1.Immune tolerance induced by bone marrow infusion combined with costimulatory blockade

Organ transplantation is the optimal treatment for patients with an end-stage organ disease.The induction of donor-specific tolerance is necessitatd because of severe side-effects of immunosuppression and chronic rejection of the transplanted organ that defy management.Bone marrow infusion and costimulatory blockade are considered to be the most effective strategies to induce immune tolerance.This review briefly discusses the mechanism and clinical application of immune tolerance induced by bone marrow infusion combined with costimulatory blockade.
2.Cloning and identification of PTCH gene involved in hedgehog signal pathway in human pancreatic cancer

Objective To clone human pancreatic cancer gene PTCH,construct the recombinant(expression) plasmid PET22b/PTCH and express the fusion protein.Methods The PTCH gene was(amplified) by RT-PCR from the total RNA extracted from human pancreatic cancer strain SW1990. The amplified product was inserted into the vector PET22b to construct the recombinant expression plasmid PET22b/PTCH,which was transformed into E.coliBL21-CodonPlus~(TM)-RP and then identified by(sequence) analysis.The expression of fusion protein was induced with IPTG and verified by Western blot method.Results A human pancreatic cancer gene with a reading frame of 789 bp was successfully cloned from human pancreatic cancer strain SW1990,which had the same sequence as that of PTCH gene in Genbank.The expression of PET22b/PTCH was proved by Western blot.Conclusion Human(pancreatic) cancer gene PTCH was successfully cloned and constructed with PET22b plasmid.The(prepared) fusion protein lays the basis for further study.
3.CLINICAL STUDY ABOUT ENDOSCOPIC TREATMENT OF PANCREATIC DUCTAL STONES

To evaluate the effect of endoscopic treatment of pancreatic ductal stones (PDS), 31 patients with PDS treated in our hospital from January 1998 to Octomber 2001 were analyzed retrospectively. PDS were successfully extracted in 10 patients by endoscopic papillotomy (EPT) or endoscopic pancreatic sphincterotomy (EPS). In 2 patients, one was treated with extracorporeal shock wave lithotripsy (ESWL) and endoscopic retrograde pancreatic drainage (ERPD), another with EPS and ESWL, the PDS were all disappeared in 6 months. Fourteen patients with pancreatic ductal stricture were treated with ERPD, and 4 with EPS+ERPD. However, 4 patients were subjected to operation for pancreatic carcinoma and recurrent symptom. Only one of them failed owing to severe pancreatic ductal stricture. The success rate of the procedure was 96 8%, and the remission rate of abdominal pain was 90% shortly after operation and 70 4% in the follow up interval. The body weight and pancreatic exocrine function were also improved. But the stent occlusion was found in 3 patients and their stents were changed. So endoscopic therapy such as EPS, ESWL, and ERPD are effective and safe methods in treatment of PDS.
4.Monitoring Instrument of Multiple Physiological Parameters Based on Wireless Internet

Objective To study a monitoring instrument of multiple physiological parameters being capable of wireless long-distance transmission,low in power dissipation and price and small in size.Methods In combination of embedded technology and mobile communication technology,S3C2410 based on ARM9 core was used as main chip,ARM-Linux as embedded operation system and SIM300 as communication module connected to Internet,to complete wireless long-distance transmission of multiple physiological parameters.Results Tele-monitoring system of embedded multiple physiological parameters based on GPRS was realized.Conclusion The instrument is small,easily-expanded and reliable in the process of transmitting data,facilitating tele-monitoring and data sharing.
5.Magnetic resonance imaging and diffusion tensor imaging of gray matter damage in multiple sclerosis

Objective To study MRI characteristics of the gray matter lesions in multiple sclerosis (MS) patients, and to investigate the occult damage in normal appearing gray matter (NAGM) by quantitative analysis using diffusion tensor imaging (DTI). Methods Conventional brain MRI and DTI were performed in 34 clinically defined MS patients and 25 non-MS healthy volunteers. Main signs of the GMlesions detected by conventional MRI were analyzed, including the distribution, numbers, shape, size,signal intensity and enhanced pattern. The apparent diffusion coefficient (ADC) and fractional anisotropy (FA) values derived by DTI were measured in normal appearing deep gray matter for all participants and the differences between the two groups were compared. Results MRI examination revealed 83 lesions in cerebral gray matter, 18. 7% of the total 443 lesions. The GM lesions distributed over all brain lobes especially in frontal lobe, temporal lobe, and thalamus. Thirty-four, 60, 78, and 36 plaques were detected on T1WI, T,2WI, FLAIR, and reconstructed DWI images, respectively. Nine small lesions were identified on DWI more easily than on T2WI and FALIR. The ADC values of the head of caudatum (8. 0±0. 7) ×10-4mm2/s, t=-3.079, P<0.05), putamen (7.4±0.5)× 10-4mm2/s, t= -2.564, P<0.05),and thalamus (7.7± 0. 4) × 10-4mm<'2>/s, t = -2. 722, P < 0. 05) in MS group were significantly higher than those in healthy controls [ the ADC values of head of candatum (7.4 ± 0. 6) × 10-4 mm2/s, putamen(7.0±0.5) ×10-4 mm2/s,and thalamus(7.2±0.7)×10-4mm2/s]. Conclusions This study confirms the presence of GM damage in MS. It shows MRI characteristics of the macro-lesions, and combination of FLAIR and DWI can improve the detection of GM lesions. Occult micro-change in NAGM can be evaluated by using DTI quantitative analysis.
6.Comparison on the clinic features and neuromyelitis optica-IgG positive rate of neuromyelitis optica and multiple sclerosis

