International Journal of Cerebrovascular Diseases 2010;18(9):646-649
Objective To investigate the risk factors of stroke-associated infection (SAI)and their correlation with immunodepression. Methods One hundred fourteen consecutive patients with acute ischemic stroke admitted with 24 hours after symptom onset were enrolled. They were divided into infection (n =45) and non-infection (n =69) groups according to whether they had infection or not within 7 days after admission.Automatic blood analyzer and flow cytometry were used to measure the peripheral blood lymphocyte count and T lymphocyte subsets on the day of admission.Univariate analysis and multivariate logistic regression analysis were used to analyze the related risk factors for SAI.Results There were statistical significances in vomiting, disturbance of consciousness, dysphagia, indwelling gastric tube, indwelling catheter on admission, and the National Institutes of Health Stroke Scale (NIHSS) score 0-3 days after admission between the infection and non-infection groups.Further more, the numbers of peripheral blood lymphocytes,CD3 + and CD4 + T lymphocytes in the infection group were significantly lower than those in the non-infection group.Multivariate logistic regression analysis showed that dysphagia, the increased NIHSS score 3 days after admission, and the decreased numbers of CD4 + T lymphocytes were the independent risk factors for SAI.Conclusions The T lymphocyte immune function decreased in patients with SAI.It was the result of the combined action of various factors.Targeted prevention and treatment should be taken to reduce the occurrence of SAI.
2.Discussion on advantages and disadvantages of frequent cervical regulating manipulation from spinal biomechanics
China Journal of Traditional Chinese Medicine and Pharmacy 2005;0(08):-
Cervical regulating manipulation is based on the extrinsic active system and intrinsic support system. Only when the former system is relaxed, can the motive force of regulating manipulation reach the latter system to extend the vertebral discs and regulate the posterior joints. Frequent cervical regulating manipulation can stretch soft tissue around the joint and enlarge activity of joint and make it soft from subjective views, but it isn’t proved by experiment. From the aspect of spinal biomechanics, cervical regulating manipulation has advantages and disadvantages, so it should be used according to certain standard, or it will cut down the self-regulation activity of spinal biomechanics.

Journal of Applied Clinical Pediatrics 1992;0(06):-
Objective To explore the effect of hyperoxia through mechanical ventilation induced lung injury(VILI) in newborn rabbit. Methods One hundred newborn rabbits aged 1-5 days were randomly divided into 4 groups: with 900 mL/L O2 and mechanical ventilation(MV),group A received high peak inspiratory pressure(HPIP),group B received moderate peak inspiratory pressure(MPIP),group C received low peak inspiratory pressure(LPIP),and group D with no mechanical ventilation with room air. There were 30 rabbits in group A,B,C,10 rabbits in D group. All rabbits were killed at 1,3,6 h after trial respectively.Wet-to-dry weight ratio(W/D) of left lung, and white blood cell(WBC)counts in broncho alveolar lavage fluid(BALF) were measured. The changes of lung histopathology were assessed by hematoxylin-eosin(HE) staining and observed under light microscope. Results Compared with group B and C,the group A demonstrated more pulmonary hemorrhage and pulmonary bullae formation, and the WBC and neutrophils counts in BALF increased and the W/D was higher compared with group B and group C.Meanwhile, alveolar epithelial cellⅡ(AECⅡ) hyperplasy transformed to AECⅠ.In group C,AECⅡswelled and lung tissue edema showed obviously, after 6 hours collagen fibers hyperplasia appeared.Part pulmonary atelectasis was obvious in group C.Conclusions HPIP ventilation can increase lung injury induced by hyperoxia in newborn rabbits, but has minimal effects on MPIP and LPIP ventilation.Pulmonary histopathologic changes participate in the newborn rabbit machine ventilation induced lung injury.
8. Determination of droplet size of propofol fat emulsion injection
Chinese Pharmaceutical Journal 2014;49(4):338-343
OBJECTIVE: To establish a method to determine the droplet size of propofol fat emulison injection for quality control. METHODS: Coulter counting and light diffraction were employed respectively to determine the droplet size of Diprivan® and Lipuro®, and the two methods were compared for their advantages and disadvantages. The optimal determining condition was built through investigating the influence of concentration. Demulsification was carried out by adding electrolyte, then several parameters were measured. According to the results, the optimal condition and the key parameter for determination of the droplet size were established. RESULTS: The optimal concentrations for droplet size determination by coulter counting were 15 μg · mL-1 for Diprivan® and 6 μg · mL-1 for Lipuro®, respectively. Coulter counting method could not provide the whole size distritbution of the emulsion, but could monitor the large droplets in the injection. Additionally, the change of the quality of the emulsion could be monitored by measuring the portion of the droplets larger than 0.6 μm. Light diffraction could offer the whole size distribution and the mean size of the emulsion. But it was not sensitive to measure large droplets in trace amount. And the detection results could not reflect the change when the emulsion was destructed. CONCLUSION: The quality of propofol fat emulsion can be effectively controlled by determining the key parameters with coulter counting under reasonable conditions.

