Journal of Shanghai Jiaotong University(Medical Science) 2006;0(09):-
Objective To determine the mechanism of nitric oxide synthase(NOS) and prostacyclin(PGI2) acting on splanchnic hyperdynamic circulation of portal hypertention(PHT). Methods Ninety-six Sprague-Dawley rats were divided into three groups, namely, intrahepatic portal hypertension(IHPH, n=31), prehepatic portal hypertension(PHPH, n=33) and sham-operated controls(SO, n=32). Animals of each group were received indomethacin(INDO) either on a short term or long term with saline as control. Portal venous pressure, together with the concentration of nitric oxide (NO) and PGI2 in serum was measured. The constitutive nitric oxide synthase(cNOS)and inducible nitric oxide synthase(iNOS)activity in the abdominal aorta and small intestine of these rats were detrmined by spectrophotometry method. RT-PCR was performed to measure the levels of iNOS and cNOS mRNA in the arteries and guts mentioned above. Results Although INDO decreased the concentration of PGI2 in serum, the long-term INDO-treated group restored splanchnic hyperdynamic circulation in both IHPH and PHPH rats, concomitant with enhanced expression of iNOS and concentration of NO(P0.05). Conclusion Overproduction of NO inducing hemodynamic abnormalities of PHT is synthesized principally by increase of iNOS. There may be a possible interaction between PGI2 and NO in hyperhemodynamics of PHT, while PGI2 may not be a mediator in the formation and development of hyperdynamic circulatory state.

Shanghai Journal of Acupuncture and Moxibustion 2019;38(2):164-168
Objective To observe the effect of bloodletting at Jing-well points on the recovery of cognitive function and prognosis in patients with post-stroke cognitive impairment. Method One hundred patients with post-stroke cognitive impairment were enrolled and divided by the random number table into two groups, with 51 cases in the observation group and 49 cases in the control group. The control group was intervened by conventional treatment, while the observation group was additionally given bloodletting at Jing-well points. The two groups were observed and compared in terms of Mini-Mental State Examination (MMSE), Montreal Cognitive Assessment (MoCA), Barthel Index (BI), and the plasma cortisol level before and after treatment. The therapeutic efficacy and occurrence rate of adverse reactions of the two groups were also compared. Result After treatment, the MMSE, MoCA and BI scores all increased in the two groups, but the scores of MMSE, MoCA and BI in the observation group were higher than those in the control group (P<0.05). The total effective rate was 94.1% in the observation group, higher than 71.4% in the control group (P<0.05). The plasma cortisol level dropped after treatment in both groups (P<0.05), and the level in the observation group was lower than that in the control group (P<0.05). The occurrence rate of adverse reactions was 7.8% in the observation group, lower than 22.4% in the control group (P<0.05). Conclusion Bloodletting at Jing-well points as assistant can effectively improve the mental state, cognitive function and activities of daily living, and to some extent reduce the happening of adverse reactions in the treatment of post-stroke cognitive impairment.

OBJECTIVE Cistanche deserticola Y. C. Ma is an official plant that grows in arid or semi-arid areas. Our early work demonstrated that Cistanche extracts protect against sperm damage in mice under bisphenol A induced reproductive damage. Echinacoside (ECH)is one of the major active components. The present study investigated the mechanisms behind the possible protective effects of ECH against oligoasthenospermia in rat and identified the interaction between ECH and AR. METH-ODS The distribution of ECH was assayed by HPLC,the quantity and quality of sperm was evaluated and hormone levels were determined by radioimmunosorbent assay. The levels of androgen receptor (AR)and key steroidogenic-related genes were reduced as determined by Western blotting and qPCR analysis. The interaction between ECH and AR were evaluated by fluorescence localization assay,indi-rect ELISA and molecular docking. RESULTS ECH significantly increased the quantity of sperm and secretions of luteinizing hormone and testosterone.ECH was distributed to the hypothalamus but not in the testes.ECH combined with hypothalamic AR in the pocket of Met-894 and Val-713 to inhibit transfer of AR from the cytoplasm to nuclei in the hypothalamus. While negative feedback of sex hormone regula-tion was inhibited, positive feedback was stimulated to increase the secretion of luteinizing hormone and testosterone subsequently enhancing the quantity of sperm. C. militaris significantly alleviated the BPA-induced reproductive damage by increasing testicular superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and glutathione (GSH); as well as by reducing serum malondialdehyde (MDA). C. militaris not only obviously enhanced the levels of serum LH and T, but it also improved the sperm count and motility compared to the BPA-treated group.CONCLUSION C.militaris and ECH protect the BPA induced reproductive damage.ECH blocks AR activity in the hypothalamus to increase the quantity of sperm and protect against oligoasthenospermia in rats.