Objective To compare the clinic features of neuromyelitis optica (NMO) and multiple sclerosis(MS).To compare the positive rate of NMO-IgG in NMO,MS and other related diseases,and determine whether it can be considered as a biomarker for differential diagnosis.Methods Detected serum NMO-IgG in 34 NMO patients,22 MS patients,24 high risk syndrome,5 clinical isolated syndrome,and 35 patients with other neumlogical diseases.Compared the clinic features(onset age,severity,prognosis,MRI lesions,autoimmune antibodies,CSF)of 34 NMO patients and 22 MS patients.Results The onset age of NMO is older than that of MS.It is more severe and with worse prognosis than MS.Longitudinal spinal cord lesions are easily found in NMO.NMO-IgG positive rate in NMO and high risk syndrome patients are 58.8% (20/34)and 45.8%(11/24)respectively,which are higher than that in MS(1/22),clinical isolated syndrome(1/5)and other neurological diseases(1/35;x2=37.2,P<0.01).The positive rate may have a relationship with the length of spinal cord lesions.Condusions MS and NMO are probably different diseases.NMO-IgG positive rate in NMO is significandy higher than that in MS,it can be considered as a biomarker for differential diagnosis.
7.The application of Montreal cognitive assessment in urban Chinese residents of Beijing

Objective To study the distribution of Montreal cognitive assessment(MoCA)scores in terms of age and educational level in Chinese residents aged 50 years and over and to offer a benchmark for a cutoff score.Methods A total of 281 residents aged 50 years or older was drawn randomly in the urban areas of Beijing,including 215 healthy elderly controls(NC)and 66 patients meeting the clinical criteria of mild cognitive impairment(MCI).The final scores for MoCA were given in the form of mean percentage distributions specific for age,sex and educational level so as to compare the validity of MMSE mini-mental state examination and MoCA in detecting MCI.By a fitting muhiple regression model the influenee of the factors on MMSE and MoCA was assessed.Results Using a cutoff score of 26.MMSE had a sensitivity of 24.2%to detect MCI.whereas MoCA detected 92.4%of the MCI subiects.We found a bivariate correlation between MoCA scores and both the factors of age and educational level(P<0.001).Conclusions MoCA iS a brief cognitive screening tool with high sensitivity and specificity for detecting MCI as currently conceptualized in patients performing normally on MMSE.Our adiustment in the cutoff scores would improve the detection of MCI and Alzheimer'S disease by reducing the number of false negatives.MoCA scores should be used to identify current cognitive difficulties but not to make formal diagnoses.
8.Transurethral electric resection plus arterial catheterization chemotherapy for T2a bladder carcinoma

Clinical data of 23 patients with T2a bladder cancer admitted from March 2008 to August 2013 were retrospectively analyzed.Among 23 patients,11 cases were treated with transurethral resection (TURBT) plus arterial catheterization chemotherapy (study group) and 12 cases were treated with radical cystectomy (control group).The overall survival time and recurrence free survival time of two groups were compared.Patients in study group were followed up for 20-68 months,recurrence occurred in 5 cases (5/11),including 4 cases of invasive recurrence;patients in control group were followed up for 2 ~86 months,1 case had superficial recurrent and underwent TURBT,2 cases dead due to bladder tumor.During the course of chemotherapy,the main adverse effects were digestive reaction (7/11),fever (4/11),bone marrow suppression (2/11),symptomatic treatment was given,which was tolerated.There were no significant differences in overall survival time and recurrence free survival time between two groups (P > 0.05).The quality of life of study group was better than that of control group (P < 0.05).It is suggested that the arterial interventional chemotherapy combined with transurethral resection for T2a stage invasive bladder cancer has a certain curative effect with the advantage of preservation of bladder function and higher quality of life.
9.A Peptide Microarray-Based Fluorescent and Resonance Light Scattering Assay for Screening Thrombin Inhibitor

A peptide microarray-based fluorescence and resonance light scattering ( RLS ) two readout assay was developed for screening thrombin inhibitors in blood samples. In this assay, the biotinylated peptide microarray was used as the platform. The peptide C-terminal fragments carried biotin sites departed from the slide when the biotinylated peptides were digested by thrombin hydrolysis reaction. The hydrolysis progress was labeled by fluorescence and 30 nm peptide-stabilized gold nanoparticles through the biotin-avidin reaction. In the presence of thrombin inhibitors, the hydrolysis reactions were blocked, and the inhibition capability of inhibitors could be detected by the fluorescent and RLS signal changes. 10.Effect of Surface Charge on Biobehavior ofPEGylated Gold Nanoparticles

Three kinds of PEGylated gold nanoparticles (PEG-Au NPs) with different surface charges are prepared by assembly of thiolated polyethylene glycol (HS-PEG) with different terminal groups including methoxy, amino or carboxyl on gold nanoparticle surface through sulfur-gold covalent bond, respectively.The experimental results of cell co-culturing and tail intravenous injection in mouse indicate that the biological behaviors of PEG-Au NPs are affected significantly by their surface charges.The cellular internalization amounts of PEG-Au NPs are following the order, positive charge > neutral charge > negative charge.The PEG-Au NPs are gradually transferred to liver and spleen from main organs through the circulation of blood after tail intravenous injection in mouse.The negatively charged PEG-Au NPs have the slowest hepatic clearance rate while the positively charged PEG-Au NPs can cause the strongest response of immune system in mice.