Objective To explore the dynamic change of serum leptin interleukin-6(IL-6),and interleukin-8(IL-8)during cerebral infarction, and to analyze the possible relationship between these factors and the severity of the disease.Method The levels of serum leptin,IL-6 and IL-8 were measured by radioimmunoassay in 60 patients with acute cerebral infarction, who were admitted to the hospital within 24 hours after stroke.Patients' neurologic impairment were evaluated by European Stroke Scale(ESS)at 1,3 and 7 days respectively after hospitalization. At the same time, another 30 normal adults were enrolled set as control group. Results The levels of serum Leptin,IL-8 in patients group after at 1 days,3 and 7 days were higher than those in the controls(P0.05).The ESS of these patients were 76.77?26.42,70.02?29.17,74.65?28.42 respectively, after 1,3 and 7 days.It shown that ESS simply correlated to the level of IL-8(P0.05).Statistic analysis was carried, but by using t test and linear correlation.(SPSS11.5 statistical package). Conclusion Leptin,IL-6 and IL-8 probably play a role in the inflammatory, reaction during acute cerebral infarction. The IL-8 may reflect the severity of the disease.

Background Recombinant hirudin variant Ⅲ(rHV3) can effectively prevent galactose-induced human lens epithelial cells LECs injury, but little is known about the molecular mechanism of its action. Objective The present study was to investigate the effects of rHV3 on the expression of apoptosis-related genes in damaged LECs induced by galactose. Methods The rHV3 was extracted by our research group, and the biological activity of rHV3 was identified by titration of thrombase according to Markwardt's method.Human LECs (SRA01/04) were cultured using 125×10-3 mol/L D-galactose+10% FBS+D/F12 medium to establish the damaged human LECs model.rHV3 was added into the medium of the damaged human LECs model.Human LECs were cultured in D/F12 medium containing 10% FBS as normal control. The expression of apoptosis-related genes, such as aldose reductase (AR),bax,bcl2 and p53,in LECs at the mRNA level was detected using RT-PCR.The abundance ratio of target genes was presented with the absorbance (A) of gene mRNA/GAPDH mRNA.Results Compared to the normal control group, the A values of AR mRNA/GAPDH mRNA,bax mRNA/GAPDH mRNA and p53 mRNA/GAPDH mRNA were significantly elevated in model group (t=3.90E-06,t=8.44E-04,t=5.15E-08,P<0.01).However,in the rHV3-treated group, the A values of AR mRNA/GAPDH mRNA,bax mRNA/GAPDH mRNA and p53 mRNA/GAPDH mRNA were lower than those of model group (t=5.90E-06,t=1.51E-04,t=3.42E-06,P<0.01).The bcl2 mRNA/GAPDH mRNA was markedly downregulated in the model group when compared with the normal control group (t=1.86E-05,P<0.01);while after rHV3 addition,bcl2 mRNA/GAPDH mRNA increased in comparison with the model group (t=8.56E-05,P<0.01).Conclusion 125×10-3mol/L D-galactose induces the damage and apoptosis of human LECs.rHV3 likely plays a protective function on D-galactose-induced damage of human LECs by inhibiting the polyol pathway and mitochondria-mediated pathway.
