explore the effect of Jiangu Erxian Pill (JGEXP) on proliferation and cell cycle of human osteoblast on the basis of previous clinical and experimental studies.
METHODSHuman primary osteoblast were isolated and cultured. The cell proliferation was tested by 3H-thymine incorporation and methyl thiazolyl tetrazolium (MMT) method and the cell cycle was determined by flow cytometry technique.
RESULTSIn the medium and high dosage JGEXP groups, the cell proliferation rate and index, and percentage of diploid synthesis phase (S phase) cells were significantly higher than those in the blank control group (P < 0.01 or P < 0.05), and similar to those in the estrogen group; and the cell apoptosis rate and percentage of G0-G1 stage cells were lower than those in the blank control group (P < 0.01 and P < 0.05).
CONCLUSIONJGEXP could effectively promote the cell proliferation and differentiation, and prevent the cell apoptosis of osteoblast in vitro.

3.The function of circulating CD4+CD25+ regulatory T cells in patients with chronic hepatitis B

Objective To investigate the induction and function of circulating CD4+ CD25+ regulatory T cells(CD4+CD25+ Treg cells)in patients with chronic hepatitis B,and their role in the progress of chronic HBV infection.Methods Twenty two patients with chronic hepatitis B(CHB),11 patients with acute hepatitis B(AHB)and 20 healthy blood donators were enrolled in the study.After stimulating with recombinant HBV antigen or anti-CD3 mAb,the fraction of CD4+ CD25+Treg cells in peripheral blood mononuclear cells(PBMC)from different subjects was analyzed using flow cytometry.CD4+CD25+ Treg cells and effector cells(PBMC-Treg)were sorted out and cultured alone or together in vitro,and the cell proliferation and cytokines production were measured. Serum HBV titers,HBV markers and liver function were also evaluated.Results The fraction of circulating CD4+CD25+ Treg cells induced in PBMC from CHB patients under the stimulation of recombinant HBsAg was much higher than that in AHB patients(F=3.47,P<0.05)and normal controls(F=4.98,P<0.05).With HBsAg stimulation,cell proliferation and cytokines production (IFN-γand IL-10)of CD4+ CD25+ Treg cells either from patients or from normal controls was very low when cultured alone,and there was no significant difference between two groups(P>0.05).Cell proliferation and IFN-γproduction of effector cells from AHB patients were higher than those from CHB patients,however,when effector cells were cultured with CD4+Cd25+Treg cells at a ratio of 5:1,the cell proliferation and IFN-γ production were significantly inhibited (F=4.66,5.24,P<0.05).Conclusion Circulating nificantly suppress the production and IFN-γ production of effector T Cells,which may be associated with the progress of chronic HBV infection.
4.Effect of spot size on the result of single-particle ploidy detection

Objective To explore the relationship between the spot size and the result of ploidy analysis in the detection technology based on single-particles, and to fix the scope of spot thickness on the basis of the experimental result.Methods The influence of spot thickness on voltage signals produced by single cells was analyzed.The parameters of the beam shap system in the incident field were designed and optimized on ZEMAX.Finally, according to the cells′diameter, the target size of spots was set.A set of spots of different thickness and of Gaussian distribution obtained from the optical experimental platform was used to conduct ploidy detection experiments.Results Target spots were both obtained from ZEMAX simulation and the optical platform.When the spot thickness was larger than both monocytes and coenocytes, the mean fluorescence intensity ratio was 2.03,which met the demand of the index.Conclusion When the height of the pulse is used to represent the fluorescence density, the relative size of spots and cells will affect the result of ploidy detection.Only when spot thickness is larger than cells is the ploidy ratio accurate.
5.Uses of fresh herbs with antioxidant effect and prospect for population ecology reconstruction in Macau.

The average life expectancy in Macau is ranking the second in the world, the consumption of fresh medicinal plants is a profound culture in Macau. The paper focus on the distribution of the antioxidant herbs, a comprehensive investigation and analysis the amount of the plant resources was carried out. The antioxidant activity of alcohol extracts was determined by using the DPPH method, and six kinds fresh herbs with high antioxidant free radical activity were screened out. Reference to adult daily dose of vitamin C, it is calculated that the daily dose amount of fresh herbs is less than 200 g. For the expected shortage of resources and the ecological status of Macau, we give some suggestions of herbal introduction in population ecology reconstruction.

6.Rapid automated analysis method of flow cytometry data

Objective A major component of flow cytometry data analysis involves gating , which is the process of identifying homogeneous groups of cells .As manual gating is error-prone, non-reproducible, nonstandardized, and time-consuming , we propose a time-efficient and accurate approach to automated analysis of flow cytometry data .Methods Unlike manual analysis that successively gates the data projected onto a two-dimensional filed, this approach, using the K-means clustering results , directly analyzed multidimensional flow cytometry data via a similar subpopulations-merged algorithm.In order to apply the K-means to analysis of flow cytometric data , kernel density estimation for selecting the initial number of clustering and k-d tree for optimizing efficiency were proposed .After K-means clustering , results closest to the true populations could be achieved via a two-segment line regression algorithm .Results The misclassification rate (MR) was 0.0736 and time was 2 s in Experiment One, but was 0.0805 and 1 s respectively in Experiment Two. Conclusion The approach we proposed is capable of a rapid and direct analysis of the multidimensional flow cytometry data with a lower misclassification rate compared to both nonprobabilistic and probabilistic clustering methods .
7.The protective effect of Yuyin Ruangan Decoction on experimental hepatic injury.

