1.THE STUDY OF RAT MYOGENIC SATELLITE CELLS DIFFERENTIATION TO MYOBLASTS IN VITRO

Objective To investigate the whole differentiating process of myogenic satellite cells in vitro,and the expression of N-ACh receptors.Methods Skeletal muscle satellite cells were isolated by trypsin digestion in vitro and purified by preferential attachment method.The cells were observed morphologically and identified by ?-sarcomeric actin,desmin and skeletal myosin antibodies.Contraction of myotubes was observed after adding ACh in the culture medium, N-AChRs in myotube were also be identified by bungarotoxin.Results Cultured satellite cells in vitro differentiated into myoblasts and fused to form myotubes later.These cells were positive by ?-sarcomeric actin, skeletal myosin and desmin antibodies' staining.Adding ACh directly in the medium stimulated the contraction of myotubes.No obvious expression of ACh receptors was observed on the surface of satellite cells.Cluster of receptors aggregated on myotubes from neonatal rats' satellite cells cultured in differentiation media.Conclusion The myotubes differentiated from myogenic satellite cells have the function of contraction.N-ACh receptors were synthesized gradually during the process of differentiation.
2.Effects of mesenchymal stem cells transplantation on the brain-derived neurotrophic factor and growth associated protein-43 after the spinal cord injury of rats

[Objective]To observe the effects of mesenchymal stem cells(MSCs) transplantation on the brain-derived neurotrophic factor(BDNF) and growth associated protein-43(GAP-43) after the spinal cord injury(SCI) of rats, and to investigate the mechanism of repairing the SCI by MSCs transplantation. [Method]Seven days after the operation of SCI,the MSCs were transplanted into the injured site. Then GAP-43 and BDNF were tested by RT-PCR. By means of above,the mechanism of repairing the SCI after the cell transplantation was investigated.[Result]Compared with group B, transplantation of MSCs enhanced the expression of BDNF mRNA and GAP-43 mRNA in the spinal cord of group A, improved the neuron regeneration.[Conclusion] The transplantation of MSCs can repair the injuried site and promote the regeneration of axon by enhanced the expression of BDNF mRNA and GAP-43 mRNA. It is one of the mechanism of repairing the SCI by MSCs transplantation.
3.Research progress in novel PA protein members of influenza A viruses.

Influenza poses a great threat to life and health in populations worldwide. Studies regarding the protein components of influenza viruses will facilitate the research and development of vaccines and diag nostic reagents. The influenza virus contains both structural and non-structural proteins. From the outset, it has been accepted that an influenza A virus possesses eight gene segments that encode eight corresponding viral proteins, respectively. Research has demonstrated that the M gene encodes the M2 ion channe! protein and the NS gene encodes the non-structural protein, NS2. In recent years, several novel viral proteins have been identified from influenza A viruses. This article will briefly describe the state of current research into PA-related proteins of influenza A viruses.

4.Performance evaluation of HCV antibody detection by bi-directional lateral flow method

Objective To analyze the clinical performance of hepatitis C virus antibody detection by bi-direetional lateral flow method and evaluate whether the bi-directional lateral flow method is suitable for clinical use.Methods Four hurdred and nineteen samples were collected and tested anti-HCV antibodies with hi-directional lateral flow and CMIA kits.These samples contain:107 patients with confirmed hepatitis C virus infection,162 patients with other diseases than hepatitis C virus infection and 150 healthy controls.Two positive reference materials for ELISA were tested at the same time.The testing results were analyzed with statistical methods.Results Compared with RIBA,the sensitivity and specificity of bi- directional lateral flow method were 98.1% and 99.7% respectively,the agreement rate with CMIA was 99.0%.The reproducibility percentage of the+/-20% at the cutoff concentration were 97% and 99% respectively.No crossing reaction with hepatitis A/B/D/E virus infection and HIV-1/2 positive individuals were found in the tests.Conclusion The hi-directional lateral flow method for HCV detection shows good clinical performance and is suitable for clinical use.
5.Effect of hypoxia on expression of angiopoietin-2 in human retinal pigment epithelium

