2. Expression of fragile histidine triad protein in lung cancer tissues and its correlation with multidrug resistance protein 1

Objective: To investigate the expression of fragile histidine triad (FHIT) protein in lung carcinoma tissues and its relationship with multidrug resistance protein 1 (MDR1), and to discuss the clinical significance of the changes of FHIT expression. Methods: The expression of FHIT protein was detected in the lung cancerous tissues and the adjacent tissues (n = 60) using S-P staining method. The expression of MDR1 in lung carcinoma tissues was also detected. The expression of FHIT was analyzed in lung cancerous tissues with different clinicopathological parameters and its relationship with MDR1 was analyzed at the same time. Results: The expression of FHIT protein in the adjacent tissues was obviously higher than that in the lung cancerous tissues(76.7% vs 50%, P<0.01). No correlation was found between FHIT expression and the age, sex, smoking, general type, TNM staging, histological type, and lymph node metastasis in the patients with lung cancer. The expression of MDR1 protein in lung cancerous tissues positive for FHIT was obviously higher than those negative for FHIT(83.3% vs 60%, P<0.01). Conclusion: The expression of FHIT protein in lung cancer is evidently down regulated and is correlated with the expression of MDR1. Potential multiple drug resistance should be considered in FHIT gene therapy.

6. The aberrant methylation of fragile histidine triad (FHIT) gene and its significance of the expression of FHIT protein in lung cancer

Objective: To detect the expression of fragile histidine triad (FHIT) protein and aberrant methylation of its gene in lung carcinoma tissue and discuss their significance in tumorigenesis of lung cancer. Methods: The expression of FHIT protein was determined by immunohistochemical SP staining and its aberrant methylation was detected by methylation-specific PCR (MSP) in lung cancer and adjacent lung tissues (n = 60). The sequencing of the amplified products of MSP was performed. The 60 patients were followed up. Results: The expression of FHIT protein in adjacent lung tissues was obviously higher than that in lung cancer tissues. There was significant difference between them (76.7% vs 50%, P < 0.05). The aberrant methylation of FHIT gene was significantly higher in lung cancer tissues than that in adjacent lung tissues (68.3% vs 35.0%, P <0.001). And the aberrant methylation of FHIT gene occurred frequently in FHIT-negative patients compared with FHIT-positive patients (83.3% vs 53.5%, P <0. 05). The expression of FHIT protein and the aberrant methylation of FHIT gene had no correlation with clinicopathological parameters (age, sex, smoking status, histological type, general classification, TNM staging, and lymph node metastasis) in the patients with lung cancer (P > 0.05). The survival time was longer in FHIT-positive patients than FHIT-negative patients (P <0.05). The FHIT protein was the risk factor associated with the disease-free survival time of patients (P <0.01). Conclusion: The aberrant methylation of FHIT gene occurrs frequently and the expression of FHIT protein is significantly down-regulated in lung cancer. FHIT may play an important role in the tumorigenesis and development of lung cancer. FHIT protein could be used as an important factor for predicting the prognosis of patients.
7. Expression pattern analysis of terpene synthases of endangered Aquilaria sinensis under different stresses

Objective: To analyze the expression of terpene synthases of Aquilaria sinensis, an endangered south medicine, and to predict the influences of the environmental factors and stresses on the synthesis of terpene in A. sinensis. Methods: Two-year old seedlings and calli were treated by different stresses. The gene expression patterns of terpene synthases were analyzed by real-time qPCR. Results: Wounding and cauterizing could induce the transcriptional expression of terpene synthases in the stems, and the effect of cauterizing was more significant. Low temperature inhibited the transcription of terpene synthases. In calli, methyl jasmonate (MeJA) treatment had the most effective induction. Conclusion: Both wounding and cauterizing could induce the synthesis of terpenes in stems, while the cauterizing treatment might have better effect. Low temperature has negative influence on agarwood formation. For calli, different treatments could induce the terpene synthesis, while MeJA has the best efficacy.
8. A Meta-Analysis of the Efficacy of Glucagon-Like Peptide-1 Receptor Agonists on Nonalcoholic Fatty Liver Disease in Patients with Type 2 Diabetes Mellitus

