To observe the alterations of oxygen delivery/consumpation, the deformability and adherence of neutrophil in canine with respiratory distress syndrom(RDS), Method:The 15 selected healthy mongrel dogs were randomly allocated to receiving intravenous oleic acid 0.06 ml. kg~(-1) (test group, n= 8)or equivalent volume of saline (control group, n=7) respectively following anesthesia induction. The hemodynamics was monitored with Swan-Ganz catheter technique, the neutrophils were collected from arterial blood with centrifugal way to measure the deformability and adherence at 1st, 2nd, 4th and 6th hour after the administration of oleic acid separately. The pulmonary pathological examination underwent at 6th hour after the administration. Result: The pathological findings of test group were compatible with the RDS pathological characteristics. Aim To study the effects of pyrrolidine dithiocarbamate(PDTC) on matrix metalloproteinase-9(MMP-9) expression in alveolar macrophages(AM) induced by TNF-?,and then to explore it's signal pathway.Methods AM were collected from bronchoalveolar lavage fluid in patients with chronic obstructive pulmonary disease.AM were pretreated with PDTC and then stimulated by TNF-? or IL-1.MMP-9 mRNA expression was detected by semi-quantitative reverse transcription-polymerase chain reaction and MMP-9 protein expression was detected by Western blot.Phospho-I?B? protein levels induced by TNF-? or IL-1 were detected by Western blot.NF-?B activity was detected by electrophoretic mobility shift assay.Results Both the mRNA and protein levels of MMP-9 induced by TNF-? in AM were significantly elevated in a dose dependent manner(P0.05).PDTC had distinct effect of inhibiting the activation of NF-?B induced by TNF-? or IL-1 in a dose-dependent manner(P0.05).Conclusions NF-?B plays an important role in MMP-9 expression induced by TNF-? in AM;PDTC can down-regulate MMP-9 expression induced by TNF? in AM possibly through preventing the degradation of I?B? via ubiquitylation-proteasome proteolytic pathway.

AIM: To investigate whether focal adhesion kinase (FAK) takes part in the intracellular signaling pathway involved in apoptosis of human pulmonary artery smooth muscle cells (HPASMCs). METHODS: Cultured HPASMCs stimulated by fibronection (40 mg/L) were passively transfected with antisense-FAK oligodeoxynucleotides (ODNs). Expression of FAK and caspase-3 proteins was detected by immunoprecipitation and Western blotting. In addition, apoptosis were measured by TUNEL. RESULTS: The protein expression of FAK in HPASMCs decreased and caspase-3 expression upregulated in HPASMCs in antisense-FAK ODNs group. At the same time, antisense-FAK ODNs transfection increased the rate of cell apoptosis. CONCLUSION: These results suggest that FAK may be related with the apoptosis of HPASMCs. Antisense-FAK ODNs inhibit HPASMC proliferation and may facilitate their apoptosis via caspase-3 signaling pathway.

Objective To analyze the status and impact factors of continuous rehabilitation nursing about stroke patients.Methods 120 medical staff in 6 hospitals and 108 medical staff in 9 community using the multi-stage random sampling method were investigated with questionnaires designed by ourselves.Results Lacking communication and cooperation between hospitals and community medical center,lacking unified and normative regulation and standards were the main influencing factors of the continuous rehabilitation nursing.Conclusions We should establish information interactive platform and perfect the corresponding policies to build continuous rehabilitation nursing model.

AIM: To investigate the effects of NF-?B decoy oligodeoxynucleotides (ODNs) on apoptosis in lung cancer cell A549. METHODS: The treatments of lung cancer cells (A549) were divided into three groups: group A (control group); group B (decoy ODN group) and group C (scramble decoy ODN group). FITC-labeled NF-?B decoy ODNs was transfected into A549 with LipofectAMINE TM 2000. The activation was observed by electrophoretic mobility shift assays (EMSA). The proliferation was observed by growth curve. The apoptosis of cells were observed by flow cytometry and TdT mediated dUTP-biotin Nick End Labeling (TUNEL). The expression of Bcl-2 and Fas were observed by Western blot. RESULTS: After FITC-labeled decoy ODNs was transfected for 1 hour, the decoy ODNs was detected in the nuclei of A549 cells. EMSA performed the depression of the NF-?B binding to the nucleus. The growth curve showed the inhibition of the A549 cell growth and the percentage of apoptosis was increased compare with control group by flow cytometry and TUNEL. The amount of apoptosis inhibitor (Bcl-2) in group A and group C were 2.0 times and 2.1 times more than that in group B, respectively. The level of apoptosis accelerator (Fas) in group B were 2.6 times and 2.3 times more than that in group A and group C, respectively via Western blot. CONCLUSION: The NF-?B decoy ODNs accelerate the apoptosis of lung cancer cell A549 and the mechanism may be due to its inhibiting the expression of Bcl-2 and increasing the level of Fas. [

