2.Comparison of cyclopentolate and atropine on cycloplegia in children

AIM: To compare the effectiveness on 10g/L cyclopentolate and 10g/L atropine on cycloplegia in children before optometry.
METHODS:Eighty eyes of 40 children among 4-12 years old with refractive error were recruited in this study. 10g/L cyclopentolate eye drops were topically administered once per 5min for 3 times and optometry was performed after 45min. Three days after that, 10g/L atropine sulfate eye gel then was used 3 times per day for consecutive 3d and again the refractive diopter was obtained at the 4th d. The differences of the results in retinoscopy refraction were compared between 10g/L cyclopentolate eye drops and 10g/L atropine sulfate eye gel.
RESULTS: Therefraction results of those given 10g/L cyclopentolate eye drops and 10g/L atropine sulfate eye gel were no statistical different in both 4-8 years group and 9-12 years group with myopia (≤-3. 00D) (P=0. 411, 0. 924). The differences of refraction results of both the drugs were significant in 4 - 8 years group with low hypermetropia, medium hypermetropia and high hypermetropia (P=0. 007, 0. 007, 0. 009). No significant difference was found in 9 - 12 years group with low hypermetropia (P= 0. 592), given 10g/L cyclopentolate eye drops and 10g/L atropine sulfate eye gel, but the differences of refraction results of both the ophthalmic preparations above were significant in 9-12 years group with medium and high hypermetropia (P=0. 039, 0. 012).
CONCLUSION:Both 10g/L cyclopentolate eye drops and 10g/L atropine sulfate eye gel have the cycloplegic effects, but their cycloplegic effects are significant different among hypermetropia children. Thus, the reasonable cycloplegic should be chosen according to the specific situation.

6.Isolation and characterization of chronic myelogenous leukemia bone marrow mesenchymal stem cells and their hematopoiesis support ability

Objective To study the characteristics of mesenchymal stem cells(MSCs)derived from chronic myelogenous leukemia(CML)patients' bone marrow and examine their hematopoiesis support in vitro.Methods MSCs from CML were obtained and cultured.Immunophenotype and in vitro differentiation capacities were investigated.Moreover,the Ph chromosome and bcr/abl gene of CML derived MSCs were detected.The expression of cytokines was detected by RT-PCR,and hema- topoiesis support of MSCs in vitro was detected by long-term bone marrow culture and the methylcel- lulose progenitor assay.Results CML derived MSCs showed a typical fibroblast like morphology. They were positive for CD29,CD44 and CD105,while negative for CD14,CD31,CD45,CD34 and HLA-DR.Under suitable conditions,CML derived MSCs could differentiate into osteoblasts and adi- pocytes.Further,CML derived MSCs showed a normal choromosome,and did not express bcr/abl gene.At last,they expressed hematopoiesis cytokines and possessed ability of hematopoiesis support in vitro.Conclusion There are MSCs with multidirectional differentiation potential in CML bone mar- row and they possess the ability of hematopoiesis support.
7.Establishment of lymphatic metastasis model for human gastric cancer in nude mice

Objective To investigate the method of establishing lymph node metastasis model for gastric cancer in nude mice.Methods Human poorly differentiated gastric cancer SGC-7901 ceils were cul- tured,inherited,amplified and collected in vitro,and were inoculated in nude mice subcu.taneously.After pas- saging six generations,the subcutaneous tumor was orthotopically implanted in nude mice.The mice were then sacrificed on the 9th week after implantation.Serum CEA density was detected using radio-immunity method. Orthotopically implanted tumors,remote organ metastases as well as lymph node metastasis were observed macrescopically and histologically.Results The successful rate of orthotopically transplantion of gastric can- cer was 100 %.The rate of perigastric lymph node metastases was 93.3 %.Local infiltration and remote metastasis were observed.The level of serum CEA in nude mice bearing tumor was significantly higher than that of normal nude mice(P
8. Inhibiting activation of ERK1/2 pathway improves atrial fibrosis and connexin40 remodeling in rats

