1.Influence of Butanol Extract from Stevia on the expression of single immunoglobulin interleukin-1 receptor-related molecule and nuclear factor kappa-B in depression mice liver

Objective To study the antidepressant-like effect of Butanol Extract from Stevia (BEFS) and its possible corresponding mechanisms.Methods Forty-five mice were randomly divided into control group,model group,model + Fluoxetine group,model + BEFS-3 g/kg group and model + BEFS-6 g/kg group.The chronic unpredictable mild stress(CUMS) for 3 weeks was used to make up depressive animal model.It was based on body weight,crossing numbers,rearing numbers and sucrose preference to assess whether the model was a success.After continuous lavage giving BEFS and fluoxetine for 2 weeks and subsequent behavioral test,the mRNA transcription of Single immunoglobulin interleukin-1 receptor-related molecule (SIGIRR) and nuclear factor kappa-B (NF-κB) in liver tissue of mice were detected by using quantitative real time-PCR.Results Three weeks after CUMS,behavior parameters were significantly decreased in mice exposed to CUMS compared with control group[(17.28 ±0.49) g vs (22.47 ± 0.82) g,P =0.000;(43.35 ± 1.68) scores vs (125.25 ± 3.07) scores,P =0.000;(3.92 ± 0.18) scores vs (15.92 ± 1.78) scores,P =0.000;(49.39 ± 2.05) % vs (67.18 ± 4.46) %,P =0.001].However,2 weeks after dosing,body weight,crossing numbers,rearing numbers and sucrose preference were (19.03 ± 0.44) g,(70.50 ± 6.56) scores,(3.08 ± 0.77) scores and (41.27 ± 7.59) %,respectively in model group.Compared to model group,behavior parameters of BEFS (3 and 6 g/kg BEFS) groups were significantly increased [BEFS-6 g/kg group:(24.42 ± 1.46) g,(99.75 ± 5.07) scores,(13.08 ± 1.60) scores,(64.31 ± 1.92) %,all P < 0.05;BEFS-3 g/kg group:(23.82 ± 1.72) g,(91.67 ± 5.69) scores,(6.92 ± 1.05) scores,(59.56 ± 5.31) %,respectively,all P < 0.05].Correspondingly,BEFS could reverse the mRNA expression of SIGIRR lowering and NF-κB rising in the liver of mice exposed to CUMS(BEFS-6 g/kg group:1.208 1 ± 0.102 2,1.437 7 ± 0.352 9;BEFS-3 g/kg group:0.768 0 ± 0.108 6,2.186 2 ± 0.203 8,all P < 0.05).Conclusions BEFS can remarkably improve depression-like behaviors in CUMS mice and its antidepressant activity is mediated,at least in part,by upregulating SIGIRR and downregulating NF-κB in the liver.
2.Molecular mechanism of a pedigree with inherited coagulation factor Ⅶ deficiency

Objective To identify gene mutations and explore the molecular mechanism of a pedigree with inherited coagulation F Ⅶ deficiency. Methods The levels of F Ⅶ: Ag in the proband and other family members were measured by ELISA assay. The values of PT, F Ⅶ: C and other coagulant parameters were determined by one-stage clotting for laboratory phenotype diagnosis. All the exons,exon-intron boundaries and 5',3' untranslated sequences of F7 gene were amplified by direct sequencing. The detected mutations were further confirmed by sequencing the other stand. The CLC Protein Workbench software was used to analyze the species conservation of the mutated site and the protein secondary structure. 100 healthy individuals were selected to exclude gene polymorphism. Results PT, FⅦ∶C and FⅦ: Ag in the proband and his sister were abnormal, which were 36. 3 s, 5.0%, 40. 7% and 33.4 s,5. 0%, 37.4%, respectively. Both PT and FⅦ∶C in the proband's father, mother, daughter and niece were slightly abnormal, which were 14.9 s, 14. 6 s, 15.5 s, 14. 6 s and 70%, 85%, 59%, 79%, respectively.The heterozygous mutations c. 784T > C and c. 964T > G in exon 8 of F7 gene were found in the proband,resulting in the substitutions of Ser269Pro and Cys329Gly respectively. Compound heterozygous mutations c. 784T > C and c. 964T > G were found in the proband's sister. The proband's mother was heterozygous for c. 784T > C. His father, daughter and niece were heterozygous for c. 964T > G. The protein biological characteristics analysis revealed that the Cys329Gly caused the change of spatial configuration, and Ser269Pro led to the change of amino acid polarity and hydrophobicity. Conclusion Compound heterozygous mutations of Cys329Gly and Ser269 Pro in F7 gene may be the underlying molecule mechanism of FⅦ deficiency in this pedigree.
3.Efficacy of etoposide capsule on metastatic breast cancer

