1.Therapeutic effect of deproteinised calf serum eye gel on corneal alkali burn

Background Corneal chemical burns,especially hazards of alkali burn become increasingly prominent.Clinically,anti-inflammatory,immuno-suppression,corneal transplantation are the common treating method for corneal alkali burn.But the research of tissue repair under the microenvironment of corneal alkali burn is necessary.Objective The aim of this study was to investigate the therapeutic effect of deproteinised calf serum eye gel on the corneal alkali burn.Methods Alkali burn model of cornea was established on the right eyes by putting the filter paper with 0.5 mol/L NaOH on the center cornea for 1 minute in 24 white rabbits.The model rabbits were divided randomly into 4 groups.Normal saline solution,deproteinised calf serum eye gel,blank matrix gel or recombinant bovine basic fibroblast growth factor(rb-bFGF)eye-gel was topically administered 4 times per day for 14days in the 4 groups,respectively.The inflammatory reaction was examined under the slit lamp and scored based on Ando' s criteria.Corneal fluorescine staining was performed to calculate the corneal ulcer area and scored based on Trousdale' s criteria.Histopathological examination of corneas was performed on the fourtcenth day after experiment.The use of the experiment animals complied with ARVO Statement.Results Corneal edema and opacification were seen in the model eyes with the modeling successful rate 100%.On the seventh day after experiment,the severe ulcer of cornea and hypopyon appeared in the normal saline solution group.Corneal epithelium was intact but the intrarocular structure was invisible in the blank matrix gel group.In th(c) rb-bFCF group,corneal new vessels were seen,however,the corneal ulcer completely regrow in the deproteinised calf serum eye gel group.In 3,5,7,10 and 14days after examination,the corneal inflammatory scores were significantly lower in the deproteinised calf serum eye gel group and rb-bFGF group than those of the normal saline solution(P<0.01).No significant difference was found in the inflammatory score between the deproteinised calf serum eye gel group and rb-bFGF group (P>0.05) but was significantly lower than the blank gel matrix group (P < 0.05).With respect to the corneal ulcer,the score was decreased in the deproteinised calf serum eye gel group compared with the normal saline solution group and blank gel matrix group (P < 0.05).Howcver,no significant difference was found in the corneal ulcer score between the deproteinised calf serum eye gel group and rb-bFGF group in various time points (P> 0.05).Conclusions Deproteinised calf serum eye gel can promote the healing of corneal ulcer and remit the inflammatory response afler corneal alkali burns with a better effectiveness than rb-bFGF.
2.Regulation of the imbalance of interferon-gamma and interleukin-4 of serum and pulmonary tissue homogenate in asthmatic guinea pig with scoparone

BACKGROUND: Scoparone,which is a major principle separated from traditional Chinese herbs artemisia scoparia, which possesses actions of immunosuppression, anti-inflammation and relaxing bronchial smooth muscle. It' s not known whether scoparone possesses the regulation of the imbalance of interferon-gamma(IFN-γ) and interleukin-4(IL-4) in asthmatic guinea pig.OBJECTIVE: To investigate the inhibition and immunoregulation of scoparone on asthmatic guinea pig.DESIGN: Randomized controlled study based on the experimental animals.SETTING: Department of ethnopharmaeology in a university.MATERIALS: The experiment was completed in the Department of Ethnopharmacology,China Medical University between July 2000 and March 2002. Totally 84 male guinea pigs were selected at sanitation grade,weighting about(200 ± 50) g.INTERVENTIONS: The ameliorative effects of scoparone on the imbalance of IFN-γ and IL-4 in asthmatic guinea pigs were investigated by using the acute and delayed asthmatic guinea pigs which were respectively challenged by the aerosol mixture of 10 g/L histamine and 1 g/L acetylcholine,and 10 mL/L ovalbumin.MAIN OUTCOME MEASURES: The asthmatic latent periods in acute and delayed asthma, and the levels of IgE, IL-4 and IFN-γ in serum and pulmonary tissue homogenate in the asthmatic animals.RESULTS: Scoparone significantly prolonged the asthmatic latent periods in acute and delayed asthma,observably reduced the levels of IgE and IL-4 and raised the content of IFN-γin serum and pulmonary tissue homogenate in the asthmatic animals (P<0. 05).CONCLUTION: The anti-asthmatic effects of scoparone result from the ameliorative effects on the imbalance of IFN-γ/IL-4 ratio thereby to inhibit the abnormal synthesis of IgE.