Background Choroidal neovascularization(CNV)has been descrihed as a main reason of visual loss in a lot of ocular diseases.Researches showed that local hypoxia and retinal pigment epithelial(RPE) cells play an important role in the formation of CNV.A closely relationship of angiopoietin-2 (Ang-2) and angiogenesis has been proved.However,whether the expression of Aog-2 in hypoxic cultured human RPE cells is associated with the pathogenesis of CNV is still below understood.Objective This study was to investigate the effects of hypoxia on expression of Ang-2 in cultured human RPE cells in vitro,and discuss the possible effects of Ang-2 in the formation of CNV.Methods Human RPE cells were cultured and passaged,and 4-7 generation of cells were used in the experiment.The cells were incubated in cultural plate at the density of 5×107 cells/L.The culture medium containing 200 μmol/L CoCl2 was used to establish the hypoxia model of human RPE cells culturecd in vitro for 0.5,1,2,4,6,12and 24 hours,and the RPE cells cultured under normoxia were as controls.Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of Ang-2 mRNA in cultured human RPE cells,and enzymelinked immunosorbnent assay(ELISA) was used to assay the content of Ang-2 protein in supernatant of cultured human RPE cells.Results The survival rate of human RPE cells was 90% after resoscitation.The fourth generation of cells showed the fusiform with the less pigment in them.The Ang-2 mRNA/β-actin mRNA value in human RPE cells was significantly different among various groups(F=1086.30,P=0.00),The Ang-2 mRN A/β-actin mRNA value in hypoxia cultured for 0.5 hours group began to increase and peaked in hypoxia culture for 4-6 hours group,with the significant differences in comparison with normoxia control group(P<0.05).The Ang-2 mRNA/β-actin mRNA value decreased to the baseline level at hypoxia for 24 hours.The ELISA analysis showed that the concentration of Ang-2 protein in supernatant of RPE cells showed significant difference among groups(F=1034.00,P=0.00).The concentration of Ang-2 protein increased at hypoxia culture for 0.5 hours and peaked at 6 hours,showing significant differences in comparison with the control group (P<0.05).Conclusions Hypoxia could significantly up-regulate the expression of Ang-2 in human RPE cells cultured in vitro.Ang-2 expresses highly in the early stage of hypoxia,implying that Ang-2 participates in the formation of CNV.
6.Comparative study of main components of ginseng on immune function of rats.

Ginseng and its effective components are famous for their influence to enhance human immunity, regulate endocrine and antioxidant action. However, the different effects of different components are not clear. In this study, Wistar rats were used to study the effects of main components of ginseng, including total ginsenoside, panaxadiol saponins, panaxtrol saponin and ginseng polysaccharide. The results showed that the effects of panaxadiol saponins and ginseng polysaccharide on improving animal immune organ weight, plasma interleukin 2 (IL-2), interleukin 6 (IL-6), plasma gamma-interferon (IFN-γ), tumor necrosis factor alpha (TNF-α) were better than that of the other groups. Total ginsenoside and panaxtrol saponin can effectively increase the concentration of spleen NK cells (NKC) while panaxadiol saponins and ginseng polysaccharide can significantly increase the concentrations of rat plasma adrenocorticotrophic hormone (ACTH), corticosterone (CORT) and thyroid stimulating hormone (TSH). As for the effect of increasing organization nitric oxide (NO) and superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA), total ginsenoside is better than that of other groups. In brief, different components in ginseng possess different effects on enhancing immunity, regulating endocrine and resisting oxidation. Panaxadiol saponins and ginseng polysaccharide are better in enhancing immune, and total ginsenoside shows advantages in resisting oxidation and stress.

7.Noninvasive positive pressure ventilation in early acute respiratory distress syndrome

10.Influence of alveolar ventilation changes on gastric intramucosal pH in neurosurgical patients

Objective To assess the influence of changes in alveolar ventilation on gastro-intestinal perfusion measured via a nasogastric tonometer.Methods Sixteen ASA Ⅰ - Ⅱ patients ( 7 male, 9 female) aged 15-67 yr, undergoing elective intracranial operation were included in this study. The patients were premedicated with oral ranitidine 150 mg and intramuscular phenobarbital 0.1 g and atropine 0.5 mg. Anesthesia was induced with midazolam 1-2 mg, fentanyl 2 ?g? kg-1 , propofol 1.5-2.0 mg ? kg-1 and vecuronium 0.1 mg ? kg-1 , and maintained with 0.5%-2.0% isoflurane inhalation and intermittent iv boluses of fentanyl and vecuronium. The patients were intubated and mechanically ventilated after induction of anesthesia. The ventilatory settings were : RR 10 bpm; FGF 1 L? min-1; I: E = 1:2; the initial VT was 7 ml? kg-1 (T1 ) which was gradually increased to 8 ml ?kg-1(T2), 9ml?kg-1(T3) and 10 ml?kg-1 (T4) and then returned to 7 ml ? kg-1 again. Each VT was maintained for 1 h. A nasogastric tonometer (Tonocap, Datex-Ohmeda, Finland) was inserted into stomach and automatically measured gastric intramucosal CO2 tension (PgCO2) every 10 min. Radial artery was cannulated for direct MAP monitoring and blood sampling. Blood gas analysis was performed every hour at various VT levels. Gastric-to-arterial pH gap and gastric-to-arterial PCO2 gap [P(g-a)CO2] were calculated.Results PgCO2 decreased during T2_4 (hyperventilation phase) (P