Objective To investigate the effects of extracellular signal-regulated kinase l/2(ERKl/2) pathway inhibitor U0126 on isopreterenol(ISO)-induced atrial fibrosis and connexin 40 (Cx40) remodeling in rats. Methods Thirty-two male SD rats were evenly randomized into control group, DMSO group, ISO (5 mg/[kg • d])+DMSO group (fibrosis group), and ISO (5 mg/[kg • d]) + U0126 (0. 5 mg/[kg • d]) + DMSO group (U0126-treated group). The corresponding reagents were given to each group once a day and the rats were killed and the myocardial tissues were collected after 7 d. The Ang IJ contents in the myocardial tissues were measured by radioimmunoassay; H-E staining and Masson staining were applied to measure the degree of atrial fibrosis; p-MEKl/2, p-ERKl/2, and Cx40 were detected by immunohistochemistry method. Results (1) The contents of Ang IJ were similar between control group ([242. 133 ± 4. 870] ng/L) and DMSO ([239. 412 ± 1. 795] ng/L) group (P>0. 05). Compared with the above two groups, Ang IJ contents in fibrosis group ([500. 250 ± 8. 869] ng/L)and U0126-treated group([498. 695 ± 9. 340]ng/L) were significantly increased (P alKO. 01). (2) Control group and DMSO group had no atrial fibrosis; the degree of atrial fibrosis in U0126-treated group was significantly lower than that in the fibrosis group (P<0. 01). (3) p-MEKl/2 and p-ERKl/2 expressions were similar in control group and DMSO group (P>0. 05), and those in the fibrosis group were significantly increased compared with control group and DMSO group(P<0. 01); the expression in U0126-treated group was similar to those in the control group and DMSO group(P>0. 05), and was significantly decreased compared with the fibrosis group(P<0. 01). (4) The contents of Cx40 were similar between control group and DMSO group (P>0. 05), and Cx40 was distributed in myocardial cell intercalated disc in a linear manner. The content of Cx40 was significantly reduced (P<0. 01) in the fibrosis group compared with the control group and DMSO group, with Cx40 distributed in disorder. The content of Cx40 in U0126-treated group was similar to that in the control group and DMSO group(P>0. 05)and most of the Cx40 was linearly distributed in myocardial cell intercalated disc. Meanwhile, the reduce degree of Cx40 content in U0126-treated group was significantly decreased than that in the fibrosis group(P<0. 01), and some Cx40 was linearly distributed in myocardial cell intercalated disc. Conclusion Long-term Ang TJ elevation in myocardium may be involved in atrial fibrosis and Cx40 remodeling, and U0126 can efficiently improve atrial fibrosis and Cx40 remodeling by inhibiting the activation of ERK1/2 pathway.
9.Endovascular embolization in managing intracranial aneurysms and prevention and cure for its complications

Objective To discuss the techniques and the prevention and cure of preoperative complications of the endovascular embolization in managing the intracranial aneurysms. Methods A total of 90 aneurysms in 88 patients were treated by detachable coils embolization. The clinical results and the management of complications were reported. Results Seventy aneurysms were occluded at 100%, 13 aneurysms at 90% to 95% and 5 aneurysms at only 80. The perioperative complications occurred in 9 patients, including the coil escaping from the aneurysm sac into its parent artery in 3 patients, re-rupturing of aneurysm in 4 patients during operation and serious vasospasm in 2 patients. Two patients died. Conclusion The endovascular embolization is a safe, effective and micro invasive method in managing intracranial aneurysms. The exact embolizing techniques chosen are very important to improve the curative results and decrease the complications of embolization of intracranial aneurysms.

Background The key premise of genetic research and treatment is to select the desired gene vector,ultrasound microbubbles as a new type of gene vector can safe,fast and effectively enhance the gene transfection and expression by reversibility increasing the permeability of cells. Objective This study was to observe the effects of ciliary neurotrophic factor(CNTF)gene mediated by ultrasound microbubbles intraocular transfer on visual function and retinal ganglion cell(BGCs)after optic nerve injury. Methods Fifty-five adult Sprague-Dawley(SD)rats were divided into 6 groups randomly,including normal control group(n=5),sham injury group(n=10),simple injury group(n=10),naked plasmid group(n=10),plasmid+ultrasonic irradiation group(n=10)and ultrasound microbubbles group(n=10).The model of optic nerve injury Was made by forceps clip on the fight optic nerve.and the corresponding intervene was performed in different groups.Flash visual evoked potential(F