OBJECTIVE: To evaluate the efficacy of glucagon-like peptide-1 receptor agonists (GLP-1RA) on nonalcoholic fatty liver disease (NAFLD) in patients with type 2 diabetes mellitus (T2DM). METHODS: The randomized controlled trials (RCTs) of GLP- 1 RA, placebo, and anti-diabetes drugs in the treatment of NAFLD in patients with T2DM were collected from PubMed, Embase, CNKI, Wangfang database, VIP, and CBM. The trials were evaluated for the quality and selected, and the results of the studies were reviewed and analyzed using RevMan 5.2 software. RESULTS: Four RCTs were included, involving 154 patients. The Meta-analysis showed that compared with the control group, GLP-1RA could significantly improve the ALT [MD: -8.36, 95% CI(-13.41-3.31), P = 0.001], HbA1c [MD: -0.43%, 95% CI(-0.73-0.31), P = 0.005], FBG [MD: -0.71%, 95% CI(-1. 39-0.03), P = 0.04], BMI [MD: -1.38%, 95% CI(-2.18-0.58), P = 0.000 8], TG [MD: -0.49%, 95% CI(-0.82-0.16), P = 0.004]. CONCLUSION: GLP-1 RA can obviously improve the metabolic index of patients with NAFLD and T2DM. Given the quality and quantity of the literature, large RCTs are still needed in the future.
9. Taste comparison of unprocessed and processed Siegesbeckiae puescens based on electronic tongue

Objective: To study the taste of unprocessed and processed Siegesbeckiae puescens (SP) and establish a taste discrimination model. Methods: The sour, bitter, astringent, salty, and sweet values of 12 batches of unprocessed and processed SP were measured by electronic tongue using a paired t-test, principal component analysis (PCA), and the linear discriminant factor analysis (LDA) to study the changes of the taste of unprocessed and processed SP. Results: The paired t-test showed that the bitterness and saltiness of processed SP decreased; Astringency, sweetness, and sourness did not significantly change. The PCA can distinguish unprocessed and processed SP. The model of the taste of unprocessed and processed SP was established through the LDA, and conduct cross validation; Its correct rate reached 100%. The fingerprint of taste of unprocessed and processed SP was established through radar-graph. Conclusion: Electronic tongue can identify sour, bitter, astringent, salty, and sweet values of traditional Chinese medicine accurately. It can reflect the numerical change of taste of unprocessed and processed SP combining with statistical method. Based on it, it can be inferred that the relationship between the change of taste and clinical efficacy before and after processing of SP. Besides, the discriminant model can be used to distinguish unprocessed and processed SP.
10. Cementing the apical and terminal pedicle screw applied in osteoporotic spine with lumbosacral degenerative disease: A finite element analysis

BACKGROUND: Cement-augmented pedicle screw is an effective fixation for osteoporotic spine, and it is important to reduce the rate of cement leakage. OBJECTIVE: To evaluate the stability of cementing the apical and terminal pedicle screw applied in osteoporotic spine with lumbosacral degenerative disease by finite element analysis. METHODS: An intact finite element model of L2-5 segment was established by using CT scan data of one normal male volunteer. After verifying the validity of the intact model, the cementing apical and terminal pedicle screw and cement-augmented pedicle screw models of double/multi-level segment fixation were established, respectively. A 150 N vertical axial pre-load was imposed on the superior surface and a 10 N·m moment was applied on the superior surface along the radial direction to simulate six different physiological motions: flexion, extension, left bending, right bending, left rotation, and right rotation. The different of range of motion, cage stress, and pedicle screw stress on fixed segments were compared between models. RESULTS AND CONCLUSION: (1) The validity showed that the range of motion of the intact model was similar to cadaveric studies in all directions. (2) The range of motion of cementing the apical and terminal pedicle screw group was slightly larger than that of cement-augmented pedicle screw group and the difference between the two groups was less than 0.15°. The two fixation methods could maintain the similar stability of the operation segment. (3) The difference of the cage stress and instrument stress was also small between the two groups. (4) These results suggest that compare with cement-augmented pedicle screw, cementing the apical and terminal pedicle screw can increase the approximate stability in double-level and multi-level segment fusion. The cementing the apical and terminal pedicle screw procedure may reduce the risk of cement leakage and patient costs, and offer a useful alternative to the cement-augmented pedicle screw procedure.