AIM: To study the effect of dexamethasone (DEX) on the adrenomedullin (ADM) and its gene expression in lung tissue of asthmatic rats. METHODS: 30 healthy male SD rats were randomly divided into three groups (10 for each). The asthmatic model was established by ovalbumin inhalation and injection. The mRNA expression of ADM was examined by RT-PCR and the protein expression was detected by immunohistochemical method. The airway wall thickness, the airway smooth muscle (ASM) thickness and pulmonary tissue changes were observed under light microscope. AIM: To investigate the effect of sodium nitroprusside (SNP) on activation of nuclear factor ?B. METHODS: The techniques of culture of human T lymphocytes, Western blot and RT-PCR were applied. The effects of NO donor sodium nitroprusside (SNP) at different concentrations on mRNA and protein expression of I?B ? in human T lymphocytes at 30 min or 120 min after stimulating with phytohaemagglutinin (PHA-P) were observed. RESULTS: SNP at middle or high concentrations reduced the degradation of I?B ? protein 30 min after stimulating with PHA-P, and increased the re-expression of I?B ? mRNA 120 min after stimulating with PHA-P significantly. CONCLUSION: The mechanism of inhibitory effect of SNP at middle or high concentrations may be due to the decrease in degradation and the increase in re-synthesis of I?B ?. The regulatory mechanism of SNP at low concentration may not be through I?B ?.

AIM: To study the role of hypoxia-inducible factor-1alpha(HIF-1?) on lung cancer cells A549 growth in vitro and in vivo. METHODS: To observe the growth rate of A549 cells after HIF-1? transfected, A549 cells (1?10~6/mouse) were inoculated subcutaneously into 20 nude mice, which were randomly divided into two groups: the control group (group A, n=10), the HIF-1? transfected group (group B, n=10). The weights of subcutaneous tumor were detected. The resected specimens were made into paraffin-embedded sections. The proliferating cell nuclear antigen (PCNA) was identified by immunohistochemistry(ISH). The expressions of HIF-1?? apoptosis-related protein survivin and bcl-2 were analyzed by Western blot. RESULTS: The growth rates of the HIF-1? transfected lung cancer cells A549 were significantly increased, and more importantly, the HIF-1? transfected lung cancer cells A549 was able to enhance lung cancer growth in nude mice(P

Objective To investigate the status of stroke patients′continuous rehabilitation care and it′s influencing factors in the community. Methods Questionnaires were used to investigate 174 stroke patients from 6 communities of Zhengzhou. Results The implementation of continuous care was not satisfactory. Occupation, medical payment method and medical resource utilization were important predicting factors. Conclusions In order to promote continuous rehabilitation care, the health resources should be fully used, the payment methods of the patients should be improved, and the occupational characteristics should be considered.

To study the effect of Erigeron breviscapini on chronic hypoxic pulmonary hypertension and mechanism，the hypoxic rats were injected intraperitoneally with Erigeron breviscapini. The mPAP，the content of serum nitric oxide (NO) and endothelin-1 (ET-1) were measured 5，14 and 28 days after intraperitoneal injection of Erigeron breviscapini in hypoxia respectively，and were compared with those in the single hypoxic group. The results showed that the contents of serum mPAP and ET-1 were higher，and serum NO was lower in single hypoxic group than in the normal rats (both P<0.01). The contents of serum mPAP and ET-1 were lower and NO was higher in the Erigeron breviscapini-treated group than in the single hypoxic group (P<0.01). It was suggested that Erigeron breviscapini could reduce the hypoxic pulmonary hypertension by increasing NO and decreasing ET-1 of serum. Erigeron breviscapini could be used to treat hypoxic pulmonary hypertension.