Objective To evaluate the efficacy and safety of etoposide capsule on metastatic breast cancer. Methods Forty-five advanced breast cancer patients treated with oral etoposide during Feb.1,2012 and Oct.31, 2016 in our department were enrolled in our study,with their general data,median number of treatment lines, treatment effect,common adverse reaction collected. Such indexes of objective response rate,rate of clinical benefit,disease control rate,progression free survival time were anlayzed. Results All patients received median five lines of etoposide capsule therapy. The objective response rate(ORR,CR + PR)was 6.7% and the clinical benefit rate(CBR,CR+PR+SD≥6.0 months)was 26.7% and the disease control rate(DCR,CR+PR+SD) was 68.9%.The median progression free survival time(PFS)was 4.0 months(95% CI:2.3~5.6).The main toxic-ities were grade 1-2 neutropenia(37.8%),grade 3-4 neutropenia(6.7%).Conclusion Etoposide capsule could be an option with efficacy and safety for metastatic breast cancer.
4.A clinical study of the position relationship between the optical fixation and the fovea in healthy adults

Objective:To observe the positional relationship between the central fixation point of the retina and the fovea in normal adults.Methods:A retrospective clinical study. From August 2019 to January 2020, 100 eyes of 100 normal adults who underwent physical examination at the Fourth People's Hospital of Shenyan g were included in the study. All patients underwent BCVA, diopter, microfield, OCT examination, and axial length (AL) measurement. There were 42 males and 58 females with the average age was 46.4±14.7 years. The average diopter was -1.02±1.99 D, the average AL was 23.22±0.47 mm, the average foveal avascular zone (FAZ) area was 0.38±0.13 mm 2. The MP-3 microperimetry was used for central fixation examination. After the examination, high-definition fundus images were automatically taken and the central fixation point of the retina were automatically calculated by the equipment. The Nidek Overlay functional multi-mode imaging platform was used to superimpose the images containing the central fixation point of the retina and the macular fovea, the positional relationship between the two was observed, and the distance between the two was measured. Pearson correlation analysis was performed on the distance between the fixation point of the center of the retina and the center of the fovea, age, diopter, and FAZ area of the macula. Results:The fixation point of the retinal center of all tested eyes was within the range of the macular fovea, which did not coincide with the center of the macular fovea. Among 100 eyes, the fixation point of the center of the retina were 53, 23, 15, and 9 eyes at the nose, lower, temporal, and upper sides, respectively. The average distance between the fixation point of the center of the retina and the center of the fovea was 158.31±71.56 μm. The distance between the fixation point of the retinal center and the center of the macular fovea and age ( r=0.140), diopter ( r=-0.009), FAZ area( r=0.038) were not correlated ( P=0.165, 0.932, 0.707) in correlation analysis. Conclusion:The central fixation point of normal adult retina is more common on the fovea nasal side.
5.Multiplexed single-cell transcriptome analysis reveals molecular characteristics of monkey pluripotent stem cell lines.

Efforts have been made to establish various human pluripotent stem cell lines. However, such methods have not yet been duplicated in non-human primate cells. Here, we introduce a multiplexed single-cell sequencing technique to profile the molecular features of monkey pluripotent stem cells in published culture conditions. The results demonstrate suboptimized maintenance of pluripotency and show that the selected signaling pathways for resetting human stem cells can also be interpreted for establishing monkey cell lines. Overall, this work legitimates the translation of novel human cell line culture conditions to monkey cells and provides guidance for exploring chemical cocktails for monkey stem cell line derivation.

6.Advances in biodegradation of macrolide antibiotics.

Macrolide antibiotics are a class of broad-spectrum antibiotics with the macrolide as core nucleus. Recently, antibiotic pollution has become an important environmental problem due to the irregular production and abuse of macrolide antibiotics. Microbial degradation is one of the most effective methods to deal with antibiotic pollution. This review summarizes the current status of environmental pollution caused by macrolide antibiotics, the degradation strains, the degradation enzymes, the degradation pathways and the microbial processes for degrading macrolide antibiotics. Moreover, the critical challenges on the biodegradation of macrolide antibiotics were also discussed.
