Gastroesophageal reflux disease and sliding hiatal hernia are chronic disease,and sometimes there are not typical clinical symptoms.Patients don't have enough cognition or put enough em-phasis on it.Determining the sliding length of the sliding hiatal hernia and gastroesophageal reflux disease respectively by means of high resolution manometry and 24 hour pH monitoring,in turn,analyzing the rela-tionship between sliding length of the sliding hiatal hernia and gastroesophageal reflux disease.

5.Construction of the Recombinant Plasmid with esat6 Gene and Its Expression in Streptococcus gordonii

To construct expressing vector carrying esat6 gene and express this protein in Streptococcus gor-donii GP251. esat6 gene was amplified by PCR with specific primer from genome of Mycobacterium tuber-culosis (MTB)H37Rv. Inserted esat6 into the pMD18-T vector by T/A clone to get recombinant vector pMD18-esat6. Then digested pMD18-esat6 with restriction enzyme, esat6 was cloned to vector PSMB104 and expressed in Streptococcus gordonii GP251. The expression of esat6 protein was detected by Tricine-SDS-PAGE and Western-blot, ELISA technique was also used to detect its secretory volume. Re-striction endonuclease, PCR, Tric ine-SDS-PAGE and Western-blot confirmed that esat6 gene was cloned into expressing vector successfully, and a 10 kD protein secreted in Streptococcus gordonii GP251, this pro-tein has a good immunogenicity. The expression vector of esat6 gene was constructed, and esat6 protein ex-pressed in Streptococcus gordonii1 successfully, it will be benefit for future study.
6.Interleukin-17 in apical exudates of periapical periodontitis treated with minocycline controlled-release formulation

BACKGROUND: Experimental proof for the efficacy, safety, and immunological assessment is needed when minocycline is used for root canal disinfection.OBJECTIVE: To investigate the effect of minocycline for root canal disinfection on levels of interleukin-17 in apical exudates of periapical periodontitis and periapical exudate volume.METHODS: Sixteen patients with acute periapical periodontitis (16 teeth) scheduled for root canal therapy were enrolled and randomly divided into calcium hydroxide and minocycline groups, respectively, followed by root canal disinfection.One week after disinfection, periapical index, periapical exudate volume and interleukin-17 level were detected prior to the root canal filling. Another 16 patients with normal pulp vitality (16 teeth) scheduled for single root canal filling were enrolled as control group, in which periapical index, periapical exudate volume and interleukin-17 level were detected.RESULTS AND CONCLUSION: The periapical exudate volume and interleukin-17 level in the calcium hydroxide and minocycline groups were significantly higher than those in the control group (P < 0.05). The periapical index and interleukin-17 level in the calcium hydroxide and minocycline groups were decreased significantly at 1 week after root canal disinfection (P < 0.05), while there was no difference between these two experimental groups in the periapical index, periapical exudate volume and interleukin-17 level. To conclude, the use of minocycline significantly reduces interleukin-17 level and periapical exudate volume, and thus achieves effective outcomes in periapical disease.
7.Overview on method and strategy of therapeutic material basis in traditional Chinese medicine by multidisciplinary approach.

Traditional Chinese medicine (TCM) has a good reputation for preventing or healing diseases in clinic due to its higher efficacy, minor toxicity and abundant resources. Screening bioactive components in TCMs is not only crucial for clarifying their action mechanisms, but also the basis of their safety and quality control. TCM is characterized by multiple components, multiple targets and multiple mechanisms, however the complex composition of TCM makes it difficult to study the therapeutic material basis which has become the bottleneck in the process of its modernization and internationalization. Recently, with the rapid development of modern technologies and the unceasing progress of various disciplines, multidisciplinary approach, such as analytical chemistry, chemistry of TCM, pharmacology, cell biology, systems biology and bioinformatics has been successfully applied to the study of TCM. Multidisciplinary approach realizes the communication and interaction of multi-discipline, and accelerates the research and development of TCM. This review summarizes the application of multidisciplinary approach which may have certain potential of bringing new thoughts to TCM research and provide references for screening and identification of therapeutic material basis of TCMs.

8.Advances in human placenta-derived stem cells for liver regeneration

Human placenta-derived stem cells (hPD-SCs) are a mixed group of stem cells.Stem cell medicine has applications for organ damage or failure through regenerative,anti-apoptotic,anti-inflammatory and anti-tumor properties in addition to cell function recovery.Presently,human placenta mesenchymal stem cells (hPMSCs) have similar characteristics to the differentiation of hepatocyte-like cells by promoting hepatocyte regeneration,anti-hepatocyte apoptosis and anti-liver fibrosis,in vitro or in animal models.To further our investigation,a summary of the origin,sorting and biological properties of hPDSCs along with a narration of hPDSCs for liver disease therapy was written.This leads to a discussion for new ideas to further explore cell treatment for liver disease.

AIM:To investigate the effects of MMP-7asODN on MMP-7 expression in human stomach cancer cell line KATOIII and explore the effect of MMP-7 on cancer invasion and metastasis. METHODS: A 15mer PS-asODNs targeted against the MMP-7 mRNA were introduced into KATOIII cells by FuGENE TM 6. The levels of MMP-7 mRNA was observed by reverse transcription polymerase chain reaction (RT-PCR). The ability of the invasion was observed by Boyden Chamber invasion assay. RESULTS: The MMP-7/?-actin ration was 0.31?0.02 for the PS-asODNs treated cells which is significantly lower compared with the control and PS-sODN and PS-mODN( 1.59?0.01, 1.14?0.03,1.51?0.02, respectivly.) 10.Changes of Surfactant Protein A and D in Bronchoalveolar Lavage Fluid in Human Fetal Lungs during Various Gestational Ages

Objective To investigate the metabolic rules of surfactant protein A and D(SP- A,SP- D )in bronchoalveolar lavage fluid(BALF)in human fetal lungs during gestational ages. Methods BALF with 30 mL saline was performed on clinically collected human fetus with induction of labor by water- bag Their BALF was respectively retrieved [total retrieval rate(85. 6% ? 13 1)% ]for analysis of protein content. The BALF SP - A and SP - D from fetus of various gestational ages or newboms were detected by RPHA and ELISA. Results The total protein in BALF gradually increased since 10th week to newborn peak during lung development. And SP - A and SP D were respectively updated from(0.34 ?0.07 ) ,(0.05?0.01) ng/L to newborn climax[ (6 42 ? 0 36),(1.22 ? 0 13)ng/L] .Conclusions The protein in BALF gradually increases with fetal growth and lung development. SP-A and SP- D may reach prenatal climax and become the main indicator of newborn lung maturity.