OBJECTIVETo investigate the protective effects of Yuyin Ruangan Decoction(YRD, traditional Chinese medicine) on experimental hepatic injury in mice.
METHODSThe mice were randomly divided into control group, model group and YRD low, middle and high dose group(n = 11). By ip injection of D-GalN, CCk or thioacetamide (TAA), three models of hepatic injury mice were established to investigate the effects of YRD through detecting the indexes of liver function in serum and, the content of antioxidant system in the hepatic tissue.
RESULTSYRD could decrease the content of alanine aminotransferase (ALT)and aspartate aminotransferase (AST) in serum and that of malonaldehyde (MDA) in the hepatic tissue, upregulate the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in the hepatic tissue. Furthermore, the above effects were dosedependent in a certain degree. CoNCLUSION: YRD has some protection effects on the model of experimental hepatic injury in mouse.

8.Adeno-associated vector mediated intracellular biological activity of human Kallistatin.

Human tissue kallikrein-binding protein (Kallistatin, KAL), a secretory protein that participates in the regulation of multiple signaling pathways by binding to the extracellular receptor, however, at present has not been reported about the intracellular activity, and whether it has the similar biological activity with extracellular activity. Here we constructed no signal peptide KAL (NSK) into the adeno-associated virus vector to explore the intracellular activity of KAL. Both the endothelial cell and lung cancer cells could express KAL, but not secreted after rAAV2-NSK transfection. The proliferation and migration of human umbilical vein endothelial cells (HUVECs) were inhibited, but the apoptosis rate was not affected. The proliferation rates, mobility and tubule formation of all the three tested lung cancer cells, such as NCI-H446, NCI-H460 and A549, were inhibited to different extents. This cellular study not only confirmed the intracellular activity, but also suggested it may serve as a kind of "balance factor" in multi-targeted controlling, which may provide a new train of thoughts to explain the regulatory contradiction in PI3K-Akt signaling pathways by KAL.

9.An algorithm of real-time peak detection for pulse signals based on FPGA

Objective To present a new algorithm of real-time peak detection for pulse signals based on FPGA , which is known as the dynamic threshold with half peak detection method ( DT-HP ) .Methods With Gaussian-shaped pulse signals as the target , the method was improved from conventional methods .The FPGA detection process of the algorithm required no more than three detection parameters: the starting point , the maximum value and the pulse width .Results The algorithm solved the floating baseline and repeating detection that occur in traditional methods .Compared with the results of polynomial fitting method and flow cytometry , the difference was only 3.2% and 9.3%.Conclusion The algorithm takes less time ,RAM, and cache while allowing floating baseline detection , which can be used as an effective method for rapid detection in FPGA .
10.Effect of Asi-antidiarrheal capsule on gastrointestinal goblet cell of thyroid hormone-induced diarrheic rats

Objective To study the effect of Asi-antidiarrheal capsule on gastrointestinal goblet cell of thyroid hormone-induced diarrhea.Methods Total of 120 SD male rats aged about 8 weeks were randomly divided into 2 groups:control group(10 rats)and thyroid hormone-induced diarrheic group(110 rats).Rats in control group were lavaged with normal saline 1 ml/d.Thyroid tablets were partly desolved into normal saline forming a 40 mg/ml suspension.Rats in thyroid hormone-induced diarrheic group were given the thyroid suspension 1 ml/d to make thyroid hormone-induced diarrheic model.Serum FT3 and FT4 were tested.Fourty thyroid hormone-induced diarrheic rats were screened out according to serum FT3 and FT4 levels,body weight and wet stool.The fourty rats were randomly divided into 5 groups,8 rats in each group:positive control group,berberine group,low-dose,mediandose and high-dose groups.Normal saline of 1 ml/d was admnistered to diarrhea control group,1.94 g·kg-1·d-1 Berberine capsule was given to positive control group,and 0.63,1.26,2.52 g·kg-1·d-1 Asi-antidiarrheal capsule to low-dose,mediandose and high-dose groups,respectively.After sever days treatment,rats are executed.Duodenum,jejunum,ileum and colon were dissected,respectively.Histology observation and cell counting were carried out under light micmscopo on HE coloration.Cell counting unit was defined as:cell/high power field of vision (cells/hpf).Results In jejunum,the number of goblet cells in berberine group,mediandose group and high-dose group[(15.32±2.53),(20.24±1.24),(14.98±1.10)cells/hpf,respectively],were all lower than that of the diarrhea control group[(25.73±4.55)cells/hpf,all P<0.05]with an exception of low-dose group[(23.98±2.28)cells/hpf].The numbers of goblet cells in berberine control group,low-dose group,mediandose group and highdose group[(18.29±1.33),(20.61±2.12),(19.38±2.01),(16.34±1.55)cells/hpf,respectively]were all less than that of the control group[(23.36±3.10)cells/hpf,all P<0.05].The numbers of goblet cells of diarrhea control group and high-dose group were obviously lower than that of the low-dose group(all P<0.05)in jejunum and colon.The numbers of goblet cells of Duodenum and ileum were not significantly different between groups(F=2.81,2.67,all P>0.05).The numbers of goblet cells in the diarrhea control group increased markedly observed under microscope,but decreased following therapeutic treatment.Conclusions The numbers of goblet cells from jejunum and colon in thyroid hormone-induced diarrheic rats are increased significantly.Asi-antidiarrheal capsule can remarkably decrease the number of goblet cells in jejunum and colon,and reduce mucus secretion.