7.Antioxidative and antinitrosative effects of volatile oil from A.longiligulare T.L.Wu on ulcerative colitis mice

AIM: To investigate antioxidative and antinitrosative effects of volatile oil from A.longiligulare T.L.Wu on dextran sulfate sodium(DSS)-induced ulcerative colitis mice. METHODS: Balb/c mice were fed with 4% DSS solution for 7 d to induce ulcerative colitis.Using biochemical method,the activity of antioxidative enzyme SOD,MDA and NO were determined in normal,model,SASP and three mouse's groups with low,moderate and high volatile oil from A.longiligulare T.L.Wu respectively.At the same time,the activity of iNOS was also measured by immunohistopathology. RESULTS: The concentration of MDA and NO were reduced and SOD increased significantly in high-and moderate-volatile oil groups compared with those in the model group.The activity of iNOS was reduced significantly in high-and moderate-volatile oil groups compared with those in the model group.The results demonstrated that the expression of iNOS was significantly inhibited in DSSinduced ulcerative colitis mice after being treated with high or moderate-dosage volatile oil. CONCLUSION: Volatile oil from A.longiligulare T.L.Wu has antioxidative and antinitrosative effects which may be one of the mechanism for treating UC.

9.Trial study on design of irradiated fields of radiotherapy in cervical and upper thoracic esophageal cancer

Objective To compare three kinds of irradiation treatment plans for cervical and upper thoracic esophageal cancer,in order to arrived at proper decision for the patient.Methods From February 2001 to June 2004,43 such patients were studied with three different simulated treatment plans made including conformal plan,conventional four-field plan and conventional two-field plan for every one.All plans were evaluated with iso- dose curve and dose-volume histogram.Results GTV on 95% isodose curve was 99.5%,98.2% and 87.4% in conforaml plan,conventional four-field plan and conventional two-field plan,respectively;PTV_1 and PTV_2 on 95% isodose with 97.8%,97.2%,94.8% and 95.8%,86.6%,73.7%.The volume of>20 Gy dose of left lung accepted was 18.6%,17.2% and 32.3%,in conformal plan,conventional four-field plan and conventional two-field plan,respectively;the right lung received 20.5%,19.9% and 35.5%.Conclusions Conformal plan is the best in radiotherapy,as it can provide ideal dose distribution of irradiated target with adequate protection of the normal tissues.Conventional four-field plan,being easy to carry out,can replace the conformal plan in most situations.Conventional two-field has the most uneven dose distribution and largest lung volume irradiated.
10.Experimental study on anti-inflammation and analgesia effect and acute toxicity of extract from Pongamia pinnata roots

AIM: To study the effect of extract from Pongamia pinnata roots on anti-inflammation and analgesia and acute toxicity. METHODS: The models of mice ear edema induced by xylene and Cotton pellet granuloma in rats to observe the anti-inflammation effect of PRE via oral administration. The effect of PRE on analgesia was tested by measuring the latent period licking hind foot with the hot plate method and counting body twisting induced by acetic acid in mice. The acute toxicity of PRE was measured by the method of Bliss. RESULTS: PRE could significantly inhibit the ear edema caused by xylene in mice, granuloma hyperplasia caused by cotton in rats. It could significantly prolong the pain threshold on hot-plate in mice, reduce the writhing times in mice. The LD50 of PRE was 6. 371 8 g/kg, its 95% confident limit was 5. 408 4-7. 723 2 g/kg. CONCLUSION: PRE has obvious effect on anti-inflammation and analgesia and the lower acute toxicity.
